1.Effects of Rutosids on Apoptosis of Retinal Capillary in Rats with Different Courses of Diabetes

Objective: To research the effects of Rutosids on apoptosis of retinal capillary in rats with different courses of diabetes.Methods: Diabetes models were prepared by injecting streptozotocin 65mg/kg via rats' abdominal cavity.Diabetes rat models were randomly divided into four groups in terms of diabetes model group(DMG),Rutosids intervention group(RIG),Rutosids therapy group 1(RTG1) and Rutosids therapy group 2(RTG2).Eyeballs were selected separately,retinals digesting stretched preparation capillary samples were prepared,and apoptosis cells were specificity marked by PAS dyeing and ribonucleotide terminal transferase mediating dUTP nick-translation(TUNEL method),to observe below light microscope.Results: Compared with the homeochronous DMG,the apoptosis of retinal pericapillary cells in RIG was not obvious;perithelial cell apoptosis of RTG1 was relatively common,a few endothelial cells were apoptosis;endotheliocyte apoptosis of RTG2 was increased after treating for 3 months,but obviously lighted than 12 months model group.Conclusion: Rutosids has the preferable inhibitory action on perithelial cell apoptosis of early metaphase retinal microangiopathy in diabetes rats.
2.Effects of Luotong on the Morphological Changes of Retinal Microvessels in the Streptozotocin-induced Diabetic Rats

Objective To study the protective and therapeutic effect of Luotong on the retinal microvessel kinetic morphological changes during various stages of streptozotocin-induced diabetic rats. Methods The diabetic rat model was setup by a single intraperitoneal injection 65 mg/kg of streptozotocin. Diabetic model rats were randomly divided into diabetic model group (6, 9, 12 months subgroups), diabetic group protected with Luotong (6 months) from the onset of diabetic model (LT group)and diabetic group treated with Luotong (3, 6 months subgroups ) 6 months after the onset of diabetic model (LT1, 2 group). The eyeballs were removed at 6, 9 and 12 months, respectively. The whole mounts of retinal digestion were prepared and stained with PAS staining method. The light microscope and transmission electron microscope morphological observations were performed and the basilar membrane thickness of retinal capillaries were quantitatively analysed by LEICA-Q550IW computer image system. Results At 6 months, the retinal microvessel morphological changes of LT group were less severe than those of 6 month model group. At 9 months of the diabetic model rat, the retinal microvessel morphological changes were prominent. 3 months after Luotong therapy, the retinal microvessel changes were partly improved in comparison with 9 month model group but still aggravated gradually. At 12 months, the retinal microvessel morphological changes of diabetic model rats were very severe. 6 months after Luotong therapy, the retinal microvessel changes were significant mitigated compared with 12 month model group. 3.Identification of liposoluble constituents in Yushu tablets by UPLC-ESI-IT-TOF/MS.

In order to identify the chemical constituents of Yushu tablets comprehensively, we studied the chemical constituents of CHCl3 extract from Yushu tablets by the ultra performance liquid chromatography-electrospray ionization-ion trap-time of flight mass spectrometry (UPLC-ESI-IT-TOF/MS). It showed that there were more than 100 compounds separated, and forty-nine peaks among these were identified on the basis of high resolution mass spectrometry data and literature data reported. Determination of twelve peaks was further confirmed by standard substances. These components assigned to the different plant sources mainly included phenylpropanoids, triterpenoids, quinones and m-trihydroxybenzene compounds. By analyzing the chemical components of CHCl3 extract from compound Chinese medicine Yushu tablets comprehensively, this study provided the foundation for studying chemical components, pharmacodynamic substance and quality control of Yushu tablets.

Jaundice is frequent clinic symptoms with high incidence especially among the critical patients. The sepsis complicated with jaundice has drawn more and more consideration. However, the pathogenesis about sepsis complicated with jaundice is not confirmed and it is sometimes identified incorrectly as cholestatic jaundice. This article summarises the pathogenesis and elucidate the management about sepsis complicated with jaundice.
7.Diagnostic value of pentraxins-3 and triglyceride/high-density lipoprotein cholesterol in acute coronary syndrome

Objective To investigate the value of serum pentraxins-3 (PTX-3) together with triglyceride (TG)/high-density lipoprotein cholesterol (HDL-C) as predictor risk factors of future acute coronary syndrome(ACS). Methods One hundred and seventy-one cases of selective coronary angiography from July 2008 to December 2009 were collected and analyzed. According to the situation on admission and coronary angiography, patients were divided into three groups: normal control group (25 cases), stable angina pectoris (SAP) group (23 cases) and ACS group (123 cases). Fasting venous blood was extracted for measuring serum PTX-3 and lipids (TG, HDL-C) in the next morning, and the statistical significanc was analyzed. Results The level of serum PTX-3 and the ratio of TG to HDL-C in ACS group [(6.39 ± 3.01)μ g/L, 2.38 ± 2.00] were significantly higher than those in SAP group[(3.87 ± 2.05 ) μ g/L, 1.70 ± 1.01] and normal control group [(2.90 ± 1.94)μg/L,0.95 ±0.35] (P <0.01 or <0.05). Conclusion Increased serum PTX-3 levels and ratio of TG to HDL-C in patients are closely related with ACS, both of which increase the accuracy of early diagnosis of ACS.

Purpose:To study the expression of COX 2,P53 and PCNA in esophageal carcinomas and its significance. Methods:Immunohistochemical method was used to examine the sections from 82 esophageal squamous cell carcinomas, 20 esophagitis and 16 normal esophageal mucosa tissues. Results:The positive ratios of COX 2,p53 and PCNA were 87.8%(72/82),82.9%(68/82)and 95.1%(78/82) in 82 esophageal carcinomas, respectively. But there was no expression in adjacent noncancerous tissues and normal esophageal mucosa tissues. 9.Study on the Anti-influenza A Virus H1N1 Effect of Aqueous Extraction of Lonicerae Flos in vitro

OBJECTIVE:To investigate the effect of aqueous extraction of Lonicerae Flos (SYHW) on anti-influenza A virus H1N1 (H1N1 virus) in vitro. METHODS:Using Madin-Darby canine kid ney (MDCK) cells cultured in vitro by H1N1 virus, half of the tissue culture infection dose(TCID50)was calculated. Culturing MDCK cells for 24 h with different mass concentrations of SYHW,the maximum non-toxic concentration was investigated. And then test was divided into normal cell group,virus control group,SYHW preventive administration group,therapeutic administration group and direct killing group (given SYHW of maxi-mum non-toxic concentration,infecting cells by 100 TCID50 H1N1 virus),and antiviral effective rate (ER) of SYHW was deter-mined. Test was divided into normal cell group,virus control group,SYHW therapeutic group and direct killing group (the same administration and infection as above),changes of cell proliferation index (PI) and cell apoptosis rate were respectively deter-mined. RESULTS:100 TCID50 of H1N1 virus was 1.26×10-7,and the maximum non-toxic concentration of SYHW on MDCK cells was 50 μg/mL(cell survival rate was 91.3%). ERs of preventive administration group,therapeutic administration group and direct killing group were 0,80.3% and 52.7%,respectively. Compared with normal cell group,PI value in virus control group was sig-nificantly reduced (P<0.05),early and late apoptotic rates were significantly increased (P<0.05). Compared with virus control group,PI value in directly killing group was significantly increased(P<0.05),and early apoptotic rate was significantly reduced (P<0.05);early apoptotic rates in therapeutic administration group were significantly reduced (P<0.05). CONCLUSIONS:SY-HW shows anti-H1N1 virus effect in vitro,therapeutic administration and directly killing are preferred in antiviral effect.

Objective To explore the association between X-ray repair cross complementing group 1 (XRCC1)gene rs25487 locus polymorphisms and nonobstructive azoospermia in Hui minority ethic population of Shaanxi Province.Methods We used polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP)method for genotyping at XRCC1 gene rs25487 locus in 79 patients with nonobstructive azoospermia and 82 healthy male controls in Hui minority ethic population of Shaanxi Province.Then we analyzed the association be-tween XRCC1 gene rs25487 locus and nonobstructive azoospermia.Results Compared with GG genotype,GA, AA and GA + AA genotypes demonstrated a significantly increased risk for nonobstructive azoospermia (OR =2.286,95% CI 1.1 5 1-4.539;OR =2.202,95% CI 0.753-6.439;OR =2.271,95% CI 1.1 71-4.403),respec-tively.Meanwhile,the A allele frequency was significantly higher in azoospermic patients than in controls (OR =1.582,95% CI 1.005-2.492,P =0.047).Conclusion G→A in XRCC1 gene rs25487 locus is correlated with nonobstructive azoospermia in Hui minority ethic population of Shaanxi province.
