Objective To investigate the applied value of adipose-derived stromal cells(ADSCs) transfected glial cell-derived neurotrophic factor(GDNF)by lentivirus in cerebral transplantation.Methods Primarily isolated and cultured, the rat ADSCs were transfected GDNF by high-titer lentivirus. Then, configuration of cells and expression of neuron-specific enolase (NSE) were observed. By stereotaxic frame, transfected ADSCs were intrastriatally transplanted.1month and 2month after transplantation, survival and migration of marked cells were detected. GDNF expression, verified by Western blot, was compared with control group.Results ADSCs, transfected by lentivirus, can differentiate into neuron-like cells and express NSE in cytolymph. 1 month after transplantation, the majority of cells survived in transplanting area. 2 month later, part of transplanted cells still survived and migrated far from the transplanting area. Objective To investigate angiogenesis and mosaic vessel in colorectal carcinoma.Methods Forty-nine patients suffering from colorectal carcinoma were studied.Endothelial cell was labelled with monoclonal anti-human CD_~31 antibody,tumor cell was labelled with monoclonal anti-human CEA antibody.Endothelial cell and tumor cell were labelled by double-stained immunohistochemistry.The quantity of microvessel was estimated by counting microvessel density (MVD) at hot spots,and the quantity of mosaic vessels (MV) and double-stained cells were estimated at the same visual field.The relationship between mosaic vessels,microvessel density and Dukes staging were analyzed.Results The MVD in colorectal carcinoma "hot spots" was counted.The MVD of Dukes staging B was 43.98?21.46,staging C was 59.54?26.95,and staging D was 70.80?19.04.MVD increased gradually accompanied by clinical staging.There was statistical difference between staging B and D (P
3.Screening of serum biomarkers and establishment of decision tree in silica-exposed populations by SELDI-TOF-MS

Objective To screen the serum biomarkers by surface-enhanced laser desorption-ionization time-of-flight mass spectrometry (SELDI-TOF-MS) and establish the decision-tree model for the silica-exposed populations.Methods In addition to 20 healthy people without silica-exposed history chosen as control group,all the subjects diagnosed as different stages (stage 0 group,silica-exposed population,n=20;stage 0+ group, doubtful silicosis,n=20;stage Ⅰ group,stage Ⅰ silicosis) of silicosis according to the national diagnostic criteria (GBZ70-2002) of pneumonoconiosis with no complications were recruited. SELDI-TOF-MS and CM10 chip assay were applied to acquiring serum protein finger printing. The bioinformatic software was adopted to perform data ananlysis and establish Wilks'lambda decision equation and decision tree.Results As compared with that of control group, differential peaks appeared in three groups of silica-exposed population, accounting for 80% of total differential peaks. Wilks'lambda decision equation was established with M/Z 3711.73, M/Z 4183.66, M/Z 5487.28 and M/Z 6124.64, which sensitivity and specificity in differentiating healthy controls and silica-exposed population was 95% and 90% respectively. The sensitivity and specificity of decision tree was 100% and 95% respectively. Neither single peak nor Wilks'lambda decision equation worked well for discrimination of silica-exposed population, while the discriminating rates of decision tree accounted for 100%, 95%, 85% and 90% for healthy control group, stage 0 group, stage 0+ group, stage Ⅰ group respectively.Conclusion M/Z 5933.63,M/Z 4183.66,M/Z 5487.28,M/Z 6124.64,M/Z 5915.14,M/Z 2745.7, M/Z 3711.73, M/Z 2954.78 and M/Z 3401.03 are potential biomarkers for discriminating healthy controls and silica-exposed population, and the decision-tree works well for discrimination of controls, silica-exposed population, doubtful silicosis patients and stage Ⅰ silicosis ones.

OBJECTIVE: To explore the effects of Gymnadenia conopsea alcohol extract (GcAE) on the collagen synthesis in rat lungs exposed to silica and the influence on antioxidase activities, level of lipid peroxidation (LPO). METHODS: One hundred and twenty rats were randomly divided into control group, silica group, and GcAE-treated group. Silicotic animal models were established by direct tracheal instillation of silica into rat lungs surgically. From the second day of model establishment, rats in GcAE-treated group were orally given GcAE [8 g/(kg x d) corresponding to raw herb]. At 7, 14, 21, 28 and 60 days after establishment of the animal model, eight rats in each group were sacrificed, and samples were collected. The malondialdehyde (MDA) content, superoxide dismutase (SOD) and glutathione peroxidase (GPx) activities in plasma were assayed by a spectrophotometer. Types I and III collagen were detected by Sirius red polarization and microscopy, and measuered by Image-Pro Plus Version 4.5 for Windows software. RESULTS: GcAE could reduce the lung/body weight ratio of rats exposed to silica, the synthesis of types I and III collagen of the lungs and the level of lipid peroxidation, increase the activities of SOD and GPx. CONCLUSION: GcAE can ameliorate the silica-induced pulmonary fibrosis by increasing the activities of antioxidase and alleviating the damage of lipid peroxidation to the lungs.
6.The expression of macrophage migration inhibition factor in pulmonary tissues of smokers with or without chronic obstructive pulmonary disease

Objective To investigate the expression of macrophage migration inhibition factor (MIF) in pulmonary tissues of the smokers with and without chronic obstructive pulmonary disease (COPD).Methods The subjects were assigned into three groups:non-smokers without COPD (control group,n =12),smokers without COPD (smoker group,n =13) and smokers with COPD (COPD group,n =16).The specimens were obtained from lung tissues as far away from cancer focus as possible (> 5cm).Real-time quantitative PCR and immunohistochemistry were used to investigate the expression and distribution of MIF in pulmonary tissues.The relationship between the severity of airflow obstruction and the differential expressions of MIF in lung tissues of the smokers with or without COPD was analyzed.Results (1) MIF mRNA expression in COPD group (4.87 ± 1.79) was higher than that in the smoker group (2.16 ±0.72;P<0.01),which was higher than that in the control group (1.09 ±0.48;P <0.01).(2)Immunohistochemistry analysis showed that MIF protein expression in lung tissues of the COPD group (0.277±0.025) was higher than that in the smokers group (0.199 ±0.034;P <0.01),which was significantly higher than that in control group (0.130 ±0.021 ;P <0.01).(3) Correlation analysis of MIF mRNA expression in the lung tissues and pulmonary function parameters of forced expired volume in one second (FEV1) percentage of predicted (FEV1 pred) and ratio of FEV1 to forced vital capacity (FEV1/FVC) suggested that MIF mRNA expression in the lung tissues was negatively related with FEV1 pred (r=-0.578,P < 0.01) and FEV1/FVC (r =-0.607,P < 0.01).Conclusions MIF expression significantly increases in the smokers with COPD,and MIF level in the lung is positively correlated with airflow limitation.The results suggest that MIF may play an important role in the pathogenesis of smokinginduced COPD.
7.Functional polarization of T cells and clinical value in children with obstructive sleep apnea hypopnea syndrome

Objective To investigate the functional polarization of T cells in children with obstructive sleep apnea hypopnea syndrome (OSAHS).Methods Forty five cases of OSAHS children were chosen to undergo tonsillectomy and adenoidectomy.Preoperative analyses of peripheral blood Th1 /Th2 ratio and Tc1 /Tc2 ratio were compared with those of normal children and those of postoperative 6 months for observation of the change of T cells.Th1/Th2 ratio and Tc1/Tc2 ratio were detected by flow cytometry.Results The preoperative ratio of Th1 cells in children with OSAHS was significantly lower than normal children [(9.66 ± 3.73) % vs (13.34 ± 4.36) %,P < 0.05],while the preoperative ratio of Th2 cells was significantly higher than normal children [(3.42 ± 1.64) % vs (2.53 ± 1.97) %,P < 0.05],which made the Thl/Th2 ratio lower than normal children (3.51 ± 1.53 vs 5.41 ±2.17,P <0.05).At the 6th month after tonsillectomy and adenoidectomy,the postoperative ratio of Th1 cells was significantly higher than preoperative ratio [(12.99 ± 3.68) % vs (9.66 ± 3.73) %,P < 0.05],while the postoperative ratio of Th2 cells was lower than preoperative ratio [(2.62 ± 1.88) % vs (3.42 ± 1.64) %,P < 0.05],which made the postoperative Th1 /Th2 ratio higher than preoperative ratio (5.15 ± 1.86 vs 3.51 ± 1.53,P < 0.05).There was no significant difference in Tc1 /Tc2 ratio between the OSAHS children and normal children.Conclusions Functional polarization of T cells was abnormal in OSAHS children accompanied by the predomination of Th2 cells and the normal Tc cell polarization.Functional polarization of T cells can return to normal at the 6th month after tonsillectomy and adenoidectomy.
8.Expression of caveolin-1 in osteosarcoma and its significance for clinicopathological features

Objective By detecting the expression of caveolin-1 in osteosarcoma tissues,to explore its possible roles in the occurrence,progress and metastasis of osteosarcoma and its relationship with clinicopathological features.Methods The expression level of caveolin-1 protein in 48 osteosarcoma tissues and 24 normal bone tissues were detected by immunohistochemistry of SP method.Its relationship with clinical and pathological features were also analyzed.Results among the 48 osteosarcomas examined,38 were stained weak or negative for caveolin-1,whereas the strong caveolin-1 staining was observed in all the 24 normal bone tissues.The absorbent optical density value of caveolin-1 protein in osteosarcoma tissues and normal bone tissues were(0.2292 ± 0.0329) and(0.6428 ± 0.0028),respectively.The expression of caveolin-1 in osteosarcoma tissues was significantly lower than that in normal bone tissues (t =21.48,P < 0.01 ).Caveolin-1 protein expression was significant correlated with the pulmonary metastasis of tumors ( t =11.84,P < 0.05 ),but had no correlation with nge,sex,tumor size and pathologic grades.Conclusion The low expression of caveolin-1 may participate in osteosarcoma invasion and metastasis,and may be a new prognostic molecular marker for osteosarcoma and a new therapeutic target for tumor.
9.Preliminary exploration of the influence of beam orientation and beam number on the optimization of IMRT plan with cervical cancer

Objective To explore the influence of the number and orientation of the beams on the optimization of IMRT plan.Methods Four IMRT plans were designed for 9 patients with cervical cancer,and 7 and 15 fields were applied.The 15-field plans had 30 segments and the 7-field plans had 55 segments.The initial beam angle degrees were 0° and 180°,respectively.Dose delivery time,MUs of plans,the dose distributions of the targets,organs at risk and normal tissues were analyzed and compared in the plans.Results Compared the plans with different beam directions under the same amount,no difference of the irradiation dosimetry in the target and organs at risk was found,except for irradiation dosimetry received by the 7-field 180° small intestine was about 4% higher than the other three plans(F=6.164,P<0.05).The terms of the volume of organs at risk got high dose irradiation(V40 and V30 of the rectum and bladder,V40 of the small intestine),which was similar in the 7-and 15-field plans.V20 and Dmean of organs at risk were significantly smaller(F=3.665-10.503,P<0.05)in the 15-field plans.The 15-field plans needed a little longer treatment time(F=0.312,P<0.05)and HI was slightly worse (F=12.933,P<0.05),but the number of MUs was significantly reduced(F=4.650,P<0.05).Conclusions Increasing the beam number will offset the negative impact of sub field reduction and get the similar dose distribution result.
10.Determination of niclosamide in water with extraction photometry

Objective To study the method and conditions of determination of niclosamide in water. Methods Niclosamide was extracted by mixed extractent, then striped by NaOH. Niclosamide in NaOH solution was determined with dual-wavelength photometry. Results The recovery of niclosamide was 93%. The linear ranger was 0-4. 0 g/m3 and the detectable limit for niclosamide was 0. 042 7 g/m3. The apparent molar absorptivity was 1. 26 ?105 L/(mol?cm). Conclusion Extraction photometry is suitable for determination of niclosamide in water in the field.
