1.Expansion of chicken bone marrow mesenchymal stem cells by laminin culture system

BACKGROUND:Bone marrow mesenchymal stem cel s from chickens are important cel models for embryonic developmental biology, immunology and oncology research. However, it is difficult to keep bone marrow mesenchymal stem cel s with good undifferentiated potential in a large-scale expansion system.
OBJECTIVE:To establish a culture system in vitro with laminin coating to expand bone marrow mesenchymal stem cel s from chickens.
METHODS:Isolated bone marrow mesenchymal stem cel s from chickens were seeded in laminin-coated plates and traditional two-dimensional plates, respectively. After expansion in vitro, the morphological characteristics, expression of surface markers, expansion characteristics and adipogenic differentiation of bone marrow mesenchymal stem cel s in both conditions were analyzed and compared.
RESULTS AND CONCLUSION:There were no statistical differences in the morphological characteristics and expression of surface markers of bone marrow mesenchymal stem cel s expanded by laminin-coated plates and traditional two-dimensional plates. But, the expansion characteristics and adipogenic differentiation of bone marrow mesenchymal stem cel s cultured in laminin-coated plates were better than those in traditional two-dimensional plates. Laminin culture system could quickly amplify out of a large number of chicken bone marrow mesenchymal stem cel s with better proliferation ability and undifferentiated performance. Al above results indicated that a more efficient expansion system with laminin coating is established.

A 25-year-old women was admitted to the department of Neurology in Affiliated 2nd Hospital of Hainan Medical University due to recurrent syncope for 8 years and return for 2 months. She had multiple episodes of syncope at onset. She presented with the feeling of weakness in both lower limbs, and fatigue in the past year. She experienced pain in the waist and limbs joint in recent three months. Physical examination showed joint hyperactivity in metacarpophalangeal joints of both upper limbs, increased skin elasticity. Active-standing transcranial Doppler (TCD) test showed that the average heart rate (HR) and the average middle cerebral artery (MCA) blood flow velocity in the supine position were 79 beats/min and 62 cm/s, respectively; while the average HR and the average MCA blood flow velocity in the standing position were 126 beats/min, 47 cm/s. Meanwhile,the blood pressure was normal during the test of supine-to-standing TCD. Genetic testing indicated LDB3 transgenation. The patient was diagnosed as postural tachycardia syndrome (joint-hypermobility-related), Ehlers-Danlos syndrome, and relieved by fluid infusion and rehabilitation therapy.
4.Research Progress of Postoperative Right Ventricle Function by Imaging Evaluation in Congenital Heart Disease

The restoration of right ventricle dysfunction is essential to the maintenance of systemic circulation in patients after cardiac surgery.It also plays a significant part in planning surgical repair following the last stage of surgical palliation.Nevertheless,the complex geometry and wall motion of the right ventricle make it difficult to evaluate the right ventricle function with the noninvasive tools.Cardiac magnetic resonance(CMR)is the gold standard of the most commonly used technology for cardiac imaging,which is necessary to be involved in regular surveillance for patients undergoing complicated cardiac surgery.The application of new CMR technology mainly includes 4D flow hemodynamic measurement,MR1 mapping and tracking technology to evaluate the right ventricular function by detecting the myocardial injury.Echocardiography is the first-line method of postoperative assessment and appropriate for all kinds of congenital heart disease.Speckle-tracking echocardiography has been further development in recent years.Cardiac computed tomography has prominent advantages in detecting the vessels and pulmonary lesions,which is often used as an alternative to MRI.The development of artificial intelligence in image processing technology has greatly improved the diagnostic and predictive value of imaging examination.By comparing the characteristics of CMR,echocardiography and cardiac computed tomography,this paper reviews the research progress of these three imaging methods in the evaluation of the postoperative right ventricle function in congenital heart disease.
5.Analysis of prevalence,related factors of orthostatic intolerance in patients with Type 2 diabetes

To explore the prevalence and related factors of orthostatic intolerance in patients with Type 2 diabetes. Methods Patients with Type 2 diabetes were enrolled,admitted to the Department of Neurology from September 2020 to October 2021.General clinical data were collected. The active standing TCD test were performed on all the patients,and the changes of blood pressure,heart rate and cerebral hemodynamics were recorded from supine to standing for 1 min,3 min,5 min and 10 min. Orthostatic intolerance questionnaire were completed. Association between Orthostatic intolerance questionnaire score and results of supine to standing TCD test were analyzed. Results A total of 161 patients were enrolled for this study,70 (43.5%) with Symptoms of orthostatic intolerance and 91 (56.5%) without. Correlation analysis showed that Orthostatic intolerance questionnaire score have positive correlation with the magnitude of systolic pressure fall,diastolic pressure fall,Cerebral blood flow velocity fall and cerebral blood flow response to tilt score.(r=0.57、P= 0.000,r=0.44、P=0.000,r=0.30、P=0.022,r=0.24、P=0.044).Conclusion The proportion of OI in patients with Type 2 diabetes is high,which is associated with orthostatic blood pressure and cerebral blood flow decline.
6.Study on the 90-day Feeding Experimental Background Data of SD Rats for Drug Safety Evaluation

ObjectiveTo establish background data for a 90-day feeding trial of SD rats to ensure the reliability of research data. MethodsBackground data from six independent 90-day feeding trials of SD rats conducted by the National Center for Safety Evaluation of Drugs from 2020 to 2023 were summarized. These studies involved a blank control group of 120 SPF-grade 4-week-old SD rats, with an equal number of males and females, which were only given standard full-nutrient pelleted rat feed. After the quarantine period, the animals were observed for an additional 90 days, followed by intraperitoneal injection of Zoletil (50 mg/mL) for anesthesia, blood sampling, euthanasia, and necropsy. By analyzing the data from the blank control group, a relevant background database for SD rats was established. ResultsBoth male and female rats exhibited steady weight gain, with a more pronounced increase in male rats. Within 90 days, the average body weight of male and female rats increased to over 500 g and 300 g, respectively. Three weeks later, the average daily food intake of male rats stabilized at approximately 25~28 g per rat, while that of female rats remained stable at approximately 16~19 g per rat. The food utilization rate of all animals gradually decreased from the first week of the experiment. In the white blood cell (WBC) differential count results, significant differences were observed in the counts of WBCs, neutrophils (Neut), lymphocytes (Lymph), and monocytes (Mono) between males and females (P<0.001). However, there were no significant differences in the percentages of neutrophil (%Neut), lymphocyte (%Lymph), and monocyte (%Mono) between the sexes (P>0.05). The average red blood cell count (RBC), hemoglobin concentration (HGB), hematocrit (HCT), platelet count (PLT), prothrombin time (PT), and activated partial thromboplastin time (APTT) were higher in male animals than in female animals (P<0.05). The average values of alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), creatine phosphokinase (CK), lactate dehydrogenase (LDH), glucose (GLU), and triglyceride (TG) in male rats were higher than those in female rats (P<0.05). The urinary pH range for male animals was 5.0 to 8.5, while for female animals it was 6.5 to 9.0. The majority of male animals had a urinary specific gravity lower than 1.020, and the majority of female animals had a urinary specific gravity lower than 1.015. The weights of various organs (excluding the adrenal glands and reproductive organs) in male animals were heavier than those in female animals (P<0.001), while the organ/body weight ratios (excluding the kidneys and reproductive organs) of female animals were higher than those of male animals (P<0.001). ConclusionThis study summarizes the background reference ranges for body weight, food intake, hematology, and serum biochemistry indicators in SPF-grade SD rats in the untreated control group from six 90-day feeding trials conducted by the National Center for Safety Evaluation of Drugs. It provides important reference data for related research. By summarizing the background and spontaneous histopathological changes in rats, this study aids in the standardization and normalization of subsequent research, as well as in the evaluation and analysis of abnormal results.
