Dual-energy X-ray absorptiometry(DEXA),scanning electron microscopy, and bone biomechanical test were used to assess comprehensively bone quantity and quality of ovariectomized rats. In OVX rats, not only bone mineral density (BMD) of lumbar vertebrae in vivo and vitro, but also BMD of femora (except for R3 region) and proximal metaphysis(R1 region) in vitro decreased obviously (P<0.01),whose bone loss rates of L5 and L6 were the highest and achieved 13%; the trabeculae of OVX rats were few, fine, and discontinued and there were lacunae on the surface;in OVX rats, both compressive strength of vertebral bodies and the mechanical properties of femora decreased;the falling degree of the former was greater;the maximal compressive power of lumbar vertebrae decreased with 33.32%. We conclude that after 4-month post-ovariectomy, the bone quantity and quality of six-month-old rats decreased, especially in the area of abundant cancellous bone such as vertebral bodies, distal femora; the assessment of the efficiency of new drugs to prevent and treat postmenopausal osteoporosis using rat models should include these drugs' effects to the bone mass, the bone structure, and the bone strength.

PurposeTo evaluate the impact of filtered back-projection (FBP) and sinogram-affirmed iterative reconstruction (SAFIRE) technique on the image quality of routine dose temporal bone high resolution CT (HRCT) scan, and to provide reference for the realization of low dose scanning.Materials and Methods Sixty patients underwent routine dose temporal bone HRCT scan, 6 groups of images were reconstructed with FBP and SAFIRE (strength grade 1-5), average CT value, noise (SD), signal to noise ratio (SNR), contrast to noise ratio (CNR), subjective score and lesion detection were compared between different groups. Results There was no significant difference of average CT value among the 6 groups (P>0.05); compared with FBP, SD of SAFIRE (strength 1-5) reconstruction decreased gradually (F=1.78,P<0.01), which was 23.1%, 39.2%, 42.4%, 54.1% and 61.2% respectively; SNR and CNR increased gradually (F=1.42 and 3.15, P<0.05), SNR increased 26.6%, 40%, 71.1%, 117.7% and 153.3% respectively, while CNR increased 17.4%, 33.1%, 63.2%, 104.1% and 147.2% respectively. The differences among the 6 groups were statistically significant (P<0.05), subjective scores of SAFIRE 2-4 were higher than those of FBP, SAFIRE 1 and SAFIRE 5 group, while the difference among SAFIRE 2-4 group was statistically insignificant, reconstruction smoothness and edge sharpness of SAFIRE 3 group was better, with highest objective score. Conclusion Routine dose temporal bone HRCT scan combined with SAFIRE reconstruction can increase the image quality of temporal bone high resolution scan effectively, with the potential for radiation dose reduction.

To localize and characterize the hypothetical protein CT358 in the chlamydial infected cells. CT358 gene from the Chlamydia trachomatis (C. trachomatis) serovar D genome was amplified and cloned into the pGEX and pDSRedCI vectors. The recombinant plasmid pGEX-CT358 was constructed and expressed as GST fusion proteins. The GST-CT358 fusion protein was used to immunize mice to raise the antibodies, which specifically recognized CT358 without eross-reacting with other unrelated proteins. The antibodies were then used to localize the endogenous CT358 protein and determine the expression pattern in Chlamydial infected cells using an indirect immunofluorescence assay (IFA). Meanwhile, pDSRedC 1-CT358 recombinant plasmid was transfected to HeLa cells to evaluate the effect of CT358 expression on the subsequent chlamydial infection. The hypothetical protein CT358 was identified in the inclusion membrane of C. trachomatis-infected cells for the first time, and it was detected as early as 12 h after C. trachomatis infection and remained in the inclusion membrane throughout the rest of the infection cycle. Cytosolic expression of CT358 via a transgene failed to affect the subsequent ehlamydial infection. These observations together have demonstrated that CT358 is a newly identified chlamydial inclusion membrane protein, giving the potentially importance for further understanding the mechanisms of chlamydial intracellular parasitism.

The heptadecatungstodiphosphate substituted by L a(Ⅲ) with the formula α2-K7P2W17O61(La.OH2 ) (P2W 17La) was prepared by vacant refilling method. The anion of single substitu ted Dawson-type tungstophosphate salt was doped into the polypyrrole (PPY) film on a glassy carbon electrode (GC) by electrochemical method to prepare the chem ically modified electrode (P2W17La/PPY/GC). It maintains not only t he electrochemical activity and electrocatalytic property of P2W17L a, but also has good stability and sensitivity. In 0.50 mol/L HCl solution, th e first cathodic peak of P2W17La doping in polypyrrole film can cat alyze well the electroreduction of nitrite. The catalytic current was proporti onal to the concentration of nitrite between 6.0×10-5 and 1.0×10-2 mol/L.
