1.Determination of hydrochlorothiazide and rutin in Zhenjujiangya Tablets by HPLC

AIM: To establish a method for determination of hydrochlorothiazide and rutin in Zhenjujiangya Tablets(Flos Chrysanthemi Indici, hydrochlorothiazide, rutin, etc.) by HPLC. METHODS: HPLC was used with Spherisorb C 18 (4.6mm?250mm 5?m) column 0.05mol?L -1 citric acid acetonitrile(80∶20) (adjusted the mixture with triethylaminum to pH 4.1?0.1) was used as a mobile phase. The flow rate was 1.0mL?min -1 . The detection wavelength was at 270nm. RESULTS: The linear response range lay in 10~50?g?mL -1 of hydrochlorothiazide( r =0.9994 n =5) and 40~200?g?mL -1 of rutin( r =0.999 n =5), respectively. The average recoveries of hydrochlorothiazide and rutin were 99.8% and 99.5%, respectively. CONCLUSION: This method is rapid, accurate and suitable for the determination of the two constitutents in the compound tablets.
2.Hereditary effects and patterns of suicidal behavior in unipolar depressed patients

BACKGROUND:Hereditary factor occupies a certain position in suicidal behavior of depression. The researches in the past are focused on the hereditary effect on bipolar depression suicide.How do hereditary patterns and effects work in suicidal behavior in unipolar depression?OBJECTIVE: To probe into hereditary patterns and effects on suicidal behavior in unipolar depression.DESIGN:Retrospective investigation.SETTING:A municipal psychiatric hygienic centerPARTICIPANTS:Unipolar depression group included 115 outpatients and inpatients diagnosed in Wuxi Psychiatric Hygienic Center from June 1st 1983 to May 31st 2002.The diagnosis tallied with Standards on Depression Onset in Categories and Diagnostic Standards on Psychiatric Disturbance in China of 3rd Edition and with Standards on Severe Depression Onset in Manual of Diagnosis and Statistics of Psychiatric Disturbance in America of 4th Edition.The attack frequency of all the cases ≥ 3 times or the cases had been relieved ≥8 years after a couple of attacks.METHODS:The patients who tallied with the standards on unipolar depression received the investigation in every family tree under the instruction of 2 physicians-in-charge and more than 2 physicians and filled up the self-made investigation form of psychiatric family tree,including mainly the data of social demography of patients and their first grade relatives,characters of disease onset,frequency of attack,history of treatment and suicide. After re-diagnosed by 2 physicians-incharge and more than 2 physicians and checked by one physician-incharge,the cases were collected in patient group. The interview was carried on for the patients with suicidal behavior among all of the survived patients (107 cases) and first grade relatives (14 cases).The interview (337 cases) and investigation with letter (380 cases) were carried on for the first grade relatives without suicidal behavior. The investigation forms of 13 dead cases (8 cases of patients, 5 cases of first-grade relatives) were provided and filled-up by one or two first grade relatives. Two researchers interviewed the cases in the control,inquired the first grade relatives and filled up the investigation form of family tree.Single factor analysis was used for all the data and Falconer pattern of polygenetic threshold-value theory was used to estimate hereditary rate and standard error in suicidal behavior.Separation analysis in medical hereditary mathematic method and polygenetic threshold-value theory were applied to discuss the hereditary patterns.MAIN OUTCOME MEASURES:Hereditary effects and patterns of suicidal behavior in unipolar depressed patients.RESULTS:Suicidal risk of unipolar depressed patients(51.30%,59/115)was higher than their first grade relatives (2.58%,19/736) (x2=283.16,P < 0.01).Suicidal risk of the first grade relatives (2.58%,19/736) of unipolar depressed patients was higher than the control (0.12%,3/2469)(x2=50.36,P < 0.01).Suicidal risk of the first grade relatives of the patients with suicidal behavior (3.8%,14/372) was higher than that of the first grade relatives of the patients without suicidal behavior (1.4%,5/363)(x2=4.14,P< 0.05).The weighted average hereditary rate and standard error was (70.16±0.79)% for suicidal behavior in unipolar depression.The predictive morbidity of suicidal behavior in the first grade relatives was 3.1% and the real morbidity was 2.6%,which did not indicate significant difference (u =0.766, P > 0.05).CONCLUSION:Suicidal behavior of unipolar depression presents obvious hereditary effects and its hereditary patterns tally with polygenetic inheritance.
3.Effects of fuzheng huayu recipe in antagonizing rat's renal interstitial fibrosis.

OBJECTIVETo observe the effects of Fuzheng Huayu Recipe (FHR) on rat's renal interstitial fibrosis induced by mercuric chloride (HgCl2), and to explore preliminarily its mechanism of action.
METHODSRats were randomly divided into four groups: the normal group, the model group, the FHR group and the vitamin E group, the latter two were treated respectively by FHR 4.6 g/kg and vitamin E 100 mg/kg. Rats model was established by oral administration of 8 mg/kg HgCl2 for 9 weeks. Serum creatinine (Cr) and urea nitrogen (BUN) content were tested with corresponding test kits; hydroxyproline (Hyp) content in kidney was assayed with hydrochloric acid hydrolysis; renal histologic change was observed with HE, Masson and methenamine silver (PASM) staining; and collagen type I (Col I), as well as protein expressions of fibronectin (FN) and alpha-smooth muscle actin (alpha-SMA) was determined with Western blot.
RESULTSCompared with the model group, the kidney/body weight ratio, serum levels of Cr and BUN, kidney Hyp content, and severity of renal interstitial fibrosis in the two treated groups were significantly lower (P<0.05 or P<0.01), and the improvements were more significant in the FHR group than those in the vitamin E group; Col I and FN protein expression was also weaker in the two treated group (Col, P<0.05; FN, P<0.01); while the expression of alpha-SMA was lower in the FHR group (P<0.01), but it wasn't in the vitamin E group (P>0.05).
CONCLUSIONFHR could improve the HgCl2-induced renal function injury in rats, decrease extracellular matrix deposition and restrain renal interstitial fibrosis, the mechanism of action might be related with its inhibitory effect on myofibroblast activation.

4.Prilimanary investigation of commence time of liver specific phase on Sonazoid contrast-enhanced ultrasound

Objective To investigate commence time of liver specific phase on Sonazoid contrastenhanced ultrasound.Methods Rats were administrated with saline (n =6),SonoVue (n =24) and Sonazoid(n =24),respectively.Liver perfusion was performed at 2 min,5 min,10 min and 20 min in situ.Changes of liver enhancement caused by perfusion were quantitatively analyzed.Results Degree of liver enhancement without administration of contrast agent increased after perfusion.In SonoVue group,liver enhancement decreased after perfusion at 2 min and 5 min and increased at 10 min and 20 min significantly.In Sonazoid group,liver enhancement decreased dramatically at 2 min after perfusion,but no changes were found at 5 min,10 min and 20 min after perfusion.Conclusions Liver specific phase on Sonazoid CEUS can begin as early as 5min after administration of contrast agent.
5.Effects of alimentary reconstruction on gastrointestinal hormones in a dog model undergoing subtotal gastrectomy

Objective To evaluate effects of alimentary reconstruction procedures (integral continual jejunal interposition, Billroth Ⅱ and isolated jejunal interposition) after subtotal gastrectomy on postoperative plasma gastrin, motilin and cholecystokinin. Methods Twenty-four dogs were divided into 3 groups undergoing distal subtotal gastrectomy and three different digestive tract reconstruction (integral continual jejunal interposition, Billroth Ⅱ and isolated jejunal interposition). The concentration of plasma gastrin, motilin and cholecystokinin were detected by enzyme-linked immunosorbent assay before and after operation. Results Two months after operation, plasma gastrin level of the integral continual jejunal interposition group (2. 2 ±0. 7 ) ng/L, ( 3.9 ± 0. 8 ) ng/L was significantly lower than that of preoperative both in fasting and postprandial state (3.8 ± 1.0) ng/L, (5.3 ± 1.6) ng/L, all P <0.05, but was significantly higher than other two groups in postprandial state (2. 7 ± 1.0) ng/L, (3.6 ±0. 6) ng/L, P <0. 05. Two months after operation, plasma motilin concentration of integral continual jejunal interposition group (577 ±204) ng/L, (1003 ± 209) ng/L were significantly higher than that of preoperative both in fasting and postprandial (429 ± 128) ng/L, (854 ± 218 ) ng/L, P < 0. 05. The postoperative plasma motilin of integral continual jejunal interposition group ( 1003 ± 209 ) ng/L was significantly higher than other two groups in postprandial state (840 ±205) ng/L, (986 ± 189) ng/L, P <0. 05. Two months after operation,plasma cholecystokinin concentration of integral continual jejunal interposition group ( 19.6 ± 2.0 ) ng/Lwere significantly higher than that of preoperative both in postprandial ( 19.0 ± 2. 0) ng/L, P < 0. 05. The postoperative plasma cholecystokinin of integral continual jejunal interposition group ( 19. 6 ± 2. 0) ng/L was significantly lower than other two groups (22.2 ± 2. 1 ) ng/L, (20. 1 ± 2. 5 ) ng/L, P < 0. 05. Conclusion Integral continual jejunal interposition after distal gastrectomy maintains the postoperative plasma motilin and gastrin in a relatively higher level and decreases the concentration of plasma cholecystokinin.
6.Role of follicular helper T cells in development and progression of liver diseases

Follicular helper T cell (TFH) is a new CD4+ T lymphocyte subset which helps B lymphocytes to produce antibodies and is closely related to the development and progression of liver diseases. This article reviews the detection and differentiation of TFH and related functional molecules, the mechanism of action of TFH in helping B lymphocytes to produce antibodies through producing interleukin-21, as well as the role of TFH in liver diseases such as chronic hepatitis B, chronic hepatitis C, autoimmune hepatitis, primary biliary cirrhosis, hepatic failure, and liver transplantation, and points out that TFH may become the new target cell in the treatment of liver diseases.
7.Protective effect of Zhizi Dahuang decoction in a mouse model of alcoholic liver disease

ObjectiveTo investigate the effect of Zhizi Dahuang decoction (ZZDHT) in the treatment of alcoholic liver disease (ALD) by improving oxidative stress in hepatic neutrophils. MethodsNetwork pharmacology was used to obtain the chemical components of ZZDHT and their corresponding action targets and analyze the potential targets and functional pathways of ZZDHT in the treatment of ALD. The non-target metabolomics technology was used to observe the changes in the metabolites of ZZDHT in mouse serum and liver. The mice were given ZZDHT at a dose twice as much as the middle dose concentration by gavage, and serum and liver samples were collected at six time points after gavage (10 minutes, 30 minutes, 1 hour, 2 hours, 4 hours, and 6 hours) and were then mixed for mass spectrometry (administration group with 18 mice), while the 18 mice in the control group were given an equal volume of normal saline by gavage. Ultra-performance liquid chromatography was used for rapid isolation and identification of the metabolites of ZZDHT in serum and liver tissue, and the effective constituents of ZZDHT were validated. Male C57BL/6J mice, aged 8 weeks, were randomly and equally divided into control group, model group, and low-, middle-, and high-dose ZZDHT groups, with 10 mice in each group. All mice except those in the control group were used to establish a mouse model of ALD (NIAAA model mice), and at the same time, the mice in the administration groups were given low-, middle-, and high-dose ZZDHT by gavage, while those in the control group and the model group were given an equal volume of normal saline by gavage. The serum levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), and triglyceride (TG) were measured; PCR was used to measure the gene expression levels of related inflammation, oxidative stress, and neutrophil indicators in the liver; ELISA was used to measure the levels of related inflammation and oxidative stress indicators in serum; superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), and malondialdehyde (MDA) were measured to observe the level of oxidative stress in the liver; HE staining, myeloperoxidase staining, and oil red staining were used to observe liver injury, neutrophil infiltration, and lipid deposition. A one-way analysis of variance was used for comparison of continuous data between multiple groups, and the least significant difference t-test was used for further comparison between two groups. ResultsA total of 53 active components and 227 target genes were obtained for ZZDHT, and there were 8685 target genes of ALD, resulting in 222 common target genes between these two groups of genes. Core pathways included the interleukin-6 signaling pathway and the TNF signaling pathway. The non-targeted metabolic analysis of ZZDHT obtained 225 metabolites in mouse liver and 227 metabolites in serum, among which there were 126 common metabolites. The core pathways of liver metabolites included glycerolipid metabolism and inflammatory mediator regulation of TRP channels, and the core pathways of serum metabolites included the AMPK signaling pathway and oxidative phosphorylation, all of which were associated with oxidative stress- and inflammation-related pathways. Compared with the model group, the low-, middle-, and high-dose ZZDHT groups had significant reductions in the serum levels of ALT, AST, and TG (all P<0.05), and the middle-dose ZZDHT group had significant reductions in the levels of Ly6g, Ncf1, Ncf2, IL-6, TNF-α, IL-1β, MDA, 4-HNE, Gp91, and P22 in the liver (all P<0.05), a significant increase in the level of SOD (P<0.05), a significant reduction in the serum level of 4-HNE (P<0.05), and a significant increase in the level of GSH-Px (P<0.05). There were significant improvements in fat deposition and neutrophil infiltration in the liver of mice in the middle-dose ZZDHT group (both P<0.05). ConclusionZZDHT significantly reduces oxidative stress and inflammatory response in NIAAA model mice.
8.Analysis of chemical ingredients of modified Shenqi Dihuang decoction by UPLC-LTQ-Orbitrap-MS

Objective To analyze the chemical compounds of Shenqi Dihuang decoction by the ultraperformance liquid chromatography coupled with linear quadrupole ion trap-orbitrap mass spectrometry (UPLC-LTQ-Orbitrap-MS). Methods Warters ACQUITY UPLC HSS T3 (2.1 mm ×100 mm, 1.8 μm) was used as chromatographic column with mobile phase: 0.1% formic acid water (A)-0.1% formic acid acetonitrile (B) with gradient elution, and flow rate was 0.3 ml/min. Electrospray ion source (ESI) and an electrostatic field orbital ion trap mass analyzer were adopted, which was used to collect mass spectrometry fragment information with positive and negative ion modes, by comparing with the relative retention time of the reference substance. In addition, the fragment information of the mass spectrum was used to identify the compounds. The accurate identification of the chemical components in Shenqi Dihuang decoction was confirmed with literature. Results The study found that UPLC-LTQ-Orbitrap-MS technology could be used to identify 62 chemical components, including 13 aromatic acids, 9 flavonoids, 8 saponins, and 5 aromatic amines, 3 keto acids, 2 phenols, 1 aromatic quinone and other ingredients in Shenqi Dihuang decoction. Conclusion The identification analysis method in this study was efficient and accurate, which could be applied to the identification and analysis of chemical components in Shenqi Dihuang decoction and provided the important experimental data for the research on the material basis and mechanism.
