As an efficient gene transfer vehicle lentiviral vector has been widely used in therapy research. Comparing with other retrovirus vectors, lentiviral vectors have the unique ablility of transfecting nondividing cells and terminal differentiated cells. In addition, lentiviral vectors can accommodate two or more promoters and can carry larger foreign gene insertions. Now the new generation of lentiviral vectors encoding transcriptional control sequence provides effective means for the regulation of foreign gene expression. The development of lentiviral vectors and its application in the gene therapy field were summarized.

Objective To explore the application of ROC curve analysis in evaluating the virtual screening of ?-secretase (BACE-1) inhibitors. Methods Virtual screening were performed for BACE-1 inhibitors using different scoring functions (Surflex-score,D-score,G-score,PMF-score,ChemScore),the screening library was composed of 50 BACE-1 inhibitors and 9 950 inactive compounds.Five scoring functions were used for ranking, and application of ROC curve analysis in evaluating the results of ranking. Results Using D-score to rank the database, the area under the curve (AUC) was 0.935.If specificity was considered primarily, more active compound were obtained by Surflex-score;if sensitivity was considered primarily, more active compounds were obtained by D-score.Conclusions ROC curve consists of sensitivity and specificity together,it can be used to set the right threshold after virtual screening to obtain active compounds as many as possible.
4.Study on relationship between CAG repeat in ITI5 gene and clinical phenotype of Huntington's disease
Chinese Journal of Neurology 2005;0(08):-
Objective To detect the relationship between CAG repeat in IT15 gene and clinical manifestations of Huntington's disease (HD).Methods Non-denaturing polyacrylamide gel electro- phoresis method was used to detect CAG repeat.Clinical manifestations were scored by UHDRS and MMSE. Results Genotypes of IT15 were heterozygous in all 29 HD patients.CAG repeat in the HD chromosome, being 42—62,13—28 in normal chromosome, was inversely correlated with age of onset (r=-0.539,P

Objective To develop an approach for a rapid extraction of genomic DNA fragments from serum. Methods Serum specimen from human was processed using high temperature denaturation and was centrifuged, then using various primer sets to amplify the DNA fragment extracted, so as to verify the efficiency of this DNA extraction method. Results The templates preparing from this DNA extraction method were able to guarantee the amplification of fragments shorter than 400 base pairs. Conclusion This method is simple, rapid and inexpensive. These genomic DNA fragment extracted can be utilized to amplify definite DNA template for polymerase chain reaction.

Taking children respiratory infectious diseases as the research object, CDSS transform from isolated clinical data to an integration Model. With integrating pharmacy and laboratory knowledge, the system establishes children typical characterization data of common infectious diseases and medical efficacy data repository to providing treatment recommendations for doctor. Based on the clinical characteristics of children infectious diseases, this paper proposes knowledge inference engine and an information integration model. decision-making inference engine, medical information integration model.

Journal of Chinese Physician 2011;13(6):783-785
Objective To discuss clinical effect and feasibility of laparoscopic pelvic lymphadendenectomy assisted radical vaginal hysterectomy (LARVH) in the treatment of cervical cancer. Methods Laparoscopic pelvic lymphadendenectomy assisted radical vaginal hysteretomy was performed in 38 patients with cervical cancer. The operative quality and postoperative recovery effects were analyzed. Results All 38 patients were successfully performed with LARVH and no complications. The radical vaginal hysteretomy were performed in all of patients, 21.4±2.2 pelvic lymph nodes were removed. The average duration of surgery was (240±25.6) min and the average hemorrhage was (340±40.2) ml. There were 7 patients who needed blood transfusion during operation. After operation, the mean time for passage of gas by anus was (2±0.26) days, and the mean recovery time of bladder function was (10.5±1.3) days. Patients were discharged after (9.2±1.1) days on average. 38 patients had no complication. Conclusions For the treatment of uterine neoplasm, LARVH decreased operative damage with more short recovery time, same operative extent as laparotomy. It is an ideal way to treat uterine neoplasm currently.
