1.Effect of self-designedFuzheng-Quxie-Jiedu decoction combined with chemotherapy on breast cancer

ObjectiveTo study the effect of self-designedFuzheng-Quxie-Jiedu decoction on transferring, relapse and liver quality of breast cancer patients after operation.Methods144 breast cancer patients after operation were divided into a study group and a control group. The control group was treated with CTF chemotherapy, and the study group was additionally treated with Chinese medicine. Both groups were treated for for 6 months. The 5-year recurrent rate and TCM syndrome scoring of the two groups were compared. The QOL was evaluated by SF-36 scale, and the related risk factors were tested by Logistic analysis. ResultsAfter treatment, the 5-year transferring recurrent rate of the study group lower than the control group (11.63%vs. 25.86%,χ2=4.892,P=4.892). The SF-36 scale body condition (72.11 ± 10.41 vs. 64.59 ± 9.83, t=3.252), social function (65.61 ± 12.54 vs. 58.94 ± 11.62,t=3.302), body role function (55.84 ± 10.65 vs. 47.46 ± 9.18,t=3.487), body pain (66.62 ± 12.53 vs. 60.68 ± 11.76,t=2.859), mental health (67.64 ± 11.35 vs. 62.65 ± 12.10,t=3.449), emotional role function (60.44 ± 9.20 vs. 55.04 ± 9.44),t=2.624), (69.77 ± 9.43 vs. 62.47 ± 10.12,t=3.557), and general health status (68.46 ± 10.25 vs. 62.45 ± 11.78,t=3.216) in the study group were all significantly increased comparing with the control group (P < 0.05). Results of logistic regression analysis showed that the transferring risk factors for recurrence were TNM staging (OR=1.984, 95%CI 1.510-2.607) and tumor size (OR=2.015, 95% CI2.015-2.847), the number of axillary lymph nodes (+) (OR=2.114, 95%CI 2.114-2.759), Chinese medicine adjuvant chemotherapy (OR=0.389, 95% CI0.235-0.644) and CD4/CD8 (OR=0.487, 95%CI 0.487-0.748).ConclusionSelf-designedFuzheng-Quxie-Jiedu decoction combined with standard chemotherapy can obviously reduce the recurrent rate of the postoperative patients with breast cancer, improve the quality of life and improve the clinical symptoms.
2.Clinical observation of Huoxue-Sanjie decoction combined with GEMOX chemotherapy regimen in the treatment of advanced pancreatic cancer for shireyudu syndrome

Objective To evaluate the clinical effects ofHuoxue-Sanjie decoction combined with GEMOX chemotherapy regimen in the treatment of advanced pancreatic cancer with dampness-heat and stasis pattern.MethodsA total of 90 patients with advanced pancreatic cancer and TCM pattern of dampness-heat and stasis were enrolled from January 2012 to May 2014 in our hospital and randomly divided into control group (45 patients) and TCM group (45 patients). The control group was treated with GEMOX chemotherapy regimen used alone, while the TCM group was treated withHuoxue-Sanjie decoction assisted treatment based on control group. The clinical effects, quality improvement rate, immune function index and coagulation index before and after treatment and survival rate in 1 year after treatment and adverse events were compared between two groups.Results The total effective rate (73.33%vs. 66.67%,χ2=1.708,P=0.135) in TCM group was not significantly different from the control group. The survival rate in 1 year was (28.89%vs. 55.56%,χ2=10.406, P<0.01) in TCM group was significantly lower than that in control group. The adverse events (15.56% vs. 35.56%,χ2=8.728,P=0.002) in TCM group was significantly lower than that in control group. Compared with the control group, the CD3+ (65.20 ± 5.21 ratio/mlvs.62.94 ± 4.73 ratio/ml,t=2.106), CD4+ (36.86 ± 3.70 ratio/mlvs. 33.88 ± 3.04 ratio/ml,t=2.482), CD4+/CD8+(1.46 ± 0.30vs.1.36 ± 0.24,t=2.972) levels in TCM group increased significantly (P<0.05 orP<0.01). The PT (13.76 ± 1.70 svs. 14.73 ± 2.12 s,t=2.842), APTT (31.84 ± 2.53 svs. 36.97 ± 3.40 s,t=2.713), D-dimer (291.68 ± 25.15 mg/Lvs. 486.21 ± 44.32 mg/L,t=2.845), PLT (298.91×109/L ± 20.75×109/Lvs. 329.44×109/L ± 24.07×109/L,t=2.693) levels in TCM group significantly decreased (P<0.01).ConclusionHuoxue- Sanjie decoction combined with GEMOX chemotherapy regimen in the treatment of advanced pancreatic cancer for Shieryudu syndrome can improve the quality of life, prolong the survival time and helpreduce the risk of adverse events.
3.THE EXPRESSION OF REGENERATION GENE PROTEIN-2 IN THE SPINAL CORD TRAVERSE INJURY MODEL

Objective To investigate the expression of regeneration gene protein-2(Reg-2) after the transection injury in rat spinal cord. Methods Forty five SD rats were randomly divided into two groups: in the injury group,the SCI was produced by complete transection of the cord at the 9(th) thoracic level,and the sham operation rats were taken as the normal control.The animals were perfused at 1h,1d,3d,7d,14d after the operation,and the spinal cord was taken out at different time points.The expressions of Reg-2 were tested by immunohistochemical analysis and Western blotting,the double staining of Reg-2 and different neural cells special markers(GFAP,Olig2,NPY,CGRP,GAP-43,et al)was done with immunofluorescence method. Results The expression of Reg-2 was found at 1h after the spinal cord injury,reached the peak on day 3 after the injury in the neurons of dorsal horn and day 7 in the neurons of ventral horn.The high expression persisted for 1 week,then decreased gradually.Conclusion Reg-2 may work as an important growth factor and participate in the regeneration and rehabilitation process after the spinal cord injury.
4.Clinical analysis of opioid drug on treating 146 case moderate or severe cancer pain

Objective To investigate the distribution of moderate to severe cancer pain and administrated dose of opioid drugs in order to provide the reference for the treatment of moderate and severe cancer pain.Methods Retrospective analyzed the clinical data of 146 patients with moderate to severe cancer pain.The information were recorded including opioid use dose,cancer pain incidence,age,gender,height,weight,body surface area and type of tumor.Results There were no significant difference among patients with opioids drug administration in terms of gender,average daily oral dose(x2 =0.473,Z =-0.185,P > 0.05).Meanwhile cancer incidence rate was same in different age groups (x2 =2.280,P =0.684).The average daily opioid dose in patients with over 80 year old was the lowest among other age groups (Z =-2.745,-1.986,-2.141,-2.162;P <0.05).Cancer pain incidence in patients with hepatobiliary pancreatic tumors were 67.50% (27/40),highest than other different types cancer including metastatic tumor(62.50% (10/16)),urinary tumor (61.54% (8/13)),lung cancer (52.63% (50/95)),breast cancer and gynecological tumor (50.00% (17/34)),gastrointestinal tumor(41.94% (26/62)),other tumors 38.46% (5/13) and head and neck tumor(25.00% (3/12)),the difference was statistically significant (x2 =23.672,P < 0.05).The average day oral morphine dose of different types tumor were listed as followed from high to low in the order:160 (80,200) mg of metastatic tumors,120 (60,160) mg of breast and gynecological tumor,100 (40,125) mg of lung cancer,90(45,115) mg of urinary tract tumors,80(60,160) mg of other tumors,70(50,90) mg of hepatobiliary pancreatic tumor,60 (40,80) mg of gastrointestinal tumor,55 (40,70) mg of head and neck tumor.There were significant differences in terms of among oral morphine dose per day of different type tumors (H=14.280,P <0.05).No correlation was found between pain patients with height,body mass,body surface area and average daily dose of morphine (r =-0.045,-0.042,-0.046 ; P < 0.05).Conclusion No significant differences were found in terms of moderate to severe cancer pain among different type tumors at different and age groups.While there is significant difference in term of cancer pain incidence among different tumor types.The average daily amount of morphine in patients with cancer pain is not related to gender,height,body mass and body surface area.The average amount of morphine of patients with age over 80 years old is significantly lower than that of the other age groups,and the average amount of morphine t is related to cancer type.

[Objective] To observe the effect of extract of Carapax et Plastrum Testudinis (ECPT) on nuclear receptor in the proliferation of rat mesenchymal stem cell (MSC) in vitro. [Methods] The rat MSC dissociated from bone marrow by density gradient method were cultured and identified by marking of bromodeoxyuridine (Brdu) and staining of CD44. Then different doses of ECPT (3333, 333.3 and 33.33 ?g/mL) were respectively added into in-vitro cultured MSC for 12, 24, 72 and 120 hours. 6.Relationship between adolescent depressive symptoms with childhood psychological trauma and maternal pregnancy

Objective:
To explore the relationship between adolescent depressive symptoms, childhood psychological trauma and maternal illness during pregnancy, so as to provide scientific busis for the development of adolescent mental health.
Methods:
A stratified cluster random sampling method was employed to select 2 092 primary and secondary school students in Wuhan City from January to July 2022. Questionnaires were administered to investigate adolescent depressive symptoms, childhood psychological trauma, and maternal illness during pregnancy. The influencing factors of adolescent depression were analyzed.
Results:
A total of 139 adolescents reported depressive symptoms. The childhood trauma questionnaire (CTQ) score of the depression group was (57.49± 6.85 ), and the score in adolescents without depression group was (46.28±5.96)( t =21.14, P <0.05). Among the maternal diseases during pregnancy, hypertension accounted for 11.51% , diabetes 9.35%, cholestasis 7.19%, hypothyroidism/hyperthyroidism 7.91% , anemia 9.35% and viral hepatitis 3.60% in depression group, 5.79%, 4.71%, 3.64%, 3.07%, 4.30%, 1.18% in the group without depression, respectively, and the differences were statistically significant ( χ 2=7.35, 5.87, 4.42, 9.28, 7.49, 5.75, P <0.05). The proportion of academic stress in the depressed group and non-depressed group were 61.15% and 46.34%, respectively, and was statistically significant ( χ 2=16.04, P <0.05). Multivariate Logistic regression analysis showed that gestational hypertension ( OR=5.04, 95%CI =2.07-12.24), diabetes mellitus ( OR=4.49, 95%CI =1.85-10.91), anemia ( OR=3.68, 95%CI =1.51-8.94), high academic stress ( OR=3.52, 95%CI =1.45-8.56) and the Childhood Trauma Questionnaire ( OR=4.63, 95%CI = 1.91 -11.26) were the risk factors of depression in adolescents ( P <0.05).
Conclusion
Adolescent depression may be due to high academic stress, childhood psychological trauma, maternal hypertension, diabetes, thyroid dysfunction and anemia in pregnancy. It is suggested that relevant departments provide appropriate measures to reduce the risk of adolescent depression.
7.Effect of Nox4/NADPH oxidase on the process of osteoblast apoptosis in vitro

Objective To investigate the role and mechanism of nico-tinamide adenine dinucleotide phosphate oxidase 4 (NAPHD oxidase 4,Nox4)-mediated reactive oxygen species (ROS) generation on high-dose dexamethasone (DEX) induced apoptosis in osteoblasts.Methods According to culture conditions,3rd passage of murine osteoblastic MC3T3-E 1 cells were divided into control group,Dexamethasone group,Dexamethasone+NAC (N-acetyl-L-cysteine) group,NAC group,Dexamethasone+DPI (Diphenyleneiodonium) group and DPI group.24 hours after culture,the morphology of osteoblasts was observed by inverted phase contrast microscope.Cell viability was determined by MTT assay.The generation of ROS in osteoblasts was measured using a fluorescent probe DCFH-DA.The apoptosis of each group was observed through Hoechst staining.The mRNA level and protein expression of Nox4 were detected by real-time quantitative PCR and Western Blot.In addition,after silence of Nox4 with small interfering RNA (siRNA),the ROS generation was further detected by a fluorescent probe DCFH-DA.Results After treatment with 1000 nmol DEX for 24 hours,compared to control group,the results of inverted phase contrast microscope and MTT showed that osteoblasts in DEX group exhibited more obvious signs of shrinkage and deformation with decreased cell viability.After intervene with NAC and DPI,morphology of osteoblasts was good with increased viability of osteoblasts.Compared to control group (5.86%± 0.28%),the production of ROS in DEX group (45.14%±1.49%) was significantly increased (P=0.000).The apoptotic rates in DEX group (29.60%± 1.52%) was significantly increased compared with control group (4.12%±0.67%) (P=0.000).Compared to DEX group,the production of ROS generation in DEX+NAC group (28.06%±1.61%) and DEX+DPI group (23.70%±1.28%) was significantly decreased (P=0.000).It presented that NAC or DPI significantly decreased the formation of ROS.Compared to DEX group,the apoptotic rate in DEX+NAC group (8.94%± 1.47%) and DEX+DPI group (12.96%±2.03%) was significantly decreased (P=0.000).It presented that NAC or DPI significantly decreased osteoblast apoptosis.In addition,the Nox4 mRNA level in DEX group was 2.67-fold compared with control group (t=-10.301,P=0.009).The difference had statistically significance.The protein expression of Nox4 in DEX group was 2.37-fold compared with control group (t=-15.542,P=0.004).The difference has statistically significance.After silence of Nox4 by siRNA,the generation of ROS in DEX+Nox4 siRNA group (14.53%± 1.00%) was decreased by 16.92% compared with DEX group 31.45%±0.72% (P=0.000).The difference had statistically significance.Conclusion Nox4-mediated ROS generation plays an important role in osteoblasts apoptosis induced by high-dose dexamethasone.It provided us the new target in the management of Nox4 to provide possible therapy for steroid-induced avascular necrosis of the femoral head (SANFH).
