1.Clinic efficacy of Cypher~(TM) stent and bare stent in elderly patients with acute myocardial infarction

Objective To investigate the effects of rapamycin eluting stent(Cypher~(TM)) and metal bare stent on the prognosis and C-reactive protein(CRP) in elderly patients with acute myocardial infarction(AMI).(Methods The) immediate angiographic and clinic follow-up outcomes of 45 elderly patients with AMI reciving Cypher~(TM) and bare stent were retrospectively analyzed.CRP levels in Cypher~(TM) and bare stent groups were determined before implantation,6 h,1 d,7 d and 30 d after implantation.Clinical follow-up and major coronary events analysis were developed after operation.Results 27 rapamycin eluting stents were implanted in patients of Cypher~(TM) group and(26 bare) stents were implanted in patients of bare stent group.The Cypher~(TM) stents were successfully implanted with 100%.In patients of Cypher~(TM) group,no serious complications were found in the term of operation and in hospital.There was no restenosis,revascularization and events.In patients of bare stent group,except for(1 patients) with sudden death,angina pectoris occurred in 2 patients,repeat angiography showed that there were new lesions in other coronary arteries of these patients.Restenosis attacked in 2 patients.The concentrations of CRP in Cypher~(TM) and bare stent groups 6 h and 7 d after operation were much higher than that of(pre-operation (P

Aim To investigate the reversal of resistance to doxorubicin by CJZ3,a lomerizine derivative,in doxorubicin-resistant human myelogenous leukemia(K562/DOX)cells.Methods The effect of CJZ3 on P-glycoprotein(P-gp)function and resistance to doxorubicin in K562/DOX cells were observed by using flow cytometry and MTT assay.Results CJZ3 increased intracellular uptake and accumulation of rhodamine123(Rh123)in K562/DOX cells in a concentration-related manner,significantly inhibited the efflux of Rh123 from the cells,and increased the doxorubicin-induced cytotoxicity,apoptosis rate,Caspase-3 activity in doxorubicin-treated cells and the intracellular accumulation of doxorubicin.Conclusions The reversed effect of CJZ3 on resistance to doxorubicin in K562/DOX cells was associated with its inhibitory effect on P-gp function in vitro.
3.Albumin overload induces ?-smooth muscle actin expression in proximal tubular epithelial cells

AIM:To study whether albumin overload in proximal tubular epithelial cells(PTCs)induces epithelial to myofibroblast transdifferentiation(EMT).METHODS:Rat renal proximal tubular cell line NRK52E was cultured on 6 well plates.When the cells reached 70% confluens or complete confluens,cells were serum starved for 24 h.Different concentrations of delipidated bovine serum albumin(dBSA),ranging from 0-30 g/L,were then added to the cells.The media was changed every 48 h until the end of 144 h.Cell shapes were monitored by light microscope during experiment.Cell structures were detected by electron microscopy.Epithelial cell markers:E-cadherin,?-catenin,and myofibroblast marker:?-smooth muscle actin(?-SMA)were detected by immunofluorescent microscopy and Western blotting.RESULTS:dBSA overload induced the expression of ?-SMA in sub-confluent NRK52E,and a few cells elongated,but the induced expression of ?-SMA was not in a dose dependent manner.dBSA overload did not induce the expression of ?-SMA in complete confluent NRK52E,cell shape did not change either.dBSA overload did not inhibit expression of E-cadherin or ?-catenin both in sub-confluent and complete confluent NRK52E.The electron microscope showed that these cells retained epithelial phenotype,with microvilli and tight junction.CONCLUSION:Albumin overload induces PTCs expressing myofibroblast marker ?-SMA and promotes EMT.However,complete EMT does not achieve.Complete confluens(cell-cell contacts)inhibits albumin induced ?-SMA expression in PTCs.
4.Determination of anthraquinone aglycones of Radix et Rhizome Rhei in Ganning Granule by HPLC

AIM:To determine Radix et Rhizoma Rhici in Ganning Granula(Radix et Rhizoma Rhei,Rhizoma Chuanxiong). METHODS: In order to determine five anthraquinone aglycones of Radix et Rhizoma Rhei,the HPLC system consisted of Nucleosil ODS column,methand-0.1% phosphoric acid(85∶15) mixture as mobile phase,with detection wavelength at 430 nm. RESULTS: HPLC showed that the linearity ranges were as follows: aloe-emodin concentration was at the range of 0.041 6-0.291 2 ?g,the rhein concentration was at the range of 0.037 1-0.259 8 ?g,the emodin concentration was at the range of 0.045 8-0.320 3 ?g,the chrysophanol concentration was at the range of 0.049 60.347 2 ?g,the physcion concentration was at the range of 0.020 6-0.144 5 ?g.The average recoveries of five anthraquinone aglycones were between 96.6%-98.4%.CONCLUSION: The method is simple,accurate,sensible and with good repeatability and can be used for quality control of Ganning Granule.
5.Effects of Different Dilutions and Fractions of TF-P on Normal Human Lymphocyte Transformation

Porcine transfer factor(TF-P) was prepared from porcine spleen by means of hom-ogenization and vaccum dialysis. Fractionized by Sephadex G-10 gel chromatography,TF-P could be separated into seven fractions but five fractions were obtained from human transfer factor(TF-h) .These fractions of TF-P and TF-h and different dilutionsof TF-P were tested for biological activity in vitro. By the use of ~3H-TdR uptake assay we found that, both of whole TF-P and TF-hpossessed suppresivs activity to PHA-induced human periperal blood lymphocyte tran-sformation. Whereas, Fraction Ⅲ of TF-P and Fraction Ⅲ and Ⅳ of TF-h containedcomponents responsible for augmentation of PHA-induced responses. When TF-P was diluted 10~(-2) to 10~(-5) and cultured with normal human peripheralblood lymphocytes, we found that the original suppression effects of whole TF-P toPHA-induced lymphocyte transformation were converted to normal. The optimum dil-ution of TF-P was 10~(-3), which were shown to augment responses. It shoud be notedthat the responsiveness of normal human lymphocytes to TF-P was dose dependent.
6.The isolation of Corynebacterium tuberculostearicum from prostatic fluid

Objective To carry out a taxonomic identification of a strain of claviform bacteria iso-lated from prostatic fluid of a patient who suffered from chronic prostatitis, and to approach its phylogenic and biologic position. Methods We undertaked an initial identification by phenotypic characters such as morphologecal, physiological and biochemical characteristics to ascertain its phylogeny by chemical composi-tion analysis of cell wall and 16S rRNA gene sequencing and alignment. Results A club-shaped gram posi-tive rod bacillus was isolated in pure culture state. Its biochemical reactions were not active. The diamino-acid of cell wall was meso-diaminopimelic acid (meso-DAP) and it had wall chemotype Ⅳ ( contained arabi-nose, galactose and maltose ). Sequence searches of the GenBank database revealed that this strain had a highest level of 16S rDNA sequence similarity (99.4%) to C. tuberculostearicurn strain ATCC35692 with only 8 nucleotides difference. Conclusion On the basis of phenotypic and phylngenetie analysis, it is rea-sonable to assign this strain to the species C. tuberculostearicum, and this is the first isolation of C. tubercu-lostearicum from prostatic fluid home and abroad.
7.The experience of women with heroin dependence in compulsory detoxification before the society-return: A qualitative study

Objective To understand the experience of women with heroin dependence in compulsory detoxification before the society-return.Methods A semi-structured interview was conducted with 15 women with heroin dependence in compulsory detoxification.Data were analyzed based on Colaizzi phenomenological research method.Results The feelings of women with heroin dependence in compulsory detoxification were classified into 3 themes,including negative self-concept,loneliness and helplessness,uncertainty about the future.Conclusions Nurses should pay attention to problems about women with heroin dependence in compulsory detoxification and provide effective measures to improve their physical and psychological health and help them to return to the society.
8.Relationship between serum 25-hydroxyvitamin D level and post-stroke depression in the elderly with acute stroke

Objective To investigate the correlation between serum 25 hydroxyvitamin D[25 (OH) D] level and post-stroke depression in elderly patients with acute stroke.Methods Depression was assessed by Beck Depression Inventory (BDI),Burns Depression Checklist (BDC),and the Diagnostic and Statistical Manual of Mental Disorders Ⅳ in 231 elderly inpatients at 2-4 weeks after onset of acute cerebral infarction.According to the depression assessment,the patients were classified into four groups:normal group),depressive symptoms group,with mild depression group,and severe depression group.The cut-off point of 25 (OH) D level for post stroke depression was analyzed by receiver operating characteristic (ROC).Results The serum level of 25(OH)D was (23.0±1.6) nmol/L in all subjects,(25.0±1.7) nmol/L in normal group,(23.7± 1.6) nmol/L in depressive symptoms group,(22.4±1.5) nmol/L in mild depression group,(16.3± 1.5) nmol/L in severe depression group.Serum 25(OH) D level had significant differences between major depression groups and other three groups,while no significant differences were found between mild depression group,depressive symptoms group and normal group.Pearson correlation analysis showed that scores of BDI and BDC were inversely related to 25 (OH)D level (r=0.24 and-0.25,both P<0.01).ROC curve analysis showed that the cut off point of serum 25 (OH) D level for severe post-stroke depression was 21.6 nmol/L.Conclusions Serum 25 (OH)D level is inversely related to depression.Serum 25(OH) D level is reduced in patients with severe post stroke depression.
9.The status and related factors of self-efficacy of convalescent patients with heroin dependence in compulsory detoxification

Objective To investigate the status and influencing factors of convalescent patients with heroin dependence in compulsory detoxification.Methods A total of 90 convalescent patients with heroin dependence in compulsory detoxification were recruited and investigated with a self-designed scale,General Self-efficacy Scale (GSES),Social Support Assessment Scale (SSRS) and Self-rating Depression Scale (SDS).Results The mean score of the self-efficacy was (20.97±2.26) points.Multiple regression analysis showed that depression,social support and daily dose of heroin use were influencing factors of the self-efficacy,which could explain 59.1% of the variance.Conclusions Improving the self-efficacy of convalescent patients with heroin dependence in compulsory detoxification can enhance physical and psychological health and decrease the relapse.
10.Effect of Cyclosporine A on Activity and Expression of MMP-2 and MMP-9 in Renal Tubular Epithelial Cells

[Objective] To observe the effect of cyclosporine A (CSA) on the activity and expression of MMP-2 and MMP-9 in renal tubular epithelial cells. [Methods] NRK52E cells were cultured until its reached confluent. Then NRK52E cells were exposed to different concentration of CSA (0, 0.42, 0.84, 4.2, and 8.4 μmoL/L) for 48 h or 72 h respectively. MMP-2 and MMP-9 activities were detected by gelatin zymography. The expression of MMP-2 and MMP-9 mRNA were detected by RT-PCR. [Results] MMP-2 activity and mRNA levels were decreased in a dose dependent manner after exposed to different concentration of CSA for 48 h or 72 h in NRK52E cells. Compared with control (CSA 0 μmoL/L), CSA 0.42, 0.84, 4.2, 8.4 μmoL/L decreased the MMP-2 activities to 27%, 24%, 11%, and 9% respectively; The differences are significant, P<0.05. But the MMP-9 activity and mRNA levels were increased after exposed to CSA for 48 h or 72 h in NRK52E cells. Compared with control group, CSA 4.2 μmoL/L exposure increased MMP-9 activity to 438% in 48 h, and 237% in 72 h; the differences are significant as well, P<0.05. [Conclusion] A dose-dependent decrease in the expression and activity of MMP-2, and the up-regulation of the expression and activity of MMP-9 by CSA in renal epithelial cells may related to CSA associated tubulointerstitial damage.
