1.The expression of HO-1,RvD1 and LXA4 in cases with ulcerative colitis

Objective In this study,the change of HO-1,RvD1,LXA4 and IL-6 in the patient with ulcerative colitis (UC)was investigated.Effects on the pathogenesis of ulcerative colitis were dis-cussed.Methods The distribution of HO-1 proteins in the colonic tissues in 60 cases of UC and 30 cases of normal control group were detected by SABC immunohistochemistry and perfusion catheter manometer. The slides were then analyzed with an Image Analyzing system to obtain the density of stained proteins.The levels of RvD1,LXA4 and IL-6 in serum by using enzyme linked immunosorbent assay(ELISA).Results The expression of HO-1 in patients with active UC was higher than that in normal group and remission of UC.The levels of RvD1,LXA4 and IL-6 in patients with active UC were significantly higher than that in normal group and remission of UC.The levels of HO-1,RvD1,LXA4 in patients with active UC were posi-tively correlated with IL-6.The levels of HO-1,RvD1,LXA4 in patients with normal group and remission of UC had no relevance with IL-6.Conclusions The expression of HO-1,RvD and LXA4 in activity of the UC may plays an important role in the pathogenesis of gastrointestinal motility disorders such as UC.

Objective To investigate the expressions of endogenous cystathionine-γ-1yase (CSE)/ hydrogen sulfide (H2S) and resolvin E1 (RvE1) in patients with ulcerative colitis (UC),and its effect on the pathogenesis of ulcerative colitis.Methods The distribution and expression of CSE proteins in the rectum mucosa in 60 cases of UC and 30 cases of normal control group were detected by Strept Avidin-Biotin Complex (SABC) immunohistochemistry.The average optical density value of CSE was analyzed with an Image Analyzing systems.The expression of CSE mRNA in the rectum mucosa was detected by real-time polymerase chain reaction (RT-PCR).The levels of H2S and RvE1 in sera were detected by spectrophotometry.Results The expressions of CSE proteins in three groups were detected in the rectum mucosa membrane epithelia.The average optical density value of CSE and the expression of CSE mRNA in patients with active UC were higher than that in normal group and remission of UC.The levels of H2S and RyE1 in patients with active UC were significantly higher than that in normal group and remission of UC.Conclusions The abnormal expressions of CSE/H2S and RyE1 in activity of the UC might play an important role in the pathogenesis of UC.
3.Effect of RNA interference of STAT1 expression on radiosensitivity and cell cycle of esophageal carcinoma cell line Eca109

Objective To inhibit the gene expression of signal transducer and activator of transcription factor 1 (STAT1) in human esophageal squamous cell carcinoma cell line Eca109 by RNA interference and investigate its effect on the radiosensitivity and cell cycle of Eca109 cells.Methods Interference vector pSTAT1-shRNA for STAT1 gene was designed and constructed.After being mixed with lentiviral packaging plasmids,the interference vectors were used to transfected 293T cells.Virus solution was collected to infect ECA109 cells.Real-time PCR and Western blot were used to measure the mRNA and protein expression levels of STAT1 in Eca109 cells.Colony formation assay and flow cytometry were used to evaluate the radiosensitivity and cell cycle distribution of Eca109 cells.Results All Eca109 cells were divided into blank control group,transfection-positive group,and transfection-negative group.The transfection-positive group showed significantly lower mRNA and protein expression levels of STAT1 than the other two groups.The values of D0,SF2,and Dq of transfection-positive group were 2.03 Gy,0.83,and 1.20 Gy,respectively,lower than those of blank control group (2.98 Gy,0.88,and 1.39 Gy) and those of transfection-negative cells (3.02 Gy,0.88,and 1.57 Gy).At 12 h,24 h,and 48 h after 4 Gy-irradiation,the transfection-positive group showed significantly higher percentage of G0 + G1 than the blank control group and transfection-negative group (34.13% vs 22.03% vs 22.27%,F =7.56,P =0.023 ; 43.80% vs 28.40% vs28.63%,F=10.01,P=0.012;53.20% vs42.2% vs41.83%,F=10.73,P=0.010) and significantly lower percentage of G2 + M than the blank control group and transfection-negative group (14.33% vs 32.23% vs 32.23%,F=16.86,P=0.003;27.73% vs 43.53% vs 44.00%,F=26.62,P=0.001;14.23% vs27.97% vs27.93%,F=40.34,P=0.000).Conclusions RNAinterference of STAT1 in Eca109 cells does not affect the proliferation ability of Eca109 cells,and it can increase the radiosensitivity of Eca109 cells probably by regulating cell cycle after irradiation.
4.Vascular endothelial growth factor in improving liver function of rats following partial liver transplantation

Objective To determine the impact of exogenous vascular endothelial growth factor (VEGF) on rat liver function after partial liver transplantation. Methods Male SD rats which had undergone 30% partial liver transplantation were divided into VEGF treatment group (n=60) and control group (n=60). Animals in each group were injected with solution of VEGF or saline, respectively. Serum ALT, AST, and hyaluronic acid (HA) levels were assessed at 0, 12, 24, 72, 168 hours after operation. Results Serum ALT, AST and HA levels were significantly decreased at 72 hours after transplantation in VEGF treatment group compared with controls (t =4.681, 4.252, 2.853, P<0.05). In accordance with liver function, treatment with VEGF minimized the damage to the liver sinusoidal endothelial cells and decreased the infiltration of inflamatory cells. Conclusion Administration of VEGF may improve the liver function of rats after partial liver transplantation by maintaining the integrality of the sinusoidal structure and reducing the inflamation reaction.
5.Ray-tracing by use of optical design software ZEMAX:Correlation of intraocular lens forward movement with human eye diopter adjustment

BACKGROUND: The intraocular lens can move forward during cataract optic capsular surgery,but the changes in the human eye diopter cannot be monitored in such a dynamically adjustment process,it is difficult to obtain clinical data which could reveal the correlation between intraocular lens forward and human eye diopter adjustment.OBJECTIVE: To observe the influence of intraocular lens movement on the adjustment of human eye diopter.METHODS: Based on Hwey-Lan Liou eye model,using optical design software ZEMAX to realize ray tracing,the relationship of intraocular lens forward in the eye with the adjustment of human eye diopter was investigated.RESULTS AND CONCLUSION: Once the intraocular lens forward for a certain distance,human eye obtained the amount of accommodating diopter was not constant; the adjustment amount depended on the axial length and the required intraocular lens implantation,especially in axial length; for the intraocular lens forward of the surgical eyes at different axial lengths,the amount of accommodating human eye diopter was negatively correlated with axial length.
6.The relationship between expression of endogenous cystathionine beta-synthase and hydrogen sulfide in rectal mucosa tissue and the activeness of ulcerative colitis

Objective To study the expression of endogenous cystathionine-β-synthase (CBS) and hydrogen sulfide in patients with ulcerative colitis (UC), and explore their possible role in the pathogenesis of UC. Methods Thirty patients with active period UC (active period UC group), 30 patients with remission period UC (remission period UC group) and 30 healthy controls (control group) were selected, and SABC method was used to observe the localization of CBS in rectal mucosal tissues. The optical density value of CBS was analyzed with image analysis systems. The relative expression of CBS mRNA in the rectal mucosa tissue was detected by reverse transcriptase-polymerase chain reaction (RT-PCR), and the expression of glyceraldehyde-3-phosphate dehydrogenase (GAPDH) was detected as an internal reference. The expression level of mRNA was detected by semi quantitative analysis with gel imaging analysis system, and the results were expressed as the ratio of the target bands to the GAPDH absorbance. The serum level of hydrogen sulfide was detected. Results The serum level of hydrogen sulfide, optical density value of CBS and relative expression of CBS mRNA of rectal mucosal tissues in active period UC group were significantly higher than those in remission period UC group and control group: (90.13 ± 3.12) μmol/L vs. (50.34 ± 2.34) and (48.13 ± 2.75) μmol/L, 0.433 ± 0.037 vs. 0.295 ± 0.064 and 0.214 ± 0.026, 1.532 ± 0.134 vs. 1.031 ± 0.107 and 0.986 ± 0.067, and there were statistical differences (P < 0.01); but there were no statistical differences between remission period UC group and control group (P>0.05). Conclusions The abnomal expression of CBS and hydrogen sulfide in aactive period UC may play an important role in the pathogenesis of UC.
7.The dose study of radiation gastritis caused by esophageal cancer radical postoperative prophylactic irradiation

Objective To study the relationship between radiation gastritis ( RG ) caused by esophageal cancer radical postoperative prophylactic IMRT and the dose of gastric. Methods A total of 49 esophageal cancer patients received postoperative adjuvant radiatherapy after esophagectomy, we analysised the dose?volume histogram of treatment plan and the the occurrence of RG, To analysis those physical indicators may be related to the occurrence of RG. The of ROC curve to analysis the physical index of the occurrence of acute radiation gastritis, Chi square test is compared between groups, Logistic method was used to multivariate analysis. Results The whole group, a total of 19 cases ( 39%) patients with symptoms of acute RG level 2 or higher. The results of ROC curve analysis showed that the physical variables associated with the occurrence of RG include stomach Max,stomach Mean,LSTT5?LSTT40 and V5?V50.According to the ROC curve,the cutoff values of the physical parameters of the patients were grouped. The incidence of acute≥2RG in the group of above boundary value patients significantly higher than below boundary value group, the physical index of the differences between groups were significant (P=0. 000?0. 022),except V5.The results of Logistic multivariate analysis showed that LSTT5 and V40 were independent predictor of≥2 RG (P=0. 026,0. 001). Conclusions According to the results of this study,We should pay attention to the protection of the stomach in develop a treatment plan for the prevention of postoperative patients with esophageal cancer,dose?volume histogram can well predict the occurrence of RG. Stomach LSTT5 and V40 were independent predictor of≥2 RG.The results of this study need to be further studied.
8.Effects of Low Molecular Weight Heparin Calcium and Rivaroxaban Combined with Atorvastatin on Relat-ed Indexes in Patients with Acute Pulmonary Embolism

OBJECTIVE:To investigate the effects of low molecular weight heparin calcium and rivaroxaban combined with atorvastatin on related indexes in patients with acute pulmonary embolism(APE). METHODS:The data of 72 APE patients were analyzed retrospectively. According to treatment plan,the patients were divided into group A(21 cases),group B(26 cases)and group C(25 cases). Group A was treated with intramuscular injection of Low molecular heparin calcium injection immediately after admission;2 days later,they were given Warfarin sodium tablets;7 days later,Low molecular heparin calcium injection was stopped while warfarin was still administrated,lasting for 3-6 months. Group B was given Low molecular heparin injection(same usage and dose as group A);2 days later,they were additionally treated with Rivaroxaban tablets;7 days later,Low molecular heparin calcium injection was stopped while rivaroxaban was still administrated,lasting for 3-6 months. Based on the treatment in group B,group C was treated with Atorvastatin calcium tablets 20 mg orally,once a day in the evening,lasting for 3-6 months. The time of dyspnea,chest pain and cyanosis disappearance were observed in 3 groups as well as the levels of HR,pa(O2),pa(CO2), CRP,D-dimer,TNF-α,IL-1,IL-6,ET-1 and NO before and after treatment. The occurrence of clinical outcome events and ADR were also observed. RESULTS:The time of dyspnea,chest pain and cyanosis disappearance in group A were longer than group B, and the group B was longer than the group A,with statistical significance(P<0.05). Before treatment,there was no statistical sig-nificance in the levels of HR,pa(O2),pa(CO2),CRP,IL-1,IL-6,TNF-α,D-D,ET-1 and NO among 3 groups(P>0.05). After treatment ,HR of 3 groups were all lower than before ,and they showed group A
9.Effects of Siwu Decoction on Memory and Spatial Learning in Mice with Blood Deficiency and Related Mechanisms

Objective To investigate the effects of Siwu decoction on the learning and memory abilities in mice with blood deficiency and the related mechanism.Methods The mice were randomly divided into five groups:normal control group,model control group,high-,medium-,and low-dose group of Siwu decoction.Blood deficiency mouse model was established by continuously cuttingtailand bleedingin model control group,high-,medium-,and low-dose group of Siwu decoction.The mice in high-,medium-,and low-dose group were intragastrically administrated with Siwu decoction of 2.5,5.0,10.0 g·kg-1 for 14 days,respectively.The erythrocyte counts (RBC),hemoglobin (HGB),red blood cell hematocrit (HCT) were detected before modeling,7 and 14 days after administration.The spatial learning and memory abilities were assessed using the T maze test.The spontaneous activities were assessed using locomotors activity detector.The levels of erythropoietin (EPO) content in serum and acetylcholinesterase (AChE) in hippocampal tissue were detected by enzyme-linked immunousorbent assay.Results 5.0 and 10.0 g·kg-1Siwu decoction could increase RBC,HGB,HCT on the 7th and 14th day,2.5 g.kg-1Siwu decoction could increase RBC,HCT on 14th day,and each dosage of Siwu decoction could significantly increase the spontaneous activities of model mice with blood deficiency on the 7th and 14th day as compared with model control group (P <0.05 or P <0.01).Compared with model control group,the number of errorsin T Maze test were decreased significantly in high-,medium-,and low-dose groups (P < 0.01).And the Siwu decoction had a tendency to reduce the hippocampal AChE levels,but when compared with the model control group there were no statistical differences (P > 0.05).Conclusion Siwu decoction is benefit formemory and spatial learning in mice with blood deficiency,which may be related with higher serun EPO and lower hippocampal AChE expression.

Data analysis poses a significant challenge to the large-scale proteomics studies. Based on the structured and controlled vocabularies-Gene Ontology (GO), and the GO annotation from related databases, a strategy composed of several programs and local databases is developed to identify the functional distribution and the significantly enriched functional categories of the proteomic expression profile. It would be helpful for understanding the overall functions of these identified proteins and supply the fundamental information for further bioinformatics exploration. This strategy has been successfully used in the Human Fetal Liver (HFL) proteomic research, which is available online at http://www.hupo.org.cn/GOfact/.
