1.Expression and clinical significance of hypoxia-inducible factor-1α,vescular endothelial growth factor and survivin in colorectal cancer

Objective To evaluate the expressions of hypoxia-inducible factor-1α(HIF-1α),vescular endothelial growth factor(VEGF)and survivin in developing colorectal cancer,and the association among them.Methods The protein expressions of HIF-1α,VEGF and survivin were detected in specimens obtained from 69 patients with colorectal cancer and 20 normal controls by immunohistochemistry.The correlations of HIF-1α,VEGF and survivin with clinicopathologic features were analyzed.Results The expression of HIF-1α,VEGF and survivin proteins in patients with colorectal cancer were 56.52%,66.67% and 46.38%,respectively,and little expressed in normal controls(P<0.01).The expressions of HIF-1α,VEGF and survivin were closely related to the differential grade of adenocarcinoma,he in volvment of penetration,lymph node metastasis and Dukes stage(P<0.05).In addition,HIF-1α protein expression had positive correlations with both VEGF and survivin(P<0.05),and the expression ofVEGF also had positive correlation with survivin(P<0.05).Conclusions HIF-1α may involve in thepathogenesis of colorectal cancer through up regulating the expressions of VEGF and survivin,which mayhave synergetic effects in the pathogenesis of colorectal cancer.
2.Correlation between psychological capital and compassion fatigue in operating room nurses

Objective To investigate the status quo of psychological capital and compassion fatigue in operating room nurses and to analyze the correlation between them.Method One hundred and ninety nurses from an operating room of four First-Class Grade A general hospitals in Beijing were engaged in the investigation by psychological capital questionnaire and compassion fatigue scales.Results The total score on psychological capital was (78.60±8.72) and the total score on compassion fatigue was (111.53 ±9.88).The mental capital was negatively correlated with compassion fatigue (P <0.05).Conclusions The psychological capital of the nurses in the operating room is at the middle level and the sympathetic fatigue is at a high level.The higher the psychological capital,the lower the sympathetic fatigue.Nursing administrators can improve the psychological capital of the nurses in the operating room to reduce their sympathy and fatigue so as to improve the level of mental health of nurses in the operating room.
3.A family-based association study of the autism and PTEN genes polymorphisms in children

Objective To explore the correlation of single nucleotide polymorphism (SNP) with PTEN gene involving in mammalian target of rapamycin (mTOR) C1 signaling genes polymorphisms and autism in children.Methods A total of 97 cases with autism were enrolled from Mar.2011 to Dec.2012 in this study,who came from the child and adolescent out-patient department in Shanghai Mental Health Center of Shanghai Jiaotong University School of Medicine.Single SNP association and haplotype association analysis were performed using the family-based association test and Haploview software.Results 1.In a family-based association test,two SNPs showed significant association with autism(rs17107001 G:Z =2.982,P =0.003 ; rs2299941 G:Z =2.524,P =0.012).After the correction of false discovery rate,they all remained significant.2.Haplotype association analysis showed significant transmission disequilibrium in haplotype T-T-G and C-T-A generated from rs532678-rs17562384-rs2299941 (block2) in LD Block,and haplotype T-T-G was over transmitted to offspring(Z =-2.986,P =0.003) while haplotype C-T-A was the opposite (Z =-2.197,P =0.028).Conclusion The SNPs of PTEN genes might have a correlation with autism in children.
4.Related factors of no or weak immune response after inoculation of hepatitis B vaccine

Objective To study the related factors of no or weak immune response after inoculation of hepatitis B vaccine .Meth‐ods 120casesofadultreceivedinoculationofhepatitisBvaccineinourhospitalwereenrolled,including60casesofnoweakre‐sponse and 60 cases of normal response ,they were divided into non response group and response group .Related factors of no or weakimmuneresponsewereanalyzed.Results (1)singlefactoranalysis:Therewerestatisticalsignificanceofage,gender,BMI, smoking history ,HBV potential infection ,CD4+ T cells and CD8+ T cell content ,IFN‐γand IL‐2 content between two groups ;(2) logistic regression analysis :old age ,male ,smoking history ,high values of BMI ,HPV latent infection ,low content of CD4+ T cells and IFN‐γand IL‐2 ,high content of CD8+ cells were risk factors of no or weak immune response after inoculation of hepatitis B vaccine .Conclusion No or weak immune response after inoculation of hepatitis B vaccine is affected by gender ,age ,smoking ,obesi‐ty ,potential infection of HPV and immune function ,and necessary measure should be taken .
5.Association study between polymorphisms of TSC1, TSC2 ,PTEN genes and autism in Chinese Han population

Objective To explore the association between the polymorphisms of TSC1,TSC2,PTEN genes and autism in Chinese Han population.Methods 274 autism patients and 386 heahh controls were recruited,and SnaPshot technique was used to genotype the 13 tagSNPs of TSC1,TSC2 and PTEN genes.The allele,genotype and haplotype frequencies of the SNPs were compared using SHEsis and SNPStats softwares.Results Mter Bonferroni correction,the allele distribution of rs2809244 (TSC1) (x2 =9.537,P=0.002,adjusted P=0.016),rs1050700 (TSC1) (x2 =9.313,P=0.002,adjusted P=0.016),rs2072314(TSC2) (P<0.01,adjusted P<0.01) and rs8063461 (TSC2) (P<0.01,adjusted P<0.01)showed significant difference between two groups (P<0.05).The genotype frequencies of rs2072314(TSC2)and rs8063461(TSC2) showed significant difference between two groups(P<0.05).Moreover,the frequency of haplotype A-G (OR =14.548,95% CI =5.450-38.830) in the haplotype block rs2809244-rs3761840 showed significant difference between two groups(P<0.05),A-G significantly increases the risk of autism.The frequencies of haplotype A-A (OR=0.608,95% CI =0.409-0.903,P=0.013),G-A (OR=7.812,95% CI =5.338-11.459,P<0.01)and G-G (OR=0.356,95% CI =0.274-0.463,P<0.01) in the haplotype block rs2074969-rs8063461 were identified,which were significant difference between two groups(P<0.05),and AA and G-G significantly reduced but G-A increased the risk of autism.Conclusion The polymorphisms of TSC1 and TSC2 genes might associate with autism in Chinese Han population.
6.Association study of dopamine D1 receptor gene and cognitive function of first-episode schizophrenic patients

Objective To investigate the relationship between cognitive function of first-episode schizophrenic patients and dopamine D1 receptor gene. Methods A total of 112 first-episode schizophrenic patients and 60 healthy controls were evaluated with Wechsler adult intelligence scale ( WAIS-R), Wechsler memory scale (WMS) and Wisconsin card sort test (WCST) ,and genotyped one polymorphism (rs4532) within DRD1 gene using TaqMan SNP genotyping assay. Results There were no significant differences on the frequencies of the genotypes and alleles of rs4532 polymorphism between patients with schizophrenia and normal controls ( x2 =2.90, P=0.35; x2 = 0.01, P= 0. 93 ). There were significant differences in all index of WCST between two groups (P <0.01 ). Patients with rs4532G allele had worse WCST performance than those without G allele ((60.9 ± 13.2)%vs (44.9 ±21.3)%, t=4.79, P=0.00002). Conclusion Rs4532 polymorphism of DRD1 gene may be associated with executive function impairment in schizophrenic patients.
7.Research about sexual knowledge nursing intervention for femal workers in Dongguan

Objective To know the master condition about sexual knowledge in female workers in Dongguan, and then carry out certain effective community nursing intervention to populize the related knowledge in female workers. Methods Divided 3000 femle workers with 18-35 years old who had selected in 9 factories in Dongguan into the DV group and the teacher education group randomly, there were 1500 cases in each group. Video displaied was used in the DV group in certain propert time, teacher education about sexual knowledge was used in the teacher education group, compared the master condi-tion of sexual knowledge before and after the interventioni by questionnair in the two groups respectively, and then summarized the results. Results There were no significant differences between the two groups about sexual knowledge before the intervention, while after the intervention, the master conditon of sexual knowl-edge in the DV group were better than those of in the teacher education group. Conclusions Video dis-play is a kind of saving, effectiveness, open-end and circular mode for female workers to pepulize sexual-related knowledge, which should be ranging used.
8.The use of percutaneous endoscopic gastrostomy and percutaneous endoscopic jejunostomy in enteral nutrition

Objective: To establish the method of percutaneous endoscopic gastrostomy(PEC) and percutaneous endoscopic jejunostomy (PEJ) for enteral nutrition. Methodes: PEG tubes were placed in 114 patients with Pull method. On the foundation of PEG, PEJ tubes were placed in 26 patients by pushing endoscopy to send tubes through Treitz ligment with usingthe the clip. Results: All PEG insertion was performed successfully. PEJ tubes were placed successfully with a new method in 26 patients. 15 patients had a little blooding and 8 patients had slight infection. 21 patients had respiratory tract infection and had been cured by using antibiotic. There was no severe complication. Conclusion: PEG is simple、safe、efficient. The new method of PEJ is feasible.
9.Liraglutide promotes proliferation and migration of cardiac microvascular endothelial cells through PI3K/Akt and MAPK/ERK signaling pathways

Objective To investigate the effect of liraglutide, an analogue of glucagon-like peptide-1, on the proliferation and migration of cardiac microvascular endothelial cells (CMECs) and explore the mechanism. Methods In vitro cultured CMECs of SD rats were purified by differential adhesion method and identified immunocytochemically using CD31 antibody and factor VIII. MTT assay was performed to assess the proliferation of the first- generation cells exposed to different concentrations (0-1000 nm/L) of liraglutide. Western blotting was used to detect the activation of PI3K/Akt and MAPK/ERK signaling pathways. BrdU fluorescent labeling and scratch assay were performed to observe the proliferation and migration of CMECs following liraglutide treatment, and PI3K/Akt and MAPK/ERK pathway inhibitors LY294002 and PD98059, respectively, were used to further confirm the role of these signaling pathways in regulating the proliferation and migration of CMECs. Results Immunocytochemical staining demonstrated a proportion of double positive cells exceeding 95%. The cells exhibited a logarithmic growth 48 h after plating. Liraglutide exposure concentration-dependently promoted the proliferation of CMECs with the optimal concentration of 100 nmol/L (P<0.05). Liraglutide exposure of the cells for 24 h significantly increased the levels of intracellular phosphorylated Akt and ERK (P<0.05), but pretreatment of the cells with Akt and ERK signaling pathway inhibitors 1 h before liraglutide obviously reversed such effect (P<0.05). BrdU and scratch assay showed that 100 nmol/L liraglutide significantly promoted the proliferation and migration of CMECs (P<0.05), but such effects were obviously suppressed by Akt and ERK inhibitors (P<0.05). Conclusion Liraglutide promotes the proliferation and migration of CMECs in vitro via PI3K/Akt and MAPK/ERK signaling pathways.
