Based on the AuNPs/Nafion composite membrane technology and immobilization of amino adenosine aptamer using carboxyl carbon nanotubes on the surface of a glassy carbon electrode, a electrochemiluminescence sensor was preparated.The sensor was characterized by cyclic voltammetry and electrochemical luminescence.The result showed that the sensor had a good stability and reproducibility.Adenosine and adenosine aptamer could form G-tetrahedral structure, leading a decrease of ECL intensity.Under the optimum experimental conditions, the relative ECL intensity showed a good linear relationship to the negative logarithm of adenosine concentration in the range of 1.0×10-11-1.0×10-7 mol/L, the linear equations was ΔIECL=-890lgC-5050 with a detection limit of 5.0×10-12 mol/L.The RSD was 2.7% in 11 times measurement of adenosine (1.0×10-10 mol/L).The recovery was 97.1%-110.0% in the determination of real adenosine sample.
2.Expression and clinical significance of PIM-1 in prostate neoplasm

Objective To study the expression of PIM-1 in prostate cancer (PCa) and its clinical significance. Methods Reverse transcription polymerase chain reaction ( RT-PCR) analysis was used to determine the expression level of PIM-1 mRNA in 2 cases of benign prostatic hyperplasia ( BPH) samples and 5 cases of PCa samples, and immunohistochemical analysis was used to investigate PIM-1 protein expression in 20 cases of BPH, 20 cases of high grade-prostatic intraepithelial neoplasia ( HGPIN) and 42 cases of PCa tissues. The immunohistochemical staining intensity was scored as negative, weak, moderate and strong positive. Results The expression level of PIM-1 mRNA in 5 cases of PCa was 0. 63 , 0. 55 , 0. 42, 0. 91 and 0. 76 ; the level in 2 cases of BPH was 0. 26 and 0. 27 , respectively. The negative rates of expression of PIM-1 protein in BPH, HGPIN and PCa tissues were 60% ( 12/20) , 20% (4/20) and 2% (1/42) ,the weak positive rates of the expression were 40% (8/12) , 20% (4/20) and 12% (5/42) , while the moderate to strong positive rates of the expression of PIM-1 protein was 0 (0/20) , 60% ( 12/20) and 86% (36/42) , respectively. Chitosan-Ru ( bpy ) 2+3 -SiO2 composite nanoparticles ( CRuS NPs ) were prepared by reverse microemulsion method, and based on the Nafion/MCNT composite membrane technology, CRuS NPs were effectively and steadily immobilize on the surface of a glassy carbon electrode to prepare the electrochemiluminescence sensor for uric acid determination. In 0. 1 mol/L PBS (pH 7. 4) buffer solution, when the actuation duration between uric acid and the modified electrode was 15 min, the electrochemiluminescence showed a good linear relationship to the negative logarithm of uric acid concentration in the range of 1. 0 × 10-10-1. 0 × 10-5 mol/L, the linear equation was IECL=-709. 52-202. 74lgC and the correlation coefficient was 0. 9936 with a detection limit of 6. 0 × 10-12 mol/L. The ECL sensor exhibited excellent repeatability and stability, and the RSD for 11 times determination of 1. 0 × 10-8 mol/L uric acid was 2. 9%. The recovery was 98. 5%-103. 5% in the determination of real Uric acid sample.
4.Observation on the effect of continuous infusion of dopamine and furosemide in treatment of 76 elderly patients with refractory heart failure

Objective To explore the clinical effect of continuous infusion of dopamine and furosemide on elderly patients with refractory heart failure.Methods 76 elderly patients with heart failure were selected as the observation subjects,they were randomly divided into observation group and control group,38 cases in each group.The control group was given oxygen inhalation,the expansion of vascular,cardiac and other conventional treatment.The observation group was given conventional treatment and continuous infusion of dopamine and furosemide.Clinical efficacy and cardiac function improvement were compared between the two groups.Restlts The effective rate of observation group was 92.1%,which was higher than that of the control group (73.7%) (x2 =10.29,P < 0.05).After treatment,the improvement of cardiac function between the two groups had significant difference (t =15.94,10.18,all P <0.05).Conclusion Continuous infusion of dopamine and furosemide in treatment of elderly patients with refractory heart failure could improve heart function,enhance curative effect.
5.Changes of lung AT1R and Mas receptor protein expression in lung tissue of mice with lung injury after limb ischemia-reperfusion

AIM: To explore the role of imbalance of local renin-angiotensin system (RAS) in lung injury by observing the changes of angiotensin Ⅱ type 1 receptor (AT1R) and Mas receptor protein expression in the lung and the degree of lung injury subject to limb ischemia-reperfusion (LIR) in the mice.METHODS: Male ICR mice (n=42, 8 weeks old) were randomly assigned into 7 groups (6 in each group), including control group and 6 model groups with LIR of 0.5 h, 1 h, 2 h, 4 h, 6 h and 12 h reperfusion.Tourniquets were used to block the blood flow of the hind limbs of the ICR mice and were released after 2 h ischemia to initiate reperfusion.The mice were sacrificed by eyeball blood withdrawal at different time points after reperfusion.The organ coefficient and wet/dry weight ratio (W/D) of the lung tissue were calculated.Bronchoalveolar lavage fluid (BALF) was taken for cell counting and protein concentration measurement.The histopathological changes of the lung tissues was observed, and the pathological score was calculated.The protein expression of AT1R and Mas receptor in the lung tissues was determined by Western blot.RESULTS: The organ coefficient, W/D of lung tissue, and cell number and protein concentration in BALF of model groups were significantly higher than those in control group after LIR.The pathological changes were found in the lung tissue of model mice, including alveolar capillary dilation and congestion, edema, inflammatory cell infiltration in peripheral vascular, alveolar and bronchial walls, alveolar septal thickening and inflammatory cell infiltration.The lung injury score was elevated gradually along with the extension of reperfusion time.The protein expression of AT1R began to increase at reperfusion time points of 0.5 h and 1 h.With the extension of reperfusion time, the protein expression of AT1R decreased gradually.Conversely, the protein expression of Mas receptor increased gradually with prolonged reperfusion.CONCLUSION: LIR induces acute lung injury gradually.The imbalance of AT1R and Mas receptor expression may be involved in the damage process.
6.Bone mineral density-related factors of adult health checkup receivers

Objective To investigate bone mineral density-related factors to decrease the prevalence of osteoporosis. Methods This croas-sectional survey enrolled 46 219 adults less than 60 years old. Anthropometry, blood pressure, serum lipid, glucose, electrolytes, uric acid and homocysteine were detected. Bone mineral density (BMD) of distal forearm was measured by using peripheral dual energy-ray detector (MestriscanTM). Our data were analyzed by Pearson's correlation analysis and stepwise multiple regression analysis. Results All BMD-related factors except age showed significant difference between male and female. BMD of female subjects was lower than that of male participants (0.50±0. 15 vs 0. 54 ±0. 15,t = 22. 38 ,P < 0. 05 ). The prevalence of osteoporosis in female was higher ( 29. 51% vs 26. 48%, χ2 =47.90,P <0. 05). BMD increased with age before 40-year old, and then decreased more rapidly in female after 50-year old. Conclusion BMD of male is higher. Cigarette smoking, waist-hip ratio, systolic blood pressure, serum sodium, total cholesterol, and ALP were negatively correlated with BMD.
7.The combination of flow cytometry and automated cell image analyzer in determining the nature of ascites and pleural effusion nature

Objective To explore the clinical value of flow cytometry( FCM) and DNA automated cell image analyzer ( AICM) in determine the character of ascites and pleural effusion.Methods This was a cross-sectional study.203 ascites and pleural effusionsamples were random selected from PLA hospital inpatients between August 2013 to June 2014 .The DNA content of sediment cells were detectedthrough the FCM and AICM respectively benign and malignant disease were differentiated according the counts and proportion of aneuploid cells.The sensitivity, specificitywere calculated byROC curves.Results The sensitivity, specificity and accuracy of flow cytometry cell in detectingtumor cells were 78.6%,80.0% and 79.2%%, while the sensitivity, specificity and accuracy of image analyzer were 83.5%,78.6% and 81. 3%respectively.When FCM and AICMwere combined ,the sensitivity, specificity and accuracyincreased to 92.2%, 86.3% and 89.6%.Conclusions Compared toconventional cytology test, the sensitivity and specificity were significantly high when the two methods were combined .Therefore, the combination method can be used to assist in clinical identification of the nature of ascites and pleural effusion and to help the diagnosis of disease.
8.Neurologic function and expression of angiopoietin-1 and its receptor at recovery stage of cerebral hemorrhage after neural stem cell transplantation in rats

BACKGROUND:Recent studies have found that bone marrow mesenchymal stem cels that culturedin vitro for a long time can naturaly differentiate into neural stem cels, which then differentiate into neurons and glial cels, thereby providing a new therapeutic thinking for Parkinson’s disease, sequela of cerebral infarction, cerebelar atrophy and brain dysplasia.
OBJECTIVE:To discuss the influence of neural stem cel transplantation on neurologic function of rats with cerebral hemorrhage at recovery stage and the relevant mechanism of action.
METHODS: Sixty male Sprague-Dawley rats were randomly divided into normal group (n=18), cerebral hemorrhage group (n=21) and transplantation group (n=21). Cerebral hemorrhage models were established in the latter two groups using VII type colagen enzyme induction method. At 21 days of modeling, rats in the transplantation group were injected neural stem cels via the tail vein, and those in the other two groups received the same volume of normal saline. At 7, 14, 21 days after cel transplantation, modified adhesive removal test (MST) was employed to evaluate the neurologic function of rats, and then the rats were kiled. RT-PCR was used to detect angiopoietin-1 mRNA expression in the bleeding tissues, and western blot assay was employed to measure tyrosine kinase receptor-2 protein expression.
RESULTS AND CONCLUSION:Compared with the normal group, the MST scores in the cerebral hemorrhage group and transplantation group were significantly decreased (P< 0.05). From the 7th day after transplantation, MST scores in the transplantation group were significantly higher than those in the cerebral hemorrhage group (P < 0.05). At 7, 14, 21 days after transplantation, expressions of angiopoietin-1 mRNA and tyrosine kinase receptor-2 protein were ranked as folows: transplantation group > cerebral hemorrhage group > normal group, and there was a significant difference among the three groups (P< 0.05). These findings indicate that neural stem cel transplantation can effectively promote the neurologic recovery of rats with cerebral hemorrhage at recovery stage, and the concrete mechanism may be related to the increase of angiopoietin-1 mRNA and tyrosine kinase receptor-2 protein in the bleeding tissues.
9.Analysis of coronary artery Z-scores of children with Kawasaki disease on echocardiography

Objective To investigate the clinical value of coronary artery Z-scores on echocardiography in diagnosing coronary artery abnormalities. Methods The echocardiography results of 612 patients with Kawasaki disease (KD) at the acute and recovery phase were retrospectively studied. Coronary artery luminal diameters were converted to body-surface-area-adjusted Z-scores. According to coronary Z-scores classiifcation, all the subjects were divided to four groups:415 cases with no dilation (ND), 133 cases with small coronary artery abnormalities (SCAAs), 47 cases with large coronary artery abnormalities (LCAAs), and 17 cases with giant coronary artery abnormalities (GCAAs). Clinical features (gender, age, typical clinical manifestations, fever duration) and laboratory results (CRP, ESR, WBC, PLT) were compared among all the four groups. Coronary artery diameters and the Z-scores were compared between acute and convalescence phase. Results Along with the increase of coronary Z-score, fever duration was prolonged [ND group:(7.75±3.12) d, SCAAs group (8.50±4.12) d, LCAAs group: (8.57±3.58) d, GCAAs group: (11.88±4.33) d, F=22.375, P<0.05]. With coronary Z-score increasing, PLT also increased (F=22.029, P=0.000), and the highest PLT was observed in GCAAs group. There were no significant differences in the CRP, ESR and WBC among all the four groups (F=0.236, 1.116, 0.121, all P>0.05). No significant different coronary diameters were found in ND cases between recovery and acute phase [(2.24±0.34) mm vs (2.33±0.36) mm, t=1.926, P > 0.05]. But there were significant difference in the coronary Z-scores of ND patients between recovery and acute phase (0.41±0.82 vs 1.17±0.75, t=8.332, P < 0.05). The coronary Z-scores in SCAAs group (1.32±0.89 vs 3.40±0.62, t=11.073, P < 0.05), LCAAs group (3.12±2.27 vs 6.20±1.28, t=4.579, P<0.05) and GCAAs group (11.88±6.77 vs 20.4±9.70, t=3.480, P<0.05) at recovery phase were smaller than values at acute phase. Conclusions The KD coronary Z-scores are the body-surface-area-adjusted standard value, and not subject to the influence of children growth and development. Therefore, it may accurately evaluate the severity of coronary artery abnormalities and its recovery process. Accurate quantitative of the coronary artery luminal dimensions is important in KD clinical management and prognosis prediction.
10.Application of flexible t medical horacoscopy in the diagnosis of pleural effusion

Objective To evaluate the application of flexible thoracoscopy in the diagnosis of pleural effusion.Methods Thoracoscopy was performed in 20 pleural effusion patients in our hospital from May to December 2007.Biopsies were performed in 16 patients,not in the other 4 patients since difinite diagnosis had been reached before thoracoscopy.Results Rate of accurate diagnosis via thoracoscopy was 93.75%(15/16).Results Of biopsy were as follows:adenocarcinoma 8 cases,squamous carcinoma 1 case,adenosquamous carcinoma 1 case,malignant pleural effusion 1 case,tuberculous pleuritis 4 cases,malignant mesothelioma 3 cases,chronic suppurative pleuritis 1 case,failed diagnosis 1 case.Presentations of lesions under thoracoscope were as follows:diffused miliary nodules 10 cases(10/20),multiple mass 7 cases(7/20),fibrous compartmentation or conglutination 9 cases(9/20).There were no severe complications.Conclusion Flexible medical thoracoscopy is a safe and efficient method of etiologic diagnosis of pleural effusion.
