1.Effect of isoflurane on expression of IL-1β mRNA,IL-6 mRNA and TNF-α mRNA In the hippocamapus of immature rats

Obiective To investigate the effect ofisoflurane on expression of IL-1β mRNA,IL-6 mRNA and TNF-α mRNA in the hippocampus of immature rats.Methods sixty-four 7-clay-old SD rats were randomly assigned into 2 groups(n=32 each):control group(group C)and isoflurane group(group S).group S was exposed to 1.5% isoflurane for 6 h while group C to air.Fore animals were killed before anesthesia(T0,baseline),at 2,4,6 h(T1-3)of isoflurane anesthesia and 4,6,12 and 24 h after anesthesia(T4-7).The hippocampi were immediately removed for determimation of the expression of IL-1β mRNA,IL-6 mRNA and TNF-α mRNA by RT-PCR.Results Compared with group C,the expression of IL-1β mRNA at T1-5,IL-6 mRNA at T2.3 and TNF-α mRNA at T1-6 in the hippocampus was upregulated in group S.Conclusion The expression of IL-1β mRNA,IL-6 mRNA and TNF-β mRNA was elevated in the hippocampus of immature rats after being exposed to isoflurane.
2.Comparison of the therapeutic effect of several drugs infused into the bladder for superficial tumors after transurethral resection of the bladder tumor

Purpose:To compare the therapeutic effect and significance of different drugs poured in intravesical instillation for superficial biadder tumor after transurethral resection of bladder tumor.Methods:117 patients of superficial bladder cancer after transurethral vesection were divided into four groups. Ballistocardiogram group(n=26);Mitomycin C group (n=27);adriamycin group (n=27);hydroxycamptothecin group(n=37).Results:The tumor recurrent rate in Ballistocardiogram group, mitomycin groups, adriamycin group, hydroxycamptothecin group were 30.77%,40.74% 33.33%,24.32% respectively. The tumor recurrent rate differed significantly between the hydroxycamptothecin group and the mitomycin and Adriamycin groups.Conclusions:Intravesical instillation of hydroxycamptothecin for patients with superficial bladder tumor after transurethral resection of bladder tumour could decrease tumor recurrent rate, prolong the tumor-free survival period.
3.Administering a second rinse solution of low-potassium-dextran before reperfusion ameliorates the donor lung

Objective To studied the effects of administering a second rinse solution of low-potassium-dextran(LPD) before reperfusion on the donor lung of rat after long time,cold ischemic preservation.Methods Twenty-four rats were randomly divided into control group(n=12) and trial group(n=12).Donor lungs of control group were flushed with LPD solution.The levels of malonaldehyde(MDA),myeloperoxidase(MPO) and interleukin-8(IL-8) were tested in the tissue of right lung and the left lungs were reperfused with venous blood for 30 minutes.The partial pressure of oxygen(PaO_2),PaP and PawP were measured at every 10 minutes intervals during reperfusion.After reperfusion,the MDA,MPO activity and interlukin-8(IL-8) were determined in the lung tissue.Results The levels of MDA,MPO and IL-8 decreased significantly in right lung of trial group than those in control group(P

Objective To investigate the effects of isoflurane anenthesia on myocyte enhancer factor 2(MEF2) signaling pathway in neonatal rat hippocampus. Methods Twenty-four 5-day-old SD rats of both sexes,weighing 10-13 g, were randomly divided into 2 groups ( n = 12 each): control group (group C) and isoflurane group (group I). In group I, 1.5% isoflurane in 100% O2 was inhaled for 6 h. Group C received no treatment.Three rata in each group were sacrificed at 2, 4, 6 h of isoflurane anenthesia and 24 h after isoflurane anenthesia (T1-4), and the hippocampi removed for determination of MEF2 mRNA, synGAP Ⅰ mRNA, Arc mRNA and synapsinⅠ mRNA expression (by PT-PCR) and synapsin Ⅰ protein expression (by Western blot).Results Compared with group C, the expression of MEF2 mRNA, synGAP Ⅰ mRNA, Arc mRNA and synapsin Ⅰ mRNA at T1-3 and synapsin Ⅰ protein at T2-4 was up-regulated in group I ( P < 0.05). Conclusion Inhalation of anaesthetic concentration of isoflurane may affect synapse formation during the development of central nervous system by actirating hippocampal MEF2 signaling pathways in neonatal rats.
5.Synthesis of novel acacetin derivatives and evaluation of their activities

Objective To improve the in vivo analgesic activity of acacetin and find leads for the development of new drugs, novel acacetin derivatives containing alkyl amide groups with different length of carbon chain were designed and synthesized according to the molecular structure of the active hit compound found in our previous work. Methods Using apigenin as the initial chemical ma-terial,the acacetin was synthesized through 3 steps,then the target compounds were prepared by conjugating hydroxy group of acace-tin at position 7 with bromoalkyl amides. The analgesic activity of the target compounds was evaluated by acetic acid writhing model of mice. Results and Conclusion Novel acacetin alkyl amide derivatives showed more potent analgesic activities than that of clinical medicine diclofenac,which could be used as leads for further development of new drugs.
6.Effects of silymarin on homocysteine-induced apoptosis in human umbilical vein endothelial cells

AIM: To investigate the effect of silymarin on homocysteine-induced cell viability and apoptosis in human umbilical vein endothelial cells (HUVECs). METHODS: Cell viability was analyzed by using MTT and LDH assay. Apoptotic cells were detected by using DNA fragmentation and flow cytometric analysis. The level of intracellular reactive oxygen species (ROS) and the potential of mitochondrial membrane were determined by flow cytometric assay. The activity of caspase-3, -6 and -9 were measured with microplate spectrofluorometer. Protein levels were examined by Western blotting. RESULTS: Treatment of cultured HUVECs with HCY for 48 h induced a significant decrease in cell viability, and the percentage of apoptosis increased to 76.8%. The level of intracellular ROS and activity of caspase-3, -6 and -9 enhanced, and the red/green ratios of mitochondrial membrane decreased. However, simultaneous treatment with silymarin exhibited cytoprotective effects, reduced formation of the DNA ladder, prevented the levels of Bax and Bcl-2 proteins and the accumulation of ROS as well as caspase-3, -6 and -9 activation, reconverted the potential of mitochondrial membrane, and the percentage of apoptosis/necrosis was significantly decreased to 12.7% in a dose-dependent manner. CONCLUSION: These results demonstrate that silymarin has the protective capacity to antagonize HCY-induced apoptosis in HUVECs. The antiapoptotic action of silymarin may be partially dependent on an anti-oxidative stress effects, inhibition of caspases activity, and maintenance of mitochondria function.
7.Pregnantal ultrasonography on fetus' complicate congenital heart diseases

Objective To discuss the value of pregnantal ultrasonography(US) on fetus' complicate congenital heart diseases(CHD). Methods The uhrasonographic fetures of fetus complicate CHD were analyzed and compared with the pathological diagnosis. Resells Twenty fetus' complicate CHD were diagnosed by pregnantal US. Four cases with single ventricle, two eases with transposition of great arteries,two eases with double outlet right ventricle,two cases with truncus arteriosus,three cases with hypoplastic left heart syndrome, three cases with endocardial cushion defect, two cases with hypoplastic right heart syndrome,one case with tetralogy of Fallot and one case with pulmonary atresia were included. The diagnosing according rate was 100%. Conclusions Pregnantal US can fully evaluate the fetus' complicate CHD.
8.Study on metabolic changes of myocardial tissue in rats exposed to macleaya cordata

Objective To analyze the metabolic changes of myocardial tissue in rats under acute exposure to macleaya cordata by gas chromatography mass spectrometry(GC-MS),explore forensic identifications of its characteristic metabolites,and verify its toxicological mechanism in poisoning cases.Methods The rats in the exposure group were given 382 mg/kg macleaya extract solution by gavage,and the rats in the control group were given the same dose of solvent.The myocardial samples were analyzed by GC-MS,and pattern recognition was conducted through partial least squares discriminant analysis(PLSDA).The differential metabolites with characteristic changes were identified by variable importance projection(VIP value>1)and Student's t test(P<0.01).Results Compared with the control group,21 potential characteristic metabolites were identified.Through KEGG pathway enrichment analysis,it was found that these metabolites were mainly involved in the pathways of glycine,serine and threonine metabolism;pyruvate metabolism and glycerolipid metabolism.Conclusion Through the study of myocardial metabolism in rats exposed to macleaya cordata,we found the information on metabolites closely related to poisoning,which provides new insight and reference for studies on the mechanisms of macleaya cordata poisoning in the field of forensic medicine.

Here, we took base construction of neurosurgery as example to discuss and analyze according to requirements and evaluation indexes of base construction in Xinqiao Hospital, and put forward the specific objectives, measures and implementations of base construction. Foremost, we summarized experiences and overcame shortcomings through interpreting and implementing scheme of our base construction, which would help to improve the construction of standardized residency training base in China.
10.Navigated transcranial magnetic stimulation in preoperative evaluation of epilepsy in functional zones

Objective To investigate the value of navigated transcranial magnetic stimulation (nTMS) in preoperative evaluation of epilepsy in functional zones.Methods Retrospective analysis was performed in 12 refractory epilepsy patients had functional cortical lesions and accepted surgery in our hospital from January 2016 to March 2017.The target surgery areas were considered in or near the functional areas by video electroencephalogram and stereotactic electroencephalography or Wada test,nTMS was used to identify the motor or language functional areas before surgery,and direct cortical stimulation (DCS) was performed during the surgery to identify the accuracy of nTMS.Treatment efficacies were evaluated 6-18 months after surgery.Results Hand motor areas were detected in all patients by nTMS,of which 72.7% patients (8/11,one severe hemispheric atrophy patient was excluded due to functional zone drift to the opposite) were coincided with intraoperative DCS (<0.5 cm),the deviation distance between 0.5 cm and 1 cm was noted in 27.3% patients (3/11);motor speech areas were detected in 11 patients with nTMS,72.7% patients (8/11) were coincided with intraoperative DCS (<0.5 cm),the deviation distance between 0.5 cm and 1 cm was noted in 27.3% patients (3/11),18.2% patients (2/11) were for adjacent relationship,and one was with large difference (>1.5 cm).And 8 patients had Engel grading Ⅰ outcome (seizure-free),2 had grading Ⅱ outcome (almost seizure-free),and one had grading Ⅲ outcome (worthwhile improvement).No persistent function deficits were noted.Conclusion The nTMS is an accurate,noninvasive and safe functional localization method,which can provide important information for epilepsy patients especially who are not suitable for invasive language evaluation in epilepsy surgery.
