1.Research progress on exosome-derived non-coding RNA in digestive system malignancy

Exosome is a kind of biological membrane structure at nanometer level. It is secreted by various cells and widely distributed in most body fluids. Biological active substances, including non-coding RNAs and proteins, have been identified in the exosomes. Exosome may be involved in cell-to-cell substance transporting and signaling. Non-coding RNAs have important function of regulating gene expression and also play key roles in the occurrence and development of many cancers. Research on exosome-derived non-coding RNA is a new hotspot at present. In this paper, we will review research progress on exosome and exosome-derived non-coding RNA in digestive system malignancy.
2.A case report of solitary fibrous tumor of the prostate

Solitary fibrous tumor (SFT) is a rare tumor originated from mesenchymal spindle cells. It is very rare to occur in the prostate. A 30-year-old male with dysuria was treated by transurethral holmium laser enucleation of the prostate. The postoperative pathological Results showed that solitary fibrous tumor.The urination symptoms of the patients were significantly improved.
3.The protective effect of tumor necrosis factor-?antibody on lung injury after cardiopulmonary bypass

Objective: To study the protective effect of anti-tumor necrosis factor-?antibody (TNF-? Ab) on lung injury after cardiopulmonary bypass(CPB). Methods: 20 patients with valve diseases underwent valve replacement under CPB. The patients were randomly divided into two groups: control group and TNF-? Ab group. In the TNF-? Ab group, human TNF-? Ab was dropped into the intracheal tube before operation and just after releasing the aortic clamp, respectively. Lung dynamic compliance, blood neutrophils count and TNF-? from the right and left atrium were determined perioperatively. Results: The dynamic compliance of the lung in TNF-? Ab group was higher than that in control group. TNF-? Ab can reduce releasing of TNF-? and restrain leukocyte accumulation in the lung. Conclusion: TNF-? Ab has markedly protective effect on lung injury after CPB.
4.Comparison of Several Methods and Their Progresses on Molecular Authentication of Lonicera japonica Thunb

Authentications of Chinese herbal medicine have a critical effect in Chinese clinical medicine. DNA molecular marker, as an important component for true or false authentication, is more and more widely used in iden-tification of Chinese medicinal materials. At the same time, many new methods for authentication of Chinese medici-nal materials are continuously emerging. But the systematically comparative analysis of these new methods is lack. The present study taking Lonicera japonica as an example, systematically compared principles, characteristics, ex-periment methods, detection time and the application scope of express sequence tag-simple sequence repeat (EST-SSR), polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP), allele-specific PCR (AS-PCR), DNA barcoding and loop-mediated isothermal amplification (LAMP), and put forward corresponding improve-ment opinions. This study can help to screen appropriate approach for rapid authentication of L. japonica and offer demonstrating to other Chinese herbal medicines.
5.The roles of different signaling pathways and their related genes in the occurrence and development of colorectal cancer

Colorectal cancer is a kind of malignancies with high incidence in the worldwide, that is seriously harmed human health. So far the pathogenesis of the disease is not fully understood, this causing many difficulties to the diagnosis and treatment of the disease, and resulting in the cure rates of disease is not ideal. With the development of molecular genetics and molecular biology, many oncogenes and tumor suppressor genes have been found to be associated with the disease, and this made it is possible to reveal the pathogenesis of colorectal cancer at the molecular level. However, it is a complex and multi-step process from normal colorectal epithelial cells transformed to colorectal cancer cells, and it is the results of polygenic and multifactorial interactions. Now it is thought that the Wnt, TGF-beta, PI3K/Akt, MAPK and p53 signaling pathways are closely associated with pathogenesis of colorectal cancer. Based on the five kinds of signaling pathways as the main line, this article reviewed the roles of different signaling pathways and their related genes in the pathogenesis of colorectal cancer.
6.Study on chemotaxis of regulatory T cells induced by different antigens in mice

Objective:To investigate the chemokine receptors expression on regulatory T cells induced by different antigens and chemotaxis of T cells conducted by CCL20 and CCL22.Methods:BALB/c mice were divided into different groups and inoculated with skin-antigen derived from C57BL/6 mice or BCG vaccine respectively.The changes of CCR4 and CCR6 expression on CD4+CD25-T cells and CD4+CD25+CD127-T cells were detected at 1st,2nd,3rd and 4th week using flow cytometer.The chemotactic effects of CCL20 and CCL22 on CD4+CD25-T cells and CD4+CD25+CD127-T cells subsets were assayed by chemotaxis assay.Results: ①In skin-antigen group,the average fluorescence intensity ( MFI) of CCR4 on CD4+CD25-T cells at 4 week was significantly stronger than that at 3 week (P<0.05).There were no significant changes of CCR4 expression in BCG group.②The MFI of CCR6 on CD4+CD25-T cells was strongest at 2 week in skin-antigen group (P<0.05) while in other groups at 4th week (P<0.05).Besides,the expression of CCR6 on CD4+CD25+CD127-T cells was stronger during the first two weeks than the later two weeks ( P<0.05) in skin-antigen group, while in BCG group,the MFI was stronger at 2nd and 4th week than 1st week (P<0.05).③The chemotactic index of CD4+CD25+CD127-T cells was highest at 4th week in BCG group (P<0.05) in CCL20 induced chemotaxis,while in other groups were higher at 2nd and 4th week(P<0.05).In CCL22 induced chemotaxis ,there were no significantly differences of chemotactic index of CD4+CD25+CD127-T cells between skin-antigen group and BCG group.Conclusion:①The expression of chemokine receptor on the surface of Treg was associated with antigenic properties.②CCL22 had a notable chemotactic effect on Treg at the early stage of post-induction, while CCL20 did that at the late stage of post-induction.
7.Effect of chemokine CCL20 and CCL22 combined with skin antigen-induced Treg on survival time of grafted skin

Objective:To study the effect of chemokines CCL20 and CCL22 combined with skin-induced Treg on survival time of grafted skin.Methods: Skin grafting mice were divided into four groups, three mice per group, namely Treg group, Treg+CCL20 group,Treg+CCL22 group and control group.C57BL/6 mice were used as donor and BALB/c as acceptor, and the Treg cells were isolated from the mice induced by skin allograft.After skin grafted,CCL20 and CCL22 were subcutaneous injection every day,which lasted for 10 day.Survival time of skin in each group were observed and recorded.The Treg colonzation experiments were performed as follows.We firstly isolated Treg with Magnetic cell sorting system( MACS) and then labled them with 99 Tcm.After that we intravenously injected them into the mice.3 hours later,the mice were sacrifced and the radioactivity of organs were detected by GC-2016γradioim-munoassay counter.Results:①After Treg treated the survival time of skin grafted in antigen-induced Treg group was signifiantly longer than control group,when treg were cooperated with CCL20 and CCL22,the skin grafted showed more longer survival time than Treg and control groups( P<0.001 ).②After injection of induced Treg, Treg in autologous and allogeneic skin grafts goups were mainly distributed in autologous and allogeneic skin,accounting for 60% and 98% respectively.When cooperated with CCL20 or CCL22,the Treg were mainly distributed in liver.Conclusion:Chemokines CCL20 and CCL22 synergistically improved the effects of skin antigen induced Treg on survival time of skin graft,which probably related with the Treg colonization into the liver.

Objective To optimize the extraction methods of mitochondrial genome DNA(mtDNA)of Oncomelania hupen-sis. Methods The pyrolysis,protein K variable-temperature digestion and high-concentration potassium acetate purification were applied to optimize the high-concentration-salt precipitation method,and then the optimized method was compared with two common extraction methods,the sucrose density gradient centrifugation method and traditional high-concentration-salt pre-cipitation method. The mtDNA samples were identified by using spectrophotometry,agarose gel electrophoresis and the amplifi-cation products of COX1. The nuclear DNA contamination was tested by the amplification products of ITS. Results The concen-tration and yield of the improved method was significantly higher than those of the traditional method(F=3032.65,10185.00, both P<0.01). The mtDNA samples extracted were essentially free of nuclear DNA and protein,meeting PCR,sequence analy-sis and other molecular biology research requirements. Conclusions The improved high-concentration-salt precipitation meth-od for isolating mtDNA is simple,and it has high yield and low cost. The extracted mtDNA can meet relevant analysis require-ments.
10.Investigation of abnormal menstruation of female workers in petrochemical industry

Objective:To explore the influencing factors of abnormal menstruation of female workers in a petrochemical enterprise.Methods:A total of 5186 female employees in a petrochemical enterprise were selected as the research objects from July 2018 to December 2019. A questionnaire survey was conducted from four aspects: basic information, occupational status, psychological status at work, reproductive and reproductive health, and a multivariate logistic regression model was used for analysis.Results:Among 5132 female workers, 676 were postmenopausal and 1117 (25.1%) among 4456 non-postmenopausal women had abnormal menstruation in the last 6 months. Logistic regression analysis showed that age (<30、30~35 years) and length of service (≤20 years) were protective factors ( OR=0.573, 0.590, 0.798, P<0.05) . Age (40~45、45~50 years old) , sleep duration (<6 h) , premature OR late menopause of maternal relatives, occupational exposure to dust, occupational exposure to chemicals, not wearing earplugs, leave due to health reasons in the last 6 months, reproductive tract infection OR symptoms in the last 6 months, spontaneous abortion, induced abortion, physical fatigue and mental fatigue were risk factors for menstrual abnormalities in female workers ( OR=2.318, 5.544, 1.231, 1.199, 1.336, 1.403, 1.351, 1.538, 1.613, 1.584, 1.199, 1.601, 1.936, P<0.05) . Conclusion:The abnormal menstruation of female workers is affected by many factors in the occupational process, and specific measures should be taken to improve the reproductive health of female workers.
