China consumes and exports traditional Chinese medicinal resources the most in the world. However, we cannot anchor our hope on field production of traditional Chinese medicinal materials and their active ingredients, due to limited land resources. Therefore, the development of biotechnology is of great importance for China to solve the problem of traditional Chinese medicinal resources. Plant cell culture is an important approach for the sustainable development of precious medicinal resources. This essary summarizes the optimization of conditions for medicinal plant cell culture, the regulation of secondary metabolic pathways and cell bioreactor culture, and realizes that the authentic commercial production of more medicinal plants requires efforts from all aspects.

2.Influencing factors on culture of medicinal plants adventitious roots.

3.Prediction of Secondary Structure and B Cell Epitope for Capsid Protein of SVDV

The secondary structure of Capsid protein was predicted by the methods of Chou-Fasman,Garnier-Robson and Karplus-Schultz based on the sepuence of capsid protein gene of Swine Vesicular Disease Virus (SVDV) and hydrophilicity. Surface probility plot and antigenic index for capsid protein were obtained by the methods of Kyte-Doolittle, Emini and Jameson-wolf, respectively, Combining the results according to these methods, the B cell epitopes for capsid protein of SVDV were predicted. The results showed that there are much flexible region such as coil region and turn region in capsid protein of SVDV, there are more predominant B cell epitopes in VP1 than in VP2 and VP3. This study would be helpful for identification of B cell epitopes for capsid protein using experimental methods and research of reverse vaccine of SVDV.
4.Cultivation of Panax ginseng adventitious roots in bubble bioreactors.

OBJECTIVETo study cultivation of Panax ginseng adventitious roots in bubble bioreactors.
METHODThe adventitious roots were obtained through tissue culture different types of bioreactors. The contents of ginsenosides Re, Rb1 and Rg1 were determined by HPLC while the contents of polysaccharides were determined by ultraviolet spectrophotometry.
RESULTThe results showed that of the three types tested, the most efficient bioreactor for cultivation of the ginseng adventitious roots was the cone-type bioreactors (with the 120 degrees ), in which, the growth curve of adventitious roots was S-shaped. The maximum biomass was obtained on the 40th day, with the fresh weight, dry weight and growth rate reaching the maximum, which were 113.15 g, 9.62 g and 63.13 times respectively, and the concomitant contents of polysaccharide and ginsenoside were 2.73% and 2.25 mg x g(-1).
CONCLUSIONThe results showed that the most efficient bioreactor for cultivation of the ginseng adventitious roots was the cone-type bioreactors (with the 120 degrees). These results provide a theoretical reference for developing an efficient production process of active metabolites of ginseng in the scale-up cultivation.

5.Advances and thinking about prevention and treatment of inflammatory bowel diseases by acupuncture and moxibustion.

OBJECTIVETo provide scientific foundations for clinical and acupoints researches on acupuncture for treatment of inflammatory bowel diseases.
METHODSReview the correlated documents of clinical experimental researches issued from 1995 to 2005, and the clinical and experimental documents about remedy of inflammatory bowel diseases by acupuncture and moxibustion were summarized and analyzed.
RESULTSAcupuncture and moxibustion had significant therapeutic effects on inflammatory bowel diseases, and the studies on the mechanisms have achieved some progresses, but the designs and the methods of these studies need to be improved.
CONCLUSIONAfterwards, the specificity of acupoints, and factors of influencing the specificity should be studied via effective diseases treated with acupuncture and moxibustion, so as sum up common regularity, rich and develop the theory about specificity of acupoints to guide clinical treatment of acupuncture and moxibustion.

6.Seroepidemical study of Coxsackievirus A 16, in four provinces, China, 2005.

OBJECTIVETo study the situation of 1- 5-years-old children's antibody against Coxsackievirus A group 16 strain (CVA16) in Guangdong, Heilongjiang,Yunnan Province and Xinjiang Uygur Autonomous Regions, China, 2005, it can offer scientific evidences for preventing and controlling CVA16 causative hand-food and mouth disease.
METHODSUsing microneutrilization test, to study 503 serum samples randomly selected from sera collected in 2005.
RESULTSPositive rate of anti-CVA16 antibody were 41.90%, 9.40%, 40.00% and 34.40% in Guangdong, Heilongjiang,Yunnan and Xinjiang, respectively. Antibody titer was relative low (average, 1: 6.1) and there was no statistical difference of geometry mean of antibody titer (GMT) among Guangdong, Heilongjiang, Yunnan (F = 0.97, 0.40, 1.06, respectively; P > 0.05), while there had statistical difference of GMT between Heilongjiang and other three regions( F = 10.61, P < 0.00).
CONCLUSIONSThere had probably existed local epidemic in some regions of Guangdong, Heilongjiang, Yunnan Province and Xinjiang Uygur Autonomous Regions, China, 2005 or even before, but the area and degree of transmission and epidemic had difference. Children aged from 1- 5-years-old were relatively susceptible population of CVA16 infection.

7.Clinicopathological classification and individualized treatment of breast cancer.

BACKGROUNDThe clinicopathological classification was proposed in the St. Gallen Consensus Report 2011. We conducted a retrospective analysis of breast cancer subtypes, tumor-nodal-metastatic (TNM) staging, and histopathological grade to investigate the value of these parameters in the treatment strategies of invasive breast cancer.
METHODSA retrospective analysis of breast cancer subtypes, TNM staging, and histopathological grading of 213 cases has been performed by the methods recommended in the St. Gallen International Expert Consensus Report 2011. The estrogen receptor (ER), progesterone receptor (PR), human epidermal growth factor receptor-2 (HER2), and Ki-67 of 213 tumor samples have been investigated by immunohistochemistry according to methods for classifying breast cancer subtypes proposed in the St. Gallen Consensus Report 2011.
RESULTSThe luminal A subtype was found in 53 patients (24.9%), the luminal B subtype was found in 112 patients (52.6%), the HER2-positive subtype was found in 22 patients (10.3%), and the triple-negative subtype was found in 26 patients (12%). Histopathological grade and TNM staging differed significantly among the four subtypes of breast cancer (P < 0.001).
CONCLUSIONIt is important to consider TNM staging and histopathological grading in the treatment strategies of breast cancer based on the current clinicopathological classification methods.

8.Study on differentially expressed genes of ulcerative colitis in the rat treated by herbs-partitioned moxibustion.

OBJECTIVETo probe the mechanism of herbs-partitioned moxibustion for treatment of ulcerative colitis.
METHODSThe rats with ulcerative colitis were randomly divided into 3 groups, normal control group, model group, and herbs-partitioned moxibustion group. The rats in the herbs-partitioned moxibustion group were treated by herbs-partitioned moxibustion at bilateral Tianshu (ST 25). BiostarR-40s gene chip was applied to detect the differentially expressed gene of their colonic tissues and fluorescence quantitative polymerase chain reaction was employed to confirm the results of the microarray analysis with interleukin-1 beta messenger RNA (IL-1beta mRNA) expression.
RESULTSOf 174 differential expression genes identified, 28 genes (including 7 known genes) elevated in rats of ulcerative colitis were down-regulated and 146 genes (including 42 known genes) reduced in the rat of ulcerative colitis were up-regulated after herbs-partitioned moxibustion treatment.
CONCLUSIONMany abnormally expressed genes are involved in occurrence of ulcerative colitis and herbs-partitioned moxibustion can regulate expression of IL-1beta and other genes to exert therapeutic effect.

9.The mechanism of sensing postural changes in human ears by numerical simulation

Objective To study the general mechanical behavior pattern of sensing angular velocity by 3 pairs of human semicircular canals.Methods Sinusoidal head rotation of a healthy subject was simulated by finite element method to analyze the dynamic responses in the above 3 pairs of canals in the left and right inner ear.Results Compared with the complicated node displacement distribution,the volumetric strain distribution of cupula was regular.Specifically,each pair of cupulae expanded or contracted within the same amplitude with the same response frequency to the angular velocity.The ratio of absolute volumetric strain among the horizontal semicircular canal cupula (HC cupula),the anterior semicircular canal cupula (AC cupula),and the posterior semicircular canal cupula (PC cupula),was approximately 1.00∶0.80 ∶ 1.72,which kept constant.In addition,the volumetric strain of HC cupula was in phase with that of AC cupula,but showing 14.4° out of phase with the angular velocity,and 180° out of phase with that of PC cupula.Conclusions The volumetric strain of cupula can preferably characterize the directional coding function of semicircular canal,and the amplitude,frequency and phase of the cupular volumetric strain can encode those of the stimulated angular velocity,respectively.These results will lay the foundation for establishing quantitative relationship of vestibulo-ocular reflex,and provide theoretical references for quantitative assessment for vestibular function by nystagmus examination.
10.Initiating Characteristics of Early-onset Type 2 Diabetes Mellitus in Chinese Patients.

BACKGROUNDType 2 diabetes mellitus (T2DM) has traditionally been considered to affect mainly the elderly; however, the age at diagnosis has gradually reduced in recent years. Although the incidence of young-onset T2DM is increasing, it is still not fully clear the onset characteristics and risk factors of early-onset T2DM. The aim of this study was to describe the initiating characteristics of early-onset T2DM in Chinese patients and evaluate the risk factors for diabetes mellitus.
METHODSThis cross-sectional controlled study was performed using a questionnaire survey method in outpatients of multiple centers in China. A total of 1545 patients with T2DM with an age at onset of <40 years were included, and the control group consisted of subjects aged <40 years with normal blood glucose level.
RESULTSIn patients with young-onset T2DM, the mean age and initial hemoglobin 1Ac at diagnosis were 32.96 ± 5.40 years and 9.59 ± 2.71%, respectively. Most of the patients were obese, followed irregular diet pattern and sedentary lifestyle, had life or work pressure, and had a family history of diabetes mellitus. Compared with subjects with normal blood glucose level, logistic regression analysis showed that waist-to-hip ratio (odds ratio [OR] 446.99, 95% confidence interval [CI] 42.37-4714.87), family history of diabetes mellitus (OR 23.46, CI 14.47-38.03), dyslipidemia (OR 2.65, CI 1.54-4.56), diastolic blood pressure (OR 1.02, CI 1.00-1.04), and body mass index (OR 0.95, CI 0.92-0.99) are independent factors for early-onset T2DM.
CONCLUSIONSWe observed that abdominal obesity, family history of diabetes mellitus, and medical history of hypertension and dyslipidemia are independent risk factors for early-onset T2DM. It is, therefore, necessary to apply early lifestyle intervention in young people with risk of diabetes mellitus.
