1.Toll-like receptor 2 and ischemic cerebral white matter lesion

Toll-like receptor 2 (TLR2) is the most w idely expressed receptor in the Toll-like receptor family, and is also an important pattern recognition receptor of the innate immune system to pathogenic microorganisms. In addition, TLR2 can also identify endogenous risk signals and participate in non -pathogenic microbial inflammatory reaction in ischemic injury. The role of TLR2 and its signal transduction in ischemic cerebral w hite matter lesions have received more and more attention. This article reviews the relationship between TLR2 and ischemic cerebral white matter lesion.
2.The Technique and Appearances of CT and MRI in Cerebrospinal Fluid Rhinorrhea

Objective To study the value of CT and MRI(3D heavy T_2WI) in cerebrospinal fluid rhinorrhea.Methods 4 cases with spontaneous cerebrospinal fluid (CSF) rhinorrhea confirmed by operation were collected in this study.There were 2 men and 2 women.All patients underwent CT thickness was 0.625 mm and no gaps.MRI (coronal 3D heavy T2WI,thickness of 1 mm,TR/TE 5000 ms/119 ms).Results All 4 cases accepted the treatment of operations.The defects bone were showed by CT,and the sign of CSF of intracranial cavity communicated with nasal cavity diretly was showed by MRI,pituitary herniated into sphenoid sinus in one case also showed by MRI.Conclusion For spontaneous cerebrospinal fluid rhinorrhea,thin-section coronal CT scan and MR coronal 3D heavy T_2WI can clearly show the location of fistulas.
3.Chemoresistance of multi-drug resistance malignant glioma cells mediated by DNA-PKcs and its molecular mechanism

Objective:To obesrve the influence of DNA-PKcs in the chemoresistance of multi-drug resistance malignant glioma cells,and to explore its molecuIar mechanism in chemoresistance.Methods:siRNA was used to construct the DNA-PKcs knockdown human glioma U251 cell line;Western blotting method was used to detect the expressions of DNA-PKcs in U251 cells (U251 cells), doxorubicin (ADM)resistant U251 cells (U251/ADM cells),DNA-PKcs knockdown and ADM resistant U251 cells (U251/ADM/siDNA-PKcs cells).CCK8 method was used to detect the cell proliferation activity in three groups;Western blotting method was used to detect the expressions of MDR1, pNF-κB/p6, total Akt, pAkt/T308 and pAKT/S473 in the cells in three groups. Results:The expression level of DNA-PKcs in U251/ADM cells was significantly higher than those in U251 cells and U251/ADM/siDNA-PKcs cells (P < 0.01 ).The IC50 values of doxorubicin (ADM),paclitaxel (PTX), gemcitabine (GEM)in U251/ADM/siDNA-PKcs cells were significantly lower than that in U251/ADM cells (P <0.05);the expression levels of pAKT/S473,pNF-κB/p65,and MDR1 in U251/ADM/siDNA-PKcs cells were significantly lower than those in U251/ADM cells (P <0.01),but the total Akt and pAkt/T308 had no significant differences between two groups (P >0.05).Conclusion:DNA-PKcs can significantly enhance the chemoresistance of multi-drug resistance malignant glioma cells,the underlying mechanism is related to up-regulation of pAKT/S473,pNF-κB/p65 and MDR1 expressions.
4.Establishment of blended readers training model with 5 D interaction

A blended readers training model which combines online training (micro-lecture, online class, etc) with traditional training ( face to face teaching plus computer training) was proposed according to the theories in re-lation with blended learning, and scientific strategies for interaction of readers were established, which include the 5 dimensions ( demand, object, content, resource and evaluation) in order to integrate online learning resources and traditional offline training, optimize the allocation of training resources, and improve the training efficiency.
5.Cloning and bioinformatic analysis and expression analysis of beta-glucuronidase in Scutellaria baicalensis.

The β-Glucuronidase gene (sbGUS) cDNA firstly from Scutellari abaicalensis leaf was cloned by RT-PCR, with GenBank accession number KR364726. The full length cDNA of sbGUS was 1 584 bp with an open reading frame (ORF), encoding an unstable protein with 527 amino acids. The bioinformatic analysis showed that the sbGUS encoding protein had isoelectric point (pI) of 5.55 and a calculated molecular weight about 58.724 8 kDa, with a transmembrane regions and signal peptide, had conserved domains of glycoside hydrolase super family and unintegrated trans-glycosidase catalytic structure. In the secondary structure, the percentage of alpha helix, extended strand, β-extended and random coil were 25.62%, 28.84%, 13.28% and 32.26%, respectively. The homologous analysis indicated the nucleotide sequence 98.93% similarity and the amino acid sequence 98.29% similarity with S. baicalensis (BAA97804.1), in the nine positions were different. The expression level of sGUS was the highest in root based on a real-time PCR analysis, followed by flower and stem, and the lowest was in stem. The results provide a foundation for exploring the molecular function of sbGUS involved in baicalcin biosynthesis based on synthetic biology approach in S. baicalensis plants.

6.Significance of expression of extracellular matrix metalloproteinase inducer in pancreatic cancer

Objective To investigate the expression of extracellular matrix metalloproteinase inducer (EMMPRIN) in pancreatic cancer and its relationship with P-glycoprotein (P-gp) and vascular endothelial growth factor (VEGF),and to study its correlation with invasion,metastasis and drug re- sistance.Methods The expressions of EMMPRIN,P-gp and VEGF in 34 cases of pancreatic cancer were detected by immunohistochemistry.The correlation analysis and Chi squared test were been used by SPSS 10.0.Results The positive rates of EMMPRIN,P-gp and VEGF in pancreatic cancer were 61.8%,55.9% and 64.7% respectively.EMMPRIN,P-gp and VEGF were associated with lymph node metastasis (P<0.05),but not associated with sex,age,size of tumor and degree of differentia- tion (P>0.05).There were close relationship between EMMPR1N and P-gp,VEGF respectively (r= 0.398,r=0.432,P<0.05).Conclusions EMMPRIN,P-gp and VEGF were higher expressed in pancreatic cancer.EMMPRIN express was correlated with both P-gp and VEGF.It has indicated that during the progression of pancreatic cancer,there are close relationship between invasion,metastasis and drug resistance,and EMMPRIN may play key role in this process.

9.Research on the value of Kang'ai injection on gastric cancer patients’ chemotherapy

Objective:To analyze the value of Kang'ai injection on gastric cancer patients’ chemotherapy.Methods: Chosen gastric cancer patients with chemotherapy between December 2012 to December 2013 in our hospital as study subject, randomly divided into control group received conventional chemotherapy and observation groups received Kang'ai injection combine with chemotherapy, compared term effect after chemotherapy, immune function and quality of life scores.Results: 1)Observation group patients’ complete remission and partial remission after chemotherapy were significantly higher than control group patients(x2=2.471,x2=2.748;P<0.05); 2)Observation group patients’ IL-6, TNF-α, CD3+, CD4+ value was significantly higher than control group after receiving chemotherapy, CD8+ value was significantly lower than control group patients(t=2.627,t=2.462,t=2.748,t=2.846,t=2.735;P<0.05); 3)two groups of patients’ life quality scores before chemotherapy was no significant,after chemotherapy observation group patients’ life quality scores were significantly higher than control group patients(t=2.637,t=2.647,t=2.983;P<0.05).Conclusion: Kang'ai injection can significantly improve the effectiveness of chemotherapy in patients with gastric cancer,and optimize immune function, while improving the quality of life after chemotherapy.
10.Role of NADPH oxidase in high glucose-induced injury in H9 c2 cardiac cells

Aim To explore the role of nicotinamide adenine dinucleotide phosphate ( NADPH ) oxidase in high glucose-induced injury in H9c2 cardiac cells. Methods H9 c2 cardiac cells were exposed to differ-ent concentrations of high glucose(5. 5 mmol·L-1 ,11 mmol · L-1 ,22 mmol · L-1 ,33 mmol · L-1 ,44 mmol ·L-1 , 55 mmol · L-1 ) for 24 h and different time pints of high glucose(0 h,12 h,24 h,36 h,48 h,72 h) . Cell viability was measured by MTT colorimetry, the protein expression of Bcl-2 , Bax and NADPH oxi-dase submits such as p22 phox , p47 phox and p67 phox were determined by western blotting. Results H9c2 cardi-ac cells exposure to high glucose for 24 h showed on decrease in cell survival and the Bcl-2 expression while an increase in the Bax expression ( P<0. 05 ) . Moreo-ver, high glucose could markedly up-regulate the activ-ity of NADPH oxidase characterized by the enhanced expression of p22 phox , p47 phox and p67 phox ( P<0. 05 ) . Conclusion Activating NADPH oxidase may play an important role in high glucose-induced injury in H9 c2 cells.
