1.Changes of plasma mitochondrial coupling factor -6 and its significance in neonatal sepsis

Objective To investigate the variation of plasma mitochondrial coupling factor -6 and explore the significance in neonatal sepsis.Methods The study of 88 cases of pediatric inpatients in our hospital were divided into non-sepsis group (46 cases)and sepsis group (42 cases).CF-6 concentration in venous blood of all study subjects was determined by enzyme linked immunosorbent assay (ELISA).Re-sults The plasma CF-6 concentrations in peripheral venous blood of the patients with sepsis were signifi-cantly higher than those in non sepsis group(107.65 ±50.38)pg/ml vs (38.96 ±29.87)pg/ml,The con-centrations of CF-6 in the infected group were higher than those in the normal control group(P <0.05). Conclusions The concentration of mitochondrial coupling factor -6 in neonatal sepsis was significantly higher,and has a certain pathological and physiological significance in the pathogenesis of neonatal sepsis.
2.Changes and clinical significances of mitochondrial coupling factor 6 and cytochrome C in neonatal sepsis

Objective To evaluate the levels of plasma coupling factor 6(CF6) and cytochrome C(Cyt-c) in neonatal sepsis,and to explore the clinical significance in neonatal sepsis.Methods A total of 88 neonates admitted to Hunan Children's Hospital from January 2015 to April 2015 were collected.Neonates were divided into non-sepsis group(n=42) and sepsis group(n=46).According to the severity of infection,the non-sepsis group was further divided into non-infection group(n=20) and common infection group(n=22);the sepsis group was further divided into general sepsis group (31 cases,no organ failure) and severe sepsis group (15 cases,combined with multiple organ failure).Femoral venous blood was collected in all patients before the use of antibiotics after admission.The levels of Cyt-c and CF6 in plasma were measured by ELISA,and the levels of C-reactive protein(CRP),procalcitonin (PCT) were measured.The changes of CF6 and Cyt-c between these groups were compared,and the sensitivity and specificity with the traditional sepsis index (CRP,PCT) were analyzed.The correlation between the levels of CF6,Cyt-c and neonatal critical illness score was analyzed.Results (1)In sepsis group,the levels of CF6 and Cyt-c[(109.7±8.9)pg/ml and (44.5±4.9)ng/ml] were significantly higher than those in the non-sepsis group[(46.3±6.0)pg/ml,(31.8±6.7)ng/ml,P<0.01,respectively].(2) In the non-infection group,common infection group,general sepsis group and severe sepsis group,the levels of CF6 were (32.1±8.9)pg/ml,(59.3±7.2)pg/ml,(79.3±5.9)pg/ml,and (172.6±6.1)pg/ml,respectively;the levels of Cyt-c were (29.3±8.6)ng/ml,(35.4±4.1) ng/ml,(43.1±5.9) ng/ml,and (44.5±5.9)ng/ml,respectively.The differences between these groups were significant(P<0.01).(3)The receiver operating characteristic curve showed that the sensitivity and specificity of CF6 were 0.761,0.732,and the Cyt-c were 0.739,0.714.(4)Cyt-C and CF6 were negatively correlated with the neonatal critical illness score(r=-0.599,P<0.001;r=-0.337,P<0.01).Conclusion The levels of CF6 and Cyt-c increase in neonatal sepsis.The damage of mitochondria may be one of the pathological mechanisms in neonatal sepsis.The levels of CF6 and Cyt-c were closely related to the severity of neonatal sepsis.
3.Evaluation on the efficacy of human umbilical cord derived-mesenchymal stem cell transplantation in liver cirrhosis patients with ascites in a prospective and control trial

ObjectiveTo evaluate the one-year follow-up of the therapeutic efficacy of human umbilical cord derived-mesenchymal stem cell (UC-MSC) transplantations in decompensated liver cirrhosis patients with ascites.MethodsFifty-four liver cirrhosis patients with ascites in Research Center for Biological Therapy in 302 Military Hospital were divided into treatment group (n=38) and control group (n=16) in a prospective controlled single-blinded trial.UC-MSC (0.5-1.0) × 106/kg and saline were intravenously transplanted into patients monthly for 3 times in treatment group and control group,respectively.The liver function,hepatitis B virus (HBV) DNA level,ascites and the model for end-stage liver disease(MELD)scores at different time points were compared between two groups.The comparison between groups was done by Mann-Whitney U test,and the data before and after transplantations were compared by Wilcoxon signed rank sum test.ResultsThere were no significant differences of alanine transaminase (ALT),total bilirubin (TBil),cholinesterase (CHE),HBV DNA positive rate and MELD scores at different time points between two groups (P>0.05).However, the albumin ( A1b)level was significantly increased after 36 weeks of UC-MSC transplantation in treatment group, which were (28.47±4.45)g/L at week 0 and ( 34.82±4.50)g/L at week 48 (P=0.046). Meanwhile, the ascites reduced markedly in treatment group with (46.6 ±30.6) mm at week 0 and (6.6±13.6) mm at week 48,which were significantly different from control group at the end of follow-up (P =0.037). Conclusion UC-MSC transplantations may help to increase A1b level and reduce ascites in patients with decompensated liver cirrhosis.
4. Effects of chronic stress during pregnancy on learning and memory ability of offsprings and its mechanisms

OBJECTIVE: To explore the effect of chronic stress during pregnancy on the learning and memory ability, amino acid neurotransmitters in hippocampus of offsprings and its mechanism. METHODS: The specific pathogen free adult Wistar rats were used. There were 10 rats each in female model group and control group, 10 rats in male mating model group and 5 in controlled mating group. A rat model of chronic stress during pregnancy of the female model group was established by the 21-day chronic unpredictable mild stress experiment. From the 3 rd day of the experiment, the model group and model mating group, control group and control mating group were caged separately.Blood was collected from the medial canthal vein from the rats on the day before stress and then on the 1 st, 7 th, and 14 th day after stress. Plasma corticosterone levels of maternal rats were determined by radioimmunoassay. Learning and memory responses of offsprings 42 days after birth were tested by Morris water maze and Y-maze. The levels of glutamate and γ-aminobutyric acid in hippocampus of offsprings were detected using enzyme linked immunosorbent assay. RESULTS: The plasma corticosterone level in maternal rats of model group was higher than that in control group at the same time points on the 7 th and 14 th day after stress(P<0.05). Compared with offsprings in the control group, the escaping latency of the model offsprings was prolonged(P<0.05). The training time needed to form correct memory increased(P<0.01), while the number of crossing platform and the correct response rate of memory retention test decreased(P<0.05), the level of glutamate in hippocampus decreased(P<0.01), and the level of γ-aminobutyric acid increased(P<0.01) in the model offsprings. The escape latency, the number of crossing platform, the training times and the correct response rate of model offsprings were associated with the plasma corticosterone levels of their mothers and amino acid neurotransmitters of offsprings(P<0.05). CONCLUSION: Chronic stress during pregnancy can reduce the learning and memory abilities of offsprings. The learning and memory ability was associated with the plasma corticosterone level of mother rats, the decrease of glutamate and the increase of γ-aminobutyric acid in the hippocampus.
5. Effect of curcumin on mitochondrial gene and high mobility group box 1 protein in sepsis myocardial injury of rats

Objective:
To observe the effect of curcumin on myocardial mitochondrial related gene (mtATP6), high mobility group box 1(HMGB1) mRNA expression and plasma HMGB1 level in rats with sepsis, and to explore a new strategy for protection of myocardial injury in sepsis.
Methods:
A rat sepsis model was established by cecal ligation and puncture (CLP). Thirty male Sprague-Dawley rats were randomly (random number) divided into the sham operation group (sham group), sepsis group (CLP group), and curcumin intervention group (Cur group), with 10 rats in each group. Rats in the Cur group were treated with curcumin by intraperitoneal injection at 100 mg/kg, and normal saline was injected into rats in the sham and CLP groups. Five rats were randomly (random number) sacrificed at 6 h and 24 h after modeling. The pathological changes of myocardial tissue were observed under light microscope. The levels of CK and CK-MB in serum were detected. The HMGB1 level in plasma was detected by ELISA. Real-time PCR was used to detect the expression of HMGB1 mRNA and mtATP6 in myocardial tissue. 6. A single center study on the clinical features and treatment of infectious mononucleosis in children

Objective:
To investigate the clinical features of children with infectious mononucleosis (IM), to compare the difference of therapeutic effects between general treatment to antiviral therapy for IM.
Methods:
This prospective study analyzed the clinical data and laboratory test results of 201 cases with IM in our hospital from January 1, 2016 to December 31, 2016. The follow-up period was 6-12 months. The patients were divided into two groups according to the order of admission. The clinical symptoms and laboratory test results of the two groups were observed after hospitalization.
Results:
Of the total of 201 patients, male to female is 1.72∶1; Age: 8 months to 13 years 6 months (average 4.8±2.8 years), The disease frequently occurred in summer and autumn, accounted for 64.18%.The major clinical manifestations was fever (97.51%), angina (79.10%), enlarged of lymph node(68.66%), eyelid swelling (67.16%), hepatomegaly (53.73%) and splenomegaly (46.77%). There was no statistical difference in duration of fever, improved angina time, lymph nodes(liver, spleen) enlargement recovery time, heterotypic lymphocytes normalization time, lymphocyte function normalization time.There was significant difference in reducing the serum/plasma or total blood EBV-DNA in the short term between antiviral group and general treatment group (
