1.Advances of gemcitabin in non-small-cell lung cancer

Gemcitabine has consistently shown activity as a new agent in non-small-cell lung cancer. In non-small-cell lung cancer, gemcitabine-cisplatin combination response rates range from 31% to 54%, with a median survival time between 8.4 and 15.4 months and a 1-year survival rate between 30% and 59%. The GP schedule has acceptable toxicity. The GP combination can be considered standard therapy for non-small-cell lung cancer. Both 21- and 28-day schedules of GP were equally effective in non-small-cell lung cancer. It is necessary to study by randomized clinical trial, the sequence of cisplatin in GP schedule, the increase of radiosensitivity by gemcitabine, the combination of gemcitabine and other new chemotherapeutic agents.
2.Expression and Prognostic Significance of CD44v6 in Advanced Cervical Cancer

The expression of CD44v6 in 105 cases of advanced cervical cancer was detected by using im munohistoehemical method and the relation between adhesion molecule CD44v6 and prognosis in advanced cervical cancer was investigated. The experimental results showed that the lymphocytic infiltration and peritumor reaction of fibrin in the stroma were milder in the patients with CD44v6 positive than those with CD44v6 negative. There were significant reductions in overall and free relapse survival in the patients with CD44v6 positive as compared with those with CD44v6 negative. It was suggested that the expression of CD44v6 might serve as a new indicator for predicting the prognosis of advanced cervical cancer. Tumor cells might interfere with the recognition of the peritumor lymphocyte and reduce the peritumor reaction of fibrin in the stroma through the expression of CD44v6.
3.The feasibility of using International Classification of Functioning, Disability and Health core set for Chinesebreast cancer patients

Objective To determine the reliability, validity and feasibility of using International Classifica-tion of Functioning, Disability and Health (ICF) core set for breast cancer in Chinese patients. Methods Quality of life (QOL) of 110 patients with breast cancer were investigated using ICF breast cancer core set and SF-36 scale. Statistical analysis tested their reliability, validity and feasibility. Results It was shown that the ICF core set for breast cancer had good reliability, validity and feasibility. Conclusions The ICF core set for breast cancer is a re-liable and valid instrument for Chinese breast cancer patients.
4.Analysis of Quality of Life in patients with uterus cervical cancer

Objective To investigate the quality of life (QOL) of cervical cancer patients and its influential factors as well as to explore the method for improving the QOL in this population of patients. MethodsA QOL questionnaire for uterus cervical cancer (QOL-UCC) was designed and used in a survey of 81 UCC patients before they got any treatment and 124 UCC patients after they were treated for a period of time. SPSS 9.0 software was used to analyze data. ResultsThe patients got poor score with psychological status items(2.11? 0.34 ) before treatment, while those who had been treated for a period of time got poor score with social characteristics items(2.01? 0.33 ), especially with the work(1.87)and sexuality (1.00) items. Economic status of the patients, unsuccessful control of progress of the disease and recurrence were the main influential factors of the QOL in the UCC patients. ConclusionPsychosocial factors play very important roles in QOL of the UCC patients. As a result, psychosocial interventions should be stressed in the rehabilitation of this population of patients.
5.Design and validation of a QOL scale for the Chinese patients with uterocervical cancer

Objective To develop and validate a qual it y of life(QOL) scale for the Chinese patients with uterocervical cancer (QOL-UC C). Methods Based on the internationally accepted de finition of QOL and the clinical characteristics of uterocervical cancer as wel l as the existing QOL scales, we designed the QOL-UCC. Then 113 patients were r ecruited and the reliability, validity and feasibility of the scale were tested. Results It was shown that the QOL-UCC had good reli ability, validity and feasibility. Conclusion QOL-U CC is a reliable and valid instrument for evaluation of the QOL in Chinese pa tients with uterocervical cancer.
6.Effects of hypoxia on apoptosis of human lung adenocarcinoma A549 cell line by adriamycin

Background and purpose:Hypoxic cells in tumors are of chemoresistant and anti-apoptoic.Our study was focused on the effects of hypoxia on expression of P-glycoprotein(P-gp) and cell apoptosis in human lung adenocarcinoma A549 cell line(wtP53),in order to explore the probable mechanism of hypoxia in apoptic tumor cells.Methods:A549 cell line(human lung adenocarcinoma) was cultured under hypoxic atmosphere(2% O_2) for 24 h,the expression of hypoxia inducible factor-1?、P-gp and P53 protein was detected by immunohistochemistry,after cells under hypoxic atmosphere were exposed to Adriamycin for 24 h,the cell survival rate was detected by MTT,and apoptosis of by flow cytometer.Results:① The expression of hypoxia inducible factor-1?、P-gp and P53 protein in hypoxic cells were higher than the expression in anoxia ,and correlation between the expression of HIF-1? and P-gp or P53 protein was observed(P
7.The Effect of Histone Deacetylases-1 siRNA on the Growth and Apoptosis of HeLa Cells

Objective To investigate the effect of histone deacetylases-1(HDAC-1)siRNA on the HDAC-1 protein expression,cell growth and apoptosis in HeLa cells.Methods The HDAC-1 protein was knocked down by HDAC-1 siRNA.HDAC-1 protein was detected by Western blot.The cell growth inhibition was assesses by MTT and clone forming assay.Apoptosis was measured by flow cytometry.Results Forty-eight h after transfection of HDAC-1 siRNA,HDAC-1 protein expression in HeLa cells was down-regulated obviously.The down-regulation of HDAC-1 significantly reduced the colony formation rate,inhibited cell proliferation and induced apoptosis in HeLa cells.Conclusion HDAC-1 siRNA may play an anti-proliferation role in HeLa cells by inducing apoptosis.
8.Clinical Analysis of 116 Cases of Keloids Treated by Combination of Surgical Resection and Immediate Postoperative Electron Beam Radiotherapy

Objective To evaluate the role of postoperative electron beam radiotherapy in the management of ke-loids. Methods Between January 2004 and December 2006,116 cases of keloids were treated with 6 MeV electron beam radiotherapy within 24 h after surgical resection of the keloids. All patients had received a total of 21 Gy in seven daily 3-Gy fractions. Treatment was started at the 24th h after surgery. Patients were followed up and the information regarding treatment results, early and late radiation toxicity,and the satisfaction level by self-assessments was recorded. Results Recurrence occurred in 17 cases of keloids(14. 7%),and was correlated with site of the lesions(χ~2 =29. 91,P<0. 01). Most recurrences were observed at site of sternum (10/43) and shoulder (5/13 ). Keloid associated symptoms, e. g. itching and pain, were improved in 88. %%. For early toxicity outcomes, 100% had grade 1~2 skin erythema,7. 8% had wound delayed union,and 4. 3% had infection. For late toxicity outcomes, 30. 2% reported grade 1~2 hyperpigmentation, 11. 2% grade 1~2 hypopigmentation, and 5. 2% grade 1 telangiectasia. No severe complications or secondary malignancies were observed. 72. 4% patients described the results of treatment as excellent or good,and 15. 5% patients were not satisfied with the treatment results. Conclusion Postoperative electron radiotherapy is well tolerated and effective in preventing keloid recurrence.
9.The effect of gefitinib on the quality of life in patients with advanced non-small cell lung cancer

Background and purpose:To improve the quality of life of patients with advanced non-small cell lung cancer is one of the problems which the oncologists have to be aware of. Gefi tinib has been used to treat advanced non-small cell lung cancer. We studied the effect of gefi tinib in the improvement of quality of life of patients with advanced non-small cell lung cancer. Methods:There were 70 patients with advanced non-small cell lung cancer treated in our cancer center. One oral gefi tinib tablet(250 mg) was administered every day without interruption unless disease progression or unacceptable toxicity occurred. The impact of treatment on disease-related symptoms and quality of life(QoL) were evaluated with the Chinese versions of European Organization for Research and Treatment of Cancer Quality of Life Questionnaires(EORTC QLQ-C30 and QLQ-LC13) . Results:58 patients finished the questionnaires. The mean scores of fi ve functional scales(physical,role,emotional cognitional and social) were 63,56,68,65,61 respectively after eight weeks of treatment,compared to 52,49,64,60,52 respectively before treatment,and the mean score of global QoL after and before treatment were 60 and 53 respectively. 10.The study on the expression of HIF-1? and its association with the expression of HER-2 and survivin in breast carcinoma

Background and purpose:HIF-1?、HER-2 and survivin are all considered as important prognostic factors of breast cancer,but the relationships among them are poorly understood.We investigated the expression of HIF-1?、HER-2 and survivin in Breast Carcinoma,and explored the relationship among the three prognostic factors.Methods:The expressions of HIF-1?、HER-2 and survivin were detected by immunohistochemistry in 51 human Breast Carcinoma samples.Results:① 28(54.88%),11(21.69%) and 35(68.62%) were positive for HIF-1?、HER-2 and survivin,respectively.② A significant difference was found in terms of the overexpression of HER-2 between the samples with HIF-1? either positive or negatiwe(P0.05).Conclusions:The positive expression of HIF-1? was strongly correlated with the overexpression of HER-2,but not survivin in breast carcinoma.
