4.Impact of citreoviridin on mRNA expression of mitochondrial respiratory chain synthesis related transcriptional regulation gene, membrane potential and reactive oxygen species in rat cardiomyocytes

Objective To investigate the impact of citreoviridin(CIT) on mRNA expression of mitochondrial respiratory chain synthesis related transcriptional regulation gene,mitochondrial membrane(MMP) potential and reactive oxygen species (ROS) in cardiomyocytes of rat.Methods Viability of rat primary cardiomyocytes treated with different concentrations of CIT (0,1,2,3,4,5,6,7,8,9,10 μmol/L) for 24 h was determined by 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) method.Based on the MTT curve,median inhibitory concentration(IC50) was calculated using SPSS 13.0.High-,medium-and low-dose groups of CIT(1.650,1.234,0.715 μmol/L) were defined corresponding to 99%,95% and 90% of cardiomyocyte viability,respectively.CIT was not added in as the control group.After 24 hours,the mRNA expression levels of peroxisome-proliferator-activated receptor γcoactivator(PGC-1α),nuclear respiratory factor 1 (Nrf1) and nuclear respiratory factor 2(Nrf2) in cardiomyocytes were detected by reverse transcriptase polymerase chain reaction(RT-PCR).Changes of MMP and intracellular ROS were determined by a fluorescence microplate reader using 5,5',6,6'-tetrachloro-1,1',3,3'-tetraethylbenzimidazolcarbocyanine iodide (JC-1) and 2,7-dichlorofluorescein diacetate (DCFH2-DA) as fluorescent probes.Results Compared with 0 μmol/L CIT group [(89.4 ± 17.6)%],viabilities of rat primary cardiomyocytes treated with 2-10 μmol/L CIT groups[(80.2 ± 20.2)%,(74.4 ± 18.7)%,(63.2 ± 8.9)%,(51.5 ± 18.8)%,(39.0 ± 15.7)%,(22.6 ± 10.5)%,(19.9 ± 4.9)%,(20.7 ± 4.8)%,(18.5 ± 3.3)%] decreased significantly(all P < 0.05).The IC50 value of cardiomyocytes after24 h treatment with CIT was 4.6 μmol/L The PGC-1α mRNA expressions ofhigh-,medium-and low-dose groups(0.431 ± 0.041,0.619 ± 0.031,0.653 ± 0.037) were significantly lower compared to that of the control group(0.776 ± 0.081,all P < 0.05).The Nrf1 mRNA expression of high-dose group(0.358 ± 0.05) was significantly lower compared to that of the control group(0.580 ± 0.098,P < 0.05).Nrf2 mRNA expressions of the high-and medium-dose groups(0.352 ± 0.041,0.472 ± 0.011) were significantly lower than that of the control group (0.667 ± 0.091,all P< 0.05).Compared with the control groups[(100.00 ± 0.00)%,(100.00 ± 0.00)%],the MMP levels of high-,medium-and low-dose groups[(55.3 ± 3.3)%,(69.8 ± 4.7)%,(81.8 ± 2.7)%] were significantly lower and the ROS levels[(606.0 ± 46.3)%,(275.0 ± 53.5)%,(158.9 ±29.5)%] were significantly higher(all P < 0.05).Conclusions CIT inhibits the biosynthesis of mitochondria in primary cardiomyocytes and induces oxidative stress.Myocardial injury is caused by cardiomyocyte apoptosis through mitochondrial pathway,which leads to myocardial injury.
5.Acute Lung Injury after Extensive BurnMerging Smoke Inhalation:Treatment in 32 Cases of Early Stage

Objective To summarize the treatment experience of extensive burn merging smoke inhalation acute lung injury(SIALI)in 32 early stage cases,and to provide a reference for the further improvement of success rate in early stage. Methods A total of 32 cases of extensive burn merging SIALI resulting from mine explosion that received clinical treatment and triage from November to December in 2014 were enrolled for the study. In addition,the experience of triage and prognosis(after injured in a month)to guide therapy was analyzed. Results Two of the 32 patients died from respiratory failure in shock period,after shock period(3 days after injured),19 bad sufferers were cured in the local hospital,and the oth?er 11 cases in stable condition were triaged to the nearby hospital. 1 month follow?up results showed that:patients who received therapy in local hospi?tal,one died from MODS in 3 weeks,one died from kidney failure in 4 weeks. The other 28 patients were alive. Conclusion For these patients who suffering from extensive burn merging SIALI received therapy in early stage,we should not only schedule positive anti?shock,tracheotomy,antibiot?ics,escharectomy and nutritional support or other treatments,but also face to the challenge of lacking medical resources from too more patients. Thus,we suggest to triage the wounded to nearby hospital after anti?shock treatment,so as to improve the success rate in early stage.
6.The comments of the application of 5.5 gauge scalp acupuncture used on the outpatients

Most of the transfusion adult outpatients are old people with chronic disease, and theirsmall blood vessels become less elastic with age. They need long course and repeated localised puncture site. On the base of that fact, the author describes the 5.5gauge scalp acupuncture and 7 gauge scalp acupuncture about their use on the patients. The 5.5 gauge scalp acupuncture is smaller and shorter, so it has the advantage of high success ratio and it is easier to be fixed. Additionally, the transfusion speed is easy to be controlled, which reduces the ratios of transfusion reaction and adverse drug reactions. The pinhole is small and can be healed faster. The congestion around the puncture location is less than that of the 7 scalp acupuncture. It can alleviate the pain for patients and increase satisfaction. To sum up, the 5.5 gauge scalp acupuncture should be used widely as we allocate“people oriented”and“All for the patients”.

8.Medpor padding for correction of sunken eyelid after hydroxyapatite sphere implantation

Objective To evaluate the clinical effect of Medpor ( porous high density polyethylene )as a padding material in correction of sunken eyelid after hydroxyapatite (HA) implantation. Methods We used Medpor in 17 patients ( 10 male and 7 female ), aged from 18 to 43 years (mean 26.6 years ), in which 9 cases had concomitant orbital fractures. The Medpor was implanted after HA sphere implantation ranged from 2 to 19 months ( average 6.7 months ). 17 consecutive patients with implantation of HA sphere were filled with Medpor under the periosteum. All of the cases were examined by CT scanning to distinguish orbital lesions with or without fracture. In the orbital fracture cases, we first repaired orbit bone defects with Medpor. Results There were no secondary infection or implant rejection occurred after following up for 1 to 18 months (average 8.7 months ). All patients' cosmesis was acceptable. 14 of 17 patients gained symmetric appearance. Although 3 patients had slight sunken eyelid, they gained a dramatic improvement of facial appearance nevertheless. Conclusions Sunken eyelid after HA sphere implant is related with orbital fracture as well as relative reduction of orbital content. Medpor presents fine biological compatibility. It is also easy to remodel and fix during operation. Medpor is a good implant for treatment of sunken eyelid after HA sphere orbital implantaion. [
9.Study on the Physico-chemical Properties of Endothelium Corneum Amylase

Objective To search for the optimal processing procedure and physico-chemical properties of Endothelium corneum amylase. Methods After dried under 50 ℃ ,amylase was extracted from Endothelium corneum with phosphate-buffered saline. The amylase activity was examined at different temperature,pH and different metal ions,and amylase isozyme was discerned on PAGE. Results The optimal pH,optimal temperature,and optimal substrate concentration for amylase activity were 8.67,50 ℃ and 1.8 g/mL,respectively. Under the experimental conditions,the amylase Km was 0.92 (starch was used as substrate of amylase). Four metal ions tested,including Ca2+,Fe2+,Zn2+,Mn2+,could inhibit the amylase activity to some extent. Furthermore,it showed one amylase isozyme band using PAGE,suggesting only one amylase isozyme existing in the Endothelium corneum tentatively. Conclusion The effects of metal ions should be considered in the process of Endothelium corneum amylase.

Xanthine-Xanthine oxidase ( X-XOD) added to the culture medium of cultured cardiac cells in rats may damage cell membrane. Electrophysiological findings indicated that APA, MDP, OS,Vmax, were decreased and the SDF was increased. Astragalus polysaccharide (APS) could protect the cells from being damaged by X-XOD. APS recoverted all the de-creased cardiac functional parameters in free-radical-damaged rats by X-XOD. APS had anti-free-radical damage action on the cultured my-ocardiocytes and the myocardial contractility of the isolated rat working heart.
