1.Selective Enhancement of Open-skill Exercise on Attention Network of College Students

Objectives To investigate the relationship between different exercise types and the attention network.Methods A total of 75 university students were enrolled and randomly divided into an open-skill exercise group,a closed-skill exercise group and an irregular exercise control group,each of 25.They underwent the exercise as their group names implied.Then the attention network test was performed to evaluate their functions of the alerting network,orienting network and executive control network.Results Significant differences were found in the function of the executive control network among the 3 groups,while no significant difference was found among the 3 groups in the functions of the alerting network and orienting network.Conclusions Both open-and closed-skill exercise are associated with better function of executive control network.Furthermore,open-skill exercise showes more benefits than closed-skill exercise in promoting the function of the executive control network.Neither opennor closed-skill exercise bring benefits to the function of the alerting or orienting network.

3.Experimental study on astrocyte-derived factors on the differentiation of neural stem cells

Objective To study the effect of astrocyte-derived factors on the differentiation of neural stem cells from neonatal rats. Methods Neural stem cells from neonatal rats were isolated, cultured and expanded by neurosphere formation. Astrocytes were isolated and purified by a standard shaking method and the differential adhesion technique. With immunocytochemical labeling, the purity of astrocytes was determined by the expression of glial fibrillary acidic protein(GFAP). Astrocytes and neural stem cells were co-cultured without contact. Immunofluorescence examination was used to detect the effects of astrocyte-derived factors on the expression of neuron specific enolase (NSE), GFAP, and tyrosine hydroxylase(TH) of the differentiated cells. 4.Diagnostic value of Mini-laparoscopy in abdominal neoplasmas

Background and purpose:Blood biochemistry and ascites test, imageology examination has low sensitivity in abdominal neoplasmas diagnosis. And also exploratory laparotomy is not suitable for final stage patients. Mini-laparoscopy has been a new diagnostic technology for abdominal disease recently. We aimed was to evaluate the value of Mini-laparoscopy in diagnosing Abdominal Neoplasms. Methods:By reviewing the clinical and operational data, a retrospective analysis of 20 cases with pathologically confirmed abdominal malignancies was performed. Among this group, 10 cases were diagnosed by mini-laparoscopy, and the other 10 cases by exploratory laparotomy. Between the two groups, the surgical expense, anesthesia cost, para-operation nursing, monitoring and treating charges, post- operative hospital days and complications were compared. 5.The expression of survivin gene in gastric cancerous tissue and its relationship with gastric cancer prognosis

Objective To explore the expression of survivin gene in gastric cancerous tissue and its relationship with gastric cancer prognosis. Methods All the samples were obtained from patients with gastric cancer who underwent surgery in Zhongshan Hospital from July 1995 to June 1996.“ Envision two steps”of immunochemistry was used to detect survivin expression in the resected gastric cancerous tissue.TNM stages were determined by pathological diagnosis and clinical status.All the cases were followed up at least 5 year or until death.The discrepancy of survivin expression was compared in different pathological types and TNM stages.Survivial curves were compared in the two groups with or without survivin expression. Results Totally 96 cases(male/female:59/37;age:29-84 years old,mean age 56) were recruited and survivin expression rate was 70.8%(68/96).Among pathological types,78 cases were adenocarcinoma,18 were non-adenocarcinoma.Survivin expression was similar in different pathological types (69.7% in adenocarcinoma vs.60.0% in non-adenocarcinoma,P=0.369).Among adenocarcinoma,survivin expression rate was higher in poorly differentiated group than that in well differentiated one(82.1% vs. 62.5%,P=0.053).As the infiltration denoted in adenocarcinoma,the expression rates of survivin were 33.3% in cases limited to mucosa and submucosa,81.3% in cases limited to muscularis layer and 75.9% in cases infiltrated to whole layers (P= 0.020).Expression rate of survivin was not related with lymph node metastasis(68.1% vs.70.8%,P= 0.771).Five-year survival rate in 68 survivin positive cases was 42.93%,lower than 52.93% of 28 survivin negative cases,but no statistical difference was observed. Conclusions Survivin is highly expressed in gastric cancer.Survivin expression is closely related with differentiation status of adenocarcinoma and degree of infiltration.Further studies are needed to evaluate its role in the prognosis of gastric cancer.
6.Construction of recombinant lentivirus vector expressing small hairpin RNA against human Bax inhibitor-1 gene expression

BACKGROUND: Lentivirus-mediated RNA interference technology has been widely used in the study of gene function because of its specificity, high inhibition efficiency and persistent effects. Virus packaging, transfection, as well as small hairpin RNA (shRNA)-coding sequence could affect the inhibition efficiency. Therefore, impact factors of RNA interference for human Bax inhibitor 1 (BI-1) was studied in this experiment.OBJECTIVE: To find the valid small interference RNA (siRNA) targeting human BI-1 gene by Lentivirus.DESIGN, TIME AND SETTING: Single sample observation was completed in Department of Medical Genetics, Basic Medical College, Peking University Health Science Center from September 2007 to December 2008.MATERIALS: 293T cell and SH-SY5Y cells were preserved in this laboratory; 4 shRNAs were designed through a use of online design tool by Ambion (www.ambion.com). METHODS: Several recombinant plasmids which expressed shRNAs targeting different regions of the BI-1 gene and labeled enhanced green fluorescent protein as a fusion gene were constructed. Four types of shRNA-expressing virions were obtained by cotransfection of shRNA-expressing plasmid and package protein-expressing plasmids into 293T cells. Green fluorescent protein expression was determined using flow cytometry to search the optimal package conditions. Real-time PCR and beta actin mRNA was used as an internal control, four types of recombinant viral supernatant and control viral supernatant were added on the SH-SY5Y cells, knock-down efficacies of the endogenous BI-1 gene were determined, and the RNA interference effective sequences were optimized. MAIN OUTCOME MEASURES: Intracellular green fluorescent protein expression rate of the cells infected with different packaging virus. RESULTS: The optimal infective packaging virus could be obtained when the ratio of plasmids pLentiLox3.7, REV, VSVG and RRE was 2:1:1:1 and the supernatants were collected 48 hours after transfection. Replacement of culture media 24 hours after transfection could increase the infection efficacy of the virions. shRNA targeting-2-17nt (start coding region) of BI-1 gene was most valid interference efficiency and could decrease the gene expression to 40%. CONCLUSION: Influencing factors of Lentivirus-mediated RNA interference technology virus packages were investigated in this study, which could be a foundation for constructing stable BI-1 knocked down neuronal cell model and for studying the abnormal expression of BI-1 related to the neuron apoptosis disease.

Objective To clarify the relation between the expression of Foxp3 in peripheral blood and plasma transforming growth factor betal (TGF-β1) and the activity of systemic lupus erythematosus (SLE). Methods Foxp3 mRNA expression of 28 active SLE patients, 13 inactive SLE patients and 16 healthy controls was determined by polymerase chain reaction. Active SLE patients were followed up; Foxp3 mRNA expression of 20 active patients was measured in the stable status and the plasma TGF-β1 was measured by ELISA in the active and stable status. Results The active SLE patients showed reduced levels of Foxp3 mRNA than the inactive SLE patients (P<0.01) and the healthy controls(P<0.01). Expression of Foxp3 mRNA and the plasma level of TGF-β1 in 20 SLE patients were both higher in stable status than in active status, Conclusion The expressions of Foxp3 mRNA in peripheral blood and TGF-β1 in plasma has significant inverse corelation with disease activity, which suggests that regulatory T cells may play an important role in the pathogenesis of SLE.
8.Proportion and tumorigenetic characteristics of side population cells in gastric cancer cell lines

Objective To study the tumorigenetic characteristics of side population (SP) cells and their exsistence in gastric cancer cell lines and tissues.Methods Flow cytometry and the DNA-binding dye Hoechst 33342 staining were used to analyze sorted SP cells in 3 gastric cancer cell lines(SGC-7901,BCIC-823 and MKN28).SP cells and non-SP cells(main population cells)isolated from SGC-7901 cell line weresubcutaneously injected into 36 nude mice with 500,5000 or 50 000 cells per mouse,respectively.Thetumorigenity was observed 8 weeks after injection.The expression of ATP-binding cassette sub-family Gmember 2(ABcG2)mRNA in three gastric cell lines and its level in gastric cancer tissues were detected by real-time PCR and immunostaining.respectively.Results The proportion of SP cells was accounted for 1.0%in SGC-7901 cells and 1.3%in BGc-823 cells.No SP cell was found in MKN28 cells.As low as 500 SP cells or 50 000 non-SP cells could initiate tumors in mice.The expression of ABcG2 mRNA was higher in SGC-7901(O.162)and BGC-823(O.096)cell lines than that in MKN28 cell line(0.005).The value of AtCG2 mRNA/beta-aetin mRNA and protein in gastric cancer tissues was different from those in gastritis tissues.Condasimm SP cells have strong ability of tumorigenesis compared with non-SP cells.The expression of ABCG2 is found in gastric cancer tissues and part gastritis tissues.The more the SP cells in gastric cell lines,the higher the expression of ABCG2.
9.The clinical characteristics and treatment evaluation of patients with Crohn's disease

Objective To analyse the clinical characteristics,follow up their prognsis and evaluate the treatment of patients with Crohn' s disease (CD). Methods The data of patients with CD were collected at Zhongshan Hospital of Fudan University from 2002 to 2007. The diagnosis was made according to the consensus recommended by Chinese Society of Gastroenterology. The disease severity, follow-up and prognsis were evaluated according to the consensus. Results Sixty-six CD patients were enrolled in this study. The ratio of males to females was 2.47 : 1. Age at diagnosis ranged from 12 to 76 years old, and the mean age was (32 ± 17)years old. The main gastroenterological manifestations were abdominal pain (80.3 %), diarrhea (54.6 %), fistula formation (31.8%) ; and the main systemic manifestation was fever (80.3 %) and defective nutrition. A total of 60.6 % of patients with CD had complications and 34. 8% of these patients had more than one complications. Extraintestial manifastation were presented in 7.6% of patients with CD. A total of 86.4% of patients with CD were active state cases and 57. 6% were suffered from inflammation of ileum and colon. Abdominal pain (P = 0. 011), hematochezia (P = 0. 008), fever (P = 0. 001), anemia (P = 0. 020), underweight (P = 0. 010) and heightening of CRP (P = 0. 033) were more common in patients who were in active state than those who were in slient state. During their inducing alleviate treatment, 33.3% of patients with CD were treated with aminosalicylate alone, 36.4% were treated with both aminosalicylate and glucocorticoids, 7. 6% were treated with aminosalicylate, glucocorticoids and immunosuppressant. After medicine treatment, 6.8% of the patients relieved, 62.7% turned better, 28.8% were ineffective, 1.7% died, 55.9% recurd. And 21.2% were treated by surgical operation. A total of 42.4% of patients with CD had histroy of surgical operation and 9.1% were carried on more than one surgical operation.ConclusionsActive state patients with CD had high rate of complications, low extraintestial manifestatons, rare rate of dysplasia and canceration, and high rates of surgical operation and recurring.

Taking global minimum essential requirements(GMER)as standard,school of basic medical science in Wuhan University paid more attention to the cultivation of students' professional skills, innovation ability and communication ability. ‘Response to injury’integrated course was carried out. Based on the immunology,this course involves pathology and pathophysiology and explores the mecha-nism of the diseases' diagnosis and treatment. It took small classes,bilingual education,literature read-ing ,experiments and discussions in various forms . This reform aimed to improve the immunology , pathology and pathophysiology teaching and make our medical education meet the process of globalization.
