Objective To explore the relationship between the plasma level of carbon monoxide and the hyperdynamic circulation of cirrhotic rats. Methods S.D. rats were divided into cirrhotic ( n =10) and control ( n =10) groups. Liver cirrhosis was induced by percutaneous administration of carbon tetrachloride. Mean arterial pressure (MAP, kPa),heart rate (HR,beat/min) and portal pressure (PP,cm H 2O) were measured by indwelling catheters. Plasma CO was measured by Chalmers method, and plasma NO 2/NO 3 was determined by the reduction of cadmium column. Heme oxygenase (HO) activity in splanchnic organs from cirrhotic rats and controls was also determined. Results Typical features of cirrhosis were histologically observed in carbon tetrachloride treated animals. Objective:To explore influence of psychological intervention on mental state in patients With rheumatic heart dis-ease (RHD)after valve replacement surgery.Methods:A total of 60 RHD patients after valve replacement surgery Were randomly and equally divided into routine nursing group and psychological intervention group according to number table,the latter received additional psychological intervention besides routine nursing intervention.Symptom checklist 90 (SCL-90) Was used to assess mental state of tWo groups 1 d before and one Week after surgery.Results:There Were no significant difference in SCL-90 scores betWeen tWo groups before psychological intervention (P>0.05).Compared With routine nursing group one Week after operation,there Were significant reduction in SCL-90 scores of somatization [(1.31±0.47) scores vs.(0.55±0.68)scores],obsessive-compulsive symptom [(2.11±0.35)scores vs.(0.78±0.35)scores],inter-personal relationship sensitivity [(1.57±0.53)scores vs.(1.22±0.46)scores],depression [(1.63±0.29)scores vs. (1.00±0.41)scores],anxiety [(0.98±0.17)scores vs.(0.76±0.13)scores],hostility [(1.11±0.31)scores vs.(0.80 ±0.37)scores]and phobia anxiety [(1.39±0.26)scores vs.(0.53±0.19)scores]in psychological intervention group,P<0.01 or P<0.05. Conclusion:Psychological intervention can alleviate the adverse psychological state and improve quali-ty of life in patients With rheumatic heart disease.

To explore the relationship between the activity of polyol pathway and diabetic microangiopathy, the level of red blood cell sorbitol(RBCS)in 106 patients with NIDDMwas determined. The results indicated that the level of RBCS in diabetic patients was significantly increased, and was positively correlated with 24-h urinary albumin excretion rate(UAER)and the degree of diabetic retinopathy(DR). Besides,the value of RBCS in patients with both pro-teinuris and DR or proteinuria was markedly higher as compared with that in patients with either proteinuria or DR,and the concentration of RBCS in the patients with DR of proteinuria was also significantly higher than those wth neither proteinuria nor DR. In addition.it was also found that the value of RBCS gradually elevated with the aggravation of UAERand DR. These findings suggest that the elevation of RBCS level plays an important role in the formation and development of diabetic microangiopathy.

Objective To study the characteristics of molecular genetics concerning Chinese mitochondr ial encephalomyopathy,lactic acidosis and stroke-like episodes (MELAS). Methods A3243G and T3271C point mutations in the mtDNA of muscle and (or) blood cells were investigated in 9 patients with MELAS and some o f their maternal relatives from 7 families by using PCR-RFLP. Furthermore,mut ant mtDNA in the sample harboring mutation was quantitatively analysed. Results The mtDN A A3243G point mutation was unanimously identified in tissues of all patients an d 1 of their relatives. However,the T3271C point mutation was identi f ied in none of series in our study. The proportion of mtDNA A3243G was 46.8%～ 61. 0% in muscle (4 cases) as well as 26.8%～50.3% in blood (9 cases). In the 3 p atie nts with muscles and blood cells available,their mutant mtDNA proportion in mus cle is consistently higher than in blood cells. The study of leukocytes of some maternal relatives from 6 families showed that,while only 1 proband had a siste r harboring A3243G mutation and none of the mothers of another 3 probands or sib lings of the other 2 probands had the point mutation. However,the sons of 2 pr o bands had not only phenotype of MELAS,but also mtDNA A3243G point mutation in t heir blood. Conclusion mtDNA A3243G mutation highly exists in the series with M E LAS syndrome in our study and can be detected in various tissues,which is consi stent with reports abroad. However,most of our cases are sporadic rather than m aternal inherited. It is presumedly caused by a de novo mutation. Whether i t is due to ethnic difference or sporadic event needs to be investigated further.

Objective To compare the cellular immune responses induced by two different vectors,eukaryotic plasmid pcDN3.1(+) and attenuated typhia live vaccine Ty21a,-associated mimic epitopes of HCV hypervariable region 1,so to find a better way for vaccine immunization.Methods The DNA sequence based on the peptides' sequence of HCV hypervariable region 1 related consensus polymimotopes was synthesize and then inserted into plasmid pcDN3.1(+) to construct recombinant plasmid pcDN3.1(+)-SP,which then was transfected into live attenuated typhia vaccine Ty21a to construct Ty21a-SP.The mice were immunized orally with Ty21a-SP and intramuscularly with pcDN3.1(+)-SP,respectively,and then sacrificed by exsanguination.The splenocytes were separated and restimulated with pooled synthesized peptides,and then collected.Flow cytometry was employed to identify CD8+IFN-?+ T cells,and non-radioactive MTS method was adopted to test T cell proliferation,and non-radioactive LDH method was used to test cytotoxic T cytolytic reaction(CTL).Results Compared with control groups,the proliferation of splenocytes was apparently enhanced,the proportion of CD8+IFN-?+ cells obviously increased,and CTL responses also significantly increased after the spenocytes of mice immunized by pcDN3.1-SP and Ty21a-SP were restimulated with synthesized peptides.And the responses mentioned above in the mice immunized by Ty21a-SP were stronger than those mice immunized by pcDN3.1-SP.Conclusion Using attenuated typhia live vaccine Ty21a as DNA vector is an effective way to induce cellular immune responses.

Objective To prepare and characterize the polyclonal antibody against N protein of SARS-CoV. Methods The polyclonal antibody against SARS-CoV N was obtained from immunized rabbit with purified GST-N. The titer of the antibody was determined by indirect ELISA, and the specificity by Western blot and immunochemical staining. Results The rabbit′s antibody against SARS-CoV N was prepared successfully. The titer of antiserum against SARS-CoV N was about 1.2?10~ -5 . Western blot and immunochemical staining analysis showed that the polyclonal antibody could bind to the expressed fusion protein specifically. Conclusion The rabbit′s antibody against SARS-CoV N has been prepared successfully, and it can be a useful reagent for clinical diagnosis and further research.

Objective Through investigation and analysis of nursing services and needs for community home visiting in Yinchuan city in order to improve the quality of nursing service. Methods Using cluster sampling method to collect 300 residents from five community for questionnaire investigation in Yinchuan city.Questionnaire included general situation and evaluation standard of home care patients and service quality satisfaction questionnaire of home care. Results The residents had different degree of nursing needs and differences existed in satisfaction degree with different project of community home visiting. Conclusions Community home visiting need is high,and further improvement of nursing service quality for community home visiting is needed in Yinchuan city.

Aim To investigate the inhibitory effect of paeonol (Pae) on the human esophageal cancer cell line Eca-109 in vitro and in vivo and its effect on apoptosis.Methods Cytotoxic effect of Pae on Eca-109 cells cultured in vitro was measured by MTT assay.Anti-tumor activity was performed on BALB/c nude mice xenografts model.The morphologic changes of tumor tissue were observed under light microscope and transmission electron microscope.The apoptosis index was assessed by TUNEL.Results Pae had significant in- hibitory effect on the proliferation of Eca-109 cells,and the IC50 value was 0.342 mmol?L-1.In the model of human esophageal cancer xenografts in BALB/c nude mice,the inhibitory rates of Pae group (25、50、100、200 mg?kg-1) were 10.67%、23.54%、27.91% and 34.46% respectively.In vivo administration of Pae 100 mg?kg-1 combined with cisplatin 5 mg?kg-1 resulted in a significant inhibition of Eca-109 tumor growth with the inhibitory rate of 77.91%,compared with cisplatin used alone (58.71%).The more apoptotic tumor cells could be seen under light microscope in every theraperutic groups than those in control.Changes of ultrastructure of tumor cells including concentration and side accumulation of the nuclear chromatin,and the fragmentation of the nuclear was observed under transmission electronic microscope.Apoptosis body was also found.The apoptosis indexes of every theraperutic groups were significantly different from the control.Conclusion Pae can inhibit the cell growth and induce apoptosis in human esophageal cancer cell line Eca-109 in vitro and in vivo.

Objective To investigate the effect of taurine on iron bioavailability in rats and its possible mechanism. Method SD rats were randomly divided into three groups:normal group (NG),taurine treated group (TG),experimental control group (EG). TG and EG were induced to be anemic experimentally for 4w before the study. Then the rats were fed iron supplements for 25 d,meanwhile TG was treated with taurine following hemoglobin(Hb) repletion bioassay. Regression analysis of TG and AG were measured following Hb repletion bioassay using iron supplements. And the relative biological value of TG following Hb repletion were calculated. The serum iron,unsaturated iron-binding capacity(UIBC),total iron-binding capacity(TIBC),transferin saturation(TS)and the expression of hepcidin were detected. Results The iron bioavailability of TG was higher than EG,and the RBV of TG was 126%. Other hematological variables of TG were close to NG,and better than EG. The expression of hepcidin was increased compared with NG,but decreased compared with EG. Conclusion Taurine can improve iron bioavailability in rats via suppressing hepcidin expression in rats.
