1.Research progress of calcification and anti-calcification of implanted silicone rubber

Calcification of silicone rubber is a pathologic mineralization phenomenon which occurs on the surface after it is implanted in vivo,and there is no effective prevention method.The important process of calcification including these steps:phosphate ions combined with calcium ions to promote nucleating aggregation,slow increasing,fusion,and gradually form a larger accumulation of hydroxyapatite calcium.The research had been developed to the level of cells and genes,such as matrix Gla protein clusters is a calcification inhibitor of the tissues around silastic prosthesis.It is necessary to obtain a theoretical breakthrough in the aspect of organic template modulation of inorganic crystals.Silicone elastomer having its own local anti-calcification property had been paid more attention.Fully revealing the mechanism of calcification and calcification-resistant of silicone rubber will probably provide the basis for building a new anti-calcification material.

3.Relationship Between Efflux Pumps and Klebsiella pneumoniae Resistance Mechanism

OBJECTIVE To investigate whether efflux mechanism is involved in fluoroquinolone-resistant Klebsiella pneumoniae strains isolated in China. METHODS We compared the ciprofloxacin accumulation in clinically isolated K. pneumoniae with or without CCCP by fluorospectrophotometry. Use reverse transcriptase-polymerase chain reaction (RT-PCR) to measure the mRNA expression level of acrAB-tolC efflux gene. RESULTS The accumulation of ciprofloxacin in resistant strains was lower than that in susceptible ones, and it could increase to a high level nearly to the susceptible strains. The mRNA level of efflux gene acrA was higher in resistant strains than in susceptible ones. CONCLUSIONS Efflux mechanism is associated with the resistance to fluroquinolones in K. pneumoniae strains isolated in China and CCCP can inhibit its active efflux mechanism , which provides a sensitive method to detect the active efflux system of K. pneumoniae.
4.The study of clinical distribution characteristics of multi-drug resistance bacteria isolated from thehospitalized patients in a hospital of Shenzhen City

Objective To study the clinical characteristics of multi-drug resistance bacterias(MDROs) isolated from hospitalized patients in the Second People′s Hospital of Longgang District,to provide strategies for the prevention of MDROs infection.Methods The MDROs data of hospitalized patients from January 2015 to December 2016 were analyzed retrospectively.The multi-drug resistance incidence of each bacterias,each types of specimens and each clinical departments were analyzed and compared by SPSS16.0.Results A total of 104 strains of MDROs were isolated,and the top five bacteria were E.coli(32 strains,30.77%),coagulase negative staphylococcus(24 strains,23.08%),pseudomonas aeruginosa(16 strains,15.38%),staphylococcus aureus(10 strains,9.62%),kiebsiella pneumonia(10 strains,9.62%) respectively.There was significant difference in the multi-drug resistance incidence of each bacterias(χ2=20.62,P<0.05),the average incidence was 25.12%,and the top three incidence were E.coli(36.78%),pseudomonas aeruginosa(33.33%),coagulase negative staphylococcus(28.24%) respectively.There was significant difference in the multi-drug resistance positive rate of each types of specimens(χ2=43.68,P<0.05),the average positive rate was 5.84%,and the highest positive rate were wound secretion and pus(11.00%),followed by urine(8.25%).There was significant difference in the multi-drug resistance positive rate of each clinical departments(χ2=40.36,P<0.05),and the highest positive rate were in department of urinary surgery(12.63%),followed by department of gynaecology and obstetrics(11.16%).Conclusion E.coli coagulase negative staphylococcus and pseudomonas aeruginosa were mainly epidemic MDROs,and the MDROs are mainly distributed in urological surgery,obstetrics and gynecolog in this hospital.The occurrence of MDROs should be for the prevention and control strongly in the hospitalized patients with all kinds of trauma or diseases of urinary system and in the clinical department of urinary surgery,gynaecology and obstetrics.
5.Analysis on Plasma Concentration Monitoring of Rifampicin and Isoniazid in Tuberculosis Patients

OBJECTIVE:To promote plasma concentration monitoring of anti-tuberculosis agents in the clinic.METHODS:Results of plasma concentration monitoring of rifampicin and isoniazid in tuberculosis patients of our hospital from Aug.2005 to Dec.2008 were analyzed retrospectively.RESULTS:Plasma concentration samples were collected from 241 patients treated with rifampicin and 53 patients treated with isoniazid 2 hours after medication.Plasma concentrations of rifampicin in 10.4% of patients were below the normal level and 31.9% were higher than the normal level.Plasma concentrations of isoniazid in 71.7% of patients were below the normal level and 15.1% were higher than the normal level.CONCLUSION:Plasma concentration monitoring of anti-tuberculosis agents is a valuable tool to adjust the dosage of drugs,which is valuable for rational use of drugs in tuberculosis patients.
6.Adult form nemaline myopathy:two cases report with clinicopathological and ultrastructural study

Objective To investigate the clinical and pathological features of adult form nemaline myopathy. Methods Biopsied muscle specimens from two patients were observed using enzymatic histochemical method and electron microscope. Results These two patients were found with an initial onset from neck muscle weakness at the age of 52 and 36 years respectively, followed by trunk and limb muscle involvement to a variant degree. Type Ⅰ fiber atrophy and numerous granules (rod) were found in both patients in light microscopy. In case 1, the centralized nuclei were seen almost in all normal sized muscle fibers. Electron microscopic observation showed marked myofibril disorganization and numerous rod-like formations. Two inclusion-carrying nuclei were detected in case 1. The intranuclear inclusion showed a lattice pattern of Z-disc line or sarcoplasmic rod. Conclusions Adult form nemaline myopathy is clinically nonspecific. Rod-like structures might be found either in sarcoplasma or in nucleus. Centralized nuclei might coexist with rods in adult form nemaline myopathy. As compared to the child form, the adult one might often show a marked muscle fiber atrophy.
7.Selection of the exogenous signal peptide that conducts the secretion of hepatopoietin

AIM:To look for a suitable signal peptide which may effectively conduct hepatopoietin(HPO)secretion,various recombinant eukaryotic expression vectors were constructed.METHODS:Different exogenous signal sequences were spliced with HPO cDNA by PCR,and the spliced genes were cloned into eukaryotic expression plasmids.The different recombinants were respectively tansfected into COS-7 cells by Lipofectamine 2000 method and the secretion of HPO was analyzed by Western blotting.RESULTS:Western blotting analysis indicated that the signal peptides from interleukin-1 receptor antagonist(IL-1ra)and an artifical signal peptide did not secret HPO directly and effectively,but the signal peptide from murine Ig kappa secreted HPO directly with great efficiency.The molecular weight of the secreted HPO was 30 kD,which means that the secreted HPO existed in homodimer.CONCLUSION:Secreted recombinant expression plasmid is successful constructed.The result may pave the way for the gene therapy of hepatic fibrosis.

9.Antiviral and Antibacterial Effects of the Effective Site of Traditional Chinese Medicine

Objective To study the antiviral and antibacterial effects of the effective site of traditional Chinese medicine(TCM-ES).Methods Chicken embryoes were infected with influenza virus A(FM1 strain) and pre-injected with TCM-ES by means of chicken-embryo inoculation technique,and the antiviral effect of TCM-ES on chicken embryo was assayed by detecting the hemagglutination titers in allantoic fluids.Mice were orally pretreated with various dosages of drugs twice daily for 3 days,then were given drugs continuously for another 4 days following FM1 infection.The protective effects of TCM-ES on mice infected with FM1 were assayed by calculating the weight,index of lung,death-protection rate,and life-prolongation rate,etc.Ribavirin was used as the positive control.In addition,the antibacterial effects of TCM-ES were observed by detecting the minimum inhibitory concentration(MIC) and minimum bactericidal concentration(MBC) by test-tube dilution method.Results In chicken embryo experiments,TCM-ES showed a potential inhibiting effect on influenza virus with the MIC of 10 mg/mL,which was weaker than ribavirin.The results of animal experiment showed that the body-weight(BW) and pulmonary index of infected model group decreased evidently compared with those of the normal group,TCM-ES groups at the dosages of 750 mg/kg and 1500 mg/kg could reverse the decrease of BW and lung index as compared with the infected models,the difference being insignificant as compared with the normal group.Moreover,TCM-ES also increased the death-protection rate and life-prolongation rate of mice in a dose-dependent manner.TCM-ES at dosage of 10 mg/mL(MIC and MBC) had an antibacterial effect on staphylococcus,while had no effect on gram-negative bacilli.Conclusion TCM-ES has obvious antivirus effect on influenza virus FM1 strain,and also has certain antibacterial effect on staphylococcus,which is worth of further development and research.
10.Use of Antimicrobials in Operating Departments of Hospital

OBJECTIVE To investigate and analyze the application of prophylactic antibiotics in perioperation period in our hospital.METHODS According to the designed table with unified contents and standards,a retrospective investigation was made in 325 patients who had been discharged from our hospital.RESULTS Of the 325 patients in our study,antimicrobials use in 281 patients was rational,44 patients had irrational antimicrobials use.CONCLUSIONS The results show that it is very important to strengthen and improve the management of the antimicrobials use in a normal and standard way in our hospital.
