1.Relationship between Yangtze River floodplain micro ecological environment and distribution of Oncomelania hupensis snails

Objective To explore the relationship between the Yangtze River floodplain ecological environment(vegetation, soil,water and light intensity)and the distribution of Oncomelania hupensis snails,so as to provide the evidence for ecological snail control. Methods Three regions(the Lu-Gang Bridge,Dragon Nest Lake in the bund,and Dragon Nest lake beach)were selected to investigate the plant characteristics(species,height,coverage,frequency and strain of clusters),soil characteristics (temperature,humidity,light intensity)and pH value. All the results were analyzed statistically with SPSS 18 software. Results A total of 920 boxes were investigated. The vegetation coverage was 3.7%-63.5%,and the dominant population was Cyperusrotun-dus L. cluster on the marshland. The soil temperature was 19.0℃-24.0℃,pH 5.0-5.7,and humidity 53%-75%. There were statis-tical significants in average number of living snails and dead snails among 3 groups( P<0.05). As the light intensity was strong in summer and weak in winter,the snails were in the back surface in summer and front surface in winter respectively. The average number of living snails was the most near the water. The difference was statistical significant(P<0.01)among three ranges(0-1 m,1-3 m and 3-5 m). Conclusion The snail survival and distribution have close relations with micro ecological environment factors,such as vegetation,soil,water and light intensity.
2.Determination of Cobalt by Spectrophotometry Using a New Reagent, 1-(6-bromo-2-benzothiazolyl)-3-(5-bromo-8-quinolyl)-triazene

Objective To synthesize a new reagent, 1-(6-bromo-2-benzothiazolyl)-3-(5-bromo-8-quinolyl)-triazene (BBTBQT) and apply to the determination of cobalt. Methods The reagent had been synthesized by diazotization and coupling reaction. After purification and characterization, BBTBQT was tested for its color reaction conditions with cobalt in the presence of cetylpyridinium bromide (CPB). Results In the presence of borax buffer solution at pH value of 9.0, the reagent reacted with cobalt to form a blue stable complex with a molar ratio of 2∶1; the complex had a maximum absorption at 640 nm. The apparent molar absorptivity of the complex is 1.55 ?105 L/(mol?cm). Beer’s law was obeyed in the range of 0-0.28 mg/L for cobalt. The method had been applied to determine cobalt in drainage sediment and vitamin B12 injection solution with a mean relative error of 1.9%-5.8% and a standard relative deviation of 1.7%-3.8%. Conclusion The present method is selective, sensitive, accurate, convenient and suitable for determination of trace cobalt in the samples.
3.Study on histochemical localization of phenol oxidase in Oncomelania hupensis

Objective To investigate the histological distribution of phenol oxidase (PO) in body of Oncomelania hupensis. Methods Oncomelania hupensis snails collected from the riverside of Wuhu section of the Yangtze River were anatomized and the unimpaired software organs were sliced in frozen condition to get the continued sections, then the sections were incubated in pH 6.8 phosphate buffer containing 25 mmol/L catechol at 37 ℃ and investigated systematically for histological distribution of PO in snail bodies. Results The light microscope showed that there were black grannles of PO (histochemical reaction) in the liver, ctenidium, head-foot, tentacle and mantle collar of snail body. Conclusion PO exists in Oncomelania hupensis and locates in the liver, mantle collar, head-foot, tentacle and ctenidium.
4.To demonstrate the change in peripheral blood T lymphocyte subgroups and cytokines of Behet′s syndrome

Objective To demonstrate the change in peripheral blood T lymphocyte subgroup and cytokines of Beh?et′s disease (BD). Methods T lymphocyte subgroup was detected by flow cytometry and serum interleukin (IL)-8, sIL-2R, IL-1?, IL-2, IL-6 and TNF-? were assayed by RIA. Results CD4, CD4/CD8, NK of BD decreased evidently and CD8 incrdased. Serum IL-8, sIL-2R increased compared to normal controls. Change of IL-2, IL-6, tumor necrosing factor (TNF)-? , IL-1? showed no statistical significance compared to normal group. Conclusion Change in peripheral blood T lymphocyte subgroup and serum IL-8, sIL-2R can be one of the activity index of BD.
5.Changes in glomerular filtration barrier in rat models of acute exhaustive exercise after intervention with sodium houttuyfonate

BACKGROUND:In high-intensity exhaustive exercise process, the body must bear the exercise intensity decreasing splanchnic blood flow“ischemia”, at the same time, along with the movement of energy and material consumption, metabolite accumulation and oxidative stress in the body cause pathological damage, leading to a decline in exercise capacity. Thus, what is the impact on kidney filtration barrier? How to adapt to the change of renal tissue? Houttuynia cordata has the functions of heat clearing and detoxifying, dieresis for treating strangurtia, hemostatic, expel ing phlegm to arrest coughing and analgesia, if it has a protective effect on the renal injury caused by acute exhaustive exercise and its mechanism has not been reported in the literature. OBJECTIVE:To explore the effect of acute exhaustive exercise on kidney filtration barrier in rats and the intervention effect of Houttuynia cordata.METHODS:After resting and watching for 3 days, Sprague-Dawley rats received adaptive running for 15 minutes at a speed of 10 m/min on a 0° treadmil . A total of 24 rats, which can finish the running, were selected. They were divided into normal control group, exhaustive exercise group and dosed exhaustive exercise group according to the weight of layer (n=8). Rats in the exhaustive exercise group and dosed exhaustive exercise group on the 10° treadmil received once exhaustive exercise. Dosed exhaustive exercise group received intraperitoneal injection of sodium houttuyfonate 10 mL/kg at 30 minutes before exercises. The normal control group did not do any exercise. RESULTS AND CONCLUSION:Compared with the normal control group, serum urea nitrogen, serum creatinine, urine protein content, malondialdehyde concentration, renal cellapoptosis and apoptosis index were significantly increased, but nitric oxide content and nitric oxide synthase activity in the renal tissue were significantly deceased in the exhaustive exercise group (P<0.05 or 0.01). Glomerular filtration epithelial cells, the kidney filtration barrier of basement membrane and podocyte damage were obvious, showing abundant cellapoptosis, occasional y necrosis. Compared with the exhaustive exercise group, urine protein content, serum creatinine, malondialdehyde concentration, renal cellapoptosis and apoptosis index were significantly reduced, but nitric oxide content and nitric oxide synthase activity were significantly increased in the dosed exhaustive exercise group (P<0.05). No obvious pathological changes were detected, but apoptosis was visible. These findings confirmed that houttuynine made a reduction in renal cellinjury induced by exhaustive exercise and possibly significantly reduced apoptosis, increased nitric oxide synthase content, decreased malonaldehyde, and apparently increased superoxide dismutase activity, and final y protected injured renal tissue induced by exhaustive exercise.
6.Effect of eucommia bark on osteopontin and osteoprotegerin expressions in rat bone marrow stem cells: Is there any difference between water extract and methanol extract?

BACKGROUND: Previous studies demonstrated that eucommia bark can promote bone marrow stern cells (BMSCs) differentiated into osteoblasts, but relative mechanism is poorly understood. OBJECTIVE: To investigate the effects of eucommia bark water/methanol extracts on expressions of osteopontin (OPN) and osteoprotegedn (OPG) in rat BMSCs. METHODS: Totally 2 g eucommia bark powder were added into water or methanol to 16 mL and oscillated for 1 hour at room temperature. After soaked overnight, both extracts were centrifuged at 15 000 r/min for 10 minutes. Water extract was obtained from supernatant in water soaked powder. In methanol soaked powder, methanol extracts was obtained by concentrated supernatant in vacuo and resolved using 16 mL water. Water and methanol extracts were then filtered by 0.22 μm membrane, and conserved at -20℃. Six SD rats, aged 2 months, were selected, and the 3~(rd)passage of BMSCs were induced by water or methanol extracts with dilution of 1 × 10~(-2), 1 × 10~(-3), 1 × 10~(-4) and 1 × 10~(-5), respectively. PBS was added in the negative control group. All cells were cultured for 6 days. Expressions of OPN and OPG was measured by immunocytochemistry at 6 days with induction. The expression of OPN and OPG induced by water and methanol at 1 ×10~(-3) and 1×10~(-4) dilution was detected by RT-PCR. RESULTS AND CONCLUSION: Immunocytochemistrical results indicated that both water and methanol extracts of eucommia bark simulated OPN and OPG expression, in particular with dilution of 1×10~(-4). The methanol extracts had a stronger effect than water extract, but the expression of OPG did not change obviously. RT-PCR demonstrated that at the 3rd day of inducement, the level of OPN expression induced by water extract was higher than that of methanol extract, and no OPG expression was detected. Osteogenic differentiation of rat MSCs induced by eucommia bark water/methanol extracts relates to stimulating expression of OPN, which has no correlation to OPG. OPN expression induced by water extract is early than that of methanol extract.
7.Preparation and Characterization of Butylaminopropyl Silica Monolithic Column for Capillary Electrochromatography

A novel silica monolithic stationary phase functionalized with butylaminopropyl ligands for capillary electrochromatography(CEC) has been presented. The monolithic capillary columns were prepared by a sol-gel) process and subsequent a chemical modification. The amino groups on the surface of the stationary phase are meant to generate a substantial anodic electroosmotic flow (EOF). The butyl and propyl groups provide) hydrophobic properties. To evaluate the column performance, effects of buffer pH and organic modifier content on the EOF and electrochromatographic retention behavior of alkylbenzenes, organic acids and anilines were investigated. The monolithic stationary phase exhibited reversed phase (RP) chromatographic behavior toward neutral solutes. The model organic acid anion solutes were separated by the mixed mode mechanism, which comprised RP interaction, weak anion-exchange, and electrophoresis. Basic compounds such as anilines) were well separated on the butylaminopropyl silica monolithic column without peak tailing.
8.Statistical analysis of quality control of automatic processor

Objective To strengthen the scientific management of automatic processor and promote QC, based on analyzing QC management chart for automatic processor by statistical method, evaluating and interpreting the data and trend of the chart. Methods Speed, contrast, minimum density of step wedge of film strip were measured everyday and recorded on the QC chart. Mean ( ), standard deviation ( s ) and range(R) were calculated. The data and the working trend were evaluated and interpreted for management decisions. Results Using relative frequency distribution curve constructed by measured data, we can judge whether it is a symmetric bell shaped curve or not. If not, it indicates a few extremes overstepping control limits possibly are pulling the curve to the left or right. If it is a normal distribution, standard deviation( s ) is observed. When ?2s lies in upper and lower control limits of relative performance indexes, it indicates the processor works in stable status in this period. Conclusion Guided by statistical method, QC work becomes more scientific and quantified. We can deepen understanding and application of the trend chart, and improve the quality management to a new step.
9.Influence of acrylamide on adult neurogenesis and expression of GSK3βin mouse

Objective:To investigate the influence of acrylamide ( ACR) on adult neurogenesis and expression of GSK 3βin mouse.Methods:Method Adult male Kunming mice were used and divided into two groups:control group and experimental poisoning groups,that were exposured to acrylamide by intraperitoneal injection.Brdu labeling and immunohistochemistry were used to investigate the proliferation of adult neural stem cells in the subgranular zone ( SGZ).BrdU/NeuN/GFAP triple labeling to investigate the survival and differentiation of newly generated cells.Detecting GSK3βexpression and distribution in Neuro-2a cells,the expression of GSK3βwas examined by using Western blot.Results:Compared with control mice ,lower number of BrdU-positive cells and less differentiated into neurons in ACR mice.Less neural stem cells survived ,but more glia cells were generated in the subgranular zone of acrylamide mice.Moreover,higher phosphorylated GSK 3β( Ser9 ) were detected in Neuro-2a cells and mouse dentate gyrus in ACR mice respectively .Conclusion:These results suggested that acrylamide inhibits neural stem cells proliferation and influences the survival and differentiation of newly generated cells.Acrylamide inhibits neurogenesis maybe through GSK 3βsignaling pathway.
10.Correlation of P-gp,bcl-2,STAT3 and the resistance of lymphoma cells to chemotherapeutic agents

Objective To investigate the correlation of P-gP,bel-2,STAT3,IL-6,IL-10 and the resistance lymphoma cells to chemotherapeutic agents.Methods There were 28 patients,among them were divided into three groups:refractory lymphoma,lymphoma which is sensitive to agents and lymphodenitis.P-glycoprotein(P-gp)on lymphoma cell membrane,STAT3,bcl-2,IL-6 and IL-10 level into lymphonla cell were detected using FCM,and the correlation between them and chemotherapy efficacy were analysed. Results In refractory lymphoma patients,P-gp and hcl-2 are significant higher than that of the group which is sensitive to agents(P:0.01,P=0.039),but STAT3,IL-6 and IL-10 were not significant different between these two groups(P>o.05).P-gp is significant higher in lymphoma than in lymphodenitis(P=0.01).STAT3 in lymphoma is significant lower than that of in lymphodenitis (P=0.04).The level bcl-2,IL-6 and IL-10 between lymphoma and lymphodenitis are not significant different(P>0.05).Conclusion The expression level of bel-2 and p-gP is correlated to resistance to the chemotherapeutic agents in lymphoma cells.STAT3 play a role in lymphoma cell signal transduction,but it is not certain in lymphoma cell muhidrug resistance.
