1.Study of the Changes of Langerhans in Cultured Human Epidermal Cells

It has been shown that epidermal Langerhans cells (LCs) were antigen presenting cells in cutaneous cell-mediated immuneresponse,and they were implicated in skin allograft rejection,If LCs can be depleted from skin grafts,the survival period of the allografts may be prolonged. Therefore,we observed the changes of LCs in cultured human epidermal cells (ECs) by ATPase staining,HLA-Dr monoclonal antibody indirect immunofluoresence and enzyme labelling avidin-biotin complex (ABC) staining.And the capacity of ECs to stimulate allogeneic lymphocyte proligeration before and after cultures was testedby mixed epidermal cell-lymphocyte culture response (MEcLR).We found that LCs in original skin explants were significantly decreased and deformed three days after culture.LCs were reduced more than 90 percent after 7 days of culture.LCs in original skin explants were approximately eliminated 14 days or more of culture.LCs were never seen within epidermal outgrowth in culture.The Ecs gained from the original skin explants which were cultured after 7 days and from epidermal outgrowth lost their ability to stimulate allogeneic lymphocyte transformation in MEcLR.These results indicate that LCs bearing HLA-Dr antigen in human are able to be depleted from epidermal grafts by epidermal cell culture,and suggest that the ECs cultured in our culture system can prolong their survival period as they are used as allografts.

Results Based on the actual cases, the improved procedure for equipment quality was realized and the management of purchase items were evaluated in quantification. Conclusion The efficiency of the management of medical equipment can be evaluated by using this method.
3.Cilnical significance of serum human kallikrein 11 in patients with ovarian cancer

ObjectiveTo investigate the diagnostic and prognostic significance of serum human kallikrein11 ( KLK11 ) in patients with ovarian cancer. MethodsThe level of serum KLK11 was determined by enzyme-linked immunoassay(ELISA) for 30 patients with ovarian cancer, 26 patients with benign gynecologic processes and 32 healthy subjects,and the KLK11 value beyond the upper-limit of 95% credibility interval( 95% CI) based on the health controls was used as the positive value. The positive rate of KLK11 was compared among the three groups.ResultsThe positive rate of KLK11 in patients with ovarian cancer was significantly higher than those in patients with benign gynecologic processes or in healthy subjects[(73.3%) and(7.7% ) or(6.3% ) ,all P <0.01 )] ,and there was no significant difference between ovarian benign tumor group and the healthy control group ( P > 0.05 ).ConclusionThe serum KLK11 should be a novel tumor marker in the clinical diagnosis of ovarian cancer.
4.Glyoxalase I-structure,function and role in Alzheimer′s disease

Glyoxalase system,a most effective defence system is present in cytosol of cells to scavenge ?-oxoaldehydes,which are reactive intermediates of AGEs formation.Glyoxalase Ⅰ is the key enzyme of this system.?-oxoaldehydes and AGEs are involved in the development of Alzheimer′s disease (AD). Therefore,the expression level and activity of glyoxalase I are essential to pathogenesis of AD.
5.Long- term evaluation of intracystic corticosteroid injection in the treatment of simple bone cysts

Objective To evaluate the long- term results of intracystic corticosteroid injection for the treatment of simple bone cysts. Methods Seventy- six children with simple bone cysts treated by intracystic injection of corticosteroid from 1985 to 1995 were followed- up for an average of 8.2 years( 5- 13 years) . Forty- one of the cysts were situated at the proximal end of the humerus, 20 at the proximal end of the femur and 15 at other sites. The mean injection times were 4.2(2- 13 times). Results Forty- two(55.3% ) cysts were healed up completely, 16(21.1% ) basically healed up, 9(11.8% ) healed up partially and 9(11.8% ) failures. Satisfactory and effective rate was 76.4% and 88.2% respectively. Conclusion Intracystic corticosteroid injection appears to be the initial choice of treatment for simple bone cysts, because it is an effective, and non- invasive method which can be carried out easily.
6.MRI Diagnosis of Primary Intracranial Teratoma

Objective To investigate the role of MRI in diagnosis of primary intracranial teratoma.Methods Nine cases of primary intracranial teratoma proved by operation and pathology were examined by MRI before operation.MRI features were analysed retrospectively.Results The localized and qualitative rates were 100% and 88.8% respectively.The characteristic MRI appearances of primary intracranial teratoma were as follows:(1)The tumor was lobular in shape with clear margin;(2)The tumor had heterogeneous signal intensity ;(3)Inhomogeneous Gd-DTPA enhancement in most of the tumors.Conclusion Multiplanar imaging and Gd-DTPA enhancement in MRI are helpful in diagnosis and differential diagnosis of teratoma.
7.CT and MRI Diagnosis of Pineal Region Germinoma

Objective To investigate the role of CT and MRI in diagnosis of the pineal region germinoma.Methods 11 cases of the pineal region germinoma proved by operation and pathology. CT and MRI features were analysed retrospectively. Results In 11 cases on CT most tumors were hyperattenuation or isoattenuation. Enhancement in most of the tumors were homogeneous. The characteristic MRI appearances of the pineal region germinoma were as follows: (1) On T 1WI, the lesions were isointense or slightly hypointense while isointense or slightly hyperintense on T 2WI. The pineal region germinoma showed no edema. (2) The lesions showed homogeneous or inhomogeneous Gd-DTPA enhancement in most of the tumors.Conclusion CT and MRI of multiplanar imaging and Gd-DTPA enhancement are helpful in diagnosis and differential diagnosis of pineal region germinoma. The tumors have sex, location and imaging characteristics.
8.The technique of autologous osteochondral mosaicplasty for repair of the cartilaginous defects under arthroscopy

Objective To evaluate the possibility for the treatment of chondral defects on the femoral bearing surface with autologous osteochondral transplantation under arthroscopy. Methods There were 17 patients with cartilaginous surface defects, including 12 males and 5 females. The average age of patients was 29 years ranging from 18 to 45 years. The defects were located at 10 left knees and 7 right knees. The functional status of patients were evaluated according to the Brittberg-Peterson scoring scale system, the results were average 80.65?9.69 points ranging from 65 to 105. 3 patients had no an obvious history of trauma, but suffered from the rheumatoid disease; however, the other 14 patients had been injured at the knee joint with pain and followed with muscle atrophy on the thigh. 3 patients experienced interlocking, and 2 had snapping in their knee joints. All the 14 patients with traumatic history had only solitary lesion, the area of local cartilaginous defects at the bearing surface of the lateral femoral condyle was of 2.5-3.0 cm2; the local defects of the other 3 patients were located at the medial femoral condyle with defect area of 2.0-2.5 cm2, which was diagnosed either as chondomalacia or corruption. The procedures of the mosaicoplasty consisted of harvesting osteochondral cylinders from non-weight bearing surface of the knee and then implanting the grafts into the cartilaginous defects under arthroscopy. Results All the patients were followed up for 10 to 20 months (mean, 15 months). The joint mobility recovered to nearly normal. According to the Brittberg-Peterson system, 14 patients had score of 0, and 3 had scores of 2-3 because of mild pain. The follow-up MRI showed adequate cartilaginous coverage in the original lesions and excellent position of the cylindrical osteochondral grafts. Conclusion The autologous osteochondral mosaicplasty is proved to have good and reliable clinical results for the cartilaginous defects in the knee bearing surface.
9.Clinical analysis of complete left bundle branch block after transcatheter closure of ventricular septal defect

Objective To evaluate the clinical features and prognosis of patients with complete left bundle branch block(CLBBB)following transcatheter device closure of ventricular septal defect(VSD)closure. Methods Clinical feathers of 11 patients with postoperative CLBBB in Department of Pediatric Cardiology,Guangdong General Hospital from January 2011 to December 2013 were collected and reviewed retrospectively. They were treated with dif-ferent protocol based on the appeared time of CLBBB occurrence and clinical symptoms. The patients were followed up, and the prognosis was recorded. Results The median age of 11 patients was 3. 9 years(3. 4 to 17. 5 years old). The median interval of intervention therapy to first attack of CLBBB was 2. 8 months(1 day to 25. 4 months). CLBBB oc-curred within 1 week to 1 month postoperatively in 4 patients,another 1 case suffered from CLBBB between 1 week to 1 month postoperatively,meanwhile 6 cases underwent CLBBB after 6 months postoperatively. The longest term of CLBBB attack postoperatively was 25. 4 months in 1 patient. The electrocardiograms on 5 patients returned to normal by only drug treatment. However,3 patients failed to recover with drug therapy,2 of them undertaken surgical procedure to re-move the occluder associated with VSD repair,1 patient recovered to normal and another converted to incomplete right bundle branch block. One of them refused to undertake surgical procedure and still bothered with persistent CLBBB. Another 3 cases did not receive special treatment due to the later attack of CLBBB(≥6 months)without clinical symp-toms. By the end of observation,the electrocardiogram(ECG)in 4 patients returned to normal,4 patients presented with persistent CLBBB. One patient's ECGs were presented with right bundle branch block. After ECG successfully returning to normal ECG by drug therapy,2 patients relapsed during follow - up,and 1 of them developed to an enlarging left ven-tricle and heart failure which led to death. Conclusions CLBBB may occur in short or long - term after VSD closure. ECG may become normal after early and appropriate treatment postoperatively. Systolic dyssynchrony and cardiac dys-function may be caused by persistent CLBBB. Therefore,patients with CLBBB after VSD closure should be treated ap-propriately without delay,and more frequent and longer follow - ups are required.
10.Effects of electroacupuncture on plasma CGRP and SP expression in rats of constipation-predominant irritable bowel syndrome

Objective To observe effects of electro-acupuncture on the expression of calcitonin gene-related peptide (CGRP) and substance P (SP) in constipation-predominant IBS rats,and to discuss the mechanism of IBS's acupuncture treatment.Methods To set WISTAR adult rats as the object and to establish models of irritable bowel syndrome rats.The experiment rats were randomly divided into a normal group,a model group,a EA Shangjuxu group and a EA Dachangshu group.The Shangjuxu group and Dachangshu group were bounded and given EA,once daily,for a total of seven times.Model group was bounded but not treatedwith EA,the normal group was done nothing.After the treatment,5ml of inferior vena blood was got inanesthesia state.The value of plasma CGRP and SP in rats' plasma were measued with Elisa.Results Compared with the normal control group,CGRP and SP content[(52.62± 15.05)pg/ml,(27.10±5.03) pg/ml] in IBS model group was obviously increased,the difference was statistically significant (P< 0.01,P< 0.01) ; the changes of plasma CGRP content [(37.82± 15.34)pg/ml,(38.93± 12.05)pg/ml] and SP content[(19.62± 5.68) pg/ml,(18.90± 5.07)pg/ml] were not obvious in EA Shangjuxu group and EA Dachangshu group (P>0.05,P>0.05).Compared with IBS model group,the content of plasma CGRP and SP was significantly descend in EA Shangjuxu group and EA Dachangshu group (P<0.05,P<0.05,P<0.01,P<0.01).The concentration of CGRP and SP in EA Shangjuxu group and EA Dachangshu group had no significant difference(P>0.05,P>0.05).Conclusion Electroacupuncture could treat IBS through regulating CGRP and SP in IBS rats.
