1.Research on the correlation of Disabled-2 gene and tumors
China Oncology 2001;0(03):-
Signal transduction is a very fine and complicated process that indudes participation in the cell propagation, division, maturation and apoptosis.Under normal circumstance disabled-2( Dab2) in human tissues participates in the signal transduction of the cell in order to allow the body to keep normal functions, but under some circumstances the absence of Dab2 influences the normal signal transduction path, leding to atypical cell changes.and the generation of tumors. This paper is a review on the construction and function of Dab2 and its related gene as well as the relation of Dab2 and tumors generation.

Objective To evaluate the expression of the GLUT-1 glucose transporter in borderline and malignant epithelial neoplasms of the ovary. Methods Histologic sections of tumor tissues from 100 epithelial neoplasms of the ovary were stained for GLUT-1 using polyclonal GLUT-1 antibody and the labeled streptavidin procedure. Chi-square analysis was used to assess if their expression was associated with clinicopathoiogic characteristics of the tumors. Results Normal ovarian surface epithelial cell were either negative or weakly positive. The intensity of the stain was significantly stronger in malignant than borderline tumors (P<0.001). In malignant neoplasms, the intensity of the stain for GLUT-1 increased with histology. Conclusion Overexpression of the GLUT-1 transporter is associated with the histology and grade of the minors. These data suggest that the expression of this transporter may be closely related to the malignant transformation of epithelial ovarian tumors.

Objective To determine whether rizatriptan has an effect on cortical spreading depression (CSD) and c-Fos expression within periaqueductal grey (PAG) induced by CSD in rats. Methods The experimental SD rats were randomly divided into group A injected with KCl, group B KCl plus rizatriptan and group C NaCL The number and amplitude of CSD were recorded after KCl or NaCl injection. C-Fos positive neurons of different layer were identified by the immunohistochemical technique 2 hours after the first injection of KCl or NaCl. Results There was no CSD in group C. The number of CSD in group A ( 10.70±3.23 ) was significantly more than that in group B (6.10±2.56, t = - 3.528, P < 0.01 ). The amplitude of CSD in group A ( 17.33 (95% CI 11.45--23.11 ) mV) was significantly greater than that in group B (11.82 (95%CI 9.24--14.70) mV, Z= -4.360, P< 0.01). There were more cFos-like immnoreactive neurons in every layer in group A than in group C (P < 0.01 ) and in group B (P < 0.05 ). Conclusion Rizatriptan has an inhibitory effect on CSD, which might induce the headache through exciting the neurons in PAG.

Objective To explore the application value of magnetic resonance molecular functional imaging diffusion weighted imaging(DWI) in the identification of pancreatic carcinoma and mass-type pancreatitis of animal model. Methods Each 8 cases of laboratory pancreatic head transplantation tumor model, chronic mass-type pancreatitis model and normal rabbits were selected and performed the MR DWI molecular functional imaging, the b values were 333,667,1 000 s/mm2 respectively. The apparent diffusion coefficients(ADC) of pancreatic carcinoma model,mass-type pancreatitis model and normal pancreas under different b values were observed. Then the change situation of ADC values of pancreatic carcinoma model,mass-type pancreatitis model and normal pancreas under different b values and difference of ADC(DADC) was analyzed. Moreover the differences in molecular diffusion, tissue perfusion among various groups were observed. Results Throughout the study period, the mortality rate of pancreatic head transplantation tumor model was 50%;the mass-type pancreatitis model and 8 normal rabbits were normally survival. The ADC value of pancreatic carcinoma under the same b value was significantly lower than that of chronic inflammation and normal pancreas area. The ADC value in each group was decreased with the increase of b value, and there was significant difference in ADC value when the b value was 333 s/mm2(F=6.662,P=0.014),in the pairwise comparison among groups, the difference between pancreatic cancer and pancreatitis (t=6.773,P=0.003) and between pancreatic cancer and normal pancreas(t=5.883,P=0.016) had statistical significance (P<0.05).The b value was increased,DADC was smaller, the difference change of DADC between pancreatic cancer area and chronic pancreatitis mass area, between pancreatic cancer area and normal pancreatic head area had statistical significance (P<0.05).Conclusion Rationally selecting the molecular functional imaging DWI technology of b value can better distinguish pancreatic cancer from mass-type pancreatitis, which may be promoted and applied in the evaluation of animal pancreatic head cancer model.

OBJECTIVESTo explore the mechanisms of neuronal loss and apoptosis in the brains of Alzheimer's disease (AD) patients, through studying the expression of proteins related to signal transduction pathways, which are important for neuron survival.
METHODS(1) Immunohistochemistry: Sections were double stained with Tunel and NSE antibodies. (2) The hippocampal tissue taken from 6 cases of AD and 6 cases of non-AD brains was homogenized. Protein estimation was done by Lowry method. Equal amounts of protein were taken from each specimen and immunoprecipitation was performed and analyzed by Western blot; color development was done by alkaline phosphatase method or luminol reagent.
RESULTS(1) Tunel positive neurons were found in both AD and non-AD brains, but the number in the former was more than the latter. (2) The AD hippocampal tissue showed diminished expression of Akt/PKB, CREB, P-CREB, increased expression of apoptosis-related protein apoptosis-inducing factor, and diminished expression of apoptosis-related protein bcl-2. The expression of bax did not change.
CONCLUSIONSDiminished expression of CREB, P-CREB, bcl-2 in AD hippocampus indicates that the neuron survival signal transduction pathway in AD brains is impaired. Neurons are in apoptotic or pro-apoptotic state. In addition, increased expression of apoptosis-inducing factor, diminished expression of bcl-2, which is an anti-apoptotic factor, promotes further neuron apoptosis.

Chinese Journal of Clinical Medicine 2024;31(5):790-794
Objective To explore the impact of sluggish cognitive tempo (SCT) on the efficacy of atomoxetine in the treatment of attention-deficit/hyperactivity disorder (ADHD). Methods A prospective study was conducted. Pediatric patients aged 6-12 years with a diagnosis of ADHD were selected from the Department of Psychiatry, Xijing Hospital, Air Force Military Medical University from May 2020 to May 2023. All the patients were treated with atomoxetine for 8 weeks. ADHD symptoms and SCT symptoms were assessed by Swanson, Nolan, and Pelham-Ⅳ rating scale (SNAP-Ⅳ) and SCT rating scale at baseline (before treatment), 4 weeks and 8 weeks after treatment. Pearson correlation analysis was used to analyze the correlation between SCT and SNAP-Ⅳ scores. Results A total of 61 pediatric patients were enrolled, including 10 cases of inattentive type and 51 cases of mixed type. Baseline SCT score was positively correlated with baseline SNAP-Ⅳ score (r=0.490, P<0.001) and reduction rate of SNAP-Ⅳ score at week 8 (r=0.670, P<0.001). The total SCT score at week 8 was positively correlated with total SNAP-Ⅳ score at baseline (r=0.320, P=0.012). The baseline SCT score of patients with mixed type was positively correlated with reduction rate of SNAP-Ⅳ score at week 8 (r=0.578, P<0.001). Conclusions The severity of SCT symptoms is positively correlated with the SNAP-Ⅳ reduction rate 8 weeks after atomoxetine treatment, which could be a predictor of the efficacy of atomoxetine.
