1.Pharmacodynamic Study of Compound Xiangqi Granules in Vitiligo Model Guinea Pigs

OBJECTIVE:To study the pharmacodynamics of Compound xiangqi granules in vitiligo model guinea pigs. METH-ODS:The vitiligo guinea pigs model was established by chemical decolorization with hydrogen peroxide. Then guinea pigs were randomly divided into 5 groups after modeling,including model control group (water),vitiligo capsules group [positive control, 50 mg/(kg·d)] and Compound xiangqi granules low-dose,medium-dose and high-dose groups [25,100,and 400 mg/(kg·d)]. They were given related medicines intragastrically once a day for 40 days. The distribution of skin melanin and the number of black hair follicle were observed,and cholinesterase (ChE) activity,malondialdehyde (MDA) content of plasma and hemorheology in-dexes were detected. Blank control group had also been established. RESULTS:Compared with blank control group,the distribu-tion of skin melanin,the number of black hair follicle and ChE activity decreased in model control group,while plasma viscosity, whole blood viscosity and hematocrit increased significantly,and erythrocyte sedimentation rate decreased significantly;there was statistical significance(P<0.05 or P<0.01). Compared with model control group,except erythrocyte sedimentation rate and hema-tocrit of low-dose group,above indicators of positive control group and Compound xiangqi granules low-dose,medium-dose and high-dose groups were all improved;there was statistical significance(P<0.05 or P<0.01),showing good effectiveness-dose rela-tionship. CONCLUSIONS:Compound xiangqi granules improve relative indicators of experimental vitiligo model guinea pigs effec-tively.
2.Effect of Bushen Yizhi Prescription on Learning and Memory in Ovariectomized Rats

Objective To observe the effect of Bushen Yizhi Prescription (BYP) on learning and memory in ovariec-tomized rats. Methods Three - month - old female SD rats were randomized into mimic operation group, model group, BYP group and estrogen control group. Ovariectomy was operated in the three latter groups. After operation, BYP group was treated with gastric infusion of BYP and estrogen group with subcutaneous injection of estrogen. The changes of refer- ence memory and working memory were tested by Morris water maze. Results & Conclusion Sixteen weeks after ovariecto-my, serum estrogen level was markedly decreased, the working memory became poor and the reference memory remained normal. B YP and estrogen can prevent the reduction of working memory in ovariectomized rats.

With the advance of hospital digitization, the computer technology and the utilization of database are greatly enhanced, and thus the information security situation of hospital has been more and more serious. The computer entering hospital's network illegally and the unauthorized application program seriously threaten the security of hospital's database and network. This article discusses a security model of medical database of LAN in practice from the aspects of authorized document library, daemon program and monitoring, reporting and the orientation of the illegal computer. With source creativity, the model is significant to the security of medical network or other network. It is suggested that a real-time medical database monitoring system be set up to monitor network.
4.Determination of CAMP and CGMP levels in the serum and breast tissues of patients with breast cancer

The levels of cyclic nucleotides(CAMP and CGMP)in the serum and braest tissues of 60 patients with breast cancer or benign breast diseases were measured.It was found that the CAMP level was significantly lower,the CGMP significantly higher and the CAMP/ CGMP ratio was reduced in the serum and breast tissues of the patients with breast cancer.Our findings suggest that CAMP and CGMP might be factors of importance in the origin and development of breast cancer.
5.Effects of different doses of simvastatin on endothelial function in elderly patients with coronary artery disease

Objective To analyze the effect of 10 mg,20 mg simvastatin on vascular endothelial function in elderly patients with coronary heart disease. Methods 96 cases withcoronary heart disease admitted in the First people's Hospital of Xiaoshan District from Janary 2009 to December 2010 were selected and divided into three groups,each had 32 cases. Control group were given expanding coronary artery,blood,anticoagulation therapy.Observation group 1 and observation group 2 were separately taking 10 mg,20 mg simvastatin on the basis of control group. Endothelium-dependent vasodilation (FMD)functional,non-endothelium-dependent vasodilation (NMD)functions,nitric oxide (NO),and endothelin (ET)in three groups before and after treatment were detected and compared. Results Before treatment,the differences of FMD,NMD,NO,ET in three groups were not significant. After treatment,FMD were increased in all three groups,but the difference between two observation group was not statistically significant.NO was also increased in all three groups,but value in two observation group was higher than control group. NMD were unchanged in all three groups.Compared with before treatment,ET in two observation groups after treatment were decreased significantly (P<0.05 ). Conclusion simvastatin can effectively rectify the metabolic imbalances of vascular endothelial cells in patients with coronary heart disease and promote the improvement of FMD,but there is no significant difference between 10 mg~20 mg dose.
6.Fugene 6——a new approach to induce the gene transfection in high efficiency for eukaryotic cells in vitro

Objective To develop a new method to induce the gene transfection in high efficiency for eukaryotic cells in vitro. Methods Four kinds of p14ARF gene primarily deleted human carcinoma cell line including H460,A549,U251,and PC-3 were transfected with the human p14ARF expression vector (pCI-neo-p14ARF) by using the new nonliposomal transfection reagent Fugene 6. The efficiency of gene transfer was determined by screening the cells in G418. Results After 21 days' selection, G418-resistant clones were shown in all the transfected plate. PCR product of p14ARF gene was positive in all the G418-resistant clones. Cytotoxicity of Fugene 6 was detected. The cell proliferation activity was not affected when it was cultured in a high dose of Fugene 6. Conclusion These results demonstrate that Fugene 6 is a rapid, feasible, reproducible, and noncytotoxic gene transfection approach for eukaryotic expression vector in vitro.
7.Therapeutic effects of exercise combined with tiotropium bromide and salmeterol/fluticasone inhaling on stable chronic obstructive pulmonary disease

Objective To observe the therapeutic effects of exercise combined with tiotropium bromide and salmeterol/fluticasone inhaling on stable chronic obstructive pulmonary diseases (COPD).Methods Sixty COPD patients with pulmonary function Ⅲ-Ⅳ grade were randomly allocated into experiment group and control group (30 cases in each group).The patients in experiment group were treated with inhaled tiotropium bromide and salmeterol/fluticasone combined with excise,while the patients in control group were treated only with inhaled tiotropium bromide and salmeterol/ fluticasone.Respiratory training with shrinkage lip respiration and steps training was given to the patients in experiment group for 8 weeks.Main outcome measures included forced expiratory volume in one second (FEV1),FEV1,in the anticipation value (FEV1%),FEV1/forced vital capacity (FVC),Medical Research Council (MRC) score,activities of daily living (ADL) score and 6-minute walking distance (6MWD).Results MRC score in control group and experiment group was both lower than that of pre-treatment (P < 0.01).ADL score,6MWD and pulmonary functional parameters in both groups were improved than those of pre-treatment (P< 0.01).FEV1,FEV1%,FEV1/FVC,MRC score,ADL score and 6MWD were (1.41 ±0.25) L,(53.76 ±11.17)%,(62.12 ±10.78)%,(1.56 ± 1.16) scores,(5.51 ±0.18) scores and (342.7 ± 19.7) m after treatment in experimentgroup,and (1.28 ± 0.23) L,(46.27 ±8.85)%,(55.76 ±8.57)%,(2.29 ±1.13)scores,(4.47 ±0.13)scores and (280.6 ± 17.8)m after treatment in control group.There were significant differences between two groups (P<0.05).Conclusion Tiotropium bromide,salmeterol/ fluticasone inhaling can improve life quality and pulmonary functional parameters in patients with stable COPD,while exercise combined with that can significantly enhance the therapeutic effects.
8.Impact of GSTM1 and GSTP1 gene polymorphisms on erythrocyte GST activity

Aim The current study was conducted to investigate the effect of GSTP1 codon 105 polymorphism, alone and in combination with GSTM1-deletion polymorphism, on erythrocyte GST activity in 196 Han Chinese. Methods GST activity was measured in healthy Chinese by a spectrophotometric method (n=196;101 males and 95 females; age range 21~81 years; median 43.5 years). GSTM1 polymorphisms were analyzed by a PCR-Multiplex procedure, whereas GSTP1 polymorphism was analyzed by PCR-RFLP. Results The frequency of GSTM1 null genotype was 56.1% and the frequency of I/I, I/V, and V/V genotypes was 60.7%, 35.2% and 4.1%, respectively, in Han Chinese. The mean erythrocyte GST enzyme activity for I/V genotype group(3.53?0.63 U?g -1Hb) was significantly lower than that for I/I genotypes (4.25?1.07 U?g -1Hb, P=0.000), while significantly higher than that for V/V genotypes (2.44?0.67 U?g -1Hb, P=0.004). In GSTM1(-) group, the GST activity of carriers of GSTM1(-)/GSTP1- I/I is significantly higher than that of GSTM1(-)/GSTP1- I/V or-V/V, however, in GSTM1(+) group, there is no difference between different subgroups. There was no significant difference in the mean GST activity among different age groups. Erythrocyte GST activities were significantly higher in females than in males, but not significantly. Conclusion The GST activity measured by CDNB-based assay is probably strongly correlated with the GSTP1 105Val genotype, although other GST enzymes would tend to dilute the GSTP1 genotype effect.
9.Correlation between MR amide proton transfer signal intensity and pathologic features in brain glioma

Objective To investigate the probable pathologic basis of amido proton transfer(APT) imaging by analysing APT signal intensity and pathologic features of different grades of glioma.Methods Twenty-eight patients with glioma confirmed by postoperative pathology underwent APT scan.All the patients were divided into two groups,including 11 cases in low grade (WHO Ⅰ and Ⅱ) and 17 cases in high grade (WHO Ⅲ and Ⅳ) group.The APT rate of tumor core was measured.The specimens were processed with routine hematoxylin-eosin (HE) staining and immunohistochemistry of Ki-67 and CD34.Independent-samples t test was used to detect the difference of APT rate,cellularity,microvessel density and Ki-67 labeling indices of tumor core between low grade and high grade group.Pearson correlation analysis and multi-variable linear regression analysis were used to detect the relationship of APT rate with cellularity,microvessel density and Ki-67 labeling indices of the tumor core.Results The APT rate,cellularity and proliferation index were (2.3±0.6) %,(9.4±2.4) % and (14.2±5.4) % in low grade group,while (3.6±0.7) %,(18.4±4.7) % and (31.7±4.5) % in high grade group,respectively.Microvessel density was (19.0±7.4) per high-power field in low grade group and (38.9±11.3) in high grade group.There were statistical differences of the APT rate,cellularity,microvessel density and proliferation index between the low grade group and the high grade group (t=-4.94,-5.89,-5.13,and-9.28,respectively,P<0.01).The APT rate was positive correlated with cellularity,microvessel density and proliferation index.The coefficient of correlation were 0.904,0.598,and 0.750,respectively,(P<0.01).Multiple linear correlative analysis showed that increasing cellularity (X1),microvessel density (X2) and proliferation index (X3) were the main factors for increasing APT rate,and the correlation equation was Y=0.801 + 0.12X1-0.003X2 + 0.026X3 (F=46.437,P<0.01,R2=0.853).Conclusions The APT signal intensity of the tumor core could reflect the pathologic features of glioma.The APT rate was positive correlated with cellularity,microvessel density and proliferation index,which indicate the higher APT rate the higher grade tumor.
10.Effect of BushenYizhi Prescription on Long-Term Potentiation in Hippocampus of Ovariectomized Rats

Objective To observe the effect of Bushen Yizhi Prescription(BYP) on long-term potentiation (LTP) in hippocampus of ovariectomized (OVX) rats. Methods Three-month-old female SD rats were randomized into mimic operation group, model group, low dosage BYP group, high dosage BYP group and estrogen control group. Ovariectomy was operated in the three latter groups. After operation, BYP group was treated with gastric infusion of BYP and estrogen group with subcutaneous injection of estrogen. The changes of LTP of dentate gyms in hippocampus of OVX rats were observed by electrophysiological method and the effect of BYP on LTP was also evaluated. Result & Conclusion The increased amplitude of population spike of LTP was significantly low and maintained a short time in model group as compared with that in mimic operation group. There were insignificant difference between the high-dosage BYP group and estrogen control group. It is indicated that synaptic transmission in hippocampus can be protected and sustained by BYP and the estrogen replacement.
