1.Changes of the function of vascular endothelial secretion and OX-LDL in obese children before and after exercise intervention

Objective To observe the changes of the function of vascular endothelial secretion, levels of OX-LDL and body-weight of obese children after exercise intervention. Methods Thirty children with obesity at the age of 8 to 14 years and 30 normal children were enrolled in this experiment. The obese children (exercise group) received a session of 1 hour's exercise each day, 5 days a week for 8 weeks. The levels of ET, 6-K-PGF1?,NO, OX-LDL and body-weight were then measured and compared with that of their pre-exercise state and that of the normal children (control group). Objective To explore the ways of repairing the large long-bone and skin defects as single stage procedures. Methods Five clinic cases were studied, firstly, the arteriae fibularis were restructuered into one or two vaso-nasa which nourished the related fibula segments and the overlying skin, then, binding all of the fibula segments and turn it into a bulky bone with a periosteal vaso-nasa wraparound. The graft was rich in blood supply so osteogenesis activity was vigorous under the periostum, the interspace in the graft soon disappear and the graft increased in diameter gradually. Results All of the harvested skin flaps survived and the wound healed on schedule, besides, the involved fractures were union 1 year later and the grafts were strong enough for weight-bearing and walking 2 year later, Neither fracture occur nor morbidity was created at the donor site, No problem was caused at the ankle. Conclusions It's appropriate way to repair large long-bone and skin defects with a vascularized fibula segments complex transfer incorporationg the overlying skin that was supplied via the same peroneal vessel pedicle.
3.Analysis on Cardiovascular Drugs Used in Our Hospital during the Period2001~2003

OBJECTIVE:To evaluate the situation of the use of cardiovascular drugs and its trend of develop?ment.METHODS:The cardiovascular drugs used during2001~2003were analyzed retrospectively in respect to the sale amount,DDDs and etc.RESULTS:The sale amount of cardiovascular drugs was increasing year by year;the antihypertensives and antianginals ranked at the first two places in respect to the sequence of the amount of money and the DDDs;the sale amount of joint-venture products was the largest,and there was a high proportions of national essential drugs consump?tion.CONCLUSIONS:There was a stable and rational structure of cardiovascular drugs in our hospital.Domestic substitute drugs should be exploited for medical researchers in the future so as to reduce the cost of medicine.
4.Molecular cloning and sequence analysis of(S)-N-methylcoclaurine-3'-hydroxylase gene in Coptis chinensis

Objective To clone and sequence the cDNA encoding(S)-N-methylcoclaurine-3'-hydroxylase from Coptis chinensis.Methods The cDNA,encoding(S)-N-methylcoclaurine-3'-hydroxylase,was amplified by RT-PCR with cDNA library of tender leaf as the template.Results The full-length cDNA of(S)-N-methylcoclaurine-3'-hydroxylase(named as CYP80B3) had 1 680 bp with an open reading frame encoding 488 amino acids of protein.The CYP80B3 had 95%,82%,70%,and 68% amino acid sequence homology to the sequence of(S)-N-methylcoclaurine-3'-hydroxylase from C.japonica,Thalictrum flavum,Eschscholzia californica and Papaver somniferum,respectively.The sequence was reported to the GenBank and coded as EF492879.Comparison of sequence with(S)-N-methylcoclaurine-3'-hydroxylase from C.japonica showed CYP80B3 possessed the same functional regions involved with 3'-hydroxylation of(S)-N-methylcoclaurine.Conclusion The cDNA encoding CYP80B3 from C.chinensis was cloned and reported.This work underlays the first step for exploring the pathway of benzylisoquinoline alkaloid biosynthesis and for improving the content of benzylisoquinoline alkaloid in C.chinensis.
5.Optimal design of portable ambulatory blood pressure monitor based on STM32L.

Based on the 32-bit ultra low power microcontroller STM32L151RBT6 using ARM Cortex-M3 kernel, the portable ambulatory blood pressure monitor powered by two AA batteries was designed. In order to insure the stability of power supply and prevent overpressure of cuff, super capacitor technology and new kind of safety logic circuits were used. The experimental result shows that: this solution is accurate and stable, which has high safety coefficient and a great clinical application value.

6.Identification of novel B(A)803 C→G allele mutation in Chinese population and induction of severe hemolytic disease of the newborn

Objective To investigate serological blood typing of the ABO locus which contradict to general law of inheritance in parentage,and the underlying reasons for severe hemolytic disease of newborn(HDN).Methods To research the family whose newborn is AB phenotypes,mother is O phenotypes and father is AB phenotypes.The familiy were genotyped by parentage tests, serological tests,PCR-SSP and direct DNA sequencing at exons 6 and 7 of ABO gene.At the salne time,HDN was detected by micro column gel Coombs (MGCT), and the primary fingerposts of the routine blood tests. Biochemical tests were dynamically observed.Results The results of parentage tests showed that three-generation pedigree have parent-child relationship. The red blood cell(RBC)of this AB phenotypes of this family members strongly agglutinated(4+)with diverse monoelonal anti-A and anti-B antibodies,and their serum did not contain anti-A and anti-B antibodies in blood anti-typing.PCR-SSP can not detect their A and B gene,but DNA sequencing at exons6 and 7 of ABO gene revealed that it had the B(A)803C→G mutation.Conclusions The genetm basis of this parentage are B(A)803G blood gene which harbored both A and B difunctionality of glyeosyhransferases.This was the first report that severe HDN resulting from a large number of A and B antigens in RBC of B(A)phenotype of a newborn,which has clinical significance on ABO locus.

BACKGROUND:The effect of dimethyl sulfoxide cryoprotectants has been got a lot of verification in the low-temperature medical applications. But there is no literature addressing microdialysis detection of dimethyl sulfoxide cryoprotectants. OBJECTIVE:To investigate the microdialysis relative recovery of different concentrations of dimethyl sulfoxide cryoprotectants used for limb reattachment. METHODS:In vitro linear probe relative recovery of different concentrations of dimethyl sulfoxide (2%, 5%, 8%) was detected by retrodialysis gain and loss method. The correction in vivo experiment was done to estimate dimethyl sulfoxide relative recovery in rabbit amputated limbs. RESULTS AND CONCLUSION:The relative recoveries of different concentrations of dimethyl sulfoxide (2%, 5%, 8%) were (49.49±3.56)%, (46.30±1.48)%, (52.66±2.54)%using retrodialysis gain method and (50.99±6.89)%, (43.86±1.35)%, (50.67±0.75)%using retrodialysis loss method. The average recoveries were (49.48±3.18)%and (48.51±4.03)%, respectively. There was no difference in the relative recovery of dimethyl sulfoxide detected using two methods. The change of dimethyl sulfoxide concentration could not influence the retrodialysis gain and loss method calibration results. The recovery was (15.45±4.8)%in vivo. These findings indicate that the microdialysis technology is suitable for dimethyl sulfoxide sampling in vivo that has no obvious influence on the relative recovery.

Objective: To explore the mechanism of congestive heart failure (CHF) by observing the effects of Tingli Shengmai Decoction on myocardial fibrosis and expression of TGF-?1 of rats with CHF. Methods: The CHF animal models were duplicated by the abdomen arteriarctia method, and 60 male wistar rats were randomly divided into the sham-operation group, the model group, the high-dose of Tingli Shengmai Decoction group, the low-dose of Tingli Shengmai Decoction group and the positive medicine (Xinbaowan) control group. After 4 weeks common fed, every group rats were given a certain dose of distilled water or medicine. OBJECTIVE:To optimize the preparation technology for enteric-coated changbiqing granules.METHODS:The concentration of acrylic resin and the four factors like the quantity of castor oil,pulvis talci and methylsilicone oil on the yield of changbiqing enteric-coated granules were determined by orthogonal test design;The effects of the enteric-coated granule plasticizer on the quantity of castor oil and the weight increment of the coating were determined by the release rate test.RESULTS:The optimized formula of the coating solution was described as follows:the concentration of acrylic resin was5%,the relative amount of castor oil to acrylic resin was35%,the dosages of pulvis talci and methylsilicone oil were3%and1%,respectively,the weight increment of the coating was30%.CONCLUSION:Changbiqing enteric-coated granules pre?pared on the optimized technology are in line with the quality requirements and which have ideal enteric soluble drug releasing characteristics.
