OBJECTIVE:To study the effects of arsenic trioxide(As 2 O 3 )on the encoding protein expressions of nm23and P53in gastric cancer cells and to study the anti-canecer mechanism of which in the tumor.METHODS:A control group and a test group were set up.The expressions of gene coding proteinum nm23and P53of the gastric cancer cells cultured in vitro by the action of As 2 O 3 were determined by immunohistochemistry method.RESULTS:The expression levels of nm23and P53in gastric cancer cells have been lowered compared with the control group.CONCLUSION:As 2 O 3 can exert its anti-tumor effect by decreasing the expression of gene coding proteinum of nm23and P53.
4.Effects of the dominant-negative I?B? plasmid on the expression of NF-?B and cyclooxygenase-2 in pancreatic carcinoma

AIM: To investigate the effects of the dominant-negative I?B? plasmid on the expression of NF-?B and cyclooxygenase-2 (COX-2) after its being transfected into pancreatic carcinoma PC-3 cell line. METHODS: The expression of NF-?B and COX-2 in PC-3 cell line was confirmed by immunohistochemistry. The effects of dominant-negative I?B? plasmid transfection on the expression of NF-?B and COX-2 were studied by reverse transcription polymerase chain reaction (RT-PCR) and Western blotting. RESULTS: Both NF-?B and COX-2 were expressed in pancreatic carcinoma PC-3 cell line, and the expression of NF-?B and COX-2 were down-regulated in a certain time-independent manner after dominant-negative I?B? plasmid transfection. CONCLUSIONS: The NF-?B and COX-2 are expressed in pancreatic carcinoma PC-3 cell lines. The expression of NF-?B and COX-2 are inhibited by dominant-negative I?B? plasmid, while NF-?B is likely to play an important role in regulating the expression of COX-2.

6.The Effects ofβ-Sheet Breaker Peptide H102 on ERK Signal Transduction Pathway in Brain of PAP Double Transgenic Mice

Objective To investigate the activation of β-sheet breaker peptide H102 on ERK signal transduction pathway in brain of PAP double transgenic mice. Methods PAP double transgenic mice were randomly divided into model group and H102 treatment group (n=10 for each group). A group of C57BL/6J mice with the same genetic background was served as controls. H102 (5.8 mg/kg) 5 μL was infused by intranasal administration to mice in H102 treatment group, and equal volume of blank solution of H102 (chitosan, BSA) was given to mice in control group and model group. The ability of spatial reference memory was tested by Morris water maze after 30 days of treatment. Then immunohistochemistry tests and Western blot technique were used to detect the content of RAS, P-MEK and P-ERK proteins in mouse brain. Results (1) The ability of learning and memory was significantly lower in model group than that of control group. The ability of learning and memory was significantly improved in treatment group than that in model group (P<0.05). (2) The contents of RAS, P-MEK and P-ERK in mouse brain were significantly lower in model group than those of control group, and these protein ex-pressions were significantly increased in treatment group than those in model group (P<0.01). Conclusion β-sheet break-er peptide H102 can activate ERK signal transduction pathway in brain of PAP double transgenic mice, increase PAS, P-MEK and P-ERK levels in nerve cells, and improve the ability of learning and memory in PAP mice.
7.Comparison of the effects between the modified and classic coracoid approach in infraclavicular brachial plexus nerve block

Objective To investigate the effects between the modified and classic coracoid approach in infraclavicular brachial plexus nerve block.Methods Sixty patients scheduled for elective surgical procedures under infraclavicular brachial plexus block were divided into observation group and control group by table of random digit method with 30 cases each.The puncture point of control group was approach via the point 2 cm medial and caudal to the coracoid process.The puncture point of observation group was modified by surface projection of the brachial plexus.Peripheral nerve stimulator was used to confirm the proper localization of the plexus.The performance time,number of puncture,anesthesia success rate,incidence of complications and 24 h patient satisfaction were recorded.Results The performance time in observation group was less than that in control group [(3.2 ± 1.1) min vs.(4.3 ± 1.4) min](P< 0.05).The chance of locating the brachial plexus in a single puncture without adjusting the block needle in observation group was significantly higher than that in control group [73.3% (22/30) vs.50.0% (15/30)] (P < 0.05).There were no statistical differences in anesthesia success rate,incidence of complications and 24 h patient satisfaction between two groups (P > 0.05).Conclusions The modified coracoid approach can significantly improve the probability of locating the nerve in one puncture that reduces the performance time.It can improve the accuracy of puncture point position on the body surface.

10.Phenylephrine-induced excessive hypertrophy responses in cardiomyocytes from endothelial nitric oxide synthase gene knockout mice

AIM: The study aimed to observe the difference in hypertrophy responses in cardiomyocytes of cultured neonatal endothelial nitric oxide synthase (eNOS) gene knockout mice and wild mice by RNA quantitation detection and cell area estimation. METHODS: The experiment was performed at the Empirical Study Center of Medicine Department in Shunde Profession and Technique Institution in August 2007. ①Male or female C57BL/6 stock neonatal eNOS knockout mice and wild mice aged 1-3 days were selected. ②Cultivated Mouse cardiomyocytes were divided into experimental and control groups 72 hours later. Adrenergic receptor agonist phenylephrine was cultured with cardiomyocytes. DMEM of the same volume as the experimental group was added in the medium in the blank control group. ③DNA, RNA quantitation detection and cell area estimation methods was used to observe the responses contrast in cell proliferation and hypertrophy of cardiomyocytes in cultured neonatal eNOS gene knockout mice and wild mice.
