Objective To study the relationship between gastrin and c-myc, c-fos expression in colorectal cancerous tissue and the mechanism of gastrin effect on colorectal cancer.Methods The gastrin and c-myc, c-fos expression in 48 cases of colorectal cancerous tissue and cancer-adjacent mucosa were detected with immunohistochemistry techniques. Results The positive rate of gastrin in colorectal cancerous tissue was 39.58%. The rate of the well differentiated adenocarcinoma was higher than that of the poorly differentiated and mucinous adenocarcinoma(P<0.05). The positive rates of c-myc and c-fos in colorectal cancerous tissue were higher than those in cancer-adjacent and normal mucosa. The positive rate of c-myc and c-fos in the group with gastrin positive expression were 78.94% and 73.68%, higher than those in the group with negative gastrin expression(37.93% and 31.04%). Conclusion Some of colorectal cancer cells formed and secreted gastrin through autocrine. The increase of c-myc, c-fos etc oncogene expression probably stimulate the cancer cells proliferation.

4.EFFECTS AND CLINICAL SIGNIFICANCE OF PROGLUMIDE IN GASTRIN-STIMULATING CELL PROLIFERATION ON HUMAN COLON CELL LINE SW480

Objective To investigate the role and clinical significance of proglumide (PGL) in gastrin-stimulating cell proliferation on human colon cell line SW480.Methods Effects of proglumide on the viable cell count (A value), IP3 (CPM value), [Ca2+]i and PKC activity of human colon cell line SW480 were evaluated in vitro by MTT assay, 3H-myo-inositol incorporation, fluorescence measurements and γ-32P-ATP incorporation. Results The results showed that A value of PGL+PG group is lower than that of PG or control group (P<0.01 in both groups); The concentration of IP3 or [Ca2+]i in PGL+PG group is lower than that in PG group (P<0.05). Conclusion These results indicate effects of proglumide may be through the phosphoinositide signalling pathway on gastrin-stimulating cell proliferation in human colon cell line SW480. It will provide an experimental evidence for antisingnal transduction therapy for patients with colon cancer.
5.Determination of CAMP and CGMP levels in the serum and breast tissues of patients with breast cancer

The levels of cyclic nucleotides(CAMP and CGMP)in the serum and braest tissues of 60 patients with breast cancer or benign breast diseases were measured.It was found that the CAMP level was significantly lower,the CGMP significantly higher and the CAMP/ CGMP ratio was reduced in the serum and breast tissues of the patients with breast cancer.Our findings suggest that CAMP and CGMP might be factors of importance in the origin and development of breast cancer.
6.Evaluation of MR Breast Coil on Axillary Lymph Nodes

Objective To study the MRI features of normal and abnormal axillary lymph nodes so as to explore the evaluation of MR breast coil on metastasis of axillary lymph nodes in patients with breast cancer before operation.Methods MR scanning was made on 119 axillas from 24 healthy volunteers,23 patients with benign breast disease and 43 patients with breast cancer with MR breast coil.The shape,border,size,number and signal intensity of axillary lymph nodes in every group were comparatively studied.The features of the axillary lymph nodes of the patients with breast cancer were compared with the histopathologic results.Results Axillary lymph nodes were detected in 15 out of the 48 healty volunteer axillas (31.25%).The shape,border,size,number and signal intensity of axillary lymph nodes had no significant difference between the patients with benign breast disease and the patients with breast cancer with no metastasis lymph nodes (LN-)and the healthy volunteers.The shape,border,size,and enhancement pattern of the patients with axillary lymph nodes metastasis(LN+)were statistically different from that of the above mentioned three groups.Conclusion MRI is an effective method to evaluate the status of axillary lymph nodes before operation.It provides useful evidences for clinical operation and treatment.
