AIM:To study of effects of Panax notoginsengnoside(PnGL) on neutrophil(Neu) functions in carrageenin(Car) induced subcutaneous air-vesicle synovitis in rats and its mechanisms METHODS:Rat air-vesicle synovitis model was established with sc Car(25mg/kg) Neu count, the phagocytosis of Neu,the realease of O2 ?- from Neu and the level of Neu-i were measured with test tube diluting method, staphylococcus aureus phagocytosis method, cytochrome C reduction method and Fura-2 fluorescence technique respectively RESULTS:At 12h after Car used,Neu count of exudates elevated, phagocytosis rate and index of Neu and the release of O2 ?- from Neu increased obviously, suggesting that Neu was in an activated state Mea_nwhile,the level of Neu-i elevated significantly PnGL 60~240mg/kg ip,30min before and 6h after Car sc,reduced Neu count of exudates, inhibited phagocytosis functions and the increase of i level of Neu in a dose-dependent manner There was a significant correlation between the reduction of Neu-i and the inhibition of Neu phagocytosis functions and the release of O2 ?- from Neu Dexamethason had similar effects on Neu functions to PnGL,but with no effect on the level of Neu-i CONCLUSION:The reduction of Neu-i level and the subsequent inhibition of Neu functions is one of the important anti-inflammatory mechanisms of PnGL,which is different from that of Dex
3.Comparison of STR polymorphism among a Kirgiz ethnic group from Sinkiang and other groups
Journal of Pharmaceutical Analysis 2007;19(1):97-100
Objective To study the genetic relationship between Kirgiz individuals living in Sinkiang China and analyze the difference among Kirgiz and the other population with STR polymorphisms. Methods PCR amplification was performed using PE9700, the PCR products were typed by automated sequencer and genescan. Results A database of nine STR loci of Kirgiz was established. It shows there are at least 73 STR alleles and 191 genotypes in Kirgiz. Genotype frequencies distribution showed no deviation from Hardy-Weinberg equilibrium by χ2-test. Kirgiz was compared with the other Chinese ethnic groups, then the American Black and the White. Conclusion These results suggested that the nine STR loci and Amelogenin locus were very useful in human identification, biological archaeology and gene resource studies.

Effects of saponin of Panax pseudo-ginseng var. notoginseng (Burk.) Hoo et Tseng(PnS) on neutrophil (Neu) count, protein and malondialdehyde (MDA) contents in the exudate of rat a-cute air pouch synovitis model induced by sc injection of 25 mg/kg carrageenan were investigated by Lowryand test tube dilution methods and thiobarbituric acid spectrophotometry. Its effects on the intracellularcAMP content in and release of O2 from Neu were further assessed by radioimmunoassay and cytochromeC reduction methods respectively. The results showed that PnS 60,120,240 mg/kg ip reduced the migra-tion of Neu and exudation of protein, lowered the content of MDA and inhibited the release of O2- fromNeu; but elevated the cAMP content of Neu in a dose-dependent manner. These results revealed that Pnshas an obvious anti-inflammatory effect and its primary action was related to the increase of cAMP contentin Neu, followed by inhibition of the free radical production.

Objective To investigate molecular biological basis associated with inflammatory and immune response in Qi deficency and blood stasis syndrome. Methods Twelve rats were randomly divided into model group and control group. Rat Genome 230 2.0 Array Gene Chip was used for examination of gene expression profiles in rats with Qi deficiency and blood stasis syndrome and normal rats. Differental expression genes associated with inflammatory and immune mechanism were analyzed. Results Compared with the control group, there were different expression genes in model group, the up-regulated were 831 genes, known functional genes 563, and down-regulated 782, known functional genes 544. The expression level increased in 50 genes and decreased in 17 genes associated with inflammatory and immune response. Conclusion Differental expression genes associated with inflammatory and immune response revealed that there was close correlation between Qi deficency and blood stasis syndrome and abnormal inflammatory and immune response.

Journal of Third Military Medical University 2003;0(08):-
Objective To investigate the effects of clonidine on the expression of myocardial ? subunit of inhibitory G protein (Gi?) in severely scalded rats and the mechanisms. Methods A scald model of 30% total body surface area Ⅲdegree burn on the back of rats was established. The basal and Gpp (NH) p, forsklin stimulated activity of adenylate cyclase (AC), and Gi? in cardiac myocytes were determined by indirect enzyme radiochemical assay and Western blotting, respectively. Results Clonidine at the dosage of 1-3 mg?kg -1 inhibited significantly the increase in the myocardial Gi? levels. Clonidine at the dosage of 0.3-3 mg?kg -1 partially reversed the decrease in the basal activity, and clonidine at the dosage of 1-3 mg?kg -1 showed the similar effect on the decrease in the Gpp (NH) p stimulated activity. Efaroxan, an I 1 imidazoline receptor antagonist, could partially reverse the inhibitory effect of clonidine on myocardial Gi? levels. Conclusion Clonidine can inhibit the elevation of myocardial Gi? levels, and the mechanisms of the inhibition may possibly be associated with the stimulatory effect of clonidine on I 1 imidazoline receptors.

Chinese Journal of Tissue Engineering Research 2015;(18):2917-2922
BACKGROUND:Breviscapine has been shown to impact the reproductive capacity in rats with type 2 diabetes mel itus, but few reports concerned its mechanism of action.
OBJECTIVE:To study the effects of breviscapine on proliferating cel nuclear antigen and proto-oncogene c-fos expression in testis of type 2 diabetes mel itus rats.
METHODS:Total y 36 healthy male rats were randomly divided into control group, model group and breviscapine group with 12 rats in each group. In the model group and breviscapine group, rat models of type 2 diabetes mel itus were established by continuous intraperitoneal injection of streptozotocin. Blood glucose reaching 16.7 mmol/L in rats was considered as the standard of model induction. In the control group, rats were given an equal volume of citrate buffer solution by single intraperitoneal injection. In the breviscapine group, rats were administered breviscapine 10 mg/(kg?d) for 4 consecutive weeks by intraperitoneal injection. Rats in the other two groups were injected with an equal volume of physiological saline at the same time point.
RESULTS AND CONCLUSION:After 4 weeks of intervention, serum testosterone testing, immunohistochemistry and PCR results showed that serum testosterone levels, proliferating cel nuclear antigen, c-Fos protein and mRNA expression:control group>breviscapine group>model group (P<0.05);blood glucose concentration:the control group
