1.Study on the linkage between the interluekin 4 receptor ? chain polymorphism and susceptibility of asthma and atopy

Objective To investigate the linkage between the mutation of IL 4R ? chain gene and susceptibility of asthma and atopy in Wuhan. Methods Thirty five samples from asthmatic children,twenty five samples from adult patients with asthma, twenty samples from health adult and children respectively, were analyzed. A coding region 1205 1928 of IL 4R ? chain gene was detected by reverse transcription polymerase chain reaction/Single Strand conformation polymorphism/Sequence. Result 57.8% cases from asthmatic children, one polymorphism (1902 A→G), were found exist in the IL 4R ? chain gene and only 8% cases from asthmatic adults and 5% cases from control have this mutation. Conclusion The SNP of IL 4R ? chain coding area 1902 correlates with susceptibility of asthma in Wuhan asthmatic children (? 2=12.54, P 0.05, ?=0.05). There may be another mutations in IL 4R ? chain coding area except 1902 or another changes in other gene related with allergy.

AIM: To study the effects of dexamethasone (DXM) on intracellular expression of TH1/TH2 cytokines and the mechanism of that during the development of asthma. METHODS: Eighteen BALB/c mice were divided into 3 groups at random: control group, asthma group, and DXM treated group, with 6 mice in each. The expressions of T-box expressed in T cells (T-bet) and GATA-binding protein-3 (GATA-3) in lung tissue were detected by Western blotting. Aim To study the effects of dexamethasone(DXM)and peroxisome-proliferation activated receptor?(PPAR?)agonist pioglitazone on intracellular cytokines expression of T_H1/T_H2 and the mechanism of that in asthma model,and compare the difference between two drugs.Methods 24 Balb/c mice were divided into 4 groups randomly: control group,asthma group,DXM group,and pioglitazone group,with 6 mice in each group.The expression of T-bet and GATA3 of lung were detected using western blot,and the intracellular cytokines interluekin 4 and interferon ? expressions of CD~(4+)Tcell were measured using flowcytometry.Results Compared with control group,the level of the expression of T-bet of lung in asthma group increased significantly(P0.05),while both of GATA3 and T-bet decreased after DXM treatment,and the decreasing degree of GATA3 was more than that of T-bet.The results of flowcytometry indicated that the ratio of intracellular cytokines IL-4/IFN? of CD~(4+) T cell in asthma group was much higher than that of control group(P
5.Association between IL-13 gene polymorphism and asthma in Han nationality in Hubei Chinese population.

In order to investigate whether Arg110Gln polymorphism in the coding region of the IL-13 gene is associated with asthma and total plasma IgE level in Han nationality in Hubei Chinese population, the allele frequency of 4257(g/a) site and Arg110Gln genotype of IL-13 was detected by using restriction fragment length polymorphism in Han nationality in Hubei Chinese population including 43 asthmatic children, 45 asthmatic adults, 31 control children and 46 control adults. Total plasma IgE was measured by Chemiluminescence assay. The results showed that the frequency of allele A at 4257 bp of IL-13 in children and adults was 0.39 and 0.32, respectively. The GlnGln form of Arg110Gln polymorphism of IL-13 gene was associated with susceptibility of asthma and elevated total plasma IgE in children (P=0.030 and 0.0009, respectively), but not with them in adults (P=0.219 and 0.174, respectively). Our results suggest that the Arg110Gln polymorphism of IL-13 gene is associated with susceptibility of asthma and elevated total plasma IgE in Chinese children of Han nationality in Hubei, but not with them in adults.

7.Genetic polymorphisms of CYP2D6 in Chinese mainland.

OBJECTIVETo observe the significant differences in the frequencies of the cytochrome P450 2D6 (CYP2D6) alleles in Chinese popoulations.
METHODSTetra-primer polymerase chain reaction (PCR), allele specific amplification (ASA) PCR and multiplex long PCR were developed to detect the CYP2D6 alleles * 2, * 3, * 4, * 5, * 6, * 8, * 10 and * 14 in 223 subjects from Chinese mainland.
RESULTSThe CYP2D6 * 5 allele was the most frequent poor metabolizer (PM) allele in Chinese (7.2%), followed by CYP2D6 * 14 (2.0%) which was only detected in orientals. There was only 0.2% CYP2D6 * 4, and no CYP2D6 * 3, * 6 and * 8 were detected. In contrast to the Caucasians, the most frequent allele in Chinese was the * 10 allele with a frequency of 51.6%.
CONCLUSIONThe frequencies of PM alleles, CYP2D6 * 5 and CYP2D6 * 14, were higher; but the frequency of CYP2D6 * 10 was lower in mainland Chinese population than that in other orientals.

8.Mechanism of Qizhenziyin mixture in the treatment of hypogonadism based on network pharmacology analysis and molecular docking

Objective To investigate the mechanism of Qizhenziyin mixture in the treatment of hypogonadism by using the network pharmacology approach. Methods The active components of Qizhenziyin mixture were obtained by searching TCMSP ,TCMID and HIT databases.The related targets of candidate compounds were obtained by searching STITCH databases. The potential targets of Qizhenziyin mixture in the treatment of hypogonadism were obtained by mapping the disease genes of hypogonadism with Genecards and DisGeNet databases. The protein interaction platform database (STRING) was used to construct the interaction relationship between action targets. The target protein interaction (PPI) network was constructed by introducing Cytoscape software. The mechanism of Qizhenziyin mixture in the treatment of hypogonadism was explained through the enrichment analysis of GO, KEGG and molecular docking technology. Results A total of 148 drug-disease chemical compounds, 96 drug-disease intersection targets, 1085 disease targets were obtained;the components for treating diseases are: quercetin,kaempferol, luteolin, etc; enrichment analysis of GO revealed 1792 biological processes (BP), 31 cellular components (CC) and 79 molecular functions (MF);the results of KEGG pathway enrichment analysis indicated such as FOXO signaling pathway, prostate cancer, AGE-RAGE signaling pathway in diabetic complications, HIF-1 signaling pathway, etc.The results of molecular docking showed that kaempferol and LEP had the best and stable binding energy. Conclusion The active components of Qizhenziyin mixture may play a role of the treatment of hypogonadism by improving insulin resistance and the expression of testosterone synthetase of Leydig cells.
