[Abstact] Circular RNAs(circRNAs) were once mistakenly considered as the transcription splicing intermediates , affiliate by-products or splicing errors , and the understanding of circRNAs has been significantly revolutionized with the development of high-throughput sequencing technology of RNA and the improvement of the corresponding bioinformatics analysis .CircRNAs with their unique closed-loop structure and undenible important biological functions have become new favorites in both molecular biology studies and clinical trials.To our knowledge , circRNAs usually arise after transcription , stably express in a variety of biological cells and regulate or interfere genetic expression , thus affecting occurrence and progression of diseases.Recently, circRNAs′influences upon various cancers have begun to edge up and we elaborates them in detail here based on the latest researches .
2.Effects and Significance of SDF-1/CXCR4 in Proliferation, Migration and Invasion of Colorectal Cancer Cell Line SW480

Objective To discuss the influence and significance of stromal cell-derived factor 1 (SDF-1) and its specific receptor CXC chemokine receptor 4 (CXCR4) in proliferation, migration and invasion ability of SW480 colorectal cancer cells. Methods The colorectal cancer cell line SW480 in logarithmic phase was divided into four groups:control group (with no any processing), SDF-1 group (added 100μg/L SDF-1), SDF-1+1 mg/L AMD3100 mixed group (added 1 mg/L AMD3100 for 2 hours, then added 100μg/L SDF-1) and AMD3100 group (added 1 mg/L AMD3100). Immunohisto?chemistry method was used to detect the protein expression of CXCR4 in SW480 cells. The expression of CXCR4 mRNA in SW480 cells was detected by RT-PCR before and after SDF-1 and AMD3100 treatment. MTT assay and transwell chamber were used to test the changes of proliferation, migration and invasion ability of SW480 cells before and after SDF-1 and AMD3100 treatment. Results The result of immunohistochemistry showed that CXCR4 protein was expressed in SW480 cells (positive rate=80%). CXCR4 mRNA was expressed in SW480 cells. The expression of CXCR4 mRNA was up-regulat?ed by SDF-1(100μg/L), which could be inhibited by AMD3100 (1 mg/L). The proliferation activity was higher in SDF-1 group (0.847±0.039) compared to that in control group (0.624±0.011) and SDF-1+AMD3100 mixed group (0.607 ±0.016). The proliferation activity was lower in AMD3100 group (0.456 ± 0.031) than that in control group and SDF-1+AMD3100 mixed group (F=108.03, P<0.05). The number of transmembrane cells was more in SDF-1 group (98.7±5.8, 33.7±6.2) than that in control group (21.0±2.2, 6.1±2.3), SDF-1+1 mg/L AMD3100 mixed group (18.5±8.4, 8.5±2.8) and AMD3100 group (12.1±3.2, 2.1±1.0) detected by transwell chamber experiment. However, there were no statistical differences between three groups. Conclusion The biological axis SDF-1/CXCR4 can promote the proliferation, migration and invasion in colorectal cancer cell line SW480.
3.Curative effect of laser sub-epithelial keratomileusis on low and midrange myopia in aged patients

Objective To study the characteristic of operation design and analyze curative effect of laser sub-epithelial keratomileusis (LASEK) on low and midrange myopia in aged patients. Method Fiftyeight patients ( 114 eyes) no less than 35 years old were treated with LASEK by monoblepsis. The subjects were examined and analyzed at 3 days, 7 days, 1 month,3 months and 6 months after operation. Results Seventy-one corneas(62.28%) recovered well at 3 days after operation. At 1 month after operation, the acuity of vision of all patients achieved or exceeded corrected vision before operation,and lasted to 6 months after operation. The break up time of 63 eyes were lower than normal at 3 months after operation,but 6 months after operation all of them recovered to normal. The satisfaction of patients raised gradually. Conclusions LASEK is effective for no less than 35 years old myopic patients. Make operation plan refer to monoblepsis and patients' needs can receive more satisfaction.
4.Nursing care for patients with mid-stage hepatocellular carcinoma treated with 131I-chTNT combined with radiofrequency ablation

Objective To summarize the nursing points for patients with mid-stage hepatocellular carcinoma who are receiving radiofrequency ablation (RFA) combined with 131I-chTNT treatment. Methods The clinical data of 12 patients with mid-stage hepatocellular carcinoma, who were encountered at authors’ hospital during the period from Aug. 2009 to Sep. 2011, were retrospectively analyzed. RFA combined with 131I-chTNT was carried out in all patients. Preoperative prophylactic anti-allergy medication and protection measure for the thyroid were employed. After the operation strengthened radiation protection and close observation for complications were conducted. The results were analyzed. Results After active treatment and proper nursing, the tumor shrank in all patients. All the patients were followed up for 5 - 48 months with a median follow- up time of 31 months. The median overall survival time was 43 months. No serious complications such as bone marrow suppression or thyroid dysfunction occurred. Conclusion For the treatment of mid-stage hepatocellular carcinoma, 131I-chTNT combined with RFA has satisfactory therapeutic effect. Close observation for the occurrence of complications and careful nursing can improve the quality of life and prolong the survival time.
5.Expression of SNCG and CXCR4 mRNAs in gastric adenocarcinoma and its clinical significance

Objective:To detect expression of SNCG and CXCR4 mRNA in gastric adenocarcinoma and evaluate its function in carcinogenesis, progression, infiltration, and metastasis, as well as the correlation of these proteins. Methods:The expression of SNCG and CXCR4 mRNAs was detected using reverse transcription-polymerase chain reaction in gastric adenocarcinoma and normal gastric mucosa. Results:The expression of SNCG and CXCR4 mRNAs was obviously higher in gastric adenocarcinoma than in normal gastric mucosa (P<0.01). The expression of SNCG mRNA was closely correlated with tumor infiltration depth (P<0.05) and lymph node metas-tasis (P<0.01). The expression of CXCR4 mRNA closely correlated with lymph node metastasis (P<0.01). The expression of SNCG and CXCR4 mRNAs positively correlated in 29 cases of gastric adenocarcinoma (r=0.346, P<0.05). Conclusion:SNCG and CXCR4 may be overexpressed in tumor tissue and may have important functions in tumorigenesis, progression, infiltration, and metastasis of gastric adenocarcinoma.
6.Detection of the aberration of X chromosome in hepatocellular carcinoma cell lin e HCC-9903 by fluorescence in situ hybridization(FISH)

Objective To study the X chromosome aberration in hepatocellular carcinoma cell line HCC-9903. Method The hepatocellular carc inoma cell line, HCC-9903, was detected by fluorescence in situ hybridization ( FISH) centromere DNA probes of X chromosome. Result Increased X chromosome copy number was observed in HC C-9903 cell line , four signals of hybridization was the major type. Conclusion Gain of X chromosome freq uently occurred in liver carcinoma. The relationship between gain of X chromosom e and liver carcinoma genesis needs further investigation.
7.Structure and function of a novel thermostable pullulanase.

Research on novel pullulanase has major significance on the domestic industrialization of pullulanase and the breakdown of foreign monopoly. A thermophilic bacteria LM 18-11 producing thermostable pullulanase was isolated from Lunma hot springs of Yunnan province. It was identified as Anoxybacillus sp. by 16S rDNA phylogenetic analysis. Full-length pullulanase gene was cloned from Anoxybacillus sp. LM18-11. The optimum temperature of the pullulanase was between 55 and 60 degrees C with a half-life as long as 48 h at 60 degrees C; and its optimum pH was between 5.6 and 6.4. V(max) and K(m) of the pullulanase was measured as 750 U/mg and 1.47 mg/mL, which is the highest specific activity reported so far. The pullulanase crystals structure showed a typical alpha-amylase family structure. The N-terminal has a special substrate binding domain. Activity and substrate binding were decreased when the domain was deleted, the V(max) and K(m) were 324 U/mg and 1.95 mg/mL, respectively. The pullulanase was highly heterologous expressed in Bacillus subtilis by P43 promoter. The extracellular enzyme activity was 42 U/mL, which increased more than 40 times compared to the initial strain. This pullulanase has good application prospects.

8.Identification and analysis of vaginal lactobacilli in patients with bacterial vaginosis patients and healthy women in nationality of Pastoral area

Objective To identify and analyze the species of vaginal lactobacilli between patients with bacterial vaginosis (BV) and healthy women at childbearing age in Inner Mongolia. Methods From Jun. 2008 to Dec. 2008, 203 Mongolian healthy women, 74 Han healthy women and 102 Mongolian patients with BV from 3 pastoral areas were enrolled in this study. Isolation and culture of lactobacilli from vaginal wall were performed by modified culture medium. DNA of lactobacilli were extracted and sequenced. H2O2 were detected by TMB-HRP-MRS. Results(1)The rate of lactobacilli identification were 76.8%(156/203) in Mongolian healthy women and 21.6% (22/102) in Mongolian patients with BV, which reached statistical difference(P<0.01).Lactobacilli identification in Han healthy women [82.4%(61/74)] did not show significant difference with that of Mongolian healthy women (P>0.05). (2) The total of 193 strains and 11 species of Lactobacillus were detected in 203 Mongolian healthy women. Meanwhile,22 strains and 4 species of Lactobacillus were found in 102 Mongolian BV cases.(3)The rate of H2O2 generating Lactobacilli was 27.3% (6/22) in Mongolian BV patients and 75.7% (56/74)in Mongolian healthy women, which showed statistical difference(P<0.05). Conclusions The rate of Lactobacillus was not related with the race of women in pastoral area in Inner Mongolian. The amount of lactobacilli and H2O2 generating Lactobacilli in the vagina of BV patients was remarkably lower than those of healthy women at childbearing age.
9.The reasons of focal liver lesions that difficult to detect by conventional ultrasound analysed by ultrasound-CT/MR fusion imaging

Objective To analyze the reasons of focal liver lesions that difficult to detect by conventional ultrasound ultrasound-CT/MR fusion imaging.Methods 101 lesions which were confirmed by pathology or clinical diagnosis standards were recruited in the research.All of them were difficult to detect by conventional ultrasound but CT/MR display clearly.Ultrasound-CT/MR fusion imaging was used to observe the size,location and internal echo of the lesions,as well as the background of the surrounding liver parenchyma.Results All cases were successfully registrated,the registration time were 2-6min [(4.1 ±0.6)min].For these 101 lesions,93.1%(94/101) of which the diameter ≤20 mm,56.4% (57/101) were located in hepatic segments near the diaphragm (such as S2,S4,S7,S8),78.2% (79/101) were internal isoecho,and 79.2%(80/101) in the background of liver cirrhosis.Conclusions The important reasons that focal liver lesions detected difficult by conventional ultrasound includes:lesion size,location,internal echo and the hepatic background.
10.Association of single nucleotide polymorphism (SNP) in micro RNA-122 with genetic predisposition and early recurrence after resection for hepatocellular carcinoma

Objective To investigate the association of single nucleotide polymorphism (SNP) in micro RNA-122 (miRNA-122) with genetic predisposition and early recurrence after resection for hepatocellular carcinoma(HCC).Methods This is a case-control study involving 173 HCC patients.DNA were exacted from cancer tissues embedded in paraffin and were amplified by PCR.The study aimed to explore SNP in gene sequence of miRNA-122 (357 base pair including extron.The outcomes of genetic predisposition were analyzed with early recurrence after resection for HCC.Results Only rs17669 was found in miRNA-122.The genetype frequence of C/C,T/T and C/T at rs17669 gene locus were 7(4.0%),110(63.6%)and 56(32.4%),respectively.When compared to T/T genetype,C/C genetype was a protective factor of risk of HCC (OR =0.213,95% CI:0.062-0.732).Genetypes had no relationship with early recurrence after resection.Conclusion For HCC recurrence,rs17669 may be associated with genetic predisposition of HCC in Hans in Jiangxi infected with HBV.
