1.A PRELIMINARY REPORT ON THE USE OF HETEROGENOUS GRAFT TISSUE (CHANG HANG-825) IN MYRINGOPLASTY

23 cases with tympanic membrane perforation were successfully treated with heterogenous tissue grafting.In six cases, the underlay grafting method was used, and the outlay method was in 6 cases. After operation, all perforations were nicely healed up.The graft tissue used is the waste product in manufacturing cat-gut unqualified for regular suturing.Segments of this cat-gut purified and chromified, whitish in colour, and tape-like in shape were used.Each segment measured 1 or 1.5 cm in width proved a little too thin, and should be double-folded or triple-folded.A segment of cat-gut sectioned and examined under the microscope, was found to contain nothing else than plenty of collagen fibers .Cat-gut could be kept in 75% alcohol under room temperature.From our limited experience, it seems to be a very -useful grafting material in ear surgery and worthy of further trial and observation. To our knowledge no similar report has been noticed in current medical literatures.
2.The experimental studies of cardioplasty and total fundoplication on dogs

The aims of this study are to obtain a suilablc condition for the cardioplasty with the fundoplication and an observation of the effect of the antircflux function following the operations. The results show that a suppcrting dcvicc of a round tube with 1 - 1.2 ctn diamcter is used as a guide to establish a good lovver esophageal sphinctcr(LES). The experimental ani-mais may survive very long. The pressure in the sphimcter is cvenly the same as that after the operation immediately. The gastroesophageal reflux is never occurred except intragastric pres-sure is the same as or higher than LES. The lower csophagcal high pressure zone formed is similar to the smooth muscle in the lower part of csophagus at the sile of opcralions . The new formed cardia plays an excellent role of anturc flux function . This report suggists that the cardioplasty with fundoplication is useful for the treatment of esophageal achalasia and somc other abdpminal disorders.
3.Clinical observation on orthokeratology combined with frameglass for treating high myopia in teenagers

Objective To investigate the clinical effect of orthokeratology (ortho-k)during night and wearing frameglass during daytime for treating high myopic in teenagers.Methods 53 teenager cases (aged 10 - 14 years old)of high myopia with spherical equivalent degree -6.00--8.00 diopters (D)in our hospital were performed the retrospective analysis,among them,23 cases were the combined wearing of ortho-k and frameglass and 30 cases were simple wearing of frameglass.The changes of eye axi-al length at 6,12,18 24 months before and after wearing frameglass were measured and compared between the two groups.The spherical equivalent degrees were compared between before wearing frameglass and at 2 years after wearing frameglass.Results The spherical equivalent degree after stopping wearing for 1 month after 2-year wearing in the ortho-k and frameglass group was(-7.28±0.57)D,which after 2 years in the frameglass group was (-7.80±0.68)D,the 2-year spherical equivalent degrees in the or-tho-k and frameglass group and the frameglass group were increased by (-0.29 ±0.22)D and(-1.07 ±0.41)D respectively,the difference between the two groups was statistically significant(P <0.01);the eye axial growths were(0.12±0.04)mm and(0.37± 0.13)mms respectively,the difference between the two groups was statistically significant (P <0.01).Conclusion The combina-tion of ortho-k during night and frameglass during daytime can effectively relieve the myopia increase and eye axis elongation in the patients with high myopia and control the myopia development to some extent.
4.Diagnosis and treatment of blunt liver injury ( a report of 135 cases)

Objective To study the effective method of improving the diagnosis and treatment of blunt liver injury.Methods The clinical data of 135 cases with blunt liver injury hospitalized in Daxing District People' s Hospital from January 2000 to October 2010 were reviewed retrospectively.Results By standard grading of American Society of Trauma Surgery (AAST) in 1994,36 cases were grade Ⅰ,42 cases gradeⅡ,33 cases gradeⅢ,14 cases grade Ⅳ,10 cases gradeⅤ.Fifty-one cases were treated by non-operative therapy,among which 42 cases were crued,9 cases underwent operative surgery.Ninety-three cases were treated by operative therapy,seven cases died.The morbidity related to blunt liver injury was 5.19%.Conclusions Non-operative therapy may be used firstly in patients with stable hemodynamics,but close monitoring should be done for changes of the life signs.Timely and reasonable choice of time and the way of surgery is important to avoid complications and reduce the mortality rate.
5.Modified culture of saphenous vein smooth muscle cells

Objective To study an effective method for culturing of human vascular smooth muscle cells (hVSMCs) in tissue engineered blood vessels (TEBVs) in vitro.Methods A 3-cm segment of human long saphenous vein was harvested under sterile conditions.The primary culture and subculture were done by modified tissue-piece inoculation and trypsin digestion respectively.The cells were purified by mechanical treatment and differential attachment.PDGF was combined with high concentration of glucose DMEM as the culture medium.The cultured cells were identified by morphology and immunohistochemistry with contrast microscope and SABC kit,and RT-PCR method detected the expression of ?-SMA and calponin 1.Results The cultured cells possessed"peak and valley"characteristics for VSMCs under microscope.Immunohistochemical staining showed positive expression of intracytoplasmic ?-actin,and RT-PCR detected positve expression of ?-SMA and calponin 1.Conclusions Culture medium of PDGF combined with high concentration of glucose DMEM,and with differential attachment method can provide highly purified hVSMCs with good structure and function.
6.Expression of p73 gene in human non-small cell lung cancer i n vitro

Objective To study the expression of p73 in human non-small cell lung cancer (NSCLC) and the relationship between p73 expression and clinico-pathological parameters. Methods Expression of p7 3 gene was detected by RT-PCR in 32 human NSCLC tissues, tissues adjacent to ca ncer and non-cancer lung tissues. Results p73 gene expression up-regulated substantially and detected in 87.5%(28/32) of human NSCLC tissues while expressed at low level in tissues adjacent to cancer and non-cancer lung tissues. Conclusion Marked up-regulation of p73 gene expres sion is found in human NSCLC.
7.The reason of operative death and operative risk factors in patients with esophageal cancer after esophagectomy by logistic regression model

Objective To explore the risk factors and causes affecting the operative mortality in esophagectomy patients with esophageal can cer. Methods 1400 cases with a curative esophagectomy for neopl asm of esophagus hospitalized from Mar,1973 to June, 2000 were reviewed. There w ere 31 died within 30 d or during hospitalization after esophagectomy as a group , and 1 369 survival cases, after operation, as another group. Sixteen factors t hat may influence the operational mortality were selected. A multi-variate anal ysis of these individual variables was performed by the computer′s logistic reg ression model. Results The operative mortality was 2.2%(31/1400 ). The causes of death included respiratory complication 17 cases (including res piratory failure caused by pneumonia or atelectasis), 15 cases, and adult respir atory distress syndrome (ARDS) 2 cases, the mortality was 54.8% in the death gro up), anastomotic leak 11 cases (34.5%), Chylothorax 2 cases (6.5%) and postopera tive digestive tract hemorrhage 1 case (3.2%). The results showed that the major risk factors that affected operative mortality in cases with esophageal cancer were history of long-herm heavy smoking, duration of operation and the year of operational (P<0.05). Conclusion To minimize operative mort ality of esophagectomy, some means must be noticed, including the reinforcemen t of the perioperative care, the improvement of anastomotic methods and surgical skill, reduing operative time as p ossible, disposing pulmonary complications in time and using respirator if neces sary.
8.Comparison of biological properties between adult peripheral blood derived smooth muscle progenitor cells and mature vessel smooth muscle cells

BACKGROUND:Vascular smooth muscle is the main cell component of constructing vascular wall tissue structure and maintaining vascular tension.Studies have shown that smooth muscle progenitor cells obtained from peripheral blood can differentiate into vascular smooth muscle cells.OBJECTIVE:To compare smooth muscle progenitor cells dedved from human peripheral blood with mature vessel wall smooth muscle cells in cell morphology and proliferation potential.DESIGN,TIME AND SETTING:Cell culture and observation trials were performed at the Key Laboratory of Molecular Medicine in Jiangxi province from October 2008 to April 2009.MATERIALS:Peripheral blood 20mL were adopted from 6 healthy volunteers,and great saphenous vein were sourced from 6 patients with varicose vein.METHODS:Mononuclear cells were isolated from human peripheral blood and cultured in fibronectin,platelet derived growth factor-BB and EGM-2 medium.At the same time,the smooth muscle cells were obtained from great saphenous vein with the collagenase digestion method.High-glucose DMEM medium was used for the culture.MAIN OUTCOME MEASURES:Cellular morphology;Cellular immumofluorescence method and RT-PCR method were used to detect the expression of α-SMA and calponinl.MTT method was used to analyze the proliferation activity.RESULTS:The smooth muscle progenitor cells denved from human peripheral blood exhibited biological characteristics of vascular smooth musc Je cells.they were found to express α-SMA and calponin1.The proliferation activity of smooth muscle progenitor cells was also greater than vascular smooth muscle cells.CONCLUSlON:Peripheral blood mononuclear cells in the stimulus of platelet-derived growth factor can differentiate into smooth muscle progenitor cells.Compared to smooth muscle cells,smooth muscle progenitor cells proliferate better,and are not easy to age in a long time of cultivation.Smooth muscle progenitor cells are a ideal ceil source of tissue engineered vessel.
9.The cardioprotective effect of the inhibitor of JAK/STAT on simulating ischemia-reperfusion injury in cultured immature rat cardiacmyocytes

Objective To investigate the cardioprotective effects of the inhibitor of JAK/STAT pathway on ischemia-reperfusion injury in cultured immature rat cardiacmyocytes.Methods The cultured immature rat cardiacmyocytes were randomly divided into normal control group,ischemia-reperfusion group,AG490 treatment group and RMP treatment group.The cell viability was measured by MTT method.The contents of lactate dehydrogenase(LDH) and malondialdehyde(MDA),the superoxide dismutase(SOD) activity and the rate of cell apoptosis were detected.Results Compared with the normal group,the cell viability decreased significantly,the contents of LDH and MDA in the supernatant of the cells increased significantly,the SOD activity decreased and the rate of apoptosis of cells increased significantly in ischemia-reperfusion group(all P
