2.Expression and role of ubiqutin carboxyl-terminal hydrolase L1 in lens with age-related cataract under oxidative stress

Background Oxidative damage is a major cause of age-related cataracts,and the ubiquitinproteasome system is involved in lens differentiation and development.Ubiquitin carboxy-terminal hydrolase L1 (UCHL1),one of key enzymes of ubiquitin-proteasome system,was discovered to participate in the age related diseases and oxidative stress damage. Objective This study was to investigate the effects of UCHL1 on the formation and development of age-related cataract. Methods Lens capsule were collected from 24 patients with age-related cataract(including 12 cases of cortical cataract and 12 cases of nuclear cataract) during the surgery.Five normal lens capsule membranes were obtained from eye bank of Tongji University.Human lens epithelial cells (LECs) line (SRA01/04) was also collected in this study.Expression of UCHL1 in the lens epithelial layer of different samples was assayed using immunofluorescence technology.UCHL1 eukaryotic expressing vector was constructed and transfected into cultured SRA01/04 by liposome,and green fluorescent protein (GFP) eukaryotic expressing vector was transfected at the same method as the control group.UCHL1 over-expressing cells were then exposed to different concentrations (0.2,0.3,0.4 and 0.5 mol/L) of tert-butyl hydroperoxide (TBHP) for 24 hours and subsequently monitored for cell viability evaluation by MTT assay. Results Immunofluorescence showed that UCHL1 was expressed in human lens epithelial layer,but significantly different expressing levels were seen among normal lens capsular membrane,cortical cataract and nuclear cataract ( F =13.411,P =0.000),and UCHL1 expressing levels were lower in cortical cataract and nuclear cataract than the normal lens (P =0.000,P =0.000).No significant difference was found in UCHL1 expressing level between cortical cataract and nuclear cataract ( P =0.164).Western blot analysis verified that UCHL1 exhibited a stranger expression in the UCHL1 transfected group compared with the GFP transfected group,illuminating a successful transfection of UCHL1 in SRA01/04 cells.MMT assay revealed that the A570/630 value in UCHL1 transfected cells was significantly elevated in comparison with GFP transfected cells following the treatment of 0.3 mol/L TBHP. Conclusions UCHL1 has an antioxidative ability,and it might plays an important role in the progress of age-related cataract.
4.Effect of Xuebijing Injection on Extracellular Release of HMGB1 Induced by Endotoxin

Objective To investigate the effect of Xuebijing injection on release of high mobility group box 1 (HMGB1) in endotoxin-induced cells. Methods HMGB1 concentration and mRNA expression were analyzed by enzyme-linked immunosorbent assay (ELISA) and RT-PCR, respectively. The effects of Xuebijing injection with 2, 10 and 50 mg/ml on HMGB 1 concentration in the culture medium of 200 ng/ml lipopolysaccharide-induced macrophage-like RAW 264.7 cells and BRL-3A hepatocytes, and HMGB1 mRNA expression in lipopolysaccharide-induced macrophages were observed. Results 50 mg/ml Xuebijing injection significantly decreased HMGB1 concentration in the culture medium of lipopolysaccharide-induced macrophages and hepatocytes(P< 0.01), and inhibited HMGB1 mRNA expression in macrophages. Conclusion Xuebijing injection inhibits endotoxin-induced release of HMGB1.
5.Regulating effects and mechanism of Laminaria japonica on serum lipid of hyperlipidemia rats

Objective To investigate the regulating effects and mechanism of Laminaria japonica (L.japonica) on serum lipid of hyperlipidemia rats.Methods Forty healthy female Wistar rats were used to establish hyperlipidemia models by feeding fat-rich forage,and the powder of L.japonica was applied as a supplement in forage for test groups.The levels of serum lipid including the triglyceride(TG),total cholesterol(TC),low-density lipoprotein (LDL),high-density lipoprotein(HDL) and the activities of lipoproteinesterase(LPL) and hepatic lipase(HL) were detected by biochemical assay.Results The levels of serum TG and TC in test group decreased significantly than those in pre-treated and model group (P
6.Progress of antimicrobial peptides in the treatment of sepsis

Sepsis is a severe systemic inflammatory response syndrome, and is common in the patients with infection, extensive burn injury and major surgery. As it may cause multiple organ dysfunction and septic shock, it is always accompanied with high mortality and poor prognosis. Currently there's no effective medication available for treatment of sepsis. During the process of killing bacteria, the classical antibiotics lead to release of a large quantity of proinflammatory cytokines, such as lipopolysaccharide, which exacerbates the malfunction of immune system. Furthermore, the growing number of multiresistant bacteria present a new challenge to the management of sepsis. Antimicrobial peptides (AMPs) are small, cationic, and amphipathic peptides with broad-spectrum microbicidal activity against bacteria, fungi and viruses. In addition, they also can neutralize endotoxin and suppress inflammatory cascade through multiple immunomodulation, which potentially serves as a promising alternative approach for sepsis treatment. This review briefly summarizes the progress of AMPs in the treatment of sepsis as well as the relevant mechanisms.
7.Effectiveness and Safety of Oral Administration of Type Ⅱ Collagen on Patients with Active Rheumatoid Arthritis

Objective To assess the effectiveness and safety of oral administration of type Ⅱ collagen(C Ⅱ) versus placebo in patients with active rheumatoid arthritis(RA).Methods Seventy patients with active RA were randomly divided into receive placebo(n=35) or bovine CⅡ(n=35) at 0 1mg/day for 12 weeks,in a double-blind study.The cumulative response rates were analyzed by utilizing the American College of Rheumatology(ACR) criteria for improvement in RA,and the requirement for ≥50% improvement of outcome measures in ACR criteria.Results After the 12-week course of treatment,much more outcome was significantly improved in C Ⅱ group than in placebo group.The rates of improvement and remarkable improvement in CⅡ group were increased significantly,as compared with placebo group(54 55% vs 12 12% and 18 18% vs 3 13%,respectively,P0 05).Conclusions Oral administration of CⅡ in treating RA was significantly effective and has slight side effects.Further investigation in the field is worthwhile.
8.Differentiation of bone marrow and umbilical cord blood mesenchymal stem cells into osteoblasts in vitro:comparison of their osteogenic potentials

BACKGROUND:Mesenchymal stem cels isolated from cord blood and bone marrow have multi-directional differentiation ability under a certain condition of induction. OBJECTIVE:To compare the difference of differentiation of umbilical cord blood and bone marrow mesenchymal stem cels into osteoblasts. METHODS:Human umbilical cord blood and bone marrow mesenchymal stem cels were isolated and cultured by density gradient method. When reached 90% confluency, mesenchymal stem cels were digested by trypsin for subculture. At the third passage, umbilical cord blood mesenchymal stem cels and bone marrow mesenchymal stem cels at 8×104/wel were incubated. When reached 80% confluency, cels were treated with low-glucose DMEM supplemented with 10% fetal bovine serum, 0.1 μmol/L dexamethasone, 50 μmol/L vitamin C and 10 mmol/L β-sodium glycerophosphate. RESULTS AND CONCLUSION:There was no significant difference in morphology and biological properties of the two kinds of mesenchymal stem cels. Cels were highly expressed CD44, CD29, but did not express CD34. They had the ability to differentiate into osteoblasts, which had a positive staining for known markers: alkaline phospatase and calciumin vitro mineralization. There was no significant difference in the activity of osteoblasts of two kinds of cels. Results verify that umbilical cord blood and adult bone marrow mesenchymal stem cels can be induced into osteoblasts with a similar ability, and they can be used as seed cels for bone tissue engineering.
9.Radiosensition of a human non-small lung cancer cell line A_(549) by endostatin

Objective To study the synergistic effect of endostatin (YH-16) and irradiation on human non-small cell lung cancer (NSCLC) cell line A549 and the expression level of vascular endothefial growth factor. Methods ① A549 cells were exposed to various concentrations of endostatin for different time. The optimal concentration giving ≤20% inhibition concentration (IC20) by MTT assay was selected. ② The cells were divided into 4 groups:control group,chemotherapy alone,radiotherapy alone,and radio-chemotherapy group. All groups were exposed in distinct treatment,the cells survival fraction and the plating efficiency of the four groups provided to select the optimal radiotherapy dosage. ③ The radio-chemotherapy group had been exposed to endostatin for 48 hours,followed by irradiation at 48 hours with various doses:0,2,4,6,8 Gy. After 14 days,the cell clonogenic survival curves and the SER were evaluated. ④ Detect the different groups' VEGF value by ELISA kit. Results Incubating cells in 200 mg/L endostatin culture medium the value of SER radiated by linear accelerator in 2 Gy after 48 h were 1.61 and 1.04. And endostatin with proper dosage and radio-exposure time could decrease the VEGF level. Conlusion It is suggested that endostatin enhances the radiosensitivity of NSCLC A549 cell line in vitro (SER=1.61,1.04). The enhancement depends on the time of exposure drug. The optimal radiation time should be 48 hour after exposure.

Anorexia is a common pediatric and multiple chronic appetite disorder disease. Long-term anorexia can easily lead to adverse effects on nutritional status, growth, intellectual development and physical and mental health of children. Professor LIU Yi-min, the national famous TCM doctor in Yunnan University of Traditional Chinese Medicine, has much experience in treating this disease, believing that the pathogenesis key is spleen and stomach disharmony and inability to transport. According to clinical experience, it should be treated from indigestion of stomach, spleen dampness, spleen qi deficiency, and spleen weakenness and liver excess. The treatment is to eliminate indigestion, tonify spleen and expel humidity, tonify spleen and nourish qi, and tonify spleen and harmonize liver. The commonly used TCM pairs were Charred Triplet and Galli Gigerii Endothelium Corneum, Plantaginis Herba and Dioscoreae Rhizoma, Hungaian Morningglory Root and Marginate Rockbell Herb and Capsella bursa-pastoris and Chrysanthemi Flos.
