1.Mechanisms on The Regulation of miRNAs Expression

MicroRNAs(miRNAs),small noncoding RNAs,are essential for posttranscriptional gene regulation and have important roles in a wide range of biological processes,including development,growth and differentiation. Thousands of miRNAs have been identified in animals,plants and microoaganisms. miRNAs regulate their target genes by mRNA degradation or translation suppression. About 30 % of genes in an organism are subject to miRNA regulation. miRNA expression and function are regulated by transcriptional factors,epigenetics,single nucleotide polymorphisms,RNA editing and so on. Additionally,the success in knocking out a specific mouse miRNA gene has provided a valuable model for studying miRNA function.
2.Establishment of Congenital Heart Disease Transposition of Great Arteries in Experimental Mice Model

Objective: To establish a mice model of congenital heart disease transposition of great arteries in order to provide a research reference for the occurrence and development of transposition of great arteries. Methods: A total of 20 pregnant ICR mice at 8-10 weeks of age were divided into 2 groups: Control group, the mice received a single dose of DMSO 70 mg/kg at 8.5 days of gestation,n=5 and Experiment group, the mice received a single dose of all-trans retinoic acid 70 mg/kg at 8.5 days of gestation,n=15. All animals were treated for 18 days and then the embryos were taken to observe cardiac morphology under stereomicroscope. Results: Compared with Control group, Experiment group had obviously increased occurrence rates of premature delivery, abortion and embryo absorption, and 61.2% phenotype for transposition of great arteries; meanwhile, combining with non-heart defect phenotypeas exophthalmos and spinal malformation. Conclusion: All-trans retinoic acid may induce transposition of great arteries in mice embryos, which is a feasible animal model in experimental research.
3.CT manifestations of retroperitoneal extra-adrenal paraganglioma

Objective To investigate the CT characteristics of retroperitoneal extra-adrenal paraganglioma(PGL).Methods CT findings of 11 patients with pathologically confirmed retroperitoneal extra-adrenal PGL were analyzed retrospectively,and the relevant literature was reviewed.All patients received plain CT as well as contrast-enhancement CT scan.Results All of the 11 tumors were located in the retroperitoneal space around large blood vessels,including 2 functional and 9 unfunctional extra-adrenal paragangliomas.Eight of 11 cases were benign tumors which presented as round or oval masses with sharp margins,separated from surrounding structures clearly;while the rest 3 cases,which with indistinct edge,even more with the invasion of surrounding structures (renal artery,renal venous and pancreas),were confirmed to be malignant.Most of the tumors were heterogeneous on plain CT.It was common to see intratumoral necrosis or cystic degeneration in the tumors (9 cases),while hemorrhage or calcification was uncommon,which was found in 3 and 2 cases,respectively.Homogeneous intensity was only observed in 2 masses with diameter less than 4 cm.On contrast-enhanced CT image, the solid part of the tumors presented continuously significant enhancement.Dilated feeding artery was found in or around the masses (8 cases) in artery phase.Conclusion There are certain characteristics of retroperitoneal extra-adrenal paraganglioma on CT scanning, which are helpful for preoperative diagnosis and treatment of this disease.
4.Double-equilibration technique for measuring liquid/gas partition coefficients of isoflurane

This study was designed to establish methods to measure partition coefficients of inhaled anesthetics at 1 atmosphere and 37 C by using syringe-flask double equilibration technique. Olive oil/gas (O/G), saline/gas (S/G), and water/gas (W/G) partition coefficients of isoflurane were 90.20?2.01, 0.6256?0.109, and 0. 741?0.025, respectively. Even though O/G was 150 folds of S/G,coefficients of variation for both O/G and S/G were less than 3%. The results indicate that the established double-equilibration technique has a very high repeatability and is applicable to wide ranges of inhaled anesthetics' solubilities.
5.On improving the professional core competencies of medical students in standardized residency training

Objective:To explore the methods to improve the professional core competencies of medical students in the standardized residency training.Methods:A total of 82 medical students who participated in the standardized residency training ("resident students" for short) in the same period were selected in this study, and the self-edited professional core competencies textbooks were used for regular theoretical study, and at the same time, clinical practice cases were collected for regular scenario reduction teaching. When the resident students entered the department and after 12 months of the standardized training, the professional core competencies training evaluation scale was used to evaluate and compare the scores of the resident students, and the value of the above-mentioned training model to improve the professional core competencies of the resident students was evaluated. SPSS 18.5 was performed for t test between groups. Results:After being trained by the above model, the selected students had significantly improved such competencies as communication, cooperation and problems solving ( P<0.05). Compared with the scores of resident students before and after the training, there was no significant difference in communication and cooperation with other students ( P>0.05), with low satisfactory communication with the leader ( P>0.05), but the problem-solving ability had been significantly improved ( P<0.05), while the score of which was low and it should be pay more attention to in the future training. Conclusion:In the process of standardized residency training, the application of the above-mentioned training model can significantly improve the professional core competencies of resident students, and at the same time, it's necessary to strengthen the cultivation of satisfactory communication with leaders and problem solving ability.
6.Comparison study on disinfection efficacy of Anerdian using swab and spray methods

Objective To explore disinfection efficacy of anerdian using swab and spray disinfection methods.Methods Hands of 30 subjects were randomly divided into the swab group and the spray group.The samples of before and after disinfection in each group were collected separately and observed their disinfection effect.Results The number of bacteria showed no significant difference between the two groups.The passing rate of disinfection reached 100%.The time of disinfection in the spray group was (2.58±0.32)s,significantly less than that in the swab group,(12.26±1.48)s,however,the drying time in the spray group was (42.37±1.79)s,significant longer than that in the swab group,(26.24±1.46)s.A subject hand disinfection used 0.5ml 0.2% anerdian in the spray group,and 0.75ml 0.2% anerdian and two cotton buds was used in the swab group.Conclusions There is identical disinfection efficacy between spray and swab methods.Spray disinfection method has short operating time,less use of disinfectant without using cotton buds,it is able to replace swab disinfection and will play an important role to reduce medical waste and health care costs.
7.ISSR Analysis for genetic diversity of Coptis chinensis

Objective To discuss the genetic diversity of Coptis chinensis.Methods The genetic diversity of 78 individuals from seven populations was analyzed by inter-simple sequence repeat(ISSR).Results Twelve primers were selected to produce highly reproducible ISSR bands.Among 106 amplified bands,72 showed polymorphism,the percentage of polymorphic bands reached to 67.92%.Nei's gene diversity index(H)was 0.180 3,Shannon information index(I)was 0.283 2,Gst was 0.681 5.The genetic distance coefficient and genetic similarity were 0.089 4—0.184 6 and 0.832 1—0.912 7,respectively.ConclusionC.chinensis holds high genetic diversity and the majority of genetic variation occurs among the populations.By cluster analysis,the geographical distribution is very obvious.The ISSR marker could be used for the analysis of the genetic diversity and genetic variation of C.chinensis.
8.A preliminary study on Norovirus variant for GⅡ.4 in Guiyang regions

Objective To study the composition of variant and point mutations of Norovirus GⅡ.4 in Guiyang regions.Methods From June to November 2010,cases information and fecal specimens were collected from guard-hospitals in Guiyang regions,who had caught the acute-gastroenteritis.Noroviruses in specimens were detected by a real-time reverse transcription polymerase chain reaction(real-time RT-PCR),and then partial genotyped norovirus-positive clinical samples (in random) were cloned and sequenced in VP1 gene code.Furthermore,the gene sequences were compared with the published variants at home and abroad of norovirus(GⅡ.4),including the phylogenetic analyses of genomes and variation of amino acids within individual sites.Results Those 267 specimens were GⅡ-norovirus-positive(62.68%) in 426 clinical samples.There were nine GⅡ.4-norovirus-positive VP1 gene-sequences available,and two subtype-norovirus variants (GⅡ.4 2008a and G Ⅱ.4 2008b variant) were epidemic in 2010,Guizhou province.The homology between and in subgroups were 95.90%-96.72% and 99.45%-100%.Two amino acids within individual sites were apt to mutate.Conclusion Norovirus GⅡ genotype were predominant in summer and fall acute gastroenteritis in 2010 for Guiyang regions,and the variants were diversity.
9.Hand Hygiene Compliance in China: An Evaluation

OBJECTIVE To evaluate the hand hygiene compliance(HHC) in China.METHODS The field observation for hand hygiene compliance,the hand hygiene material and equipment were performed in parallel in 8 hospitals from Beijing,Shanghai and Guangzhou.RESULTS It was shown that the average hand hygiene compliance before contacting patient was 35.3%,much lower than the compliance after contacting patient(56.5%).The nurse′s HHC(39.3%) was the best among groups before contacting patient,but the physicians took the first place of HHC after contacting patient.The best discipline in HHC was gynecology and obstetrics departments,especially before contacting patient and after contacting other surface,comparatively the worst was the surgeons.Most of hospitals(83.3%) provided the unmedicated detergent for handwashing.The alcohol-based hand rub(ABHR) for hand disinfection was available(56%) in many hospitals,in contrast the HHC with ABHR was quite low(14.2%).CONCLUSIONS These results indicated that the HHC is relatively low and improving adherence to hand hygiene practice is a multidisciplinary and systematic approach in China.It might be essential for promoting the HHC of health care workers,especially the physicians and surgeons,to strengthen the education and enlarge the continuous investment for some hard-ware improvement.
10.Effects of leptin on endoplasmic reticulum stress related proteins after focal cerebral ischemia

Objective To investigate the effect of leptin on endoplasmic reticulum stress related protein after focal cerebral ischemia in rats. Methods Ischemia was induced by occluding the middle cerebral artery in rats brain using the filament occlusion method. Forty SD rats were randomly divided into sham operation group, cerebral is-chemia group and leptin-preconditioning group. Leptin was injected subcutaneously before occlusion of blood vessel. Longa 5 score neurological function scale, body weight and brain edema changes were measured 6 hours after MCAO, and the brain was removed to detect the endoplasmic reticulum marker protein: glucose-regulated protein 78 (GRP78) and C/EBP-homologous protein (CHOP) by immunohistochemical method. Results There was no difference in the body weight changes between leptin-preconditioning group and ischemic group. In the leptin-preconditioning group, the neurological function score (1.90±0.31 vs. 2.50±0.52, P<0.05) and the degree of brain edema (3.60±0.52 vs. 7.70±0.94, P<0.001) were significantly lower than those in the cerebral ischemia group. Moreover, the expression of GRP78 in leptin-preconditioning group was significantly higher than that in ischemia group (48.69 ±5.06 vs. 35.78± 4.35, P<0.01), and the expression of CHOP was significantly lower than that of ischemia group (60.24 ±4.11 vs. 38.81±5.34, P<0.01). Conclusion Leptin can reduce the neurological deficit and may be associated with the up-reg-ulation of GRP78 protein, and down regulation of CHOP protein to weaken the endoplasmic reticulum stress caused by cerebral ischemia
