1.Study on the effects of OPN on migration of MSCs and its molecular mechanism in mice

Objective To investigate the effects of exogenous osteopontin (OPN ) the migration of bone marrow mesenchymal stem cells(MSCs) in mice .Methods Using the OPN -/- and wild-type C57 mice ,the MSCs were isolated and cultured .These cells were analysed and sorted by flow cytometry .Then ,Transwell migration assay detected whether OPN can induce the migration of these MSCs .Finally ,the method of Western blot was used to detect the changes of expression of OPN ,CD44 and Integrinβ1 pro-teins .Results The morphology and features of MSCs were proved correctly on 3 passages ,which these cells expressed the proteins of CD44 and CD105 ,but not CD34 .The surface marker antigens were accorded with the general features of MSCs .Compared with OPN -/- mice ,the MSCs significantly increased cell migration in groups of 0 .5 μg/mL OPN and wild type C57 mice ,which was positively related to the times using OPN .Using recombinant OPN protein(0 .5μg/mL) ,expression of related proteins was signifi-cantly increased ,but still lower than the group of wild type mice (P<0 .01) .Compared with OPN -/- mice ,expression of OPN , CD44 ,and Integrinβ1 protein increased significantly different (P<0 .01) in groups of 0 .5 μg/mL OPN and wild type C57 mice . Conclusion OPN can up-regulate the expression of CD44 and Integrinβ1 proteins and promote MSCs migration .

3.Comparison of repair distal soft tissue defect with two kind of reverse digital artery island flaps

ObjectiveTo analysis the clinic outcome(similarities & differences, indication and caution)of repairing fingertip soft tissue defect with two kind of reverse digital artery island flaps with the palmar cutaneous branch of proper digital nerve.MethodsFrom January 2000 to Auguest 2008, all 500cases (514 fingers) were repaired with reverse homodigital artery island flap (263 fingers of 250 cases) and reverse cross finger digital artery island flap(251 fingers of 250 cases).All cases were followed up from 3-8years,compared with flap range,finger extension,sensory rehabilitation,et al. ResultsAll flaps (A & Bgroup) of 500 cases (514 fingers) survived.According to flap range,cross finger flap was better than homodigital flap (P < 0.05); according to finger extension,homodigital flap better than cross finger flap (P <0.05); according to sensory rehabilitation,homodigital flap little better than cross finger flap(P < 0.05); according to long-term follow-up, all had no obviously different (P > 0.05).ConclusionHomodigital flap can cover distal soft tissue defect less than 2.0 cm× 2.5 cm, cross finger flap is a better choice for more than 2.0cm×3.0cm of distal soft tissue defect. According to the flap cover area, cross finger flap is first choice,and to finger extension & sensory rehabilitation,homodigital flap is first choice; accoding to finger movements,two kinds of flaps are all good choice.
4.The effects of venom peptide liquor on adjunctive arthritis in mice

Objective To investigate effect of the venom peptide liquor on adjunctive arthritis (AA) in mice .Methods The male Kunming mice were randomly divided into 6 groups ,named as the control group ,model group ,positive control group(1 mg/kg dex-amethason) ,wine group(10 mL/kg) ,low dosage of venom peptide liquor group(8 .3 mL/kg) and high dosage of venom peptide liq-uor group(33 .2 mL/kg);the right toes ,ankle diameter and whole body weight were measured at 7-day intervals ;the spleen index , serum level of circulating immune complexes(CIC) were determined after the thirty-fourth day when the mice was put to death .Re-sults The level of CIC in AA mice decreased significantly (P<0 .05) ,the decrease of spleen index and the reduction of toe and an-kle swelling in the low dose group were significant(P<0 .01) .Conclusion The venom peptide liquor exhibited apparent inhibitory effect on AA in mice .
5.Comparison of therapeutic effect between skin flap-extension auricular reconstruction and improved Brent au-ricular reconstruction

Objective To compare the rate of postoperative complications and satisfaction between skin flap -extension auricular recon-struction ( hereinafter flap-extension ) and improved Brent auricular reconstruction ( hereinafter improved Brent ) .Methods From October 2009 to October 2014, totally 199 patients who have been diagnosed as congenital microtia and received auricular reconstruction were divided into two groups.Group A (159 patients) received flap-extension and Group B (40 patients) received improved Brent.The medical records, pre-and post-surgery photos of all patients were analyzed and satisfaction of patients was collected by the telephone follow -up.Results Pa-tients in both of the two groups were satisfied with their therapies .There were 47 cases of complications in Group A (29.6%, 47/159) and 5 cases of complications in Group B (12.5%, 5/40).The postoperative complications were significantly different between the two groups (P<0.05).Conclusion There was no difference in patients ’ satisfaction between the two methods ,but the postoperative complications of improved Brent were much lower .
6.Influence of radiofrequency in skin collagen secretion

Objective To evaluate the histologic changes in the dermis and the changes of the rate of type Ⅲ and type Ⅰ collagen by the radiofrequency device. Methods The effects of radiofrequency current on the dermis were observed. Ten rabbits were treated by radiofrequency, and the histologic change in the dermis were observed by H-E staining and Sirius red staining. Results After RF treatment, the fibers in the dermis appeared more compact and the quantity of the type Ⅲ (red) and type Ⅰ (green) collagen were both increased. The fibers in the dermis appeared more compact and the rate of type Ⅲ and type Ⅰ collagen was increased. It was also found that a significant proliferation of dermal collagen was observed in 8 days after treatment. As time went by, the proliferation of dermal collagen was more pronounced, and the rate of type Ⅲ was increased. Conclusion The radiofrequency current can increase the quantity of collagen in the dermis and increase the rate of type Ⅲ and type Ⅰ collagen, which may be one of the key mechanisms of facial rejuvenation by RF.
7.Transfection of Vascular Endothelial Growth Factor Gene VEGF165 Mediated With PEG-PEI Copolymers and Its Effect on The Growth of Endothelial Cells

In order to investigate the ability of PEG-PEI copolymers as gene carriers for delivery of VEGF165. A series of PEG-PEI copolymers with different PEG grafting was prepared and the cytotoxicity was evaluated. Simultaneously,the VEGF165 gene segment with HindⅢ and BamHⅠ site was obtained by PCR, which was cloned into pEGFP-C1. PEG-PEI/ pEGFP-VEGF165 complexes were formed by self-assembly and transfected HUVEc. Transfection efficiency was evaluated by measuring the percentage of cells expressing green fluorecensce protein. The VEGF expression was detected by ELISA, RT-PCR, and the effect of transfection on growth of endothelial cell was evaluated by MTT. The results suggested that the formation of PEG-PEI copolymers could help to reduce the cytotoxicity of PEI. After transfection, the strong expression of green fluorescence protein was observed by fluorescence microscopy. The transfection efficiency was influenced by the number of PEG side chains and N/P ratio. Of all copolymers tested, the transfection efficiency of PEG-PEI(5-25-1) at N/P = 30 reached a maximum, which was much higher than that of PEI. The expression of VEGF protein and mRNA increased significantly, and HUVEc proliferation was accelerated after transfection.These results indicates PEG-PEI copolymers can be used as effective gene carriers for delivery of pEGFP-VEGF165 gene.
8.Clinical study of benazepril combined with amlodipine in the treatment of essential hypertension

Objective To investigate the clinical efficacy of benazepril combined with amlodipine in the treatment of essential hypertension.Methods From January 2016 to January 2017,90 patients with coronary heart disease in Taizhou Central Hospital were selected in the study,and they were randomly divided into three groups,with 30 cases in each group.The observation group(A group)was treated with benazepril combined with amlodipine,the control group (B group )was treated with benazepril,and the control group (C group)was treated with amlodipine. The clinical efficacy,blood pressure,blood lipid changes,blood glucose changes before and after treatment in the three groups were compared.Results The total effective rate of A group was 96.367%,which of B group was 66.67%, which of C group was 70.00%,and the clinical efficacy among the three groups had statistically significant difference (χ2=11.96,P<0.05).Before treatment,the systolic blood pressure(SBP)and diastolic blood pressure(DBP)of the three groups had no statistically significant differences(F=0.98,0.85,all P>0.05).After treatment,the SBP, DBP of the three groups were significantly lower than those before treatment(t=5.68,7.21,3.52,3.16,3.64,3.16, all P<0.05),the SBP and DBP of A group were significantly lower than those of B group and C group(F=6.24, 3.27,all P<0.05).After treatment,only the TC,TG,HDL-C levels of A group had statistically significant differ-ences compared with those before treatment(t=5.29,6.28,2.31,all P<0.05),and the TC,TG levels of A group were significantly lower than those of B group and C group(F=3.18,2.86,all P<0.05),while the HDL-C level of A group increased significantly compared with that of B group and C group(F=3.78,P<0.05 ).Before treatment, the FPG,FINS among the three groups had no statistically significant differences (F=0.85,0.95,all P>0.05 ). After treatment,only the FPG,FINS of A group had statistically significant differences compared with those before treatment(t=5.14,3.65,all P<0.05),and the FPG,FINS of A group were significantly lower than those of B group and C group(F=5.27,2.86,all P<0.05).The adverse reaction rate in A group was 6.67%,which in B group was 30.00%,which in group C was 26.67%,and there was no statistically significant difference among the three groups (χ2=1.72,P>0.05).Conclusion Benazepril combined with amlodipine in the treatment of essential hypertension is effective,safe,and has less adverse reactions.
9.An observation of the morphology and the degradation of hypertrophic scar collagen.

OBJECTIVETo explore the role of collagen degradation and scar morphology and structure in the formation of hypertrophic scar.
METHODSSDS-PAGE (sodium dodecyl sulfate polyacrylamide gel electrophoresis) plus collagen substrate electrophoresis, amino acid analysis and compound staining were employed to observe the collagenase activity within hypertrophic scar, collagen degradation and the tissue morphology of the scar.
RESULTSThere exhibited deranged collagen fibres within hypertrophic scar, and large amounts of acid mucopolysaccharide closely surrounded the collagen fibres. All these led to an obvious decrease in collagenase activity and reduction of collagen degradation.
CONCLUSIONThe decrease of collagen degradation and the formation of hypertrophic scar might be closely related to the decrease in collagenase activity and the inhibiting activity of acid mucopolysaccharide on collagenase.

10.A study of the influence of temperature and humidity on skin friction property.

To investigate the influence of temperature and humidity on skin friction property and to unveil the mechanism therein involved, a test of friction coefficient for four volunteers was carried out on a multi-specimen friction tester. The temperature and humidity of skin were measured with infrared temperature instrument and dermohygrometer. The results showed that the fluidity and ductility of skin were affected by the change of skin temperature. The skin temperature decreasing friction coefficient and the normal displacement decreased first, and then remained unchanged, deformation friction and adhesive friction being the major underlying mechanism. Humidity significantly affected the skin friction properties. The friction coefficient increased with the increasing of humidity. When skin humidity reached to 42% or so, the friction coefficient increased to 1.0 and higher. Meniscus effect was noted to be the major cause of moist skin surface with high friction coefficient.
