1.Protective effect of edaravone against renal ischemia/reperfusion injury and compared with ischemic postconditioning in rats.

The aim of this study is to clarify whether edaravone postconditioning had protective effect against renal ischemia/reperfusion injury and to compare the protective effect between ischemic postconditioning and edaravone postconditioning. Rats were subjected to 45 min ischemia followed by 24 h reperfusion. The rats were randomly assigned to seven groups: a sham-operated control group, an ischemia/reperfusion group, an ischemic postconditioning group, a normal saline vehicle postconditioning group and an edaravone postconditioning (1, 3, and 6 mg x kg(-1)) group. Renal function was assessed by serum creatinine and BUN concentration, while histological damage of renal tissue was assessed with HE staining. MDA content and SOD activity of renal tissue were determined. TUNEL staining was performed to analyze the apoptosis of the tubular epithelial cells, the protein level of Bcl-2 and Bax in renal tissue was examined by Western blotting. Compared to the ischemia/reperfusion group, edaravone postconditioning significantly decreased serum creatinine and BUN concentration, and ameliorated histological damage of renal tissue. MDA was less after 24 h reperfusion in the edaravone postconditioning group than that in the ischemia/reperfusion group, consistent with an increase in SOD activity. In addition, edaravone postconditioning decreased TUNEL-positive cells and Bax expression, and increased Bcl-2 expression. Results detected in the edaravone postconditioning group showed no significant difference from the ischemic postconditioning group. Edaravone administered during the last 3 min of ischemia, prior to reperfusion induces a pharmacological postconditioning in vivo against renal ischemia/reperfusion injury in rats. This protection is similar to that observed with ischemic postconditioning.

Aim To investigate the effects of ginaton on tubular cell apoptosis in a rat model of renal ischemia/reperfusion(I/R) injury.Methods The model of bilateral post-ischemic renal injury was established by clamping renal pedicles for 45 minutes.To explore the effects of ginaton on activation and expression of JNK and c-Jun,p-JNK1/2,JNK,p-c-Jun,c-Jun were examined by Western blotting.Apoptosis of renal tubular epithelial cells was assessed by terminal deoxynucleotidyl transferase mediated dUTPnick end labeling(TUNEL) and flow cytometric analysis.Results Phosphorylation levels of the JNK and the c-Jun were significantly increased at 20 min after renal I/R injury.This corresponds to a significant increase of apoptotic tubular cells induced by renal I/R injury.Ginaton attenuated the increased phosphorylation of p-JUK at 20 min(vs I/R 20 min,P

Objective To observe the influences of Yangxinshi pill on ET, NO, CRP in patients with angina pectoris belonging to manifestation of Qi-deficiency and blood stagnation. Methods 78 patients with angina pectoris were randomly divided into two groups. 40 patients in treatment group were treated with Yangxinshi pill, 4 pills once, three times a day 8 weeks. 38 patients in control group were treated with isosorbide mononitrate 8 weeks. The symptoms and the ET, NO, CRP level before and after treatment were recorded. Results Obvious drop of ET, NO, CRP were observed after treatment in the treatment group. The level of ET was reduced from (67.47?15.72)pg/mL to (37.97?7.82) pg/mL. The level of NO was increased from (78.24?13.81)?mol/mL to (96.25?12.72)?mol/mL. The level of CRP was reduced from (5.31? 2.02)mg/L to (3.42?2.17) mg/L. There was a significant difference between the two drugs (P 0.05). The total efficacy rate in improving symptom was 95%. 5.Clinical observation of coronary artery bypass grafts in the same period of heart valve replacement on the treatment of coronary heart disease with heart valve disease

Objective To analyze and discuss the value and significance of coronary artery bypass grafts in the same period of heart valve replacement in the process of the coronary heart disease with heart valve disease treatment, in order to improve the effect of treatment and reduce the incidence of adverse events. Methods 60 cases with coronary heart disease with heart valve disease , which admitted in our hospital during February 2013-February 2015, were selected as the research objectaccording to random number table method and divided into the-experimental group and the control group , each group of 30 cases according to the principle of double blind control. The experimental group was given the coronary artery bypass grafts in the same period of heart valve replacement and the control group was given the coronary artery bypass grafts without heart valve replacement. The curative ef-fects of different schemes and effects on cardiac function after surgery of the two groups were compared. Results After statistical analysis found thatthe observation indexes of the control group was significantly increased compared with the experimental group, the difference was significant statistically significant (P<0.05). The postoperativecar-diac function indexes of the experimental group was significantly improved then control group , the difference was significant statistically significant (P<0.05). Conclusion Coronary heart disease with heart valve disease in the process of treatment can also take heart valve surgery combined coronary artery bypass surgery. By preoperative and postoperative symptomatic treatment, intraoperative protect myocardium, can effectively prevent and reduce the op-eration of various complications, which is worthy of clinical popularization and application.
6.The study of serum retinol-binding protein 4 and related factors in chronic hepatitis C

Objective To investigate the level of serum retinol-binding protein 4 (RBP4) and related factors in chronic hepatitis C (CHC). Methods Fifty-six patients with CHC (CHC group) and 35 healthy volunteers (control group) were selected. Serum RBP4 level was measured by ELISA method.Fasting blood glucose ( FBG ), triglycerides (TG), total cholesterol ( TC ), alanine aminotransferase (ALT),γ-glutamyl transpeptidase (γ-GT) were measured, HCV-RNA level was tested by qualitative polymerase chain reaction(q-PCR). Results There were no significant difference in FBG, TC,TG, γ-GT between two groups (P > 0.05 ). Serum RBP4 level in CHC group [(33.38 ± 6.43 ) mg/L] was higher than that in control group [(26.11 ± 3.35) mg/L](P< 0.01),the CHC patients with ALT normal (26 cases) had significantly higher RBP4 level [( 38.96 ± 4.09) mg/L] compared with ALT abnormal [30 cases, ( 28.53 ± 3.43 ) mg/L](P < 0.01 ). ALT level was negative with RBP4 in CHC group (r = -0.6368, P < 0.05 ). Conclusion Serum RBP4 level is significantly associated with CHC and negatively correlated with ALT level,but not associated with FBG, TC,TG, γ-GT and HCV-RNA.
7.The effects of diammine silver fluoride solution on demineralized dentine adhesion ability

Objective:To investigate the effect of silver diamine fluoride(SDF)on the bonding strength between dentine and glass ion-omer cement(GIC).Methods:1 2 extracted sound molars were prepared into dintine samples and distributed into sound dentine group and demineralized dentine group.According to the treatment methods,the samples in each group were respectively divided into 3 sub-groups:A(control group),B[coated with 38% Ag(NH3 )F2 ]and C(SDF treatment with additional lighting-curing)(n =20).Then a hand-mixed conventional glass ionomer cement Fuji IX was placed on the dentine surface.After 24 h,micro tensile bond strength test and scanning electron microscope (SEM)analysis were conducted.Results:The bonding strength of demineralized dentine was higher than that of sound dentine(P <0.01 ).SDF with additional lighting-curing treated dentine showed a higer bonding strength value than only SDF treated dentin(P <0.01 ).Conclusion:SDF may improve the bonding between dentine and GIC.
8.Mechanical strength and in vitro degradation of a silk scaffold for tissue-engineered ligaments

BACKGROUND: Presently, the biomaterial used in ligament tissue engineering such as collagen protein, polylactic acid, polyglycolic acid, small intestinal submucosa, glycan and nanomaterial are characterized by rapid degradation, resulting in inflammatory reaction after applying in host.OBJECTIVE: To investigate mechanical strength and in vitro degradation of silk scaffold and explore the reaction to macrophages.DESIGN: Controlled experiment.SETTING: Experiments were performed at the Department of Orthopaedics, Union Hospital, Huazhong University of Science and Technology from September 2004 to January 2005.MATERIALS: White raw Bombyx mori silkworm fibers of size 20/22 (according to the manufacturer) were obtained from the market. Bundles of 30 parallel fibers were prepared for a bundle of scaffold, which was put into fervens 5g/L Na2CO3for degumming. Ratio of Na2CO3 solution (Ml) to raw silk (g) was 1000.METHODS: In vitro degradation: 8cm long silk scaffold was weighed after drying. Subsequently, the silk scaffold was separately dipped into phosphate buffer saline (PBS) and 1.0g/L collagenase prepared with PBS. Twelve weeks later, silk scaffold was weighed to calculate weight loss rate. Simultaneously, tensile test was performed to detect the ultimate tensile strength (UTS) of samples. Culture of monocyte strain RAW264.7:2×108L-1 macrophage suspension (1mL) were separately added in a silk scaffold group, a control group and a lipopolysaccharide (LPS) group. At days 1 and 7, cell supernatant was collected from each group. Tumor necrosis factor-α(TNF-α) levels were measured by enzyme linked immunosorbent assay (ELISA).MAIN OUTCOME MEASURES: ① Changes in weight loss rate and UTS of the silk matrices after incubated with collagenase and the PBS. ②TNF-αlevels in the supernatant of each groups at days 1 and 7.RESULTS: Mass of silk matrices reduced by over 50% after incubated with collagenase for 8 weeks, but no change was found in PBS. UTS decreased by over 50% 8 weeks after incubated with collagenase, but no change was detected in PBS. At days 1 and 7, TNF-α levels in the supernatant was less in the silk scaffold group; TNF-α levels in the supernatant was significantly higher in the LPS group than in the silk scaffold group (P<0.01), but no significant difference in TNF-α levels was measured between the silk scaffold group and the control group (P>0.05).CONCLUSION: After 12-weeks degradation, silk scaffold still has good mechanical properties. Macrophages possess immunological inertia at days 1 and 7 after inoculated with macrophages.
9.Early clinical features of severe cases with hand-foot-mouth disease

The clinical features of 120 severe cases with hand-foot-mouth disease (HFMD) were retrospectively analyzed compared with those of the regular cases. Clinical data showed that the age of mostsevere cases ranged from 1-3 years. Sustained high fever ( 55% ) , hypertension ( 40% ) , fewer rash (73.3%) were important clinical features. Lethargy, somnolence or restlessness (70. 8% ), vomiting (30. 8% ) were early signs of central nervous system ( CNS) ; and the CNS involvement in severe cases was characterized by body trembling or muscle spasm (80% ). High white cell count(75. 8% ) and high blood glucose levels(32. 5% ) were important makers to diagnose the severe HFMD at early stage. There were significant differences in presentation of above features between severe cases and regular cases of HFMD ( P < 0. 01 or P < 0. 05 ) . Early treatment with mannitol, gamma globulin and glucocorticoids were effective, no death was reported and no side effects were found in this case series.
10.Screening for Y chromosome azoospermia factor region microdeletions using modified multiplex PCR

Objective To establish an universal primer-multiplex PCR system for diagnosis of Y chromosome AZF region microdeletions in 262 patients with non-obstructive azoospermic and severe oligozoospermic male infertility. Methods In each panel of multiplex PCR, YUP and YCP containing a fragment of non-human DNA sequence at their 5' ends were designed. The universal primers and chimiric primers were employed for the amplification at the same multiplex PCR system to screen for the Y chromosome AZF region ( a, b and c) microdeletions in 262 non-obstructive azoospermic and severe oligozoospermic male infertility patients. Results Thirty-three out of 262 patients (12. 60% ) were detected with Y chromosome AZF microdeletions. Among them, 27 cases were AZF c microdeletions and 6 ones were AZF b + c microdeletions. These results were in agreement with the results from EMQN method. There was no false-positivity. The gel electrophoresis for detection of multiple STS from both methods showed that the sY84,sY86, sY127, sY134, sY254, sY255, SRY bands were homogeneous and clear with similar brightness. Conclusion The modified multiplex PCR is suitable for screening of Y chromosome AZF microdeletions in non-obstructive azoospermic and severe digozoospermic male infertility patients.
