1.Study on Professor Wang Kungen's Therapeutic Principles in Treating Functional Dyspepsia Based on Data Mining

Objective]To study Professor Wang Kungen's therapeutic principles in treating functional dyspepsia by data mining therapy. [Method]First, establish Wang Kungen's medical records database;then do data standardization and data mining.[Result]There are total y 408 cases that meet the demand. Postprandial distress syndrome is the major subtype of senile group while epigastric pain syndrome is the major subtype of non senile group. The major patterns of syndrome of FD in Professor Wang Kungen's medical records are as fol ows: liver qi attacking the stomach, deficiency and stagnation of spleen Qi, disharmony of stomach qi, stagnation of qi and accumulation of dampness, phlegm-heat harassing the inner body. The major prescriptions are as fol ows:ErChen Decoction, ZuoJin Pil s, SiJunZi Decoction, SiNi Powder, LiuJunZi Decoction, XiangSu Decoction, WenDan Decoction. There is close correlation between prescriptions and patterns as fol ows:liver qi attacking the stomach and SiNi Powder, ZuoJin Pil s;stagnation of qi and accumulation of dampness and ErChen Decoction,PingWei powder;phlegm-heat harassing the inner body and WenDan Decoction, ErChen Decoction; There is close correlation between herbs as fol ows: Processed Rhizoma Pinel iae and Tangerine Peel, Processed Rhizoma Pinel iae and Poria cocos; Rhizoma Coptidis and Processed Evodia rutaecarpa; Fructus Aurantii and Processed Mangnolia officinalis; Radix Bupleuri and Radix Paeoniae Alba; Poria cocos and Atractylodes;Tangerine Peel and Caulis Peril ae; Poria cocos and Codonopsis pilosula; Radix Bupleuri and Curcuma aromatica. [Conclusion]The incidence and subtype of FD between female group and male group is different. Professor Wang Kungen's therapeutic principles in treating FD are clearing liver and adjusting stomach, soothing liver-qi stagnation,harmonizing stomach and lowering adverse qi, promoting the circulation of qi and resolving dampness and clearing phlegm heat.
2.Study on Professor Wang Kungen's Therapeutic Principles in Treating Chronic Constipation Based on Data Mining

Objective] To study Professor Wang Kungen's therapeutic principles in treating chronic constipation by data mining therapy. [Method]First, establish Wang Kungen's medical records database; then do data standardization and data mining. [Result]There are totally 207 cases that meet the demand. The major patterns of syndrome of CC in Professor Wang Kungen's medical records are as follows: gastrointestinal heat syndrome, stagnation of liver qi and spleen deficiency, Yin deficiency syndrome, Qi deficiency. The major prescriptions are as follows: Liumo Decoction, Xiao Yao San, Pi Yue Pill, Buzhong Yiqi decoction. There is close correlation between prescriptions and patterns as follows: gastrointestinal heat syndrome and Liumo Decoction; stagnation of liver qi and spleen deficiency and Xiao Yao San; Yin deficiency syndrome and Pi Yue Pill; Qi deficiency and Buzhong Yiqi decoction. There is close correlation between herbs as follows: rhizoma atractylodis macrocephalae and fructus arctii; rhizoma atractylodis macrocephalae and radix paeoniae alba; fructus cannabis and elecampane; rhizoma atractylodis and fuling; peach kernel and cortex moutan; semen raphani and fructus aurantii.[Conclusion] The evidence type of chronic constipation is relatively purging heat and keeping body fluid, soothing the liver and stomach, tonifying spleen and moistening intestines, tonifying Qi.

4.THE UPPER AIRWAY CT SCAN OF OBSTRUCTIVE SLEEP APNEA PATIENTS

Compared with CT scan results of OSAS patients and normal adults to explore the value of CT diagnosis of upper airway stricture in patients with OSAS, 35 patients with OSAS and 35 normal adults were included in the study. 22, 19, 12, 8 cases had obstruction at palate, uvula, lingua and epiglottis region respectively, the awaked CT scan results of 61 obstructive sites were compared with those of normal adults. Obvious difference was found between patients and normal adults in area, left right dimension and coronal dimension. difference was also found in thickness of lateral pharyngeal tissue of palate, uvula and lingua region and retropharyngeal tissue of palate and uvula. The normal extent was determined by unilateral 95% confidence interval in normal adult group. And 43 sites had abnormal CT scan results, the positive ratio was 72 13%. The results sugget that there are obvious differences between the patients with OSAS and normal adults, the CT scan measure can show the upper airway obstructive site of patients with OSAS.
5.Ultrastructural Alterations of Adult Schistosoma haematobium Harbored in Mice Following Artemether Administration

Objective To perform a temporal examination of ultrastructural alterations in adult Schistosoma haematobium due to artemether Methods Eight mice infected with 100-120 S. haematobium cercariae for 81 days were treated intragastrically with 400 mg/kg artemether. At 24 hours, 3, 7 and 14 days post-treatment, groups of 2 mice were sacrificed and schistosomes collected by the perfusion technique. Worm samples were fixed and examined by transmission electron microscopy. Schistosomes were also obtained from 2 untreated mice that served as control.Results Typical ultrastructural alterations included swelling, lysis and vacuolization of the tegumental matrix, and disappearance of basal membrane. In sensory organelles and tubercles, there was extensive or local lysis of internal structure. In the musculature, parenchymal tissues, syncytium and gut epithelial cells, focal or extensive lysis, decrease in granular endoplasmic reticulum, vacuolization and degeneration of mitochondria were observed. These alterations became apparent both in male and female worms 24 hours post-treatment. In female worms, severe damage to the vitelline cells was also observed, resulting in the emergence of vacuoles, a decrease in granular endoplasmic reticulum,fusion of vitelline balls or even collapse of damaged vitelline cells. The most extensive tegumental alterations were observed 3-7 days post-treatment. Whilst 14 days post-treatment ultrastructural damage was still apparent, the tegument of some worms showed similar features to those recovered from untreated control mice. Conclusion Administration of artemether to mice infected with adult S. haematobium results in extensive damage to the ultrastructure in the tegument and subtegument tissues of the worms, confirming previous results with other schistosome species.
6.Effect of Artemether on the Tegument of Adult Schistosoma haematobium Recovered from Mice

Objective To assess the effect of artemether on the tegument of adult Schistosoma haematobium harbored in mice. Methods Ten mice were infected subcutaneously with 100-120 S. haematobium cercariae each. At day 81 post-infection, 8 mice were treated orally with 400 mg/kg artemether. Mice were sacrificed at 1, 3, 7 and 14 days post-treatment, and schistosomes were collected by the perfusion technique, fixed and examined under a scanning electron microscope. Schistosomes obtained from the 2 untreated mice served as a control. Results Twenty-four hours post-treatment, tubercles on the tegument of male worms showed lesions, characterized by enlargement, collapse and partial peeling off from the border with the tegument. In both male and female worms, the tegument showed focal or extensive swelling, fusion, vacuolization, erosion, peeling, and destruction of sensory structures. Three days post-treatment,tegumental alterations further aggravated; particularly severe damage was the swelling or collapse of the oral sucker observed in both sexes. In addition, extensive swelling, erosion and peeling of tegumental ridges and destruction of discoidlike sensory structures were seen in female worms. Seven to 14 days post-treatment, moderate-to-severe damage was still evident in some worms, whereas other worms surviving the treatment showed apparent recovery in most parts of their tegument. Conclusion Artemether causes extensive and severe tegumental damage in adult S. haematobium.
7.Efficacy and safety of bronchoalveolar lavage through flexible fiberoptic bronchoscope in treatment of neonatal atelectasis

Objective To evaluate the clinical efficacy and safety of flexible fiberoptic bronchoscopy (FFB) and bronchoalveolar lavage (BAL) in treatment of neonatal atelectasis.Methods Eligible patients, who were diagnosed as neonatal pulmonary atelectasis and admitted consecutively to the neonatal intensive care unit of the Children's Hospital of Hebei Province from January 2013 to January 2015, were included in the study.They were randomly assigned to FFB group (n=30) and control group (n=28).Newborns in the FFB group received BAL under FFB, while those controls received tracheal irrigation after intubation.The duration of lung recruitment, oxygen exposure and antibiotic administration, hospital stay, culture results of respiratory secretions, prognosis and total expenses during hospitalization were compared between the two groups.Complications of FFB were also recorded.Chi-square test and t-test were performed for statistical analysis.Results (1) In the FFB group, atelectasis occurred in the upper fight lobe (n=26), upper lobes of both sides (n=1), lower right lobe (n=2) and lower left lobe (n=1), while in the control group, atelectasis occurred in the upper right lobe (n=26), lower left lobe (n=1) and middle right lobe (n=2) (x2=0.094, P > 0.05).(2) The positive rate of bacteria culture results showed no difference between bronchoalveolar lavage fluid in FFB group and tracheal secretions in the controls [43%(13/30) vs 32%(9/28), x2=0.770, P > 0.05].(3) The duration of lung recruitment, antibiotic administration and hospital stay of the FFB group were all shorter than those of the control group [(4.7±3.4) vs (7.4±6.6) d, (14.0±4.5) vs (20.3±10.9) d, (15.1±4.7) vs (21.8±12.3) d, t=-5.718, 8.604 and 7.733, all P < 0.05].(4) Among babies in the FFB group, nine experienced fever and returned to normal after physical cooling;three showed more shadow in chest X-ray with aggravated dyspnea during a short period, and relieved 12 hours later;two had minimal hemorrhage from tracheal mucous membrane;one showed crying hoarse.Serious complications, such as pneumothorax, massive bleeding or cardiac arrest, did not occurred.No death or refuse of treatment was reported.Conclusion FFB and BAL is much more effective than tracheal irrigation after intubation in treatment of neonatal atelectasis without any severe complications.
8.Construction and expression of recombinant plasmid pCD-rbFGF in osteoblasts.

To construct basic fibroblast growth factor (bFGF) eukaryotic expression vector and to evaluate the possibility of bFGF gene therapy in orthopedic disease, the pCD-rbFGF recombinant plasmid was constructed by cloning rat basic fibroblast growth factor (bFGF) cDNA into an eukaryotic expression vector, pcDNA3. Rat osteoblasts were transfected with pCD-rbFGF plasmid by lopofectin mediated gene transfer, the transient expression was detected by streptavidin-biotin-enzyme complex (SABC) method. It was observed that the expression of rat bFGF gene was detected 72 h after transfected distinctly. Basic fibroblast growth factor gene therapy is a method of potential for a wide array of orthopedic diseases.

Rabbit osteoblasts were transfected with pCD-hVEGF165 plasmid by lipofectin mediated gene transfer. The transient expressive results were detected by immunohistochemical method. It was observed that the expression of human VEGF gene was detected 72 h after transfecting distinctly.
