Objective To use a new simple Reverse Transcription Loop-mediated isothermal amplification(RTLAMP) method was applied to detect rubella virus nucleic acid and compared with Reverse TranscriptionPolymerase Chain Reaction (RT-PCR).Method Comparing the detection rate of the RT-LAMP method with that of RT-PCR for detecting rubella virus nucleic acid from rubella virus.Results The nucleic acid positive rates of all 11 strains of rubella virus were 100% by the two methods,the positive rate was 55%.Conclusion RT-LAMP is more simple and convenient than RT-PCR.

3.Thermal Desorption Low Temperature Plasma Ionization Mass Spectrometry for Rapid and Sensitive Detection of Pesticides in Broomcorn

A thermal desorption low temperature plasma (TD-LTP) ionization was developed for rapid and sensitive detection of pesticides by direct mass spectrometry.The thermal desorption sampler was added in fount of the plasma generator.The sample was desorbed in the thermal desorption sampler firstly,and then the gas molecules were transported to the plasma generator by the carrier gas to be ionized.The utilization of thermal desorption sampler helps to shift the interaction of the gas phase plasma with the sample form gas-solid or gas-liquid to gas-gas,which increases the sensitivity and stability especially for non-volatile sample (e.g.pesticides) greatly compared with the traditional LTP ionization source.Under the optimal parameters of the thermal desorption LTP ionization source,the characteristic ions of 12 kinds of pesticides were investigated.Then the thermal desorption LTP ionization source was connected with the commercial ACQUITY TQD mass spectrometer to evaluate the pesticide residue level in broomcorn.
4. Development of real-time fluorescence quantitative RT-PCR assay for β-actin gene of Panax ginseng

Objective: To construct a real-time fluorescence quantitative RT-PCR method for the β-actin gene of Panax ginseng. Methods: According to the β-actin gene of other higher plants available in Genbank, A pair of primers weredesigned and the amplified fragment of β-actin gene was linked with pMD20-T vector to construct recombinant plasmids. Then the positive plasmids were diluted and the standard curve was established. The sensitivity, specificity, and repeatability were detected. Results: The results showed that the lowest copy number for detection of β-actin gene with this method was 43.0 copies/μL, and there was a good linear relationship in a wide range from 43 to 4.3 × 107 copies/μL (R2 = 0.995 3). The melting curve showed a single peak with the temperature of (84.51 ± 0.01) ℃. The coefficient of variation (CV) of five different concentration of positive plasmids was 0.58% to 2.79% and 2.61% to 4.41% in intra-assay and inter-assay, respectively. Conclusion: The method established in this paper has the advantage of rapidity, sensitivity, specificity, high throughput, and good repeatability, which provides a methodological basis for the quantitative analysis on the functional genes of P. ginseng when β-actin gene is taken as a reference gene.

Objective To assess the association of three polymorphisms in Megsin (rs1055901,rs1055902 and rs2689399) and susceptibility of IgA nephropathy in Asian population.Methods We conducted a comprehensive search of electronic CNKI,VIP,WangFang Data,CBM,Pubmed,Web of Science and Google Scholar database on the association between Megsin rs1055901,rs1055902 and rs2689399 polymorphism and susceptibility of IgA nephrology in Asian population (last search update on 2 May 2016).Stata 12.0 software was used to calculate the odds ratio (OR) and 95 % CI (confidence interval),as well as sensitivity and publication bias analyses.Results Six publications encompassing mine case-control studies were finally included,including 2 179 cases and 1 769 controls.Finally,no significant association between Megsin rs1055901 and rs1055902 polymorphism and IgA nephrology in Asian population was identified,while a significantly decreased risk of IgA nephrology for rs2689399 polymorphism,was identified in Asian population (G and C:OR=0.754,95%CI 0.592-0.961,P=0.022;GG and CC:OR=0.506,95%CI 0.287-0.892,P=0.019;GG and GC+CC:OR=0.551,95%CI 0.316-0.961,P=0.036).Conclusion Rs2689399 G allele and GG genotype of Megsin may be the protective factors for IgA nephropathy in Asian population.
6.Efficacy of alfentanil and remifentanil in minimizing propofol injection pain

Objective To compare the efficacy of alfentanil and remifentanil in minimizing the propofol injection pain. Methods A total of 175 adult female patients undergoing gynecological procedures with general anesthesia were randomly divided into four groups.Patients received alfentanil 1mg(2 mL,AL group,n=43),remifentanil 0.01 mg(2 mL,REM1 group,n=43),remifentanil 0.02 mg(2 mL,REM2 group,n=45) or normal saline(2 mL,control group,n=44) 30 seconds prior to propofol administration.Visual analogue scale(VAS) was employed to evaluate the subjective feelings of pain due to propofol injection,and adverse effects were recorded. A novel rapid method for detection of the illicit beta2-agonist additives in health foods and traditional Chinese patent medicines was developed with the desorption corona beam ionization mass spectrometry (DCBI-MS) technique. The DCBI conditions including temperature and sample volume were optimized according to the resulting mass spectra intensity. Matrix effect on 9 beta2-agonists additives was not significant in the proposed rapid determination procedure. All of the 9 target molecules were detected within 1 min. Quantification was achieved based on the typical fragment ion in MS2 spectra of each analyte. The method showed good linear coefficients in the range of 1-100 mg x L(-1) for all analytes. The relative deviation values were between 14.29% and 25.13%. Ten claimed antitussive and antiasthmatic health foods and traditional Chinese patent medicines from local pharmacies were analyzed. All of them were negative with the proposed DCBI-MS method. Without tedious sample pretreatments, the developed DCBI-MS is simple, rapid and sensitive for rapid qualification and semi-quantification of the illicit beta2-agonist additives in health foods and traditional Chinese patent medicines.
9.Provision capacity research of primary healthcare services in rural China

Objective Studying the status quo and constraints for rural healthcare service in grassroot rural healthcare units,for policy recommendations.Methods Using data from the fourth healthcare service investigation,by means of quantitative interview and qualitative interview,for an investigative interview of 348 township hospitals and 251 village clinics in 31 provinces in China.Results Deployment percentage of primary heahhcare services at township hospitals level is 49.1% (28.0/57),and that for village clinics is 60.6%(5.4/9).Conclusions The key to upgrading rural healthcare service system at grassroots level is to deploy better diagnostic equipments,upgrade the diagnostic competence of grassroots healthcare personnel and build a continuous service system for primary healthcare service.
