In this research, the zuelacmycin-producing strain Streptomyces venezuelaevar. qinlingensis RL-2 was used as original strain, the spore suspension of which was induced by UV, LiCl and the compound treatmeat (UV+LiCl) respectively. The zuelacmycin-high-yield strain UVL-108 was obtained by the treatment that the exposure time under UV irradition was 45 s and the concentration of LiCl was 0.4%. The heredity characters of mutant UVL-108 were stable in succesive six generations. The antibacterial activity and the fermentation titer of mutant UVL-108 were determinded by bidirectioned culture and mycelial linear growth respectively. The results demonstrated the antibaterial activity of mutant UVL-108 was improved by 77%, and the relative toxicity of fermentation to Phyricularia grisea was improved by two times compared with the original strain.

The study provide a theoretical basis for the industrial production of a elastase-high-yield strain which was isolated from the straw, and the selected strain was identified. The screening strategy included casein(skim milk) plate selecting and elastin(beef tendon) re-screening. And then, the morphological, physiological and biochemical characteristics as well as 16S rRNA sequence homology of the selected strain were studied. Finally, the strain LSF-97 which had a excellent decomposing ability to beef tendon was obtained. The results showed that the strain LSF-97 is relative to the Bacillus pumilus with 100 % similar in sequence under the phylogenetic tree, the morphological and physiological and biochemical characteristics are also consistent with pattern of bacteria. So it was identified as Bacillus pumilus.

Background Nearly over 40 years have elapsed since the original findings of visual cortical plasticity, but none of drug has been found for curing amblyopia effectively. Objective The goal of this study was to investigate the effects of different dose of levodopa on flash visual evoked potential(F-VEP)and morphology of visual cortex cells in monocular deprivation rat and explore the possible mechanism of curing amblyopia. Methods Monocular deprivation model were established by suturing eyelids of 30 2-week-old Sprague Dawley(SD)rats for 4 weeks. The 30 SD rats were then divided into 3 groups randomly and 10 rats for each group.Normal saline.20 ms/kg levodopa,80 ms/kg levodopa were intragastrically administered once per day after modeling respectively for 4 weeks.F-VEP was recorded after establishment of model and administration of drug respectively. The rats were sacrificed and the visual cogex was obtained for histological examination, and TUNEL technique was used to assess the structural change of visual cortex. Results The latency of P1 wave was significantly longer in the deprived eye than the normal eyes(P<0.05).After administration of levodopa, the latent periods of Pl wave in the deprived eye were obviously shortened in comparison with before administration of levodopa in 20 ms/kg and 80 mg/kg levodopa group (P<0.05).The difference values of latent period of P1 wave between before and after administration of drug showed statistically significant change in three groups(P<0.05).No evidently alterations were found in the amplitude differences of N1 P1 and P1 N2 waves among three groups(P>0.05).The number and structure of neurons in contralateral visual cortex of non-deprived eye were normal. However, the numbers of neurons in deprived eye were significantly less and presented the signs of para-apoptosis in normal saline group. In 20 mg/kg levodopa groups, the alterations of number and morphology in neurons of rat visual eogex were slight.TUNEL assay revealed that the numbers of positive neurons in contralateral visual codex of non-deprived eye were 2.20±1.23.while those in deprived eye were 53.7±9.36,27.20 4±5.96 and 10.70±3.23 in normal saline group,20 mg/kg and 80 mg/kg levodopa group respectively, showing a significant difference among them(P>0.05).After usage of levodopa, the numbers of positive neurons was negatively correlated with the difference value of P,latent period of VEP(r=-0.815,P=0.000).Conclusion Levodopa has a therapeutic effect on rat deprived eye,and its possible mechanism is inhibiting the para-apoptosis of neurons and participating in the development and plasticity of visual system.

Objective To compare the prophylactic efficacy of combination of ganciclovir and acy- clovir or acyclovir alone against cytomegalovirus pneumonia in renal transplant recipients. Methods A to- tal of 217 renal transplant recipient(124 men and 93 women;mean age,32 years;age range,16-72 years) were divided into 3 groups randomly. In 51 cases, acyclovir was taken orally at a dose of 400 mg,3/d,from the third d to 3 months after transplantation. In 74 cases, ganciclovir was administered at a dose of 250 mg/d intravenously from the 21st d to 27th d to replace Acyclovir. In 92 cases,no prophylaxis against eytomegalov- irus pneumonia was performed. All patients were followed 3 months after transplantation.Comparison of the incidence rates of cytomegalovirus pneumonia among the 3 groups was performed using Fisher's exact test. Results Cytomegalovirus pneumonia developed in 20 cases in the 3 groups, including 4 cases(5.4%) in combined use group,2 cases(3.9%)in acyclovir alone group, and 14 cases(15.2%)in control group. Significant difference existed between the 2 experimental and control groups(P<0.05).However,no signifi- cant difference existed between the 2 experimental groups(P>0.05).Of the 20 cases,17(85.0%)were cured, and 3 died of respiratory failure. Conclusions Ganciclovir and acyclovir have prophylactic effect a- gainst cytomegalovirus pneumonia in renal transplant recipients. These 2 medications are inexpensive, and the patients have good compliance.

Objective To investigate the current status of mouse euthanasia methods in China, and to provide the reference and basis for the administrative authorities of laboratory animal management to master the implementation of"euthanasia"and formulate related policies. Methods Research papers containing the terms of"mouse"and"execution"in Chinese characters during the period from 2015 to 2016 were searched in Wanfang database, and statistical analysis was performed with the articles meeting the searching criteria. Results A total of 890 research articles met the searching criteria,of which 351 articles clearly described the killing method, accounting for only 39.44%. The mouse-killing methods included cervical dislocation, decapitation, exsanguination and sampling after anesthesia, excessive anesthesia, abdominal aorta bleeding and carbon dioxide asphyxiation, among them cervical dislocation accounted for the highest rate,75.78%. Conclusions The current implementation of mouse euthanasia methods in our country has been far from optimistic. The mouse euthanasis methods have often been ignored in scientific articles and the description of the methods is not standardized. In order to promote the effective implementation of the regulations related to mouse euthanasia,it is needed to promote the study of related techniques and to strengthen personnel training.

This study is to evaluate the therapeutic effect of fibroblast growth factor 21 (FGF21) on type 2 diabetic mice model and to provide mechanistic insights into its therapeutic effect. Type 2 diabetic animal model was established with high calorie fat diet and low dose streptozotocin (STZ) injection. Mice were then randomized into 5 groups: model control, FGF21 0.25 and 0.05 μmol x kg(-1) x d(-1) groups, insulin treatment group. Ten age-matched normal KM mouse administered with saline were used as normal controls. Serum glucose, insulin, lipid products and the change of serum and liver tissue inflammation factor levels between five groups of mouse were determined. The results showed that blood glucose, insulin, free fatty acids (FFAs), triglycerides, and inflammatory factor average FGF-21 of type 2 diabetes model group and normal control group were significantly higher (P < 0.01), while compared with insulin group, no difference was significant. Average blood glucose, insulin, blood lipid and inflammatory factor of FGF-21 treatment group compared with type 2 diabetes group was significantly lower (P < 0.01) and insulin group has no difference with the model control group. The results of OGTT and HOMA-IR showed that insulin resistance state was significantly relieved in a dose-dependent manner. Thus, this study demonstrates that FGF-21 significantly remits type 2 diabetic mice model's insulin resistance state and participates in the regulation of inflammatory factor levels and type 2 diabetes metabolic disorders.

OBJECTIVETo investigate peripheral blood monocyte (PBMC) gene expression profile in patients with fulminant hepatic failure (FHF) by cDNA microarray.
METHODSMicroarrays consisting of 8,192 human cDNAs and labelled cDNAs prepared from PBMC in both 10 FHF patients and 10 asymtomatic surface antigen carriers (ASC) were applied to analyze gene expression. Relative ratios of gene expression in individuals were obtained by comparing the hybridization results, by GenePix 4000B scanning and by ImaGene3.0 software analysis, of Cy5-labelled cDNA from FHF patients with those of Cy3-labelled cDNA from ASC.
RESULTS249 genes out of 8,192 were identified differently, at least two times. Most of the genes (79%) involved in cell signaling transduction, cell cycles, metabolism, inflammatory response and apoptosis, whose mRNAs were differently regulated.
CONCLUSIONSThese results suggest that HBV infection alters a broad range of cellular genes expression during developing into FHF and provide a framework for future functional study on the genes expressed differently.
