1.Classification and characteristic of the acupuncture medical literature during the period of the Republic of China.

The acupuncture medical literature during the Republic of China is divided into four categories, including theoretical literature, acupuncture channels and acupoints drawing literature, translations, and teaching materials and medical cases. It is found that the theoretical literature focuses on inheriting the traditional theories as well as combining traditional Chinese and Western medicine. The drawings are becoming more and more precise, and in these drawing works anatomy begins to appear at that time. Translations are the achievements of communication between TCM and Western medicine, which promote the scientization of acupuncture greatly. Teaching materials are also influenced by Western medicine, presenting a rough frame of modern acupuncture disciplinary system. As important clinic data, medical cases have great clinical value. Overall, acupuncture medical literature during the Republic of China is widely published, which is influenced greatly by the Western medicine and presents a significant tendency of scientization.

2.ZHU Lian's New Acupuncture Academic System and acupuncture science initialization.

Acupuncture scientization was a consensus of most of acupuncture scholars who had long-term perspectives in the 20th century, among them Ms. ZHULian was the important one. Ms. ZHU Lian built a systemic new acupuncture" academic structure in practice and theory aspects. At the same time, as the main architect of Institute of Acupuncture-moxibustion of China Academy of Traditional Chinese Medicine, Ms. ZHU Lian was the first one who began to carry out the acupuncture clinical trail and laboratory experiment in modern way, which meant "acupuncture therapy" was transformed into "acupuncture science" by Ms. ZHULian's endeavor.

3.The role of RET proto-oncogene methylation in the pathogenesis of Hirschsprung's disease

Objective To investigate the relationship between the methylation of RET (proto-oncogene, RET) and Hirschsprung disease (HD), and understand its significance in the development of intestinal wall ganglion cells. Methods Twenty-one surgical removal specimens, which were all dilation segment of HD in Tianjin Children’s Hospital, were used as experimental group, and 5 samples of non-HD normal colon tissues were used as control group. The bisulfite sequencing (BSP)-direct detecting method was used to detect RET CpG island methylation status. The expression of RET protein was detected by immunohistochemistry in experimental group and control group. Results In the experimental group 12 cases (57.14%) were found methylation, but no methylation was found in control group. The average optical density of methylated RET protein was 0.201±0.015 in 12 cases. The average optical density of un-methylated RET protein was 0.364±0.023 in 9 cases (P<0.05). Conclusion RET CpG island methylation reduced protein expression levels of RET. The corollary RET gene methylation may influence the expression levels of RET protein, thereby affecting the ganglion cell development, and thus participating in the occurrence of HD.
4.Screening of optimal embryonic time for in vitro separation and culture of Kunming mouse embryonic stem cells:Comparison among 2.5,3.5,and 4.5 pregnant days

BACKGROUND:Genotype of Kunming mice was similar to human population,thus an establishment of embryonic stem cell line is beneficial for research of transgenic animal.However,the best time to collect embryo has been less reported yet.OBJECTIVE:To find the best time to collect embryos from Kunming mice.METHODS:The embryos were collected from mother mice of 2.5,3.5,and 4.5 pregnant days.Microscope was used to evaluate the growth condition of embryos,embryo attaching rate(A/C),inner cell mass(ICM)growing rate(I/C),embryonic stem cells (ESCs)clone growing rate(P1/C)and ESCs subclone growing rate(P2/C).The cells were then stained with alkaline phosphatase.RESULTS AND CONCLUSION:Most of the 2.5-pregnant-day embryos were 16-cell-phase embryos.The 3.5-pregnant-day embryos were morulas while the 4.5-pregnant-day embryos were blastulas.There were no significant differences in A/C,I/C,P1/C,P2/C between 2.5 and 3.5 pregnant days(P > 0.05).The 4.5-pregnant-day indicators mentioned above were significantly greater than those two groups;therefore,4.5-pregnant-day embryos were the best source to culture,clone,isolate and passage ESCs.
5.Value of clinical parameters in predicting the initiation of renal replacement therapy in acute kidney injury patients with cardiorenal syndrome

Objective To investigate the value of clinical parameters in predicting the initiation of renal replacement therapy(RRT) in acute kidney injury (AKI) patients with cardiorenal syndrome (CRS).Methods A total of 75 AKI patients hospitalized with CRS were enrolled.All patients received pharmacologic therapy on the beginning 3 days.The patients whose heart function improved were divided into control group (n=39),and the patients whose heart function worsened were divided into RRT group (n=36).Clinical and laboratory data on the first day and the fourth day were collected and analyzed.The factors on the first day were labeled asⅠ ,and those on the fourth day were labeled asⅡ. The ratio of some parameters calculated were labeled asⅡ/Ⅰ .Area under curve (AUC) of receiver operating characteristic curve (ROC) of these factors was used to evaluate the sensitivity and specificity in predicting the initiation of RRT.Results The patients in RRT group had significantly higher levels of BNP-Ⅱ,BNP Ⅱ / Ⅰ and creatinine Ⅱ / Ⅰ (P < 0.01),and lower levels of 24 hours urine volume-Ⅰ and 24 hours urine volume-Ⅱ (P < 0.01).From ROC curve analysis,the AUC of 24 hours urine volume-Ⅰ,24 hours urine volume-Ⅱ,creatinine Ⅱ / Ⅰ,BNP-Ⅱ levels and BNP Ⅱ/Ⅰ to predict RRT were 0.736,0.875,0.747,0.779 and 0.894 respectively.When the cutoff values of 24 hours urine volume-Ⅰ,24 hours urine volume-Ⅱ,BNP-Ⅱ levels,BNP Ⅱ / Ⅰ and creatinine Ⅱ / Ⅰ were 905 ml (sensitivity 75%,specificity 94.9%),1450 ml (sensitivity 75%,specificity 100%),3360 ng/L (sensitivity 72.2%,specificity 100%),1.37 (sensitivity 75%,specificity 100%) and 1.25 (sensitivity 72.2%,specificity 94.4%) respectively,the value of the parameters to predict RRT was high.Conclusions The 24 hours urine volume,BNP levels after treatment and the dynamic changes of BNP levels and creatinine levels can be used as predictors of the initiation of RRT in the AKI patients with CRS.
6.Diffusion-weighted imaging in differentiating benign and malignant lesions of the tongue

Objective To observe the value of diffusion-weighted imaging (DWI)and ADC value in the differential diagnosis of benign and malignant lesions of the tongue.Methods 75 patients with lingual lesions,including 32 benign lesions and 43 malignant tumors,underwent conventional MRI,contrast-enhanced MRI and DWI with b values of 0 and 1 000 s/mm2 before therapy.ADC maps were reconstructed,and the ADC values of the lingual lesions were calculated.Diagnostic performance of ADC was compared using receiver operating characteristic curves (ROC).Results The mean ADC values of benign lesions and malignant tumors were (1.84±0.47)×10 -3 mm2/s and (1.12±0.21)×10 -3 mm2/s,respectively.Malignant tumors had lower ADCs than benign lesions (t=-8.038,P <0.000).The areas under the ROC curves of ADC diagnosing benign and malignant lesions of the tongue was 0.957±0.022.The optimal cutoff values of ADC for differentiating benign and malignant lesions of the tongue was 1.30 × 10 -3 mm2/s with sensitivity of 90.7%,specificity of 93.8% and accuracy of 92%.And ADC had a high consistency compared with pathological results (Kappa values were 0.813).Conclusion Different features between benign and malignant lesions of the tongue are able to be identified with DWI,which can be applied as a complementary tool in the detection of benign and malignant lesions of the tongue.
7.Applied anatomy of anterior and dorsal root nerve tracts in the improved high selective dorsal rhizotomy

BACKGROUND: Anterior and dorsal root nerve tracts should be separated to small tracts in high selective dorsal rhizotomy, because detailed tract separation will benefit electrostimulation, thereby helping correctly cutting the lower-threshold Ia nerve fibers that cause convulsion, and meanwhile sensory nerve fibers in dorsal nerve root can be reserved as many as possible.OBJECTIVE: To meet the needs of limited and high selective spinal dorsal rhizotomy, anterior and dorsal root of spinal nerve were microanatomized to be certain of the separation standard and the number of small nerve tracts, so as to provide reliable basis and novel operative standard for clinical operation.DESIGN: Single sample experiment with adult corpses as subjects.SETTING: Orthopedic Department of the First Affiliated Hospital and Department of Anatomy, Jinzhou Medical College.PARTICIPANTS: This study was carried out at the Anatomical Laboratory of Jinzhou Medical College in December 1999. Fifteen adult corpses, 11 males and 4 males, were donated, and the donators signed informed consent when alive.tained from the 15 adult spinal cords (30 sides) and subjected to morphoanterior and dorsal roots of L5 spinal cord were obtained from a fresh corpse for immunohistochemical staining. The starting part, middle part and the exterior of intervertebral foremen was cut into slices, and the total number of nerve fibers, the number of Ia nerve fibers responsible for convulsion, and their percentage in the total fibers were counted. Meanwhile the distribution and the number of Ia nerve fibers in the three parts were compared.ber of nerve fibers per 100 μm2, the percentage of Ia nerve fibers in the total nerve fibers at the starting part, middle part and exterior of intervertebral foremen of spinal nerve dorsal root.root filaments. Microsurgical observation proved that dorsal root could be divided into 10-18 small tracts and anterior root 6-11 tracts; the diameter number of nerve fibers in the three parts of spinal nerve dorsal root was (3 243±143) fibers per 100 μm2, including (1 702±85) Ia nerve fibers that constituted about 52.5% of the total nerve fibers. Ia nerve fibers were found to be evenly distributed in dorsal root and no gathering could be observed.CONCLUSION: The tracts in anterior and dorsal roots of spinal cord should be separated as minutely as possible during improved dorsal rhizotomy, which is beneficial for cutting off Ia nerve fibers correctly. Generally,the anterior root consists of 6-11 small tracts and dorsal roots of 10-18small tracts, and nerve tracts should not be cut off over 1/2 of total dorsal nerve fibers.
8.Effects of ZHX3 gene silence on the expression of osteoblast-related factors in BMSCs

Objective To investigate the effects of zinc fingers and homeoboxes 3 (ZHX3) silence on expressions of smad3, smad4 and RUNX2 in bone marrow mesenchymal stem cells (BMSCs). Methods ZHX3 low expression vector (ZHX3 silent group) was constructed and was transfected to rat BMSCs. Empty vector was transfected into BMSCs and was used as vehicle control group, and wild type BMSCs was used as the control group. The cell transfection rate was measured under a fluorescence microscope, and then the successful transfection was identified. The immunocytochemistry and immu?noblotting methods were used to detect the expression levels of smad3, smad4 and RUNX2. Results (1) Cells with BMSCs phenotype can be obtained by recovery culturing. (2) After transfection, the green fluorescent protein was found in ZHX3 si?lence group and vehicle control group. Blank control group showed no significant fluorescence. The expression level of ZHX 3 was significantly lower in ZHX3 silence group than that of vehicle control group. (3) Results of immunofluorescence asssay showed that the positive expressions of smad3 and smad4 were located in nucleus and cytoplasm, the positive expression of RUNX2 was mainly located in nucleus. Positive cells were observed in three groups. There was no significant difference in fluorescence intensity between the control group and the vehicle control group, but the fluorescence intensity was significant?ly lower in ZHX3 gene silence group than that of two control groups. (4) There were no significant differences in expressions of smad3, smad4 and RUNX2 betweem control group and the vehicle control group, but they were significantly higher than those of ZHX3 silence group(P < 0.05). Conclusion ZHX3 gene silence can delay vitro osteogenesis of BMSCs, which may play a role by the down-regulated expression levels of smad3, smad4 and RUNX2.
9. Effect of hyperoxia on NRP-cGMP signaling pathway in brain tissue of newborn rats

Objective: To construct the brain tissue damage models of newborn rats induced by hyperoxia, and to explore the effect of hyperoxia on the natriuretic peptide receptor-cyclic guanosine monophosphate (NRP-cGMP) signaling pathway in brain tissue of the newborn rats. Methods: A total of 60 newly born Wistar rats were divided into control group and hyperoxia model group, and there were 30 rats in each group. At the 1st, 3rd and 7th days after hyperoxia exposure, the brain tissues of 10 rats randomly selected from each group were gotten, and the body weights of the rats in two groups were detected. HE staining was used to observe the morphology of brain tissue of the rats in two groups; the ultrastructures of brain tissue of the rats in two groups were observed under transmission electron microscope. Western blotting method was used to detect the expression levels of natriuretic peptide receptor A (NPR-A) and natriuretic peptide receptor B (NPR-B) in brain tissue of the rats in two groups. ELISA method was used to determine the levels of cGMP in brain tissue of the rats in two groups. Results: The body weight of the rats in hyperoxia model group was significantly lower than that in control group at the 1st, 3rd and 7th days after hyperoxia exposure (P<0. 05). The HE staining results showed that at the 3rd day after hyperoxia exposure, the volume of the hippocampal pyramidal cells of the rats in hyperoxia model group was reduced, and the arrangement was sparse; at the 7th day after hyperoxia exposure, the hippocampal cells showed deep staining and shrinkage, and the cell boundaries were not clear. Under electron microscope, at the 3rd day after hyperoxia exposure, the mitochondrial double membrane structure of the rats in hyperoxia model group was destroyed, a few crest disappeared, and the number of mitochondria and synapses was decreased; the above damage conditions were aggravated at the 7th day after birth. The Western blotting results showed that at the 1st, 3rd and 7th days after hyperoxia exposure, the expression level of NPR-A in brain tissue of the rats in hyperoxia model group was higher than that in control group (P<0. 05). But only at the 1st day after hyperoxia exposure, the expression level of NPR-B in brain tissue of the rats in hyperoxia model group was higher than that in control group (P<0. 05). 10.Value of color Doppler echocardiography in diagnosing anomalous origin of coronary artery

Objective To explore the diagnostic value of color Doppler echocardiography in the patients with anomalous origin of coronary artery.Methods Six patients were studied using color Doppler echocardiography.The origin sites,running courses,blood flow directions of coronary arteries, other accompanied cardiovascular abnormalities and valvular regurgitations were investigated.The diameters of coronary artery trunks and left ventricular ejection fraction (LVEF) were measured.And the results were compared with those of coronary angiography and surgery.Results Left coronary artery(LCA) originated from pulmonary artery(PA) in 4 cases,left anterior descending(LAD) branch originated from PA in 1 case,and accessory coronary artery originated from PA in 1 case.The main accompanied cardiovascular abnormalities included ventricular septum defect,atrial septum defect,complete endocardial cushion defect,and so on.For all the patients,the diagnostic results by echocardiography were consistent with catheterization and surgery.High correlations existed between echocardiographic and catheterization measurements for left and right coronary artery diameter and LVEF (r= 0.94 , 0.96 , 0.89 ,respectively,all P 0.05 ).Conclusions Color Doppler echocardiography can accurately diagnose the patients with anomalous origin of coronary artery, provide reliable information for surgical treatment.
