2.Effects of Ophiocordyceps xuefengensis on proliferation of DC-CIK cells and activity of killing HepG-2 cells by DC-CIK cells

Objective:To study the effects of Ophiocordyceps xuefengensis on proliferation of DC -CIK cells and the activity of killing HepG-2 cells by DC-CIK cells.Methods:Peripheral blood mononuclear cells were routinely isolated and induced into DC and CIK.DC and CIK co-cultured by 1∶5 for 7 days,then Ophiocordyceps xuefengensis were added into medicine group ,observed the mor-phology of the cells on the tenth day and counted the DC-CIK number of each group.DC-CIK cells acted as effector cells and the HepG-2 cells as target cells , cck-8 method for the detection of DC-CIK in the killing rate of HepG-2.Results: The Ophiocordyceps xuefengensis was able to proliferate the DC-CIK dramatically ,the optimal concentration was 0.1 mg/ml.Cultivation of Ophiocordyceps xuefengensis induced DC-CIK cells on HepG-2 cells killing effect was better than that of the routine method of DC-CIK cells; the effection of Ophiocordyceps xuefengensis killing HepG-2 cells was not obviously.Conclusion: Ophiocordyceps xuefengensis can enhance the anti-tumor activity of DC-CIK mainly by promoting the proliferation of it.
3.Determination of trace gallium by graphite furnace atomic absorption spectrometry in urine

Objective To establish a method for determination trace gallium in urine by graphite furnace atomic absorption spectrometry (GFAAS).Methods The ammonium dihydrogen phosphate was matrix modifier.The temperature effect about pyrolysis (Tpyr) and atomization temperature were optimized for determination of trace gallium.The method of technical standard about within-run,between-run and recoveries of standard were optimized.Results The method showed a linear relationship within the range of 0.20~80.00 μg/L(r=0.998).The within-run and between-run relative standard deviations (RSD) of repetitive measurement at 5.0,10.0,20.0 μg/L concentration levels were 2.1%~5.5% and 2.3%~3.0%.The detection limit was 0.06 μg/L.The recoveries of gallium were 98.2%~101.1%.Conclusion This method is simple,low detection limit,accurate,reliable and reproducible.It has been applied for determination of trace gallium in urine samples those who need occupation health examination or poisoning diagnosis.
4.Determination of trace bismuth by atomic fluorescence spectrometry in blood

Objective To establish a method for determination trace bismuth in blood by atomic fluorescence spectrometry (AFS).Methods 4.0 ml nitric acid and 1.0 ml perchloric acid was added into 1.0 ml blood sample then through automation graphite digestion instrument digested,after that 1.0 ml thiocarbamidevitamin (10%) was injected,8% HC1 constant volume to 10.0 ml,the bismuth was detected by atomic fluorescence spectrometry with 5.0 ml digestive sample.Results The method showed a linear relationship within the range of 0.4-50.0 μg/L (r=0.999 7).The within-run and between-run relative standard deviations (RSD) of repetitive measurement at 10.0,20.0,40.0 μg/L concentration levels were 2.2%-4.9% and 3.0%-4.0%.The detection limit was 0.032 μg/L.The recoveries of bismuth were 93.0%-103.9%.Conclusion This method is low detection limit,good accurate and high sensitivity.It has been applied for determination of trace bismuth in blood samples those who need occupation health examination or poisoning diagnosis.

7.Liquid chromatography for determination of disulfiramin workplace atmosphere.

OBJECTIVETo establish a method for determination of disulfiram in the workplace atmosphere by liquid chromatography.
METHODSSampling with glass fiber filter, eluting with methanol, separating with C18 column, and determination with liquid chromatography.
RESULTSThe bearing capacity of glass fiber filter exceeded 3.45 mg per piece. The elution efficiency was 97.8%∼101.0% The relative standard deviation varied from 1.09% to 1.44%. The limit of detection was 0.1 µg/ml. The minimum detectable concentration was 0.011 mg/m³ (with sampled air volume of 45 L).
CONCLUSIONThe method has high selectivity, accuracy, and precision and strong applicability.

8.Application of transrectal contrast-enhanced ultrasound in the diagnosis of prostate cancer in different tPSA intervals

【Objective】 To investigate the clinical value of transrectal contrast-enhanced ultrasound (CEUS) in the diagnosis of prostate cancer in different total prostate specific antigen (tPSA) intervals. 【Methods】 According to serum tPSA levels, 96 patients meeting the inclusion criteria were divided into 3 groups:4-10 ng/mL, >10-20 ng/mL and >20 ng/mL groups. All patients underwent transrectal CEUS. With pathological results as reference, the diagnostic value of transrectal CEUS in different tPSA intervals was evaluated. 【Results】 Of the 96 cases, 62 were confirmed by pathology as prostate cancer and 34 as benign prostatic hyperplasia (BPH). The main perfusion characteristics of prostate cancer under CEUS were rapid enhancement (64.52%), rapid clearance (70.97%), uneven enhancement (83.87%) and high enhancement (61.29%);the main characteristics of BPH were non-rapid enhancement (70.59%), non-rapid clearance (73.53%), uniform enhancement (76.47%) and non-high enhancement (52.94%). There were significant differences in terms of enhancement speed, clearance speed and enhancement uniformity between prostate cancer and BPH (P<0.05), but no significant difference in the enhancement intensity. The sensitivity of transrectal CEUS in the diagnosis of prostate cancer in low, medium and high tPSA groups were 58.33%, 70.37% and 95.65%, the specificity were 83.33%, 76.92% and 66.67%, and the accuracy were 73.33%, 72.50% and 92.31%, respectively. Transrectal CEUS showed consistency at different serum tPSA levels for the diagnosis of prostate cancer, with statistical significance. Moreover, in the 4.0 ng/mL ≤tPSA<10.0 ng/mL group, the diagnostic specificity was the highest. 【Conclusion】 Transrectal CEUS is helpful in the differential diagnosis of benign and malignant prostatic lesions, especially for patients with different serum tPSA levels.
