BACKGROUND:Studies have shown that theβ-tricalcium phosphate can be combined with other macromolecular compounds to improve the mechanical strength and histocompatibility, meeting the requirements of clinical application. ＜br> OBJECTIVE:To evaluate the selection and preparation methods of composite scaffold in bone tissue engineering, and to describe the problems we are facing. ＜br> METHODS:The first author retrieved PubMed, Google Scholar, CNKI, Wanfang, and VIP databases by computer using the keywords of“β-tricalcium phosphate,β-TCP, PLGA, PLA, scaffold, prepare”in English and Chinese, respectively. ＜br> RESULTS AND CONCLUSION:β-tricalcium phosphate combined with polylactic acid-glycolic acid/polylactic acid as bone tissue engineering scaffolds has achieved a great progress. At present, the organic polymer composite scaffolds can be prepared by microsphere sintering, fiber bonding, solvent casting/particulate leaching, emulsification/freeze drying technology, gas foaming method, phase separation technology, rapid prototyping technology and electrostatic spinning method.β-tricalcium phosphate composite scaffolds with polylactic acid-glycolic acid/polylactic acid can be customizable so as to meet different requirements for bone tissue engineering, which can improve the porosity, mechanical properties and biodegradation by improving fabrication techniques and processings, ratio of raw materials and fil er proportion.

Objective:To construct the expression vectors of VH and VL from an anti -CD71 monoclonal antibody (line7579) , and express an anti-CD71 mouse/human chimeric antibody (D2C) in vitro.Methods:The variable regions of heavy and light chains from an anti -CD71 monoclonal antibody (line7579) cloned in the pGEM-T vectors were constructed respectively into the expression vectors,p?-Expr(including human IgG1 constant region) and p?-Expr(including human Ig? constant region).The expression plasmid vectors (Chi7579-p?-Expr and Chi7579- p?-Expr) ,confirmed by a clone-PCR and a further sequence analysis, were co-transfected by electroporation into the non-Ig-producing murine hybridomas SP2/0.Results:Screening for transfectomas secreting chimeric antibody was done by cross sandwich ELISA, which identified the transfectomas to secrete a chimeric antibody (D2C) with the human heavy constant region(C?) and light constant region(C?). FCM analysis of culture supernatants of the transfectomas demonstrated that the binding capacity of the chimeric Ab (D2C) to the antigen (CD71) was retained.Conclusion:The construction of the V H and V L expression vectors and the expression of an anti-CD71 mouse/human chimeric antibody (D2C) were accomplished successfully.

Objective Effect of preventing recurrent pterygium excision combining amnion transplant by using recombinate interferon aIb drip eyes. Methods 105 cases of pterygium excision and combining amnion transplant patients were divided two groups,one group was treated by interferon aIb dripping eyes for 50 pterygium, another group was treated by 0.1%fu mi long drip eyes liquid to drop eyes as a contrast group. Results 50 patients in treatment group were followed up a case by regular visits to months after the operation and none of them had not been seen pterygium recurrent after passing 12～24 months and average time was 15 months;55 patients in contrast group had 6 recurrents,recurring rate was 10.9%,the statistic difference was significant( P

Objective To study the clinical significance of a novel marker of platelet clumps count provided by hematology analyzer in differentiating true thrombocytopenia from EDTA-dependent pseudothrombocytopenia (EDTA-PTCP). Methods Samples from 65 cases of thrombocytopenia (including 15 EDTA-PCTP samples and 50 random samples of true thrombocytopenia) and 50 healthy controls were analyzed using hematology analyzers, and samples with low platelet counts were checked by replacing citric acid and using manual microscope observation to identify true thrombocytopenia from EDTA-PTCP. A novel marker of platelet clumps count was used to differentiate the two diseases for samples anficoagulated with EDTA or citric acid. Results In 65 patients with thrombocytopenia, platelet counts were (48±11)×109/L detected by automatic hematology analyzers. Fifty of 65 cases were true thrombocytopenia which showed low platelet counts [(48±10)×109/L by automated analyzer and (46±11)×109/L by manual assay]. No significance was observed between them (t=-1.26, P0.05). Platelet clumps counts were 86±15. No platelet clamps were detected under microscope. The other 15 cases were EDTA-PTCP [platelet counts were (48±12)×109/L and platelet clumps counts (840±184) were increased significantly by automated analyzer and using EDTA anticoagulant] which showed obviously platelet clumps and no less platelet counts under microscope. After replacing citric acid, platelet counts [(141±13)×109/L by automated analyzer and (134±17)×109/L by manual microscope assay] were increased significantly. No significance was observed between them (t=-1.29, P0.05). Platelet clumps counts (75±12) were decreased obviously compared with EDTA anticoagulant method (t=-6.82, P<0.001). No platelet clumps were detected under microscope. Conclusion Platelet clumps counts may be a useful clinical indicator for monitoring of platelet aggregates, especially for EDTA-PTCP caused by platelet clumping.

OBJECTIVE To identify the mechanisms by which the formyl peptide receptor 2 (FPR2) mediates both inflammatory and anti-inflammatory signaling in an agonist-dependent manner. METHODS Cells expressing FPR2 were incubated with weak agonists, Aβ42 and Ac2-26, before stimulation with a strong agonist, WKYMVm. Calcium mobilization, cAMP inhibition and MAP kinase activation were measured. Intramolecular FRET were determined using FPR2 constructs with an ECFP attached to the C- terminus and a FlAsH binding motif embedded in the first or third intracellular loop (IL1 or IL3, respectively). RESULTS Aβ42 did not induce significant Ca2 + mobilization, but positively modulated WKYMVm-induced Ca2 + mobilization and cAMP reduction in a dose-variable manner within a narrow range of ligand concentrations. Treating FPR2-expressing cells with Ac2-26, a peptide with anti-inflam?matory activity, negatively modulated WKYMVm-induced Ca2 + mobilization and cAMP reduction. Intra?molecular FRET assay showed that stimulation of the receptor constructs with Aβ42 brought the C-terminal domain closer to IL1 but away from IL3. An opposite conformational change was induced by Ac2-26. The FPR2 conformation induced by Aβ42 corresponded to enhanced ERK phosphorylation and attenuated p38 MAPK phosphorylation, whereas Ac2-26 induced FPR2 conformational change corresponding to elevated p38 MAPK phosphorylation and reduced ERK phosphorylation. CONCLUSION Aβ42 and Ac2-26 induce different conformational changes in FPR2. These findings provide a structural basis for FPR2 mediation of inflammatory vs anti-inflammatory functions and identify a type of receptor modulation that differs from the classic positive and negative allosteric modulation.

Objective To observe the clinical efficacy of the combined therapy of super-laser radiation and plum acupuncture on alopecia areata.Methods Altogether 178 alopecia areata patients were randomly divided into 3 groups:65 patients in the treatment group being given super-laser radiation and plum acupuncture,55 in the control group A solely with super-laser radiation and 58 in the control group B solely with plum acupuncture.The therapeutic effects of the treatment group and the control groups were compared by rank sum test.Results At the end of the 1st treating period,the cured cases,markedly effective cases,ordinary effective cases and invalid cases in control group A were 1,4,10 and 40;those numbers of control group B were 1,3,11 and 43;those numbers of the treatment group were 3,12,14 and 36.At the end of the 2nd treating period,those numbers of control group A were 8,8,16 and 23;those numbers of control group B were 7,8,18 and 25;those numbers of the treatment group were 11,20,19 and 15.At the end of the 3rd treating period,those numbers of A control group were 21,11,12 and 11;those numbers of B control group were 20,14,11 and 13;those numbers of the treatment group were 33,17,10 and 5.Comparing the performance of each groups at the end of the 3 periods,we found that effects were always better in the treatment group than the other two control groups.The difference between the treatment group and the other two control groups was statistically significant (P＜0.05).Conclusions It is effective and safe to use super laser radiation combined plum acupuncture on alopecia areata.The clinical efficacy of this joint method is better than either of the two separate ways.

OBJECTIVE The chemokine-like receptor 1 (CMKLR1, ChemR23) is a functional receptor for chemerin, the chemerin-derived nonapeptide (C9), and the amyloid β peptide 1-42 (Aβ42). Because these peptides share little sequence homology, studies were conducted to investigate their pharmaco?logical properties and regulation at CMKLR1. METHODS Cells expressing CMKLR1 were incubated with Aβ42 before stimulation with a strong agonist, the C9 peptide. Calcium mobilization, cAMP inhibition and MAP kinase activation were measured. Intramolecular FRET were determined using CMKLR1 constructs with an ECFP attached to the C- terminus and a FlAsH binding motif embedded in the first intracellular loop (IL1). RESULTS Binding of both Aβ42 and the C9 peptide induced CMKLR1 internal?ization, but only the Aβ42-induced receptor internalization involved clathrin-coated pits. Likewise, Aβ42 but not C9 stimulated β-arrestin 2 translocation to plasma membranes. A robust Ca2+ flux was observed following C9 stimulation, whereas Aβ42 was ineffective even at micromolar concentrations. Despite its low potency in calcium mobilization assay, Aβ42 was able to alter C9 -induced Ca2+ flux in dose-dependent manner: a potentiation effect at 100 pmol·L-1 of Aβ42 was followed by a suppression at 10 nmol·L-1 and further potentiation at 1 μmol·L-1. This unusual and biphasic modulatory effect was also seen in the C9-induced ERK phosphorylation but the dose curve was opposite to that of Ca2+ flux and cAMP inhibition, suggesting a reciprocal regulatory mechanism. Intramolecular FRET assay confirmed that Aβ42 modulates CMKLR1 rather than its downstream signaling pathways. CONCLUSION These findings suggest Aβ42 as an allosteric modulator that can both positively and negatively regulate the activation state of CMKLR1 in a manner that differs from existing allosteric modulatory mechanisms.

Objective To compare the safety and oncological results of robot-assisted laparoscopic radical prostatectomy performed between transperitoneal and extraperitoneal approach.Methods 79 patients underwent transperitoneal robot-assisted laparoscopic radical prostatectomy (Tp-RLRP).The other 86 consecutive patients went through extraperitoneal robot-assisted laparoscopic radical prostatectomy (Ep-RLRP).Results All procedures were performed successfully without conversion.Total procedure time for Ep-RLRP was shorter than it for Tp-RLRP[(218.1 ± 54.4) min vs.(180.2 ± 47.1) min,P ＜ 0.001].Docking time for Ep-RLRP was longer than Tp-RLRP [(21.5 ± 7.1) min vs.(19.5 ± 5.7) min,P ＜ 0.001] Blood lost of Tp-RLRP was lesser than Ep-RLRP[(122.4 ± 71.0) ml vs.(140.4 ± 90.7) ml,P ＜ 0.05].There were no significant differences between two groups in terms of bowel function recovery,drainage tube reserve,foley catheter duration,etc.There were 19 patients (24.1％)and 4 patients (4.7％)were suffered lymph leakage in two groups separately,P ＜ 0.001.One delayed vesiclurethra hemorrhagic patient and one mesenteric torsion patient were happened in Tp-RLRP group.There were no significant differences between two groups in terms of surgical margin,biochemical recurrence and urine continence.Conclusion Compared to Tp-RLRP,Ep-RLRP can provide similar short-term oncological control,recovery of postoperative urine continence,shorter operation time and lower complication rates.

Objective To assess the safety and feasibility of utility of the third arm in robotic partial nephrectomy(RPN) through retroperitoneal approach for complex renal tumor.Methods 36 roboticassisted partial nepbrectomy were performed by one surgeon between November 2015 and January 2017,including hilar tumors in 12 cases,central tumors in 11 cases,endophytic tumors in 7 cases and multiple tumors in 6 cases.A 12 mm camera port is placed 2 fingerbreadth above iliac crest.The lateral and medial robotic trocars are placed in the posterior axillary line and anterior axillary plane respectively parallel to the cameraport trocar.Under direct visualization,the peritoneum is swept medially towards the paramedianplane.The fourth arm trocar is placed in the most medial and inferior aspect of the field approximately 7 cm to 8 cm across and parallel to the medial robotic trocar.Descriptive statistics on patient characteristics,operative parameters,and oncologic outcomes are analyzed.Result The 4-arms retroperitoneal approach was used in all patients without any conversion.Mean console time was 127 ± 21 (98-357) min.Mean ischemia time was 25.5 ± 8.3 (12-38) min,Mean estimated blood lost was 198 ± 201.5 (50-510) ml.No patients required blood transfusion,except one case underwent selective intra-arterial embolization with DSA (digital subtraction angiography) and blood transfusion post-operatively.Pathology revealed renal cell carcinoma in 12 patients,angiomyolipoma in 16 patients,chromophobe renal cell carcinoma in 6 cases,oncocytoma in 2 cases and all had negative surgical margins.Under 3 months follow-up,Mean decrease in eGFR was 4.3 ml/(min · 1.73m2).No patients found recurrence or metastasis.Conclusions The third robotic arm provides the console surgeon maximal independence from the surgical assistant when performing kidney retraction,ligation or clamping of renal hilar vessels in complex renal morbidities.It would be beneficial for patient with merits of retroperitoneal approach and overcomes limited space during RPN.

We investigated the third-generation cephalosporins-resistant Shigella and its genotype in Ningbo,China,providing a basis for disease prevention and control.Pathogenic bacteria were analyzed by direct isolation combined with enrichment culture isolation.Antimicrobial susceptibility was determined by K-B disk diffusion method and PCR was used for detecting multidrug resistance genes like CTX-M,OXA,TEM and SHV.BLAST analysis was used to determine the genotype.Results showed that 69 strains of third-generation cephalosporins-resistant Shigella were detected by drug sensitivity screening,accounting for 74.19％ of ESBLs Shigella.Drug resistance gene CTX-M(CTM-M-1 and CTM-M-9),OXA and TEM were detected.The detection rate were 79.71％,79.01％ and 26.09％ respectively.With no CTX-M-2 and SHV,DNA sequence alignment showed CTX-M-1 group were mainly of CTX-M-15 type besides seven other types;CTX-M-9 group were mainly of CTX-M-14 type besides six other types;49 strains of OXA and 18 strains of TEM were sequenced to be type 1 (OXA-1 and TEM-1 type).The 21 Shigella strains carrying more than two drug resistance genes accounts for 30.43 ％.Shigella in Ningbo has high third-generation cephalosporins-resistance rate and many kinds of ESBLs enzymes were detected.The mainstream enzyme type was CTX-M,meanwhile they also carried a variety of drug resistance genes,which could bring difficulties to disease prevention and control.The high carrying rate of OXA-1 type suggests that we should pay more attention.The detection rate of group B was higher than that of group D,including not only the phenotype resistance but also the drug-resistance genes;these findings will be useful in the study of the drug resistance prevalence of Shigella.
