1.Identification and Characterization of The Arabidopsis fumonisin B_1 resistant fbr136

Plant programmed cell death (PCD) plays an important role in plant growth and development as well as defensive response against biotic and abiotic stresses. Fumonisin B1 (FB1) is a fungi toxin, which is a competitive inhibitor of ceramide synthase in de novo sphingolipid biosynthesis. FB1 can induce PCD in both animal and plant cells. To dissect this pathway, a genetic screen for fumonisin B1 resistant (fbr) mutants, and identified 11 fbr mutants was carried out. Genetic analysis showed that these 11 mutants belonged to 9 complementary groups or genetic loci. Here a detailed phenotypic analysis of fbr136 was reported. In addition to the resistance to FB1, fbr136 also showed resistance other PCD-inducing compounds, including H2O2 and paraquat. Furthermore, FB1-induced PR1 expression was reduced in fbr136, suggesting that a PCD pathway is likely impaired in the mutant. When stained with nitroblue tetrazolium (NBT), fbr136 showed a reduced accumulation of reactive oxygen species (ROS) induced by FB1. The fbr136 mutation was roughly mapped onto chromosome Ⅲ, where no other fbr mutants have been previously identified. It is proposed that FBR136 may act as an important regulator in a sphingolipid-mediated PCD pathway, involved in the generation of ROS.
2.Analysis of clinical pathological characteristics of senile recurrent ovarian cancer

Objective To discuss and analyze clinical pathological characteristics of senile recurrent ovarian cancer and its influences on prognosis.Methods 140 senile ovarian cancer patients who have been admitted in ourhospital were selected as study subjects.103 cases with recurrent ovarian cancer were selected as observation group (group A),and the rest 37 cases who did not suffer from recurrent ovarian cancer were selected as control group(group B).Pathological characteristics of carcinoma tissues after their first admission in our hospital were compared between the two groups.And pathological characteristics of carcinoma tissues of group A after their second admissionin our hospital were analyzed,comprehensively analyzed prognosis situations and clinical pathological characteristics of patients in these two groups.Results In group A,the number of poorly differentiated carcinoma tissues was obviouslymore than group B,and the number of patients whose neoplasm staging was M stage was more than that in group B,the differences between the two groups were significant (x2 =13.48,15.87,all P < 0.05).The incidence rate of adverse reactions between the two groups also had statistical significance(x2 =13.8,11.6,14.5,13.9,14.9,13.4,all P <0.05).Conclusion The clinical pathological characteristics of senile recurrent ovarian cancer can reflect patients'prognosis situations in some degree,and has certain clinical value in evaluation of patients' disease recurrence and prognosis.
3.Enzyme electrode for the determination of sodium lactate in sodium lactate injection

OBJECTIVE: To determine the concentration of sodium L-lactate in sodium lactate injection. METHODS: Lactic oxidase was immobilized at a hydrogen peroxide electrode and the enzyme electrode was used for the amperometric determination of sodium L-lactate in sodium lactate injection. Sodium D, L-lactate in injection was analyzed by that technique according to Chinese Pharmacopoeia. RESULTS: The quantity of sodium L-lactate was about 10% in total amount of sodium D, L-lactate in sodium lactate injection. CONCLUSION: Sodium L-lactate in sodium lactate injection could be determined specially, rapidly and inexpensively.
4.Influence of Acupuncture on Cephalic Acupuncture Points Region Over the Brain-stem Evoked Potential of Patients Suffering Insufficient Blood-Supply of Vertebral Basilar Artery

Objective From the angle of electrophysiology, go further into the mechanism of action of acupuncture treatment to the insufficient blood-supply of vertebral basilar artery and at the same time, make a comparison of the effects between the acupuncture on cephalic meridian points and zonation cephalic acupuncture. Methods Through the contrast before and after adopting the self-acupuncture on the testees of 28 cases (including healthy persons of seven cases and TIA patients of 21 cases), observe the change of immediate effect of all waves latent period values of BAEP. Results The BAEP wave latent period of healthy persons (exclusive of I wave of auditory fainting region) assumes a relative delay within the physiological range and by comparison between points and region, the delay of V-wave Baihui point is evidently higher than that of auditory fainting region. However, the latent period of all waves of TIA patients is relatively moved up, especially there are evident differences before and after the I-wave acupuncture is carried out. Viewing from the comparison between points and region, there is no significant difference. Conclusion The function of acupuncture lies in the adjustment of antagonistic body state and it is most effective under the pathological state. Acupuncture will play the main role of excitation to the auditory nerve conduction of BAEP of TIA patients (from cochlea to midbrain) and the influence over the peripheral nerves is more sensitive than over the center ones. Although points and region have their own relative specificities, there is no significant difference between them.
5.Study on enhanced resist to hypoxic/hypoglycemic condition by IL-32β in cervical carcinoma C33A cells

Objective To explore the enhancement effects and mechanisms of IL-32β on human cervical carcinoma cells C33A to hypoxic/hypoglycemic condition.Methods After cultured in hypoxia/hypoglycemic circumstance and normal circumstance for 20 hours respectively, the mRNA and protein expression of IL-32β in C33A cells were detected by real time-polymerase chain reaction (RT-PCR) and Western blotting respectively.Trypan blue stain was used to detect C33A cells viability in hypoxia/hypoglycemic circumstance and adding 10, 100,500 ng/ml IL-32β circumstance.The xenografted tumor of nude mice was established by intraperitoneal injection, and their volumes were tested for a given time after injecting 0, 1.0 mg/kg IL-32β.siRNA was used to construct IL-32β knockdown cells and detect the expression of VEGF.Results Under the hypoxia/hypoglycemic circumstance, the expressions of IL-32β mRNA were (6.12 ± 0.03) times of the normal circumstance (F =43.16, P < 0.05), the expressions of IL-32β protein were (2.23 ± 0.04) times of the normal circumstance (F =22.32, P < 0.05).The C33A cells viability in hypoxia/hypoglycemic circumstance was (51.92 ± 3.41) %, whereas, viability in 10 ng/ml IL-32β group was (55.23 ± 3.92) % (F =14.25, P < 0.05), viability in 100 ng/ml IL-32β group was (62.52 ± 4.14) % (F =35.53, P < 0.01), viability in 500 ng/ml IL-32β group was (69.14 ± 2.45) % (F =56.28, P < 0.01).After 28 days, the volume of xenografted tumor of 0 mg/kg IL-32β group was (578 ± 64)mm3, and 1.0 mg/kg IL-32β group up to (1 402 ± 142) mm3 (F =27.84, P < 0.01).In addition, compared with control group, the expression of VEGF in IL-32β knockdown C33A cells was significantly decreased (F =36.85, P < 0.05).Conclusion IL-32β can enhance the resistance to hypoxic/hypoglycemic condition of C33A cells, which is associated with the increase of VEGF.
6.Determination of gene polymorphism and serum concentration of mannose-binding lectin in patients with psoriasis

Objective To determine the gene polymorphism and serum concentration of mannose-binding lectin (MBL) in patients with psoriasis,and to analyze the relationship between MBL and psoriasis.Methods Totally,67 patients with psoriasis vulgaris and 69 healthy human controls were enrolled in this study.Venous blood samples were obtained from all the subjects.Genomic DNA was extracted,and PCR-restriction fragment length polymorphism (PCR-RELP) analysis was conducted to determine the polymorphism at codon 54 of the MBL gene.Enzyme-linked immunosorbent assay was performed to measure the serum level of MBL.A chi-square goodness-of-fit test was carried out to evaluate Hardy-Weinberg equilibrium,t test to compare the serum concentration of MBL,and chi-square test to compare the frequency of genotypes and alleles of MBL gene codon 54.Results The patients with psoriasis showed higher frequency of GGC/GAC heterozygote but lower frequency of GGC/GGC homozygote (x2 =10.36,P < 0.05),together with increased frequency of GAC allele but decreased frequency of GGC allele (x2 =8.31,P < 0.05),at codon 54 of the MBL gene compared with the healthy controls.The variant allele GAC at codon 54 of the MBL gene was markedly associated with psoriasis (OR =3.383,95% CI 1.585-7.211,P < 0.05).The serum concentration of MBL was (2.193 7 ± 0.816 3) mg/L in patients with psoriasis,significantly lower than that in the healthy controls ((3.269 5±1.205 8) mg/L,t=6.11,P< 0.05).Conclusion MBL might be associated with the pathogenesis of psoriasis to some degree.
7.Expression of mannose-binding lectin in lesions of psoriasis vulgaris

Objective To investigate the expression of mannose-binding lectin (MBL) in lesions of patients with psoriasis vulgaris,and to explore the relationship between MBL and psoriasis pathogenesis.Methods Immunohistochemistry and Western blot were performed to detect the expression of MBL in lesional and normalappearing perilesional skin of 30 patients with progressive psoriasis vulgaris,as well as in normal skin of 30 healthy human controls.Statistical analysis was carried out by t test using SPSS13.0 software.Results Immunohistochemistry showed that MBL was expressed in lesional psoriatic skin,but weakly expressed or absent in normalappearing perilesional skin and normal control skin,with the relative expression level of MBL in lesional skin significantly higher than that in perilesional skin and normal control skin (0.636 7 ± 0.515 1 vs.0.416 3 ± 0.160 1 and 0.381 6 ± 0.310 9,t =2.24,2.32,respectively,both P < 0.05).Western blot revealed a positive expression of MBL protein in all the skin specimens,and the expression intensity of MBL protein in lesional psoriatic skin was significandy increased compared with perilesional psoriatic skin and normal control skin (0.273 1 ± 0.129 4 vs.0.186 3 ± 0.193 1 and 0.149 2 ± 0.268 7,t =2.05,2.28,respectively,both P< 0.05).No significant difference was shown in the expression of MBL protein between perilesional psoriatic skin and normal control skin by immunohistochemistry (t =1.51,P > 0.05) or Western blot (t =0.61,P > 0.05).Conclusion There is a high expression of MBL protein in lesions of patients with psoriasis vulgaris,which may be somewhat associated with the pathogenesis of psoriasis.
8.Effect of FCGR3A Polymorphisms on NK Cell Function

Objective To investigate the effect of FCGR3A polymorphisms on NK cell function. Methods Peripheral blood samples from can?cer patients were collected and FCGR3A polymorphisms were confirmed by PCR. In vitro proliferation rates,ADCC activity,and expression of NK cell activating receptors were compared under trastumab stimulation. Results This study showed that the wild?type FCGR3A exhibited a higher affinity to trastumab along with better NK cell proliferation and ADCC activity than the mutant type. Compared to the patients with wild?type FC?GR3A,the proliferation rates of NK cells in patients with the mutant type were reduced by approximately 8?fold. In addition,the expression of NK cell activating receptors in patients with wild?type FCGR3A was higher than in patients with the mutant type. Conclusion Mutations in FC?GR3Areduce NK cell function,causing a poor reaction to monoclonal antibody.
9.Effect of delayed cord clamping on neonatal resuscitation

ObjectiveTo investigate the effect of delayed cord clamping on outcomes of neonatal resuscitation.MethodsTotally, 7 429 full term infants born in the Maternal and Child Health Care Hospital of Jilin from January 2013 to January 2015 were randomly divided into two groups, the observation group (n=3 727) and the control (n=3 702). The cords were clamped 2 min delayed in the former group, while those in the latter group were clamped within 10 s after birth. The rate of asphyxia and successful resuscitation, Apgar score at 1 and 5 min after birth, and pH value, base excess (BE), blood glucose, cardiac troponin (cTn) I at 1 h after birth were compared witht-test orChi-square test.ResultsThe incidence of neonatal asphyxia in the observation group was lower than in the control [0.75%(28/3 727) vs 1.21%(45/3 702),χ2=4.115,P<0.05], but no significant difference was found in the rate of mild or severe asphyxia between the two groups [0.62% (23/3 727) vs 0.89% (33/3702), 0.13% (5/3 727) vs 0.32% (12/3 702), allP>0.05]. In observation group, those newborns who suffered from severe asphyxia had higher Apgar score at 1 and 5 min than control (1 min: 2.80±0.45 vs 2.08±0.67,t=2.181; 5 min: 8.00±1.00 vs 6.25±1.66,t=2.176; bothP<0.05). Higher blood pH value and lower glucose level were shown in the observation group than in the control group [7.16±0.41 vs 7.13±0.72, (5.91±1.19) vs (6.60±1.56) mmol/L,t=2.068 and 2.046, bothP<0.05]. However, no difference was found in BE and cTnI levels between the two groups (bothP>0.05). For those babies with severe asphyxia, the blood pH, BE, blood glucose and cTnI levels in the observation group were significantly different from the control group [7.08±0.29 vs 7.02±0.56, (-16.82±0.60) vs (-17.43±0.35) mmol/L, (7.93±0.78) vs (8.02±0.53) mmol/L and (0.203±0.041) vs (0.249±0.035) ng/ml,t=2.270, 2.387,-2.371 and-2.341, allP<0.05].Conclusion Two minutes delayed in cord clamping could help to reduce the incidence of asphyxia and improve the effect of resuscitation in severe asphyxia.
10.The chest radiological manifestation in psittacosis

Objective To summarize the cl inical characteristics and imaging features of psittacosis. Methods The clinical features and imaging appearances of 3 cases with acute psittacosis were retrospectively analyzed. The related literature was reviewed . Results The clinical manifestation of psittacosis was high fever in the patients. Physical findings included pulse-temperature dis sociation, localized lung crackles, hepatomegaly, and splenomegaly. Laboratory findings showed elevation of ESR in all cases, and liver dysfunction was present in 2 cases. The counts of white blood cells were normal, but the percent of ne utrophils might be increased. The chest X-ray and CT scan showed air-space co nsolidation and ground-glass attenuation in the lung, and miliary, nodular, or consolidated shadows were found in pathological areas. Pleural effusions were a lso present in 2 cases. Psittacosis was diagnosed from the history of exposure to infected parrots and elevation of the IgG and IgM titer for Chlamydia psittac i. Erythromycin was effective in all 3 patients. Conclusion Although the appearance of psittacosis on clinical findings and chest X-r ay and CT scan is not characteristic, psittacosis can be diagnosed with the comb ination of the history of exposure to infected parrots and laboratory findings. CT scan can reveal the focus earlier and accurately, and catching the imaging f eatures of psittacosis is helpful in differential diagnosis.
