Objective To determine the changes of gene expression profile in small intestinal allograft in rats after cold preservation/reperfusion and investigate the mechanism of allograft injury. Methods Heterotopic segmental small bowel transplantation in rats was performed with a modified technique. Total RNAs was extracted from the allografts (experimental group) and normal intestines (control group) 1h after cold preservation /reperfusion, and then purified to mRNA. Equal mRNAs of the two groups were as then reversely transcribed to cDNA and to prepare hybridization probes. The mixed probes were hybridized to the cDNA microarray . After high-stringent washing, the fluorescent signals on cDNA microarray were scanned and analyzed . Results Among the 4096 target genes, 82 differentially expressed genes were identified between the two groups. There were 18 novel genes and 33 expression sequence tags. 31 genes reported and differentially expressed could be related to the preservation/reperfusion injury of grafts. Conclusions Abnormal adhesion between polymorphonuclear cells and microvascular endothelial cells, and failure in energy, glucose and protein metabolism of grafts may contribute to cold preservation/reperfusion injury of small intestinal allografts.

The aim of this study was to investigate the effect of a synthetic fifteen-residue peptide, Gly-Thr-Pro-Gly-Pro-Gln-Gly-Ile-Ala-Gly-Gln-Arg-Gly-Val-Val(P-15),on islet cell insulin secretion and insulin gene expression.Wistar rat islets,isolated by collagenase and trypsin digestion,were exposed to 0,0. 72,7.2,36?mol/L P-15 with 16.7mmol/L glucose.Insulin release increased approximately twofold in re- sponse to the concentration of 7.2?mol/L P-15 and Northern-blot analysis of islet mRNA with a rat pre- proinsulin cDNA probe showed a concomitant increase in mRNA level as compared with levels observed in islets treated with control.It is concluded that P-15 not only can stimulate the release of insulin but also in- crease the preproinsulin mRNA expression of islet cells.

The clinical electroencephalogram (EEG) monitoring systems based on personal computer system can not meet the requirements of portability and home usage. The epilepsy patients have to be monitored in hospital for an extended period of time, which imposes a heavy burden on hospitals. In the present study, we designed a portable 16-lead networked monitoring system based on the Android smart phone. The system uses some technologies including the active electrode, the WiFi wireless transmission, the multi-scale permutation entropy (MPE) algorithm, the back-propagation (BP) neural network algorithm, etc. Moreover, the software of Android mobile application can realize the processing and analysis of EEG data, the display of EEG waveform and the alarm of epileptic seizure. The system has been tested on the mobile phones with Android 2. 3 operating system or higher version and the results showed that this software ran accurately and steadily in the detection of epileptic seizure. In conclusion, this paper provides a portable and reliable solution for epileptic seizure monitoring in clinical and home applications.

Objective To analyze the effect of miR-497 high expression on the gene expression profile of colon cancer cell line HCT116. Methods MiR-497 high expressing colon cancer cell model HCT116-497 and negative control HCT116-CON were established by lentiviral transduction. The human (V2) gene expression microarray was used to identify genes that were differentially expressed between colon cancer cells overexpressing miR-497 and the controls. The candidates were subjected to the gene ontology (GO) and KEGG pathway enrichment analysis by Molecule Annotation System 3.0 (MAS3.0). The differential expression of representative genes relative to inflammation were confirmed by real-time quantitative reverse transcription polymerase chain reaction (qRT-PCR). Results Of all the differently expressed genes, 582 genes were down-regulated by at least 3-folds, which were enriched in inflammation-related signaling pathways in colon cancer cells overexpressing miR-497. The decrease in 15 representative genes was validated by qPCR. Compared with those in HCT116-CON cells, expressions of 10 genes in HCT116-497 cells, including CACNB1, FOS, IL-29, RPS6KA2, TNFSF15, IL-11, INHBC, CSF1R, JAK3 and IL-2Rβ, were decreased significantly, and there were statistical differences (all P< 0.05) Conclusion MiR-497 inhibits the mRNA expression of inflammation-related genes in colon cancer cell line HCT116.

Objective To analyze the safety of laparoscopic splenectomy for traumatic splenic rupture.Methods The clinical data of 62 patients with traumatic splenic rupture treated by laparoscopic splenectomy in our hospital from Jan 2013 to Jan 2018 were retrospectively analyzed.Results All 62 patients successfully underwent laparoscopic surgery except for 1 case who was converted to open surgery.One case suffered from postoperative bleeding.There was no infection,serious pancreatic leakage and other major complications.The total amount of intraoperative bleeding was 1 600 ml on average,the average operation time was 135 min,the average splenectomy time was 55 min,and the average hospital stay was 12.5 d.Conclusion Laparoscopic splenectomy is a safe and reliable minimally invasive operation for traumatic splenic rupture.

Objective To investigate the effects of silkworm excrement ironing on blood levels of glucose,lipids and Hcy in diabetic peripheral neuropathy patients with cold coagulation and blood stasis.Methods A total of 260 diabetic peripheral neuropathy patients with cold coagulation and blood stasis were randomly divided into the control group(n=130)and the observation group(n=130).Both groups were treated with oral methylcobalamin(500 μg,tid),and in addition,the observation group was given silkworm excrement ironing on areas of the limb with numbness and pain.Patients underwent one single course of treatment lasting 4 weeks.Blood levels of glucose,lipids and Hcy preand post treatment were analyzed.Results Blood lipid profiles and hemorheology improved in both groups after treatment,compared with pre-treatment data,and greater improvement was seen in the observation group than in the control group(all P＜0.05).Plasma Hcy levels decreased in both groups after treatment,compared with pre-treatment levels,and deeper decline was seen in the observation group than in the control group after treatment(all P＜0.05).Conclusions Silkworm excrement ironing can effectively improve the blood glucose profile and lipid metabolism,reduce plasma Hcy levels and may exert beneficial preventive and therapeutic effects on diabetic peripheral neuropathy.

Objective To explore the influences of continued nursing care on the recovery of intra-articular fractures of joint function of limbs and the quality of life after operation. Methods A total of 493 intra-articular fracture of limbs postoperative patients,who were discharged from our department from January 2015 to December 2016,were randomly divided into observation group and reference group.After operation,the reference group used self-management mode to practice joint functional exercise,while the observation group uses continued nursing mode to guide physical therapy to recover joint functions.After they discharged 12 weeks,we compared these two groups of patients with satisfaction of nursing care,joint function recovery and quality of life. Results After 12 weeks of operation, in observation group, the patients' joint function recovery rate was close to 79.27%(195/246), which was significantly higher than the rates 55.47%(137/247)in reference group,the difference was statistically significant(χ2=32.947,P<0.01). Respectively, the physiological function score,vigor score, mental health score, physiology role score, body pain score, health condition score, social function score and health score of the observation group in the quality of life assessment were (72.66 ± 4.41), (89.27 ± 4.10), (93.01 ± 3.05), (88.03 ± 3.19), (91.68±3.99),(76.78±4.86),(79.11±4.68),(85.57±7.07).The scores of control groups were(65.71±3.41),(64.02±4.51),(61.43±4.29),(66.49±4.52),(64.24±4.30),(62.02±6.24),(62.94±4.50),(65.02±7.72).The comparative difference of the two groups has statistical significance (t=12.48- 60.15, P<0.05). Meanwhile,in the satisfaction rate of nursing care,the great satisfaction rate of observation group(70.73%, 174/246) was much higher than reference group (37.65%, 93/247), the difference was statistically significant (χ2=59.789, P<0.01). Conclusion Offered outside continued nursing care to the intra-articular fracture of limbs postoperative patients can effectively improve the joint function of patients,their qualities of life and can considerably increase satisfaction of nursing care. Thus,it should to be clinically promoted.
