1.Analysis of the therapeutic effects of orthodontic therapy with mandibular first molar extraction

Objective:To evaluate the therapeutic effects of orthodontic therapy with mandibular first molar extraction.Methods:77 cases of malocclusion were divided into 3 groups:Bilateral mandibular first premolar extraction group(A,30 cases),unilateral man-dibular first molar extraction group(B,27cases)and bilateral mandibular first molar extraction group(C,20 cases).The OGS indexes were measured before and after orthodontic treatment and statistically analysed.Results:Remarkable occlusal improvement was seen in all three groups after treatment(P<0.01 ),but the variation of OGS indexes had no significant difference among the 3 groups(P>0.05).OGS indexes among the three groups after treatment were significantly different(P<0.05).The improvement in group A was more significant than that in group C(P<0.01)and the improvement in group C was more significant than that in group B(P<0.01). Conclusion:Orthodontic therapy with bilateral mandibular first molar extraction is effective in orthodontic treatment of malocclusion, but is not as effective as that with bilateral mandibular first premolar extraction.
2.Endoscope Disinfection in 8 Hospitals of One City:Present Situation and Countermeasure

OBJECTIVE To know the present situation of management of endoscope disinfection in order to improve the disinfection quality.METHODS A questionnaire and on-spot-survey were sent to 8 hospitals equipped with endoscope.RESULTS Eight different grade hospitals had a system of endoscope management established and integrated into their quantity management system.One hospital had isolated cleaning and sterilizing rooms,of which 3 hospitals were well ventilated;6 hospitals had special storage cabinets for endoscope,for endoscope as well as cleaning and sterilizing troughs.All of the hospitals used multienzymes and timers when doing cleaning.CONCLUSIONS There are some problems in the present situation.The way of management monitoring and occupational protection must be further improved to reduce the nosocomial infection.
3.Analysis of the changes of serum throid hormones in post thoracic surgery patients

Objective To investigate the relationship between thyroid hormone levels and the attack of arrhythmia after thoracic surgery.Methods Information of 44 patients after thoracic surgery form September 2012 to February 2015 in General Hospital of Tianjin Medical University was collected, and divided in to the arrhythmia group and non-arrhythmia group.A retrospective analysis was carried on their ECG, thyroid hormone levels, and physical health, and the parameters were then compared between the two groups.The correlation of triiodothyronine (T3) level and Acute Physiology and Chronic Health Evaluation (APACHE Ⅱ) was analyzed.Results The arrhymia patients' serum T3 and free triiodothyronine (FT3) levels were below normal levels, while the serum T3 and FT3 levels of the non-arrhythmia patients were within normal range, and the differences between the two groups were significant (P<0.05).No notable change of other thyroid hormones levels and correlation of T3, FT3 and APACHE Ⅱ score were found.Conclusions There is a significant difference between the arrhythmia and non-arrhythmia groups in serum T3 and FT3 levels.Perioperative thyroid hormone replacement therapy is worthy of consideration and trial in the clinical prevention and treatment of arrhythmia and prognostic improvement of patients.
4.Effect of rapamycin on apoptosis of acute myeloid leukemia THP-1 cells induced by idarubicin

Objective:To investigate the effect of rapamycin (Rapa) on apoptosis of acute myeloid leukemia THP-1 cells induced by idarubicin (IDA) and its molecular mechanism.Methods:The THP-1 cells were treated with 10, 20, 40 and 80 nmol/L Rapa for 1 h, and the cells without Rapa treatment were set up. Western blot was used to detect the conversion of autophagy marker LC3 protein in THP-1 cells (the ratio of LC3Ⅱ/LC3Ⅰ), flow cytometry was used to detect the apoptotic rate, and the pretreatment concentration of Rapa was determined. THP-1 cells were treated with different concentrations of IDA for 24 h, the cell proliferation inhibition rate of IDA for THP-1 cells was detected by CCK-8 method, and the half maximal inhibitory concentration ( IC50) was calculated. THP-1 cells with or without Rapa treatment were treated by IDA with the concentration of lower than IC50 for 24 h, CCK-8 method was used to detect cell proliferation inhibition rate, flow cytometry was used to detect cell apoptosis, real-time fluorescent quantitative polymerase chain reaction was used to detect the expression changes of autophagy-related genes Beclin-1, LC3 and p62, and Western blot was used to detect the conversion of autophagy marker LC3 protein. Results:The ratio of LC3Ⅱ/LC3Ⅰ in THP-1 cells treated by 20 nmol/L Rapa was higher than that in the untreated cells ( P=0.002 4). The apoptotic rate in THP-1 cells treated by 80 nmol/L Rapa was higher than that in the untreated cells ( P=0.007 3). According to the results of Western blot and flow cytometry, 20 nmol/L Rapa was selected as the pretreatment concentration. The IC50 of IDA for THP-1 cells treated with IDA for 24 h was 59.874 nmol/L. After treated with 50 nmol/L IDA for 24 h, the proliferation inhibitory [(69.67±5.03)% vs. (41.67±3.51)%] and apoptotic rates [(74.35±4.83)% vs. (41.25±5.24)%] in THP-1 cells pretreated by Rapa were higher than those in the unpretreated cells (both P<0.05); the Beclin-1 and LC3 mRNA expression levels and the ratio of LC3Ⅱ/LC3Ⅰ in THP-1 cells pretreated by Rapa were higher than those in the unpretreated cells, and the expression of p62 mRNA was lower than that in the unpretreated cells (all P<0.05). Conclusion:Rapa can enhance the apoptosis of THP-1 cells induced by a relative low dose of IDA, which may be achieved through inducing excessive autophagy in THP-1 cells.
5.Research progress of miroRNA in the mechanism of cardiac hypertrophy and its role as new drug targets

Cardiac hypertrophy is a common physiological or pathological process, and pathological cardiac hypertrophy can lead to heart failure, sudden death, etc. The role of microRNA (miRNA or MIR) in myocardial hypertrophy has gradually attracted public attention. miR-1 plays a certain protective role in the occurrence of cardiac hypertrophy. miR-133 is a key factor in the establishment of mast gene program, which is very important for the development of myocardial hypertrophy. Carvedilol and other drugs can regulate the expression of miR-133. miR-208a plays an important physiological role in the cardiovascular system, and its expression level changes dynamically in a variety of cardiovascular diseases such as cardiac hypertrophy, which is closely related to the progression and prognosis of the disease. The expression of miR-199a is up-regulated in pressure-overload cardiac hypertrophy, and it is found that miR-199a can inhibit autophagy of cardiomyocytes and induce the occurrence of cardiac hypertrophy. miR-200c can protect cardiomyocytes through a variety of pathways. miRNA may become an important biomarker or drug therapeutic target for cardiac hypertrophy. With the deepening of the research on non-coding RNAs including miRNA, its regulation on the occurrence of cardiac hypertrophy and the pathological process of heart failure will be further revealed.
6.Tetrahedral framework nucleic acid carrying angiogenic peptide prevents bisphosphonate-related osteonecrosis of the jaw by promoting angiogenesis.

The significant clinical feature of bisphosphonate-related osteonecrosis of the jaw (BRONJ) is the exposure of the necrotic jaw. Other clinical manifestations include jaw pain, swelling, abscess, and skin fistula, which seriously affect the patients' life, and there is no radical cure. Thus, new methods need to be found to prevent the occurrence of BRONJ. Here, a novel nanoparticle, tFNA-KLT, was successfully synthesized by us, in which the nanoparticle tetrahedral framework nucleic acid (tFNA) was used for carrying angiogenic peptide, KLT, and then further enhanced angiogenesis. TFNA-KLT possessed the same characteristics as tFNA, such as simple synthesis, stable structure, and good biocompatibility. Meanwhile, tFNA enhanced the stability of KLT and carried more KLT to interact with endothelial cells. First, it was confirmed that tFNA-KLT had the superior angiogenic ability to tFNA and KLT both in vitro and in vivo. Then we apply tFNA-KLT to the prevention of BRONJ. The results showed that tFNA-KLT can effectively prevent the occurrence of BRONJ by accelerating angiogenesis. In summary, the prepared novel nanoparticle, tFNA-KLT, was firstly synthesized by us. It was also firstly confirmed by us that tFNA-KLT significantly enhanced angiogenesis and can effectively prevent the occurrence of BRONJ by accelerating angiogenesis, thus providing a new avenue for the prevention of BRONJ and a new choice for therapeutic angiogenesis.
