1.Effect of Ultraviolet Blood Irradiation and Oxygenation(UBIO)on Ischemic Strokes

The paper was a study on the effect of UBIO on ischemic strokes. 80 Cases with acute ischemic strokes were randomly divided into two groups:UBIO group and control group. Clinic effectiveness,hemorheology and in vitro infarction were studied before and after 14 days treatment. Clinical results showed that the excellent,effetive rate of UBIO group was respectively better than that of control group (P<0.05). The results of hemorheology and in vitro infarction showed UBIO has a faster effectiveness than control group. The results indicate that UBIO therapy is a hopeful assistant method for the treatment of acute ischemic strokes.
2.Effect and mechanism of quercetin from total flavones of Bidens Bipinnata L on the proliferation of hepatic stellate cells induced by TGF-β1

Objective To observe the effect and mechanism of quercetin on the proliferation of rat hepatic stellate cell(HSC)induced by TGF-β1.Methods HSC was isolated by orthotopic hepatic perfusion with collagenase Ⅳ.MTT assay was used to detect the inhibitory effect of quercetin on the proliferation of HSC.The expression of typeⅠcollagen in HSC was detected by ELISA.The effect of quercetin on the expression of typeⅠcollagen,Smad2 and Smad7 mRNA in HSC was detected by real-time quantitative PCR.The expression of type Ⅰ collagen,Smad2 and Smad7 protein in HSC was detected by Western blot.Results Quercetin could inhibit the proliferation of HSC,down-regulated the expression of type Ⅰ collagen,Smad2 mRNA and protein,and Smad7 mRNA and protein expression were significantly up-regulated(all P<0.05).Conclusion The anti-hepatic fibrosis effect of quercetin may be related to its blocking TGF-β1/Smad pathway of HSC and blocking the activation and proliferation of HSC.

Objective To observe the effect of BSD 2000 deep thermotherapy plus chemotherapy in treatment of malignant seroperitoneum of advanced epithelial ovarian cancer patients with drug resistance.Methods 36 advanced epithelial ovarian cancer patients with malignant seroperitoneum for drug resistance were randomly divided into two groups,trial group (18 cases) and control group (18 cases).Cases in trial group were treated with BSD 2000 deep thermotherapy plus GT regimen (gemcitabine 1 000 mg/m2 iv d1,d8,taxinol 80 mg/m2 ip d1,d8.28 days for a cycle),while control group with GT regimen alone.Effect,survival time (median) toxicity and Karnofsky score were evaluated after 2 cycles.Results Response rate (RR) was strongly higher in trial group compared with control group [55.6 % (10/18) vs 22.2 % (4/18),P < 0.05],the same to disease control rate (DCR),but there was not significant difference between two groups (P > 0.05).The improvement rate of Karnofsky score in trial group was higher than that in control group,which had no significance (P > 0.05).The toxicity were similar in both groups,which had no stage 3 to 4 side-effect.The differences of survival time (median) and survival rate had no statistical significance between two groups (P > 0.05).Conclusion It is useful to eliminate seroperitoneum,improve quality of life and decrease the toxicity for the regimen of BSD 2000 deep thermotherapy plus chemotherapy in treatment of malignant seroperitoneum of advanced epithelial ovarian cancer patients with drug resistance.
5.ABSORPTION MECHANISM OF THE ANTIHYPERTENSIVE PEPTIDE VAL-LEU-PRO-VAL-PRO IN CACO-2 CELL MODEL

Objective To study the absorption mechanism of the antihypertensive peptide,Val-Leu-Pro-Pro-Val(VLPVP) in Caco-2 cell model. Method Caco-2 cell model was used in vitro to investigate the effects of time,pH,drug concentration,enhancers and inhibitors on the absorption of VLPVP. VLPVP concentration was measured by HPLC. Results The transport rate of VLPVP reached the maximal level at pH7.4,and was positively correlated to transport time and drug concentration. The peptide transporter inhibitors(Gly-Pro,arphamenine A) ,the endocytosis inhibitor(phenyl arsenoxide) didn’t influence the transport of VLPVP,while the paracellular transport enhancer(sodium deoxycholate) ,the inhibitor of multi-drug resistant protein(MK-571) and the energy inhibitor(sodium azide) enhanced it from apical to basolateral side significantly,but the latter two didn’t influence the transport from basolateral to apical side. Conclusion Paracellular diffusion was suggested to have been the main mechanism for the absorption of intact VLPVP and MRP2 had strong eflux effect on the transepithelial transport of VLPVP.
6.The Acute Antihypertensive Effects of Recombinant Angiotensin Converting Enzyme Inhibitory Peptides on Spontaneously Hypertensive Rats

Objective To study the effect of angiotensin converting enzyme inhibitory peptides (ACEIP) on blood pressure of spontaneously hypertensive rats.Methods One hundred and four spontaneously hypertensive rats(SHR) were randomized to thirteen groups (n= 8,each group) and received one of following approaches:saline,captopril (10 mg/kg),low-dose (200 ?g/kg),medium-dose(400 ?g/kg) and high-dose (800 ?g/kg) of the three ACEIPs:VLPVP,KVLPVP,VLPVPR respectively.Thirty-two WKY rats were randomized to 4 groups(n=8 in each group),VLPVP,KVLPVP,VLPVPR (800 ?g/kg by gavage) and saline were administrated.The systolic blood pressure was measured before and after the administration of ACEIP.Results Compared with the control group,medium-dose and high-dose of VLPVP,KVLPVP and VLPVPR reduce blood pressure in hypertensive rats and the effect can last for above 6 hours [decline of blood pressure:high-dose:(40.9?15.7),(38.0?21.3) and (43.2?12.6) mmHg;medium-dose:(28.3?23.0),(24.7?21.5) and (26.2?9.9);control:(8.5?5.1),(6.2?4.1) and (7.1?5.5)mmHg;all P0.05].Conclusion Recombinant ACEIP acutely reduce high blood pressure in SHR,the antihypertensive effects persist for at least 6 hours,with no effect on normotensive WKY rats.
7.Efficient Derivation of Mesenchymal Stem Cells and Neural Precursor Cells From Human Embryonic Stem Cells Through Teratoma Formation

Many somatic cell typos were obtained by in vitro differentiation or teratoma formation of human embryonic stem ceLls (hESCs). However, it is unclear whether specific cell types can be obtained from hESCs-derived teratoma. It was reported that many kinds of cells, including neural progetfitor/precursor cells (NPCs) and mesenchymal stem cells (MSCs) were isolated efficiently from the teratoma of hESCs through in vitro selection. The teratoma-derived NPCs and MSCs showed specific characteristics of molecular markers similar to the primary NPCs and MSCs. Moreover, these teratoma-induced NPCs and MSCs can be further induced to differentiate into neurons, astrocytes, or adipose and bone cells, reflecting their inherent multi-potencies. Given that teratoma normally contains a mixture of ectoderm, mesodenn, and endoderm lineage cells at different differentiation stage, it was suggested that hESCs-derived teratoma could be an alternative source to generate a variety of uncommitted progenitor cells or terminally differentiated somatic cells, which may be otherwise difficult to obtain through direct in vitro differentiation.

OBJECTIVE:
To explore the curative effect and origin of antrochoanal polyp (ACP) with various approaches.
METHOD:
Fifty-seven patients with ACP were included in the study. All the ACP patients were examined by preoperative endoscopy and computer tomographic (CT) scans. The patients were treated by various endoscopic approaches including endoscopic middle meatus antrostomy, inferior meatus antrostomy combined with endoscopic middle meatus antrostomy or endoscopic medial maxillectomy combined with endoscopic middle meatus antrostomy respectively. The relationship between polyp location in middle meatus and lesions in the antrum was explored during the surgery. Pathological examination was carried out and patients were regularly followed up after operation.
RESULT:
Fifty-seven ACP develops from antral cyst. In 22 cases of endoscopic middle meatus antrostomy, two patients relapsed. In 17 cases of inferior meatus antrostomy combined with endoscopic middle meatus antrostomy, one patients relapsed. In 18 cases of endoscopic medial maxillectomy combined with endoscopic middle meatus antrostomy, no one relapsed.
CONCLUSION
Our data indicated that the ACP mainly originates in antral cyst, and capsule wall herniates to middle meatus through the antral ostium. ACP are common in unilateral, rare in both sides. The endoscopic approaches of middle meatus antrostomy and inferior meatus antrostomy combined with endoscopic middle meatus antrostomy might guarantee good prognosis. If the cyst is on the anterior wall of maxillary sinus, the approach of endoscopic medial maxillectomy can obtain a better vision and completely remove the lesions.

9.Diagnostic value of me clinical grading methods in acute pulmonary embolism

Objective To evaluate the significance of clinical grading methods in acute pulmonary embolism (APE). Methods Clinical data of 259 patients suspected APE were retrospectively analyzed. The clinical probability was classified into low, intermediate and high grade by the Geneva score, the Wells score and the SYSU score. The result was contrasted with gold standard. Results Through the three, methods, pa-tients were classified into low pmbability (43.9%-52.5%), intermediate probability (38.0%-42.1%) and high probability (9.5%-14.0%), and the actual frequencies of APE in each category were 6.2%-14.4% in low probability, 65.9%-76.2% in intermediate probability, 88.5%-90.5% in high probability. The SYSU score had the lowest rate of missed diagnosis in low probability (P<0.05 ).The Geneva score was the most accurate in predicting the intermediate probability (P<0.05). But in high probahility, three prediction rules had no significant difference (P>0.05). Combined with D--dimer test, the rote of missed diagnosis in low probability can be lowered. Conclusions The clinical grading methods can predict the clinical probability of APE. It exists similar accuracy, but has different scope of application. Clinical doctor should choose the ap-propriate grading methods in different patients.
10.Rab5a promotes LPS-induced cytokine expression in macrophages

Objective:To establish cell lines stably expressing Rab5a and its the inactive mutant Rab5aN133I,analyze the effect of Rab5a on the expression of cytokines in LPS-stimulated RAW264.7 cells .Methods:RAW264.7 cells were transfected with Rab5a and its inactive mutant vector Rab5a N133I separately,and then screened by G418.Rab5a stable expressing cell lines were identified by Real time-PCR.The growth of the stable cell lines was analyzed by MTT assay.After the stable cell lines were stimulated by LPS for different time periods,the expression of iNOS,TNF-αand IL-6 was detected.Results:Rab5a and Rab5aN133I transfection resulted in elevated Rab5a mRNA expression compared with the control cells ( P<0.05 ).Rab5a overexpression enhanced the proliferation of RAW264.7 cells.However,the proliferation of Rab5aN133I cells was significantly slower than the control cells ( P<0.05).Overexpression of Rab5a promoted LPS-induced production of iNOS,TNF-αand IL-6 in RAW264.7 cells (P<0.01). Conversely,overexpression of Rab5aN133I abolished the stimulating effects of Rab5a.Conclusion: Rab5a promoted LPS-induced expression of iNOS,TNF-αand IL-6 in RAW264.7 macrophages in a GTP-binding ability-dependent manner.
