1.Effects of TGF-?_(1,2) on basal and FSH-stimulated production of estradiol and progesterone by human granulosa cells in vitro

Objective To investigate the possibility that TGF ? may also influence the basal and FSH stimulated production of estradiol and progesterone by granulosa cells in vitro. Methods Granulosa cells were obtained from infertile patients undergoing in vitro fertilization and embryo transfer treatment and cultured with serum free supplemented HAM′s F 10 medium(control) or increasing concentrations of TGF ? 1,2 in the absence or presence of FSH (96 U/ml). The media were collected in 24, 48, 72 and 96 h intervals and assayed for estradiol and progesterone. Results Increasing amounts of TGF ? had a significant effect on estradiol production, especially in the presence of FSH,The estradiol value was evaluated from (12 077.9?5 835.3) pmol/L and (13 656.1? 8 404 3) pmol/L to (16 606.8?7 993.3) pmol/L and (19 480.4?913.8) pmol/L. 2.Effect of estrogen on murine vaginal epithelial cell anti-Candida activity

AIM: To investigate the growth inhibition of Candida albicans mediated by vaginal epithelial cells and determine if estrogen affects the anti-fungal activity. METHODS: C_ 57BL/6J mice vaginae from estrogen treatment group, diestrus group and ovariectomized group were excised, respectively. The vaginae were dissociated into single cell suspension by dispase and collagenaseⅠ. The epithelial nenriched cells were used as effector cells. Blastoconidia of C. albicans were used as target cells. After coincubation of effector with target for 9 h, the target cells growth density was observed, percent growth inhibition was calculated, and ultra-structural changes were observed. RESULTS: After coincubation of effector cells with target cells for 9 h, growth density of C. albicans was visibly reduced and it's growth activity was inhibited. Compared to ovariectomized group and diestrus group, vaginal epithelial cells from estrogen-treated mice had less ability to inhibit the growth of C. albicans (P
3.Detection of tumor cells in the peripheral blood of the patients suffered from gestational trophoblastic tumor

Objective To found sensitive and reliable method to identify trophoblastic tumor cells in the peripheral blood of the patients suffered from gestational trophoblastic tumor.Methods Given numbers of JAR cell from ten to million were mixed into 10ml non pregnant peripheral blood as a model. Detection of ? hCG mRNA with fluorescence quantitative reverse transcription polymerase chain reaction (FQ RT PCR) and then estimation of the numbers of tumor cell in the blood. Nine cases of peripheral blood were collected from the pretreatment patients of gestational trophoblastic tumor to assay ? hCG mRNA with FQ RT PCR, then to estimate the numbers of tumor cell in the circulation blood. Results FQ RT PCR could detect ? hCG mRNA when ≥10 2 JAR cells were mixed into 10ml non pregnant peripheral blood. Four cases of bloods had been detected ? hCG mRNA expression in 9 cases of gestational trophoblastic tumor, and the numbers of tumor cell from 10 4 to 10 7 per 10ml blood. Conclusion FQ RT PCR of which primers and probe are designed for ? hCG had been proved to be very sensitive detection means, it could be used to detect gestational trophoblastic tumor cells from patient preipheral blood; With FQ RT PCR the tumor cells had been detected in some patients of gestational trophoblastic tumor.

Objective To compare the differences of laboratory rodents endolymphatic sac (ES) in histology , and to provide an anatomic basis for the clinical research of ES -related diseases .Methods The temporal bones of guinea pigs ,SD rats and KM mice were fixed by heart perfusion ,sectioned by paraffin at the horizontal position and stained by hematoxylin -eosin (HE) .Results All 3 rodents ES contained sac ,epithelium ,sub-mucosal tissue and saccal stroma .The cells were bulky ,stained deep and protruded into the cavity to form a large number of nipples and wrinkles in the middle ES epithelial of guinea pigs .But the nipples or wrinkles was rarely observed in the middle ES epithelial in rat and mouse .Conclusion The histology of ES has significant difference among guinea pig ,mouse and rat .While the difference betweeen mouse and rat is not obvious .

Objective To study the distribution and properly of the transparent globules within Hensen cells (HC) of guinea -pig Corti organ .Methods The cochlear epithelial cells were isolated from 10 guinea pigs .The cells of cochlea were marked by Bodipy493/503 ,sudan III ,oil red O ,and osmium tetroxide .Results The transpar‐ent globules within the HCs of the guinea -pigs were green staining by Bodipy493/503 ,jacinth staining by Sudan III ,ruby red by oil red O .And they were black globules stripe as post -fixed in 1% osmium tetroxide .Conclusion The results indicate that the transparent globules within guinea -pigs HCs'lipid droplets by four methods .
6.Preoperative "second eye" ultrosound guided orientation in non-palpable breast lesions surgery

Objective:To investigate clinical application of preoperative "second eye" ultrosound guided orientation in non-palpable breast lesions surgery.Methods:100 patients were involved in the present study, with 136 impalpable breast lesions. Patients were divided into treatment group and control group by using random number table method. Patients of the control group were operated according to the preoperative ultrasonic location alone, while lesions of patients from the treatment group were located not only by preoperative surface location, but also by the "second eye" ultrasonic examination performed by surgeons. Volume of resected specimen and the operation time and the average length of incision were observed and compared between the two groups.Results:All lesions were completely resected. The average operation time of the treatment and control groups was 20 minutes vs 28 minutes ( P<0.05) , the average volume of resected specimens was 3.0 ml vs 4.1 ml ( P<0.05) , and the average length of incision was 25 mm vs 30 mm ( P=0.21) . Conclusions:It is an accurate, safe and effective method to remove clinical non-palpable breast lesions by the "second eye" ultrasound operated immediately before the surgery. It can obviously shorten the operation time, precisely remove the specimen, and reduce damage to normal tissues. In clinical practice, breast ultrasonic characteristics are recommended to be mastered by surgeons so that it could be helpful to the surgery.
7.Ultrasonographic features and pathological diagnosis of breast disease with ultrasound classification as BI-RADS 4a

Objective To summarize the characteristics of BI-RADS 4a breast diseases under ultrasonic examination referring to patients' clinical data and pathological reports.Methods 82 patients who were firstlydiagnosed and treated in the Affiliated Hospital of Jiangnan University were included.The ultrasonograms were retrospectively reviewed along with the correlated pathological and clinical findings.Results 48 (58.5%) cases were benign,among which fibroadenoma was the most common.34 cases (41.4%) were malignant,among which invasive ductal carcinoma was the most common.68 of the 82 cases were neoplastic disease (82.9%),6 (9.8%)were inflammatory disease,and 8(7.3%) were adenosis.Conclusions The patients who are graded BI-RADS 4a by ultrasonic examination may have completely different diseases.Beyond the most common benign diseases,few of them are malignancies with atypical,even ambiguous sonographic features.Therefore,in clinical practice,sonologists should set appropriate parameters of ultrasonic devices appropriately,and evaluate the sonographic features carefully,in order to make the correct grading and diagnosis.While for clinicians,it is necessary to learn to read the ultrasonographic features along with patients' manifestations,and finally reduce the misdiagosis and missed diagnosis of such diseases.
