1.Attenuated Salmonella as tumor vaccine vectors

Attenuated Salmonella can be used not only as live vaccines but also as vectors for exogenous genes. As vectors, attenuated bacteria can deliver genes directly to tumor tissues, leading to the expression of the cloned genes and induction of specific anti-tumor immunologic responses to inhibit tumor growth. Using attenuated Salmonella as a vector offers a new approach for vaccine development.
2.Study on clinical features of hepatitis G virus infection and HGV invasion to peripheral blood mononuclear cells

Objective To explore the clinical features of hepatitis G virus (HGV) infection and HGV replication sites in vivo. Methods A sensitive and specific reverse transcription-nested polymerase chain reaction (RT-nested PCR) was set up to detect the plus-stranded and minus-stranded HGV RNA in sera of 129 high risk population as well as in peripheral blood mononuclear cells (PBMCs) from 59 cases of them. The clinical features of HGV infection were analyzed in the meantime. Results Among 9 patients with single HGV infected, 3 cases were diagnosed as acute hepatitis, 3 cases as chronic hepatitis and the other 3 cases as healthy carriers. The detection rate of HGV RNA in PBMCs is much lower than that of HCV RNA (35.7 % vs. 77.3 % , P<0.05). In all 59 PBMCs and 129 serum samples no minus-stranded HGVRNA was found. Conclusion HGV infection can cause acute or chronic hepatitis with mild clinical manifestation. HGV can invade PBMCs but can not replicate in PBMCs, and the ability of HGV to invade PBMCs is lower than that of HCV.
3.Multidisciplinary approach in the management of diabetic retinopathy

Diabetic retinopathy (DR) is one of the microvascular complications of diabetes mellitus (DM).Like other macrovascular complications of DM,the development and progression of DR is influenced by a variety of systemic and local factors.It is essential to understand the importance of multidisciplinary collaboration.Systemic risk fators such as hyperglycemia,hypertension,dyslipidemia and diabetic nephropathy should be treated before effective DR management can be implemented.Through multidisciplinary collaboration,we can prevent the development of DR,slow the progression of DR,and improve the safety of perioperative care.Thereby enhancing the level of prevention and control of DM complications,including DR.
4.Exploring laser and lucentis treatment of diabetic retinopathy

Objective: To analysis the effects of laser and lucentis treatment of diabetic retinopathy. Methods:Fifty-three patients with DR were treated with laser and lucentis in our hospital from October 2012 to October 2013 as observer group. Forty-nine patients with DR were treated with laser in our hospital from September 2011 to September 2012 as control group. Results:The results indicated that visual acuity improved rate was 67.9%and decreased vision rate was 13.2% in observer group. Vision conditions in the observation group was better than that in the control group with significant difference (x2=4.60, x2=3.87; P<0.05). Simultaneously, improve time of hemorrhage, exudates, edema and macular thickness in the observation group was better than that in the control group with significant difference (t=4.17, t=3.92, t=4.06; P<0.05). Conclusion: The laser and lucentis treatment of diabetic retinopathy can significantly improving symptoms of retinal and promoting visual recovery.

Methods:492 community peopl e were sampled from community and tested by Generic Quality of L if e Inventory-74.Results:(1) Generic quality of life of comm unity people was well, and their body and mind were in good health; (2) Effects of mental health、physic health and social function on quality life were over ef fect of material life conditions;(3) Qualities of life of elders、workers、non-p rofession people were worse than qualities of young、profession people and cadre (P<0.01).Conclusion:Quality of life of community peop le of Hangzhou was in good condition, but attentions should be paid to qualities of life of elders and workers.

7.Investigation of severe skin infection of 38 elderly diabetic patients during the rehabilitation

Objective To investigate the mechanism,clinical features of severe skin infection developed in the rehabilitation of the elderly diabetic patients and the commonly seen bacterium.Methods The causes of skin injury,the places of infection and bacterium were analyzed retrospectively.Meanwhile,chemical indicators on admission were analyzed.Results Skin injury was caused by the improper family care and the lake of self protection knowledge.The common places for infection were feet and then back and hip skin.Almost all the infections were caused by opportunists,among which staphylococcus was the most common one.The major separated risk factors were hypoalbuminemia,hyperglycemia,and high level fibrinogen and anemia.Conclusion The elderly diabetic patients are subject to skin infection.Diabetic education and careful nursing are the key to the prevention of severe skin infection.
8.Interaction between HCV core protein and HCBP6 in mammalian cells

Objective To study the interaction between HCV core protein and HCBP6 in mammlian cells using CheckMateTM Mammalian Two-Hybrid System.Methods cDNA fragments encoding HCV core protein and HCBP6 were amplified by PCR and subsequently cloned into pGEM-T vector.After verified by sequencing,the target fragments were subcloned into mammalian two-hybrid plasmids,pBIND and PACT,respectively.The recombinant plasmids,pBIND-core and pACT-HCBP6 were co-transfected into HepG2 cells with reporter plasmid pG5luc.pBIND+pACT were induced as background controls,pBIND-Myod+pACT-Id as positive controls,and pBIND-core+pACT,pBIND+pACT-HCBP6 as blank controls.The expression of G5luc,which indicated the interaction between HCV core protein and HCBP6 in mammlian HepG2 cells,was assayed through Dual-Luciferase Report Assay System and Turner Biosystems Veritas Microplate Lunimometer.Results The recombinant vectors pBIND-core and pACT-HCBP6 were successfully constructed.When co-transfected into HepG2 cells with reporter plasmid pG5luc,there were significant differences in the luciferase activity in the pBIND-core and pACT-HCBP6 groups compared with every control group.Conclusions HCBP6 can interact with HCV core protein in HepG2 cells,which provides clues for further study on the function of HCBP6 and core proteins,and on the mechanism of HCV core in cell apoptosis and cancer transformation.
9.Effect of eucommia bark on osteopontin and osteoprotegerin expressions in rat bone marrow stem cells: Is there any difference between water extract and methanol extract?

BACKGROUND: Previous studies demonstrated that eucommia bark can promote bone marrow stern cells (BMSCs) differentiated into osteoblasts, but relative mechanism is poorly understood. OBJECTIVE: To investigate the effects of eucommia bark water/methanol extracts on expressions of osteopontin (OPN) and osteoprotegedn (OPG) in rat BMSCs. METHODS: Totally 2 g eucommia bark powder were added into water or methanol to 16 mL and oscillated for 1 hour at room temperature. After soaked overnight, both extracts were centrifuged at 15 000 r/min for 10 minutes. Water extract was obtained from supernatant in water soaked powder. In methanol soaked powder, methanol extracts was obtained by concentrated supernatant in vacuo and resolved using 16 mL water. Water and methanol extracts were then filtered by 0.22 μm membrane, and conserved at -20℃. Six SD rats, aged 2 months, were selected, and the 3~(rd)passage of BMSCs were induced by water or methanol extracts with dilution of 1 × 10~(-2), 1 × 10~(-3), 1 × 10~(-4) and 1 × 10~(-5), respectively. PBS was added in the negative control group. All cells were cultured for 6 days. Expressions of OPN and OPG was measured by immunocytochemistry at 6 days with induction. The expression of OPN and OPG induced by water and methanol at 1 ×10~(-3) and 1×10~(-4) dilution was detected by RT-PCR. RESULTS AND CONCLUSION: Immunocytochemistrical results indicated that both water and methanol extracts of eucommia bark simulated OPN and OPG expression, in particular with dilution of 1×10~(-4). The methanol extracts had a stronger effect than water extract, but the expression of OPG did not change obviously. RT-PCR demonstrated that at the 3rd day of inducement, the level of OPN expression induced by water extract was higher than that of methanol extract, and no OPG expression was detected. Osteogenic differentiation of rat MSCs induced by eucommia bark water/methanol extracts relates to stimulating expression of OPN, which has no correlation to OPG. OPN expression induced by water extract is early than that of methanol extract.
10.Progress in infection pathway and intracellular trafficking of adenovirus.

The research on intracellular trafficking of adenovirus has been described mainly through observations of subgroup C adenoviruses in transformed cell lines. The basic elements of the trafficking pathway include binding to receptors at the cell surface, internalization by endocytosis, lysis of the endosomal membrane, escape to the cytosol, intracellular trafficking along microtubules, nuclear pore docking, and viral genome translocation into the nucleus. More than 80% of the adenovirus genome is delivered to the nucleus in a highly efficient manner in approximately 1 h. However, exceptions to this trafficking pattern have been noted, including: variations based on target cell type, cell physiology, and adenovirus serotype. This review summarizes mechanism of adenovirus infection pathway and intracellular trafficking, providinging a foundation for the development of clinical adenoviral vector.
