1.The Epidemiological Survey of Hypertension among Inhabitants in Shanxi Province

Objective To comprehend the prevalence rate,awareness rate and the risk factors of hypertension among inhabitants in Shanxi Province. Methods With cluster random sampling,41 committees and villages among nine areas of Shanxi Province were selected as the investigative spots,including Taiyuan,Datong,Jinzhong,Yuncheng and etc. In each spot,permanent residents aged above 15 yrs were investigated on the correlation study of hypertension. Results In this study,10 792 people were investigated,male 4 721 and female 6 071. The prevalent rate of hypertension was 25.4%,the standardized rate was 23.6%,28.0% in male and 23.4% in female. The awareness rate for hypertension was 44.8%,38.6% in male and 50.5% in female. The multivariate Logistic regression analysis showed the risk factors for hypertension in this population included male,the aged people,lacking of knowledge,drinking alcohol,and obesity. Conclusion There is high prevalent rate of hypertension in Shanxi Province. So we should take the measures aiming at the present situation and risk factors of hypertension.
2.Hypoparathyroidism-sensorineural deafness-renal dysplasia syndrome and mutation anaiysis of GATA3 gene

Objective To explore the clinical manifestation of a patient with hypoparathyroidsmsensorineural deafness-renal dysplasia (HDR) syndrome and to sequence the related GATA3 gene of the patient.Methods A 22 year old person with HDR syndrome was reported in regard to clinical manifestation,laboratory examination,and genetic mutation.Some related literatures were reviewed.Results The patient showed tetany,deafness,and positive Chvosteks' and Trousseau' s signs.The initial laboratory studies showed that serum concentration of calcium was lowed and the iPTH level were lower than normal.Binaural pure tone audiometry showed Binaural sensorineural deafness.Colour doppler ultrasound revealed that his right kidney was not observed and the level of creatinine was increased,indicating renal insufficiency.GATA3 mutations on DNA sequence analysis indicated that the 6 exon IVS6-1G-A (G/A heterozygosis splicing),showed the mutation of G to A is in the upstream of the first base in the six exon.After treating with calcium carbonate and vitamin D,the symptoms and signs were improved.Conclusion HDR syndrome is a rare endocrine disease,that should receive more attention in order to avoid missing diagnosis;The IVS6-1G-A as a novel mutation of GATA3 gene,has not been reported so far.
3.Restriction endonuclease digest - melting curve analysis: a new SNP genotyping and its application in traditional Chinese medicine authentication.

Single nucleotide polymorphisms (SNP) is an important molecular marker in traditional Chinese medicine research, and it is widely used in TCM authentication. The present study created a new genotyping method by combining restriction endonuclease digesting with melting curve analysis, which is a stable, rapid and easy doing SNP genotyping method. The new method analyzed SNP genotyping of two chloroplast SNP which was located in or out of the endonuclease recognition site, the results showed that when attaching a 14 bp GC-clamp (cggcgggagggcgg) to 5' end of the primer and selecting suited endonuclease to digest the amplification products, the melting curve of Lonicera japonica and Atractylodes macrocephala were all of double peaks and the adulterants Shan-yin-hua and A. lancea were of single peaks. The results indicated that the method had good stability and reproducibility for identifying authentic medicines from its adulterants. It is a potential SNP genotyping method and named restriction endonuclease digest - melting curve analysis.
4.Conversion of corncob into biohydrogen by anaerobic fermentation.

Biohydrogen production from corncob by dark fermentation was reported for the first time. The effects of the pretreatment condition, substrate concentration and initial pH on the hydrogen production were investigated in batch cultivations. The maximum hydrogen yield of 107.9 mL/g-TVS and hydrogen production rate of 4.2 mL/g-TVS .h(-) were obtained under the condition of 1% HCl pretreating substrate for 30 min, 10 g/L substrate concentration and initial pH8.0. The content of hemicellulose in corncob decreased significantly from 42.2% to 3.0% after HC1 pretreatment. The contents of cellulose, hemicellulose and lignin in the acid pretreated corncob decreased slightly in hydrogen producing process. The results indicate that the acid pretreatment of the substrate plays a key role in the conversion of corncob into biohydrogen. Fourier transform infrared spectroscopy (FTIR) was used to study the changes in the corncob composition during the treatment of chemical-microbial process. It was shown that the amorphous domains of cellulose and hemicellulose were hydrolyzed into fermentable asccharides through HCl pretreatment and the microorganisms had a devastating effect on the crystallinitiy of the cellulose.

5.Study on the interaction between presenilin 1 and carboxyl terminus of Hsc70 interacting protein

ObjectiveTo understand the pathological and physiological roles of Presenilin 1 (PS1) in Alzheimer's disease (AD) recurrence, and the interaction between PSI and carhoxyl terminus of Hsc70 interacting protein (CHIP). MethodsThe yeast two-hybrid system was applied to identify a novel PS1 interacting protein as CHIP. After pGBKT7-PS1-C203 bait plasmid and full fragement CHIP of pACT2-CHIP expression vector were constructed, the interaction between PSI and CHIP was tested by β-galactosidase assay, pGBKT7-PS1-C203 was co-transfected with pACT2-CHIP into 293T cells and the interaction between PS1 and CHIP was tested by co-immunoprecipitation and Western blot. ResultsSpecificity of the interaction between PS1 and CHIP was identified by β-galactosidase assay and co- immunoprecipitation. ConclusionsCHIP is able to modulate chaperone functions and the pathway of protein ubiquitination/degradation. CHIP may regulate a proper assembly of the γ-secretase complex through its interaction with PSI, which is helpful to elucidate the mechanism of AD pathology.
6.The epidemiological survey of hypertension among the elderly population in Shanxi Province

Objective To investigate the prevalence rate of hypertension among the elderly population in Shanxi Province. Methods According to the random sampling, 41 residents committees of city blocks and villages of 9 areas (including Taiyuan, Datong, Jinzhong, Yuncheng, et al) of Shanxi Province were extracted as the investigative spots. All the people were older than 55years, and risk factors for hypertension were identified. Results A total of 3702 people were surveyed, including 1,782 men and 1,920 women. The total hypertension prevalence rate was 39.0%(the standardized rate: 39. 5 %), with 38. 7 % in men (the standardized rate: 38. 5%), 39.3 % in women (the standardized rate: 41.2%), and no statistic significance was found between men and women (χ2= 0. 143, P>0. 05). Aging, lacking of knowledge, and obesity were risk factors for hypertension. Conclusions The hypertension in Shanxi Province has a high prevalence rate among the elderly population, a comprehensive intervention should be taken in the prevention of hypertension.
7.Effects of different hypoglycemic treatments on glucagon-like peptide-1 and its correlation with β cells function in newly diagnosed type 2 diabetes mellitus

Objective To observe different hypoglycemic treatments on glucagon-like peptide-1 (GLP-1) and its correlation with β cells function in newly diagnosed type 2 diabetes mellitus (T2DM).Methods A total of 76 patients with newly diagnosed T2DM were randomly divided into insulin aspart 30 group and oral drugs group.OGTT test were measured at 0,30,120 min,the serum levels of GLP-1 and corresponding C peptide,and insulin levels were measured,HOMA insulin secretion and insulin resistance index were calculated.Then 30 healthy cases were as the control group,GLP-1 levels were analyzed,and the changes of the index were compared.Results At each time point,blood glucose and glucose area under the curve (AUCg) was significantly decreased (P <0.05).Insulin and C-peptide and the AUC (AUCins,AUCc-p) were increased (P < 0.05).HOMA-IR was statistically decreased compared with pre-therapy (P < 0.05),HOMA-IS and △INS30/G30,△ INS120/G120 were significantly increased (P < 0.05).Compared with pre-therapy,GLP-1 levels and AUCglp (area under the curve GLP-1) were significantly increased (P <0.05),and reached the peak at 120 min,closed to secretion curve of the control group.Conclusion GLP-1 levels were lower than normal in newly diagnosed T2DM patients,and different hypoglycemic drugs makes GLP-1 levels increase.Insulin,C-peptide and AUCins,AUCc-p,HOMA-IS,HOMA-IR,etc.,suggest that β-cell function is restored and insulin resistance is reduced.
