Tumor stem cells (TSCs) are a group of cells which have self-renewal and differentiation potential in tumor tissues.TSCs not only play an important role in the occurrence and development in tumors , but also are closely related to tumors′invasion,metastasis,heterogeneity and chemo-resistance.Further research of TSCs has become a hot spot .This review intends to explain the formation of TSCs ,the relationship between TSCs and the microenvironment , as well as the model of the TSC microenvironment .Signalling pathways and regulatory factors of TSCs are also discussed .
2.Effect of Pirfenidone on Activity of Hepatic Cytochrome P450 in Rats

Objective To study the effects of pirfenidone on total enzyme and isoenzyme of liver microsomal cytochrome P450 in rats. Methods The activites of liver microsomal dimethyl nitrosamine N-demethylase( NDMA )and erythromycin demethylase( ERD ) were determined. Fifty-six SD rats were randomly divided into six groups,in which they received CMC,dexamethasone 100 mg·kg-1 ,ketoconazole 40 mg·kg-1 ,pirfenidone 25,50,100 mg·kg-1 ,respectively. After administration for 6 and 12 days,livers were prepared liver microsome,the concentration of proteinum in microsome and shade selection to plasma samples were determined by spectrophotometer. Results After administration of pirfenidone for 6 days, cytochrome P450 was significantly increased in 50 and 100 mg·kg-1 pirfenidone groups,as compared with solvent control group (1%sodium carboxymethylcellulose)(P﹤0. 01). After administration of pirfenidone for 6 and 12 days,NDMA of liver microsome was not changed significantly(P﹥0. 05). ERD of liver microsome was significantly increased in 100 mg·kg-1 pirfenidone group after administration for 12 days(P﹤0. 01). Conclusion Pirfenidone can induce P450 and ERD activities in a dose-and time-dependent manner.

Objective To investigate the effect and mechanism of decitabine (DAC) on the proliferation of human cholangiocarcinoma CCLP1 cells in vitro and in vivo.Methods After treated with various concentrations of DAC,cell growth inhibition rates were determined by CCK-8 assay.Cell cycle and cell apoptosis were analyzed by flow cytometry.Cell autophagy was observed under fluorescence microscope.The effect of DAC on the growth of cholangiocarcinoma in vivo was determined in a CCLP1 mice xenograft model.Results The proliferation rate of CCLP1 cells in the DAC-treated group decreased in a time-concentrated dependent manner.After treatment with DAC,the cell cycle of CCLP1 cells was arrested at the G2/M phase.The apoptosis rate was significantly higher in the treatment group over the control group.Cell autophagy was observed after treatment with DAC in CCLP1 cells.The tumor growth of implanted CCLP1 cells significantly slowed down after the mice were treated with 0.8 mg/kg DAC,6 times weekly for 2 weeks.Conclusion DAC can inhibit the proliferation of cholangiocarcinoma in vitro and in vivo.
5.Polymorphisms of (AC)n (AT)xTy in the upstream of β-globin gene in patients with mild β-thalassemia

ObjectiveTo explore linkage relationship between polymorphisms of (AC)n (AT)xTy and mutations in the β-globin gene in patients with mild β-thalassemia.MethodsThe subjects were 89 mild β-thalassemia patients with known mutations and 110 healthy subjects from People's Hospital of Baoan District of Shenzhen from February 2009 to July 2010.Genomic DNA was extracted from peripheral leukocytes.Sequence of the BP1 binding site upstream of the β-globin gene was amplified by polymerase chain reaction,polymorphisms of (AC)n (AT)xTy were determined by DNA sequencing.Allelic frequencies of (AC)n (AT)xTy between mild β-thalassemia patients and healthy subjects were compared using x2 test.Mutation rates between two groups were also compared using x2 test for subjects carrying same haplotype. Linkage relationship was conducted according to allelic frequencies and mutations. Results Analysis of the (AC)n(AT) xTy polymorphisms of the BP1 binding site upstream of the β-globin gene showed 9 different genotypes: (AC)2( AT)7T7,( AC)2( AT)8T5,( AC)3( AT)7T5,( AC)2( AT)9T5,( AC)2(AT)8T9,(AC)3(AT)8T5,(AC)2(AT)10T3,(AC)2(AT)7T5 and (AC)2(AT)11T3.The (AC)2(AT)7T7 and (AC)2 (AT)8T5 genotypes were common for patients with mild β-thalassemia.Allele frequencies of (AC)2(AT)7T7,(AC)3 ( AT)7T5 and ( AC)2( AT)8T9 were 38.8% (69/178),11.8%(21/178),9.0% ( 16/178 ) for mild β-thalassemia patients,and 24.1% ( 53/220),5.4% ( 12/220),3.2%(7/220)for healthy subjects, respectively, there were significant differences between mild β-thalassemia patients and healthy subjects (x2 =9.966,4.371,6.093,P < 0.05 ).Allele frequency of (AC)2(AT)9T5 was 10.1% (18/178) and 33.2% (73/220) for mild β-thalassemia patients and healthy subjects,frequency of (AC)2 (AT)9T5 was significandy lower in mild β-thalassemia patients than in healthy subjects (x2 =29.691,P <0.01 ).Allele frequency of (AC)2(AT)8T5 was 25.3% (45/178) and 29.1%(64/220) for mild β-thalassemia patients and healthy subjects,there wasn't significant difference between patients and healthy subjects (x2 =0.718,P >0.05).The mutation rates of codon41/42(-TTCT) and IVSⅡ-654(C→T) were 59% (10/17) and 29% (5/17) for mild β-thalassemia patients carrying (AC)2(AT)7T7 allele,and 29% (4/14) and 57% (8/14) for patients carrying ( AC)2 (AT)8T5 allele.There were not significant differences between codon41/42(-TTCT) mutation rate and IVS-Ⅱ-654(C→T) mutation rate (x2 =2.982,2.333,P > 0.05 ) for mild β-thalassemia patients carrying ( AC)2 ( AT)7T7 and ( AC)2(AT)8T5 allele.ConclusionsAllele of (AC)2(AT)7T7,(AC)3(AT)7T5 and (AC)2(AT)8T9 are in linkage disequilibrium with β-thalassemia.Most mild β-thalassemia patients carrying (AC)2 (AT)7T7 allele are caused by codon41/42 (-TTCT) mutation in the β-globin gene,and IVS-Ⅱ-654 (C→T) is a major mutation for patients carrying (AC)2(AT)8T5 allele.
6.Effect of Jieyuyubao Prescription on Endometrial Receptivity for the Chronic Stress Rats

Objective To study the effect of Jieyuyubao prescription on endometrial pinopodes and HOXa-10 mRNA in chronic stress rats , and reveal the regulatory mechanisms of the endometrial receptivity. Methods Totally 48 SPF SD rats were randomly divided into three groups with weight stratified: normal control group (group A), model control group (group B) and experimental drug group (group C). Rat model of chronic stress was established in group B and group C. On the first day of stimulation, Jieyuyubao solution (1 g·mL-1 ) was intragastrically administered (11. 8 mL·kg-1 ) to the rats of group C at 9:00 am, once daily, and stopped giving at the fourth day of pregnancy (pd4) after being confirmed by vaginal smears. Purified water (11. 8 mL·kg-1 ) was intragastrically administered to the rats of group A and group B at 9:00 am. All the rats were mated at the sex ratio of female:male = 2:1 at 16:00 pm on the 21st day. The rats were sacrificed at the pd4. Maturity of endometrium pinopodes was assessed under transmission electron microscope. HOXa-10 mRNA content was evaluated by real-time fluorescence-PCR. Results The maturity of pinopodes was as follows: the maturity score of normal control group, model control group and experimental group was (2. 7 ± 0. 3), (1. 3 ± 0. 3), and (2. 1 ± 0. 2), respectively, with significant difference between experimental drug group and model control group (P<0. 05). HOXa-10 mRNA expression was significantly decreased in model control group (0. 658±0. 031) as compared with normal control group (0. 965±0. 102) (P<0. 05). Conclusion Jieyuyubao prescription maybe increase the number and maturity of pinopodes through promoting HOXa-10 mRNA expression in endometrial epithelium cells, and then improve endometrial receptivity.
7. Expression of FNDC5/Irisin in pancreatic cancer and its relationship with clinicopathological features

Objective:
To investigate the expression and distribution of FNDC5/Irisin in pancreatic cancer tissues and adjacent tissues, and to analyze its correlation with clinicopathological features.
Methods:
Collection of archived wax blocks from 64 patients diagnosed with pancreatic cancer after surgical treatment from January 2015 to December 2018 in the Department of Pathology, Affiliated Qingdao Municipal Hospital of Qingdao University, and 30 tissues collected intraoperatively from January 2016 to December 2018 Samples, all collected samples included tumor tissue and corresponding adjacent tissues (>2 cm from the tumor edge). Real-time quantitative PCR (qRT-PCR) and immunohistochemistry were used to detect the expression of FNDC5/Irisin mRNA and its positivity in pancreatic cancer tissues and adjacent tissues, and to analyze its relationship with clinicopathological features of pancreatic cancer.
Results:
qRT-PCR showed that the expression of FNDC5/Irisin mRNA in pancreatic cancer tissues was higher than that in the corresponding adjacent pancreatic tissues, the difference was statistically significant (
